Abstract
본 발명의 방법에 따라 제조된 실로스타졸은 수율이 높아 경제적이고, 순도가 높아 잔존하는 불순물의 함량이 낮으며, 부작용이 적고 방출 특성이 우수하다. 또한 본 발명의 방법에 따른 실로스타졸 제조방법은 단일촉매를 사용하여, 종래 방법 대비 환경오염이 적은 것을 특징으로 한다.

Claims (13)
- 6-하이드록시-3,4-디하이드로-1H-퀴놀린-2-원을 용매에 용해시키고 1종의 단일촉매를 투입하여 혼합 용액을 제조하는 단계(s1);
상기 혼합 용액에 5-(4-클로로부틸)-1-사이클로헥실-1H 테트라졸을 투입하고 교반하여 반응액을 제조하는 단계(s2);
상기 반응액을 0 내지 10℃의 온도로 냉각한 후 교반하는 단계(s3);
상기 냉각된 반응액을 여과하고 세척한 후 건조하여 크루드 실로스타졸을 제조하는 단계(s4);
상기 크루드 실로스타졸을 용매에 용해하여 교반한 후 0~20℃로 냉각하여 결정을 생성하는 단계(s5);
상기 결정을 여과한 후 세척하고 건조하여 실로스타졸을 제조하는 단계(s6);를 포함하는 실로스타졸의 제조방법.
- 제1항에 있어서,
상기 s1 단계의 촉매는 수산화나트륨, 수산화칼륨으로 이루어진 군에서 선택된 1종의 단일촉매인 것인 실로스타졸의 제조방법.
- 제1항에 있어서,
상기 s1단계의 용매는 에탄올과 정제수의 혼합물인 것인 실로스타졸의 제조방법.
- 제3항에 있어서,
상기 정제수의 부피는 에탄올의 총 부피에 대하여, 3 내지 20부피%인 것인 실로스타졸의 재조방법.
- 제3항에 있어서,
상기 정제수의 부피는 에탄올의 총 부피에 대하여, 6 내지 10부피%인 것인 실로스타졸의 재조방법.
- 제1항에 있어서,
상기 s1 단계의 단일촉매는 6-하이드록시-3,4-디하이드로-1H-퀴놀린-2-원의 총 중량에 대하여 10 내지 50중량% 투입되는 것인 실로스타졸의 제조방법.
- 제1항에 있어서,
상기 s1 단계의 단일촉매는 6-하이드록시-3,4-디하이드로-1H-퀴놀린-2-원의 총 중량에 대하여 20 내지 30중량% 투입되는 것인 실로스타졸의 제조방법.
- 제1항에 있어서,
상기 s2 단계는 60 내지 90℃ 온도 조건에서 36 내지 60시간 교반하는 것인 실로스타졸의 제조방법.
- 제1항에 있어서,
상기 s5 단계의 용매는 디클로로메탄, 메탄올, 에탄올, 이소프로판올로 이루어진 군에서 선택된 1종 또는 2종 이상인 것인 실로스타졸의 제조방법.
- 제1항에 있어서,
상기 s6 단계에서 제조된 실로스타졸의 수율은 s4 단계에서 제조된 크루드 실로스타졸에 대하여 75% 이상인 것인 실로스타졸의 제조방법.
- 제1항에 있어서,
상기 s6 단계에서 제조된 실로스타졸의 순도는 99.7% 이상인 것인 실로스타졸의 제조방법.
- 제1항의 방법에 의하여 제조된 실로스타졸을 포함하는 실로스타졸 약학제제.
- 제12항에 있어서,
상기 실로스타졸의 결정 형태는 A형 결정인 것인 실로스타졸 약학제제.
