Abstract
Description
도 2는 나노복합체의 크기, 다분산 지수 및 zeta potential을 측정한 결과를 각각 보여주는 그래프이다.
도 3a 내지 도 3e는 나노복합체의 유방암 세포에의 선택적 투과를 확인하기 위해 유세포 분석기를 이용하여 분석한 그래프이다. siRNA만을 처리한 그룹을 붉은색, RS 융합 펩타이드를 이용한 그룹을 초록색으로 표시하였다. 도 3a는 MDA-MB-231, 3b는 MCF-7, 3c는 BT-474, 3d는 SK-BR-3, 3e는 RAW264.7 세포에서 진행한 결과이다.
도 4는 FITC가 표지된 siRNA가 세포 내부로 전달된 비율을 정량화하여 나타낸 그래프이다.
도 5a 내지 도 5b는 나노복합체의 세포 독성을 측정한 결과를 나타낸다. 모든 값은 4번 측정하였고, 평균 ± 표준편차로 나타내었다. 도 5a는 HDF-n, 5b는 MDA-MB-231 세포에서 진행한 결과이다.
Claims (10)
- 서열목록 제1 서열: QLEFYTGLAKLI을 갖는 암 세포 진입(Cell Permeating, SP82)펩타이드 및 서열목록 제2 서열: RRRRRRRRRRR (R11)을 갖는 폴리아르기닌(polyarginine) 펩타이드를 포함하는 암 특이적 핵산 분자 전달용 조성물.
- 제1항에 있어서,
상기 암 세포 진입(Cell Permeating, SP82)펩타이드 및 폴리아르기닌(polyarginine) 펩타이드는 융합하여 RS 융합 펩타이드를 형성하는 것을 특징으로 하는 암 특이적 핵산 분자 전달용 조성물.
- 제2항에 있어서,
상기 RS 융합 펩타이드는 상기 SP82 펩타이드와 상기 폴리아르기닌(polyarginine) 펩타이드가 서열목록 제3 서열: GGGG을 갖는 링커에 의해 서로 연결된 형태인 것을 특징으로 하는 암 특이적 핵산 분자 전달용 조성물.
- 제2항에 있어서,
상기 RS 융합 펩타이드는 비극성 아미노산(A, G, I, L), 전하를 띠지 않는 극성 아미노산(Q, Y, T) 및 양전하를 가지는 아미노산(K, R)으로 이루어져 있는 것을 특징으로 하는 암 특이적 핵산 분자 전달용 조성물.
- 제2항에 있어서,
상기 RS 융합 펩타이드는 염기성 부분이 전체의 44±11%, 산성 부분이 전체의 4±1%, 소수성 부분이 전체의 22±6.5% 및 중성 부분이 전체의 30±7.5%로 차지하는 것을 특징으로 하는 암 특이적 핵산 분자 전달용 조성물.
- 암 세포 진입(Cell Permeating, SP82)펩타이드 및 폴리아르기닌(polyarginine) 펩타이드가 융합하여 형성된 RS 융합 펩타이드에 핵산 분자가 결합된 암 특이적 핵산 분자 전달용 나노 복합체.
- 제6항에 있어서,
상기 핵산 분자는 gDNA 및 cDNA를 포함하는 DNA 분자와 mRNA 및 siRNA를 포함하는 RNA 분자 중 어느 하나 이상인 것을 특징으로 하는 암 특이적 핵산 분자 전달용 나노 복합체.
- 제6항에 있어서,
상기 RS 융합 펩타이드의 아민 그룹(N)과 상기 핵산 분자의 인산 그룹(P)의 비율(N/P ratio)는 5:1 이상인 것을 특징으로 하는 암 특이적 핵산 분자 전달용 나노 복합체.
- 상기 제1항 내지 제5항 중 어느 한 항의 암 특이적 핵산 분자 전달용 조성물을 포함하는 암 및 염증 치료용 조성물.
- 상기 제6항 내지 제8항 중 어느 한 항의 암 특이적 핵산 분자 전달용 나노 복합체를 세포와 접촉시키는 단계를 포함하는 암 특이적 핵산 분자 전달용 나노 복합체를 세포에 전달하는 방법.
