Abstract

L'invention porte sur un procédé de production d'organoïdes rénaux comprenant des néphrons, des bourgeons urétraux et des vaisseaux sanguins et / ou des progéniteurs de ceux-ci. Dans un mode de réalisation, les procédés comprennent la mise en contact de cellules de mésoderme intermédiaires avec: le facteur de croissance des fibroblastes 9 et / ou le facteur de croissance des fibroblastes 20 et / ou le facteur de croissance des fibroblastes 2 et éventuellement un ou plusieurs choisis dans le groupe constitué par: la protéine morphogénique osseuse 7; héparine; un agoniste wnt; l'acide rétinoïque; et un antagoniste de la ra dans des conditions favorisant la formation d'organoïdes rénaux vascularisés. Un autre mode de réalisation comprend la production de cellules mésodermes en mettant en contact séquentiellement des cellules souches pluripotentes avec un agoniste wnt et un facteur de croissance fibroblastique 9 et / ou un facteur de croissance fibroblastique 20 et / ou un facteur de croissance fibroblastique 2, suivie d'une ré-exposition relativement courte à l'agoniste wnt. Les organoides rénaux peuvent avoir des utilisations finales telles que la réparation et la régénération du rein, la réimpression des reins ou des composants fonctionnels de ceux-ci, les réseaux de cellules rénales et les composés de criblage pour la néphrotoxicité.
