Abstract
Una composición para expresar un polipéptido dentro de un órgano objetivo, la composición comprende una partícula de suministro, y al menos una primera secuencia de ARNm en complejo, encapsulada o asociada de otro modo con la partícula de suministro. La secuencia de ARNm comprende una secuencia de codificación que codifica para el polipéptido, al menos una primera secuencia de región no traducida (UTR) y al menos una secuencia del sitio de unión de micro ARN (miRNA), en donde la secuencia del sitio de unión de miRNA se ubica dentro, 5' inmediatamente a, o 3' inmediatamente a, la primera secuencia UTR. La secuencia del sitio de unión de miRNA se selecciona para proporcionar una expresión diferencial de la secuencia de codificación entre el primer y el segundo tipo de células comprendidas dentro del órgano objetivo. La composición puede usarse en combinación con o para suplementar otros enfoques terapéuticos, que incluyen la quimioterapia, la terapia viral oncolítica y terapias celulares.
