Abstract
Description
도 2의 A는 ACP-ADA 또는 ADA-ACP 발현 벡터와 pDsRed 벡터를 공동으로 형질주입한 후 형광신호를 확인한 결과를 나타내며, B는 ACP-ADA 및 ADA-ACP의 활성을 측정한 결과를 나타낸다.
도 3의 A는 대장균에서 ACP-ADA를 발현시킨 후 분리 정제한 결과를 나타내며, B는 분리 정제한 ACP-ADA의 활성을 측정한 결과를 나타낸다.
도 4의 A는 분리 정제한 ACP-ADA를 HeLa 세포에 처리한 후 형광 신호를 확인한 결과를 나타내며, B는 형광 신호를 수치화한 그래프를 나타낸다.
도 5는 ADA가 결핍된 세포에 ADA를 전달하고, 세포 내의 ADA 를 확인한 결과를 나타낸다. A는 세포 내 투과된 ACP-ADA 단백질 관찰 결과를 나타내며, B는 ADA 활성 측정 결과를 나타낸다.
도 6은 ADA 유전자 발현 hetero (+/-) 마우스에 bovine ADA와 ACP-ADA를 주입하고, 혈액에서 단핵세포(Mononuclear cells)를 분리하여 ADA 활성을 측정한 결과를 나타낸다.

서열번호 2의 아미노산 서열로 기재되는 아데노신 탈아미노화효소를 포함하는 융합 단백질.
- 제1항의 융합 단백질을 코딩하는 폴리뉴클레오티드.
- 제2항의 폴리뉴클레오티드를 포함하는 재조합 벡터.
- 제3항의 재조합 벡터를 포함하는 숙주세포.
- 하기 단계를 포함하는 융합 단백질을 생산하는 방법:
⒜ 제4항의 숙주세포를 배양 배지에서 배양하는 단계; 및
⒝ 배양 배지로부터 융합 단백질을 회수하는 단계.
- 제1항의 융합 단백질을 유효성분으로 포함하는 중증복합면역결핍증 치료용 약학적 조성물.
- 제6항에 있어서, 상기 중증복합면역결핍증은 아데노신 탈아미노화효소 결핍증인 것인 약학적 조성물.
