Išradimas susijęs su Berlyno mėlynojo dengimo metodika ir šios metodikos panaudojimu nusodinant dangas ant titano pagrindo, siekiant suformuoti gliukozės biologinius jutiklius.BACKGROUND OF THE INVENTION The present invention relates to a Berlin blue coating technique and to the use of this technique for deposition of coatings on a titanium substrate to form glucose biological sensors.
Technikos lygisState of the art
Biologiniai jutikliai apibūdinami kaip analizinės sistemos, sudarytos iš biologinio atpažinimo elemento, sujungto su signalo vertikliu, teikiančios analizinius duomenis, kurie yra proporcingi nustatomojo objekto koncentracijai (L. Su et ai., 2011). Pagal naudojamą signalo vertiklį biologiniai jutikliai skirstomi į elektrocheminius, pjezoelektrinius, jonams selektyvius, optinius, magnetinius bei akustinius biojutiklius (R. Monošfk et ai., 2012). Biologiniuose jutikliuose naudojami įvairūs biologiniai atpažinimo elementai: antikūnai, ląstelės, DNR, RNR, fermentai ir kitos struktūros (M. Park, 2013).Biological sensors are defined as analytical systems consisting of a biological recognition element coupled to a signal transducer that provides analytical data that is proportional to the concentration of the target (L. Su et al., 2011). According to the signal transducer used, biological sensors are divided into electrochemical, piezoelectric, ion-selective, optical, magnetic and acoustic biosensors (R. Monošfk et al., 2012). Biological sensors use a variety of biological recognition elements: antibodies, cells, DNA, RNA, enzymes, and other structures (M. Park, 2013).
Fermentinių biologinių jutiklių (juose biologinio atpažinimo funkciją atlieka fermentas) veikimas pagrįstas fermento katalizuojama analitės chemine reakcija (A. Mulchandani, 1999).The activity of enzymatic biological sensors (in which the biological recognition function is carried out by an enzyme) is based on the enzyme-catalyzed chemical reaction of the analyte (A. Mulchandani, 1999).
Amperometriniai biologiniai jutikliai - tai elektrocheminiai biologiniai jutikliai, kurie atlieka elektros srovės, susijusios su analitės redokso reakcijomis, matavimus (A. Chaubey ir B.D. Malhotra, 2002). Amperometriniai fermentiniai biologiniai jutikliai yra skirstomi į 3 klases (S.V. Dzyadevych et ak, 2008):Amperometric biological sensors are electrochemical biological sensors that measure the electrical current associated with the redox reactions of the analyte (A. Chaubey and B. D. Malhotra, 2002). Amperometric enzymatic biological sensors are divided into 3 classes (S.V. Dzyadevych et al., 2008):
1. Biologinius jutiklius, kuriuos naudojant vyksta tiesioginis fermentinės reakcijos produktų koncentracijos nustatymas.1. Biological sensors for the direct determination of the concentration of enzymatic reaction products.
2. Biologinius jutiklius, naudojančius elektronų pernašos tarpininką (EPT), pernešant) elektronus iš aktyvaus fermento centro į elektrodą.2. Biological sensors using an electron transport mediator (EPT) to transfer electrons from the active enzyme center to the electrode.
Biological sensors which transmit electrons directly between the enzyme and the electrode.
Gliukozės koncentracijos kraujyje matavimas yra svarbiausias diagnostinis kriterijus, skirtas pacientų, sergančių cukriniu diabetu, stebėjimui. Efektyvi cukrinio diabeto kontrolė priklauso nuo tikslios gliukozės koncentracijos kraujyje stebėjimo sistemos, kurioje gliukozės biologiniai jutikliai užimą vieną svarbiausių vietų.Blood glucose measurement is the most important diagnostic criterion for monitoring patients with diabetes mellitus. Effective diabetes control relies on an accurate blood glucose monitoring system, where glucose biological sensors occupy a central place.
Amperometriniai gliukozės biologiniai jutikliai gali priklausyti kiekvienai iš išvardintų amperometrinių biojutiklių klasių. Pirmos klasės gliukozės biologinių jutiklių veikimas pagrįstas vandenilio peroksido, susidarančio gliukozės oksidacijos metu, nustatymu (J. Wang, 2008).Amperometric glucose biological sensors may belong to each of the aforementioned classes of amperometric biosensors. The performance of first-class glucose biological sensors is based on the detection of hydrogen peroxide formed during glucose oxidation (J. Wang, 2008).
Formuojant antros klasės gliukozės biologinius jutiklius naudojami elektronų pernašos tarpininkai (pvz.: ferocenas, fericianidas), leidžiantys sumažinti analizės metu naudojamą darbinio elektrodo potencialą (E.H. Yoo ir S.Y. Lee, 2010).Electron transfer mediators (eg, ferrocene, ferricyanide) are used in the formation of second-class glucose biological sensors to reduce the potential of the working electrode used in the assay (E.H. Yoo and S.Y. Lee, 2010).
Trečios klasės amperometriniuose gliukozės biologiniuose jutikliuose naudojamos laidžios organinės medžiagos, pvz., polipirolas, užtikrinančios tiesioginę elektronų pernašą tarp elektrodo ir fermento (C. G. J. Koopal et ai., 1994).Third-class amperometric glucose biological sensors use conductive organic materials, such as polypyrrole, to provide direct electron transfer between an electrode and an enzyme (C. G. J. Koopal et al., 1994).
Berlyno mėlynasis yra vienas dažniausiai naudojamų elektrocheminių mediatorių analiziniam taikymui biologiniuose gliukozės jutikliuose (F. Ricci ir G. Palleschi, 2005). Biologiniais jutikliais registruojama vandenilio peroksido redukcijos srovė, todėl tyrimai orientuoti į vandenilio peroksidui atrankius elektrodus. Siekiant išvengti pašalinių medžiagų, esančių kraujyje (tokių kaip askorbo rūgštis, šlapimo rūgštis ir kt.), oksidacijos poveikio analizės rezultatams, darbiniam elektrodui turi būti suteiktas santykinai žemas potencialas. Vienas iš būdų darbinio elektrodo potencialui sumažinti yra peroksidazių (pvz., krienų peroksidazės) naudojimas (T. Ruzgas et ai., 1996) - naudojant peroksidazes, vandenilio peroksido redukcija vyksta esant žemesniems darbinio elektrodo potencialams (Z. Gao et ak, 2014). Tačiau šis būdas turi ir trūkumų, tokių kaip didelės gamybinės išlaidos, žemas stabilumas, krienų peroksidazės ribotas nusodinimas ant kietų paviršių. Berlyno mėlynasis - tai vandenilio peroksido redukcijos reakcijos katalizatorius, pasižymintis dideliu selektyvumu bei efektyvumu vandenilio peroksido redukcijos reakcijos katalizei, todėl Berlyno mėlynasis kartais vadinamas „dirbtine peroksidaze“ (A. A. Karyakin, 1999; Z. Gao et ai., 2014). Taigi, gliukozės biologinių jutiklių, neturinčių vandenilio peroksido redukcijos katalizatorių, naudojimas turi trūkumų:Berlin Blue is one of the most commonly used electrochemical mediators for analytical applications in biological glucose sensors (F. Ricci and G. Palleschi, 2005). Biological sensors record the hydrogen peroxide reduction current, so research is focused on hydrogen peroxide-selective electrodes. The working electrode must be given a relatively low potential to avoid the effects of oxidation analysis on foreign matter (such as ascorbic acid, uric acid, etc.). One way to reduce the potential of a working electrode is to use peroxidases (e.g., horseradish peroxidase) (T. Ruzg et al., 1996) - using peroxidases, hydrogen peroxide reduction occurs at lower working electrode potentials (Z. Gao et al., 2014). However, this process also has disadvantages such as high production costs, low stability, limited deposition of horseradish peroxidase on hard surfaces. Berlin Blue is a catalyst for the hydrogen peroxide reduction reaction with high selectivity and efficiency for the catalysis of the hydrogen peroxide reduction reaction, which is why Berlin Blue is sometimes called "artificial peroxidase" (A.A. Karyakin, 1999; Z. Gao et al., 2014). Thus, the use of glucose biological sensors without hydrogen peroxide reduction catalysts has the disadvantages of:
1) tiesioginis analičių amperometrinis nustatymas tradiciniuose elektroduose reikalauja santykinai aukšto potencialo, kuriam esant tirpale gali lengvai oksiduotis kitos medžiagos.1) Direct amperometric determination of analytes in conventional electrodes requires a relatively high potential at which other substances can readily oxidize in solution.
2) siekiant išvengti pašalinių medžiagų oksidacijos srovių įtakos matavimų rezultatams, naudojami papildomi apsauginiai sluoksniai (pvz., celiuliozės-acetatinės membranos, nafiono danga, sintetiniai polimerai), tačiau dangos nepilnai apsaugo nuo nepageidaujamų jonų ar molekulių, pvz., askorbo, šlapimo rūgščių, prasiskverbimo (A. Ambrozy et ak, 2013), prailgina biojutiklio atsako laiką (M.S. Alaejos ir J.G. Montelongo, 2004) taip pat turi būti suteiktas santykinai aukštas darbinis potencialas.(2) additional protective layers (eg cellulose-acetate membranes, naphthoe coating, synthetic polymers) are used to prevent the influence of oxidation currents on the measurement results, but the coatings do not fully protect against unwanted ions or molecules such as ascorbic, uric acid, penetration (A. Ambrozy et al., 2013), extends the response time of the biosensor (MS Alaejos and JG Montelongo, 2004) must also be given a relatively high working potential.
Nafiono danga apsaugo biojutiklį nuo anijoninių elektrochemiškai aktyvių medžiagų patekimo prie biojutiklio paviršiaus - šios medžiagos nėra oksiduojamos biojutiklio paviršiuje ir todėl nedaro įtakos matavimų rezultatams (S.H. Lim, 2005; L. Yang, 2009). A. Ramanavičius, 2017).The Nafion coating protects the biosensor against the anionic electrochemically active substances entering the biosensor surface - these substances are not oxidized on the biosensor surface and therefore do not affect the measurement results (S.H. Lim, 2005; L. Yang, 2009). Although the nafion coating protects the measurement results from oxidation currents that affect the accuracy of the assay (caused by the oxidation of impurities on the surface of the biosensor), the biosensor should provide a relatively high working potential while maintaining a low working potential during analysis, positively influence cell proliferation (A. Vaitkuviene et al., 2013). In order to reduce the working potential of the biosensor and thus prevent the oxidation of foreign electrochemically active substances, a graphite electrode modified with polypyrrole, Berlin blue and glucose oxidase composite layer was recently described by A. Ramanavičius and co-authors (A. Ramanavičius, 2017).
Pastaruoju metu didelį susidomėjimą šioje srityje sukėlė neorganiniai vandenilio peroksido redukcijos reakcijos katalizatoriai, tame tarpe - elektrokataliziniai pereinamųjų metalų heksacianoferatiniai kompleksai, ypatingai Berlyno mėlynasis, kuris pirmą kartą kaip „dirbtinė peroksidazė“ biojutiklyje gliukozės nustatymui buvo panaudotas 1995 metais Karyakin (A.A. Karyakin, 2001). H2O2 redukcijos srovė, matuojama naudojant Berlyno mėlynuoju modifikuotą elektrodą, dukart didesnė nei O2 redukcijos srovė, tuo tarpu platina ar kiti taurieji metalai nepasižymi selektyvumu vandenilio peroksido redukcijai esant molekuliniam deguoniui (A.A. Karyakin et ai., 2001; A.A. Karyakin et ai., 1998; A.A. Karyakin and E.E. Karyakina, 1999).Recently, inorganic hydrogen peroxide reduction reaction catalysts, including electrocatalytic transition metal hexacyanoferrate complexes, in particular Berlin Blue, which was first used as an "artificial peroxidase" in the biosensor to detect glucose in Karyakin (A.), have recently attracted considerable interest in this field. Molecular oxygen and hydrogen peroxide reduction may occur on the blue surface of Berlin. The hydrogen peroxide reduction reaction can be catalyzed by Berlin blue (K. Itaya et al., 1984). Other inorganic hydrogen peroxide reduction reaction catalysts, not only among hexacyanoferratic complexes but also among other known H2O2 reduction mediators, do not exhibit the same efficiency as Berlin Blue. The H2O2 reduction current, measured using the Berlin blue modified electrode, is twice that of the O2 reduction, whereas platinum or other noble metals do not exhibit selectivity for hydrogen peroxide reduction in the presence of molecular oxygen (AA Karyakin et al., 2001; AA Karyakin et al., 1998; AA Karyakin and EE Karyakina, 1999).
When forming glucose biosensors, it is important to select a substrate suitable for the formation of the electrochemically active layer. The substrate must have high electrochemical activity as well as chemical and mechanical stability. Titanium oxide has recently been used extensively in the development of glucose biosensors. Titanium is characterized by biocompatibility, corrosion resistance and low cost (comparing titanium with other materials used in the formation of glucose biosensors in the substrate, such as gold and platinum). In addition to the useful properties of titanium mentioned above, titanium also has the ability to be modified by a blue or blue coating by chemical or electrochemical deposition.
Kai kurių gliukozės biojutiklių formavimo metu, Berlyno mėlynojo danga buvo padengta ant titano oksido nanodalelių ar titano oksido zolio-gelio pavidalo junginių, skirtingų elektrodo medžiagų (SnO2, platinos, aukso, stiklo, anglies ir kt.). Tačiau šie dengimo metodai turi ir trūkumų:During the formation of some glucose biosensors, the Berlin blue coating was applied to titanium oxide nanoparticles or titanium oxide sol-gel compounds, different electrode materials (SnO 2 , platinum, gold, glass, carbon, etc.). However, these coating methods also have drawbacks:
1) T1O2 nanodalelių ar zolių-gelių sintezė pati savaime yra brangi procedūra.1) The synthesis of T1O2 nanoparticles or sol-gels is itself an expensive procedure.
2) Titano oksido nanodalelių sintezės procesai yra daugiapakopiai ir sudėtingi, todėl metalurginio Ti panaudojimas keliais žingsniais supaprastina ir atpigina procesą.2) The synthesis of titanium oxide nanoparticles is multi-step and complex, so the use of metallurgical Ti simplifies and lowers the process in several steps.
One method of immobilizing enzymes in biosensors is by immobilization using electrically conductive polymers, which can be used as a medium for covalent attachment of enzymes or their adsorption. Immobilization of biomolecules using electrically conductive polymers (eg polypyrrole) also facilitates the transfer of the electron from the enzyme to the electrode. In the present invention, a polypyrrole layer was used to immobilize glucose oxidase.

The pyrrole was purchased from Alfa Aesar. Ultrafiltration water was used to prepare the reconstituted solutions.
A computer-connected pAUTOLAB potentiostat / galvanostat with GPES software installed was used for electrochemical measurement and electrochemical deposition procedures. Electrochemical processes were carried out in a three-electrode cell using a titanium plate (surface area 1 cm 2 ) as a working electrode, a reference Ag / AgCI / KCI (saturated) electrode, and a titanium plate as an auxiliary electrode.
Electrochemical deposition of Berlin blue (BM), polypyrrole (PP) and composite BM / PP / GO layer and chronoamperometric biosensor assays were performed using pAUTOLAB potentiostat / galvanostat purchased from ECO-Chemie (Utrecht, The Netherlands). Electrochemical processes were carried out in a three-electrode cell using a titanium plate (surface area 1 cm 2 ) as the working electrode, an Ag / AgCI / KCI (saturated) electrode for comparison, and a titanium plate as an auxiliary electrode. For chronoamperometric measurements, the working electrode was given a potential of 0.05 V with respect to the Ag / AgCI / KCI (saturated) electrode. Measurements were made in pH 7.3 buffer containing 0.1 M NaCl and 0.01 M NaH2PO4. Before addition of glucose to the buffer solution, a steady-state current in the electrochemical cell was allowed to settle. Then, 1M glucose solution was added to the buffer solution and stirring (magnetic stirrer) was started to maintain a uniform glucose concentration throughout the volume. After 1 - 2 min. the mixing was switched off, a steady current was recorded after switching off, which was recorded. Changes in electrical current (occurring after the addition of glucose to the buffer solution), which is proportional to the glucose concentration in the solution, were measured during the studies.

Gaminant gliukozės biojutiklį pirmiausia ant titano plokštelės buvo padengiamas Berlyno mėlynojo (BM) pasluoksnis. Tai buvo atliekama:For the production of the glucose biosensor, a layer of Berlin Blue (BM) was first applied to the titanium plate. This was done by:
a) elektrochemiškai nusodinant Berlyno mėlynojo sluoksnį ant titano plokštelės ciklinės voltamperometrijos būdu, skleidžiant potencialą nuo 0,4 V iki 0,8 V(Ag/AgCI/KCI(sotus) atžvilgiu) 1 mM FeCb ir K3[Fe(CN)6] tirpale(a) Electrochemical deposition of the Berlin blue layer on a titanium plate by cyclic voltammetry with a potential of 0.4 V to 0.8 V (relative to Ag / AgCl / KCl (saturated)) in 1 mM FeCl 2 and K 3 [Fe (CN) 6]

100 °C arba naudojamas be iškaitinimo(c) The Berlin Blue Layer was heated in the oven for 1 hour. 100 ° C or used without heating
d) ant Berlyno mėlynojo pagrindo elektrochemiškai dengiamas polipirolo sluoksnis, po to imobilizuojamas fermentas - gliukozės oksidazė.(d) Electrochemically coat a polypyrrole layer on a Berlin blue substrate followed by immobilization of the enzyme glucose oxidase.
Gliukozės oksidazės imobilizacija atlikta:Immobilization of glucose oxidase was performed:
a) elektrochemiškai nusodinant polipirolą pirolo (30 mM), FeCl3(1 mM), K3[Fe(CN)6] (1 mM) bei gliukozės oksidazės (1 mg ml’1) mišinyje skleidžiant potencialą nuo 0,4 iki 0,8 V (Ag/AgCI/KCI(sotus) atžvilgiu) ciklinės voltamperometrijos būdu(a) Electrochemically precipitating polypyrrole in a mixture of pyrrole (30 mM), FeCl3 (1 mM), K3 [Fe (CN) 6] (1 mM) and glucose oxidase (1 mg ml ' 1 ) to give a potential of 0.4 to 0.8 V (relative to Ag / AgCI / KCI (saturated)) by cyclic voltammetry
b) užlašinant ant elektrodo, padengto Berlyno mėlynojo pasluoksniu, 40 mg ml'1gliukozės oksidazės tirpalo (pagaminto ph 7,3 fosfatiniame buferyje), jį išdžiovinant ore bei laikant virš gliutaro aldehido 25 % tirpalo 15 min.(b) Applying 40 mg ml ' 1 glucose oxidase solution (made in ph 7.3 phosphate buffer) to the electrode covered with a blue layer of Berlin, air drying and placing it over 25% glutaric aldehyde solution for 15 minutes.
Ciklinės voltamperometrijos metu pagalbiniu elektrodu buvo naudojama titano plokštelė, palyginamuoju - Ag/AgCI (KCI (kone.)) elektrodas. For cyclic voltammetry, a titanium plate was used as an auxiliary electrode, with the Ag / AgCI (KCI (almost)) electrode as comparator.
Amperometrinių tyrimų metu darbiniam elektrodui buvo suteiktas 0,05 V potencialas Ag/AgCI/KCI(sotus) elektrodo atžvilgiu. Pagalbiniu elektrodu naudota Ti plokštelė.For amperometric studies, the working electrode was given a potential of 0.05 V with respect to the Ag / AgCI / KCI (saturated) electrode. Auxiliary electrode used Ti plate.
Gaminant skirtingus biojutiklius, buvo pastebėta, kad biojutikliai, suformuoti imobilizuojant fermentą - gliukozės oksidazę - su gliutaro aldehido garais bei cheminiu būdu padengiant Berlyno mėlynojo pasluoksnį, analizės metu pasižymi didesniu srovės tankiu (siekė iki 33,9 pA-cm-2), gaunamu po gliukozės {lašinimo į analizuojamą tirpalą, nei biojutikliai, suformuoti elektrochemiškai padengiant Berlyno mėlynojo pasluoksnį bei elektrocheminės pirolo polimerizacijos metu imobilizuojant gliukozės oksidazę (jų generuojamas srovės tankis siekė iki 23,8 pA-cnr2).In the manufacture of different biosensors, it was observed that biosensors formed by immobilization of the enzyme glucose oxidase with glutaric aldehyde vapor and chemically deposited on the Berlin Blue layer exhibited higher current densities (up to 33.9 pA-cm -2 ) during analysis. glucose {drip into the solution to be analyzed, than the biosensors formed by electrochemically coating the Berlin blue layer and by immobilizing glucose oxidase during electrochemical pyrrole polymerization (generating current density up to 23.8 pA-cnr 2 ).
Iškaitinus Berlyno mėlynojo pasluoksnį, padidėja tyrimo metu biojutiklio generuojamos srovės tankis: tiriant biojutiklius, suformuotus padengiant titano elektrodą Berlyno mėlyno pasluoksniu elektrochemiškai ir elektrocheminio polipirolo nusodinimo metu imobilizuojant gliukozės oksidazę, didžiausias registruotas srovės tankis buvo 12pA-cm'2, o tiriant biojutiklius, suformuotus padengiant titano elektrodą Berlyno mėlyno pasluoksniu elektrochemiškai, iškaitinant Berlyno mėlynojo pasluoksnį krosnyje ir elektrocheminio polipirolo nusodinimo metu imobilizuojant gliukozės oksidazę, didžiausias tyrimų metu registruotas srovės tankis buvo 23,8 pA-cm-2.Ashing Berlin blue substrate increases study biosensor generated current density: testing biosensors that are formed by coating titanium electrode with Prussian blue substrate electrochemically and electrodeposition of polypyrrole deposition during the immobilization of glucose oxidase at the highest recorded current density was 12pA-cm-2 while exploring the biosensors that are formed by coating The titanium electrode with Berlin Blue Layer electrochemically, by heating the Berlin Blue Layer in the furnace and immobilizing glucose oxidase during electrochemical polypyrrole deposition, had a maximum current density of 23.8 pA-cm -2 recorded during the studies.
Tiriant biojutiklius, suformuotus padengiant titano plokštelę Berlyno mėlynuoju chemiškai, iškaitinant dangą ir imobilizuojant gliukozės oksidazę elektrocheminio polipirolo nusodinimo metu, buvo pastebėta, kad gliukozės mėginio analizės metu šio tipo biojutiklių generuojama srovė pasiekė dideles vertes - viršijo 40 μΑ/cm2. Šio tipo biojutikliams būdingas ir platus nustatomos gliukozės koncentracijos intervalas elektros srovės stipris dažniausiai ženkliai didėjo po gliukozės koncentracijos padidėjimo mėginyje tol, kol gliukozės koncentracija pasiekdavo 100 mM.In biosensors that are formed by coating titanium plate Berlin blue chemically Carbohydrates covering and immobilising glucose oxidase electrochemical polypyrrole during precipitation, it was observed that blood sample analysis, this type of biosensor current generated achieve capital - over 40 μΑ / cm second Typical and wide ranges of detectable glucose concentrations in this type of biosensor usually increased significantly after increasing the glucose concentration in the sample until the glucose concentration reached 100 mM.
Tiriant biojutiklius, suformuotus padengiant titano plokštelę Berlyno mėlynuoju elektrochemiškai, iškaitinant dangą ir imobilizuojant gliukozės oksidazę su gliutaro aldehido garais, buvo pastebėta, kad gliukozės mėginio analizės metu šio tipo biojutiklių generuojama srovė dažniausiai neviršydavo -15 μΑ/cm2. Šio tipo biojutikliams būdingas siauras nustatomos gliukozės koncentracijos intervalas elektros srovės stipris dažniausiai ženkliai didėjo po gliukozės koncentracijos padidėjimo mėginyje tol, kol gliukozės koncentracija pasiekdavo 6 mM. Taip pat šio tipo biojutikliams būdingas stabilumas laike - tiriant gliukozės mėginį šio tipo biojutikliais, praėjus savaitei po biojutiklio pagaminimo, generuojamos srovės stipris dažnai būdavo neženkliai mažesnis ar didesnis nei atliekant tyrimą biojutiklio pagaminimo dieną.Biosensors formed by coating the titanium plate with Berlin blue electrochemically, heating the coating and immobilizing glucose oxidase with glutaric aldehyde vapor showed that the current generated by this type of biosensor during the analysis of the glucose sample did not usually exceed 15 μΑ / cm 2 . In this type of biosensor, a narrow range of detectable glucose concentration is usually followed by a significant increase in the current following a rise in glucose concentration in the sample until the glucose concentration reaches 6 mM. Also, this type of biosensor is characterized by stability over time - when testing a glucose sample with this type of biosensor, one week after the biosensor was manufactured, the current generated was often not significantly lower or higher than on the day of biosensor production.
