Claims (34)
- 생체분자 생산 시스템으로서, 시스템은:
표적 생체분자를 함유하는 액체를 수용하기에 적합한 챔버를 포함하는 생물반응기; 및
농축기를 포함하고, 상기 농축기는, 농축물을 수집하기에 적합하고 상기 생물반응기의 입력으로 또는 상기 농축기와 상기 생물반응기의 사이에 배치된 중간 용기의 입력으로 농축물을 재순환시킬 수 있는 농축물 도관을 구비하는, 생체분자 생산 시스템.
- 제 1 항에 있어서,
상기 생물반응기와 상기 농축기는 상기 생물반응기로부터 상기 농축기로의 액체 이송을 가능하게 하는 도관에 의해 연결되는, 생체분자 생산 시스템.
- 제 1 항에 있어서,
상기 생물반응기와 중간 용기는 상기 생물반응기로부터 상기 중간 용기로의 액체 이송을 가능하게 하는 도관에 의해 연결되고, 상기 중간 용기와 농축기는 상기 중간 용기로부터 상기 농축기로의 액체 이송을 가능하게 하는 도관에 의해 연결되는, 생체분자 생산 시스템.
- 제 1 항 내지 제 3 항 중 어느 한 항에 있어서,
생물반응기는 관류 생물반응기인, 생체분자 생산 시스템.
- 제 1 항 내지 제 4 항 중 어느 한 항에 있어서,
생물반응기는 배치 생물반응기인, 생체분자 생산 시스템.
- 제 1 항 내지 제 5 항 중 어느 한 항에 있어서,
상기 도관에는 상기 농축기를 통한 상기 액체의 교차-유동을 제공하기 위해 하나 이상의 펌프가 장착되는, 생체분자 생산 시스템.
- 제 1 항 내지 제 6 항 중 어느 한 항에 있어서,
상기 농축기는 한외-여과 또는 정밀-여과 장치인, 생체분자 생산 시스템.
- 제 1 항 내지 제 7 항 중 어느 한 항에 있어서,
상기 농축기는 접선 유동 여과 장치인, 생체분자 생산 시스템.
- 제 2 항에 있어서,
상기 생물반응기와 상기 농축기 사이에 필터가 배치되는, 생체분자 생산 시스템.
- 제 3 항에 있어서,
상기 생물반응기와 상기 중간 용기 사이에 필터가 배치되는, 생체분자 생산 시스템.
- 제 1 항 내지 제 10 항 중 어느 한 항에 있어서,
시스템은 오염 제거 용기를 더 포함하는 생체분자 생산 시스템.
- 제 1 항 내지 제 11 항 중 어느 한 항에 있어서,
시스템은 정화 격실을 포함하는 생체분자 생산 시스템.
- 제 1 항 내지 제 12 항 중 어느 한 항에 있어서,
시스템은 크로마토그래피 격실을 포함하는 생체분자 생산 시스템.
- 제 1 항 내지 제 13 항 중 어느 한 항에 있어서,
시스템은 바이러스 불활성화 격실을 포함하는 생체분자 생산 시스템.
- 제 1 항 내지 제 14 항 중 어느 한 항에 있어서,
시스템은 봉쇄 구역을 포함하는 생체분자 생산 시스템.
- 제 1 항 내지 제 15 항 중 어느 한 항에 있어서,
상기 격실은 칸막이에 의해 서로 연결되거나 분리될 수 있으며, 상기 칸막이의 구성은 개방형 구성 또는 폐쇄형 구성일 수 있는, 생체분자 생산 시스템.
- 제 1 항 내지 제 16 항 중 어느 한 항에 있어서,
시스템은 시스템의 자동 제어를 위한 공정 제어 장치를 포함하는 생체분자 생산 시스템.
- 바이러스와 같은 생체분자의 생산 시스템으로서, 시스템은:
- 표적 생체분자를 함유하는 액체를 수용하기에 적합한 챔버를 고정층 관류 생물반응기; 및 농축기, 바람직하게는, TFF를 포함하는 상류 공정 격실로서, 상기 농축기는, 상기 농축기로부터의 농축물과 같은 액체 배출물을 수집하고 상기 생물반응기의 입력으로 또는 상기 농축기와 상기 생물반응기 사이에 배치된 중간 용기의 입력으로 상기 액체 배출물을 재순환시키기에 적합한 농축물 도관을 구비하는, 상류 공정 격실과;
- 정화 격실과;
- 크로마토그래피 격실; 및
- 선택적으로 바이러스 불활성화 격실을 포함하고,
상기 격실은 하나 이상의 격리실 내에 배치되는, 생체분자의 생산 시스템.
- 생체분자 생산 방법으로서, 상기 생체분자는 액체를 포함하는 생물반응기에서 생산되고, 상기 방법은 농축 단계를 포함하고, 상기 생물반응기로부터의 배출물은 농축기에서 농축되며, 상기 농축기로부터의 배출물은 상기 생물반응기로 또는 상기 농축기와 상기 생물반응기 사이에 배치된 중간 용기로 재순환되는, 생체분자 생산 방법.
- 제 19 항에 있어서,
상기 생물반응기는 관류 모드 및/또는 배치 모드에서 작동되는, 생체분자 생산 방법.
- 제 19 항 또는 제 20 항에 있어서,
상기 액체의 방향성 유동을 유도하기 위해 펌프가 사용되는, 생체분자 생산 방법.
- 제 19 항 내지 제 21 항 중 어느 한 항에 있어서,
상기 농축기는 한외-여과 또는 정밀-여과 장치인, 생체분자 생산 방법.
- 제 19 항 내지 제 22 항 중 어느 한 항에 있어서,
상기 농축기는 접선 유동 여과 장치인, 생체분자 생산 방법.
- 제 19 항 내지 제 23 항 중 어느 한 항에 있어서,
상기 액체는 상기 농축기로 이송되기 전에 필터를 통과하는, 생체분자 생산 방법.
- 제 19 항 내지 제 24 항 중 어느 한 항에 있어서,
상기 액체는 상기 중간 용기로 이송되기 전에 필터를 통과하는, 생체분자 생산 방법.
- 제 19 항 내지 제 25 항 중 어느 한 항에 있어서,
상기 농축기의 재순환된 배출물은 상기 중간 용기로부터 수확됨으로써, 농축된 세포 배양 수확물을 수득하는, 생체분자 생산 방법.
- 제 19 항 내지 제 26 항 중 어느 한 항에 있어서,
상기 재순환된 세포 배양 수확물은 정화됨으로써, 정화된 세포 배양 수확물을 수득하는, 생체분자 생산 방법.
- 제 19 항 내지 제 27 항 중 어느 한 항에 있어서,
상기 생체분자는 상기 정화된 세포 배양 수확물에 대해 크로마토그래피 단계를 수행함으로써 더 정제되는, 생체분자 생산 방법.
- 제 19 항 내지 제 28 항 중 어느 한 항에 있어서,
상기 생체분자는 바이러스인, 생체분자 생산 방법.
- 제 19 항 내지 제 29 항 중 어느 한 항에 있어서,
상기 바이러스는 포름알데히드를 사용하여 불활성화되는, 생체분자 생산 방법.
