Abstract
La presente invención se refiere a un procedimiento para preparar un conjugado de proteína fisiológicamente activa y un polímero no peptídico, caracterizado porque comprende: (1) hacer reaccionar la proteína fisiológicamente activa con el polímero no peptídico en la solución de reacción que comprende un disolvente orgánico para unir la proteína fisiológicamente activa con el polímero no peptídico; y (2) aislar y purificar el conjugado de proteína fisiológicamente activa unido covalentemente al polímero no peptídico de la mezcla de reacción de la etapa (1), en donde el disolvente orgánico es un alcohol que tiene de 1 a 10 átomos de carbono; la solución de reacción comprende el disolvente orgánico en una cantidad de 30% a 55% en volumen basado en el volumen total de la solución de reacción; la solución de reacción tiene el p H de 4.5 a 6.5; el polímero no peptídico tiene dos o tres grupos de extremos reactivos; y la proteína fisiológicamente activa tiene dos extremos N y es insulina.
