Abstract

La presente invención se refiere a un método que incluye realizar una ampliación de ácido nucleico de una muestra de ácido nucleico usando una sonda de detección, en donde la amplificación de ácido nucleico ocurre sobre uno o más periodos de interrogación, y, de la amplificación de ácido nucleico, adquirir datos de amplificación que indican una cantidad de ácido nucleico presente en cada uno de uno o más periodos de interrogación. El método también incluye, en base a los datos de amplificación, determinar un valor de corrección de diafonía asociado con un vecino espectral a la sonda para reducir la diafonía espectral del vecino espectral; y aplicar el valor de corrección de diafonía a los datos de amplificación recolectados de amplificaciones de ácido nucleico multiplexados de muestras de ácido nucleico.
