Abstract

Se proporcionan métodos y composiciones para tratar un trastorno convulsivo que incluyen administrar al paciente un vector de virus adenoasociado que codifica a hM4Di para la entrega de hM4Di a una ubicación objetivo, el vector incluye un promotor de CaMKII humano o murino, un elemento regulador postranscripcional del virus de la hepatitis Woodchuck, y una secuencia de poliadenilación de la hormona de crecimiento bovina, y administrar al paciente un ligando sintético que activa hM4Di. La administración del vector y el ligando sintético a un paciente con un trastorno convulsivo se asocia con síntomas reducidos del trastorno convulsivo. En las realizaciones, el ultrasonido se aplica a una ubicación objetivo en el cerebro del paciente para mejorar la permeabilidad de la barrera hematoencefálica del paciente al vector y/o ligando sintético.
