Abstract
La presente invención se refiere a un método para producir una molécula de ARN de hebra única en horquilla capaz de inhibir la expresión de un gen objetivo, el método comprende: (i) una etapa de hibridación para hibridar una primera molécula de oligoARN de hebra única y una segunda molécula de oligoARN de hebra única; y (ii) una etapa de ligación para ligar el extremo 3´ de la primera molécula de oligoARN de hebra única y el extremo 5´ de la segunda molécula de oligoARN de hebra única por una ligasa de la familia Rnl2, en donde una secuencia producida por ligación de la primera molécula de oligoARN de hebra única y la segunda molécula de oligoARN de hebra única comprende una secuencia que inhibe la expresión del gen el gen objetivo.
