Description
Foreliggende oppfinnelse angår en ny fremgangsmåte for fremstilling av carbohydrater med en oligosaccharid-karakter.
I løpet av de siste år har fenomenet med bakterieved-heftning på epiteliale vertsceller vunnet stor interesse (ref. 1). To svenske grupper (ref. 2 og 3) har foreslått at di-galactoseforbindelsen av formel 1 nedenfor bør virke som reseptor ved fiksering av fimbrierte uropatogene colibakterier til overflaten av epitelceller i urinveien hos mennesker. Dessuten antas di-galactoseenheten å være tilstede på overflaten av epitelcellene i form av trihexosylceramid (formel 2 nedenfor) og globosid.
Dessuten er det di-galactoseholdige glycolipid "gala-biosylceramid" (formel 3 nedenfor) tilstedeværende i nyrevev (ref. 4), skjønt forbindelsens funksjon ikke er kjent.
Enkle glycosider (eksempelvis methyl- og p-nitrof enyy
av forbindelsen 1 og av trisaccharidenheten i forbindelse 2 har vært kjent for å inhibere agglutinasjonen av uropatogene colibakterier med visse røde blodlegemer (ref. 1). Dette indi-
kerer anvendelsen av forbindelse 1 for fremstilling av nye typer av diagnostisk reagens og også av terapeutiske midler med ny type av aktivitet sammenlignet med tradisjonelle
antibiotika, dvs. inhibering av bakteriefiksering til epitel istedenfor å drepe bakteriene., -I litteraturen er det blitt rapportert metoder for fremstilling av 4-0-a-D-galactopyranosyl-D-galactose (1) og derivater derav.
Forbindelse 1 er blitt fremstilt fra di-galacturonsyre (4) ifølge reaksjonsskjerna 1 nedenfor (ref. 5).
De uegnede reagenser som anvendes (eksempelvis diazo-methan, lithium-aluminiumhydrid) forhindrer imidlertid fremstilling av forbindelse 1 i stor målestokk på denne måte. En variasjon av denne reaksjonssekvens er også blitt rapportert (ref. 6).
Det fullt ut acetylerte derivat av forbindelse 1 er blitt fremstilt (ref. 7, 8, 9) ved reaksjonssekvenser i flere trinn ut fra galactose, slik som eksemplifisert i reaksjonsskjerna 2 nedenfor (ref. 7). Også i dette tilfelle gjør de anvendte
reagenser (eksempelvis kvikksølv(II)-cyanid) i kombinasjon
med lavt totalutbytte fremstilling i stor skala upraktisk.
Det fremgår av den ovenfor angitte kjente teknikk at det foreligger et behov for nye metoder for syntetisering av oligosaccharider av forskjellig type ut fra lett tilgjenge-lige utgangsmaterialer. Et hovedmål ved oppfinnelsen er således å tilveiebringe en ny fremgangsmåte for fremstilling av mellomprodukter for fremstilling av forbindelser som er anvend-bare for medisinske og diagnostiske formål, som ovenfor angitt.
Ifølge oppfinnelsen tilveiebringes en fremgangsmåte
for fremstilling av forbindelser av formel (VI):
hvori n er et helt tall opp til 10, innbefattende 0, R er hydrogen eller en alkylgruppe, fortrinnsvis med fra 1 til 6 carbonatomer, og X er hydrogen, deuterium eller tritium, hvilken fremgangsmåte innbefatter reduksjon av en forbindelse av formel (V):
hvori n har den ovenfor angitte betydning og R er en alkylgruppe, fortrinnsvis med fra 1 til 6 carbonatomer, hvoretter eventuelt den dannede forbindelse av formel (VI) hvori R er en alkylgruppe, fortrinnsvis med fra 1 til 6 carbonatomer, hydrolyseres under dannelse av en forbindelse av formel (VI), hvori R er hydrogen.
Den første del av angitte fremgangsmåte utføres fortrinnsvis i vannløsning, og reduksjonen av den fullt forestrede forbindelse (v) utføres hensiktsmessig under anvendelse av natriumborhydrid eller natriumcyanoborhydrid som reduksjonsmiddel .
En av fordelene med fremgangsmåten ifølge oppfinnelsen som ovenfor angitt, er det forhold at pektin kan anvendes som opprinnelig utgangsmateriale. Som kjent er pektin en av de få naturlig forekommende forbindelser som inneholder ønsket a-1,4-binding, og det er dessuten et billig og lett til-gjengelig utgangsmateriale. Pektin er egnet for fremstilling av forbindelsene med formel (VI) i industriell skala, og nedenfor er det i reaksjonsskjerna 5 vist en sammen-stilling av totalprosessen for fremstilling av a- og (3-glycosider for interessante medisinske og andre anvendelser.
I skjema 5 eksemplifiseres hydrolyseproduktet av pektin
ved digalacturonsyre, men det bør observeres at hydrolyse av pektin også kan resultere i andre oligomerer, eksempelvis fra trimerer opp til dekamerer.
Det som utgangsmateriale anvendte pektin hydrolyseres ved enzymatisk eller sur hydrolyse under dannelse av polygalacturonsyre (a-1,4), som deretter, også her ved enzymatisk eller sur hydrolyse, hydrolyseres til eksempelvis di-galacturonsyre, dvs. forbindelse 13 i skjema 5.
En erholdt blanding av oligogalacturonsyrer kan lett separeres i stor skala ved ionebytterkromatografi (ref. 10).
Den erholdte di-galacturonsyre (forbindelse 13 i
skjema 5) underkastes deretter samtidig forestring og glycosidering med methanol (eller andre enkle alkoholer) og syre på i og for seg kjent måte for andre uronsyrer (ref. 14) under dannelse av tilsvarende nye methylesterglycosider (forbindelse 14 i skjema 5). Forbindelse 14 er en anomer blanding (a/fJ, 3/1) .
Methylesterglycosidet 14 reduseres deretter med en vann-løsning av natriumborhydrid under dannelse av methyl-glycosidet (forbindelse 15 i skjema 5).
Denne forbindelse 15 kan acetyleres under anvend-
else av eddiksyreanhydrid under dannelse av et per-O-acetylert glycosid (forbindelse 16 i skjema 5)
Angitte forbindelse 16 behandles deretter med eddiksyreanhydrid inneholdende konsentrert svovelsyre, hvilket resulterer i spaltning av methylaglyconet uten vesentlig ned-brytning av resten av molekylet, under dannelse av den fullt ut acetylerte di-galactose (forbindelse 5 i skjema 5). Alternativt kan forbindelse 5 dannes i ett trinn fra forbindelse 15 under anvendelse av eddiksyreanhydrid og konsentrert svovelsyre.
Den per-O-acetylerte di-galactose 5 isoleres under anvendelse av den skisserte prosedyre og anvendes for fremstilling av forbindelse 1 og forskjellige, glycosidér derav.
Eksempler
Oppfinnelsen illustreres ytterligere i de etterfølgende konkrete eksempler.
Eksempel 1
Hydrolyse av polygalacturonsyre
50 g poly-D-galacturonsyre (Sigma) ble suspendert i natriumacetatbuffer (1000 ml, 20 mM, pH 4,5), og pH ble jus-tert med natriumhydroxyd (5 M). Suspensjonen ble omrørt ved 40°C, og pektinase (1,0 g, PV 8, Miles) ble tilsatt. Hydro-lysen ble fortsatt i 24 timer ved 40°C og ble deretter av-sluttet ved oppvarmning til 100°C i 5 minutter. Prøver ble tatt ut med intervaller og analysert ved tynnskiktskromato-grafi. Silicagel 60, Merck. Elueringsmiddel: butanol/maursyre/vann 4/6/1. Fremkalling: platene ble tørket, sprøytet med svovelsyre i ethanol (5%) og ble oppvarmet (105°C). Hoveddelen av hydrolysatet besto av di-, tri-, tetra- og pentamerer.
Rensing av hydrolyseproduktene ( jfr, ref. 10)
En del av hydrolysatet inneholdende 1,4-ct-koblede oligomerer av D-galacturonsyre ble ledet gjennom en kolonne av "Zerolite" 225 (H<+>) kationbytterharpiks (600 ml harpiks pr. 1000 ml hydrolysat), og eluatet ble lyofilisert. Den erholdte blanding av oligomerer (6 g) ble oppløst i 20 ml destillert vann, og løsningen ble overført på en kolonne av anionbytter-harpiks (Dowex® 1 x 2, 50-100 mesh, formiatform, brakt i like-vekt med destillert vann). Kolonnen ble eluert med maursyre-løsning (vasking: 350 ml, 0,2M) etterfulgt av en konkav gradient (0,2 M, 2000 ml til 0,65 M, 1200 ml) under dannelse av fraksjoner inneholdende di- og tri-galacturonsyre. Tetra-galacturonsyre ble eluert med 1 M maursyreløsning. Fryse-tørking ga forbindelsene i temmelig ren form (>90%). Utvinn-ingen av D-galacturonsyreoligomerene er angitt i tabell 1.
Fremstilling av 1, 2 , 3 , 6-tetra-O- (2 ,3 , 4 , 6-tetra-O-acetyl-cc-D-galactopyranosyl) -ct-D-galactopyranose (forbindelse 5a i skjema 5) .
20,0 g di-D-galacturonsyre (13) ble tilsatt til en løs-ning av 20 g calciumklorid i 1,0 1 methanol. 15 g Duolite®
(H<+>) ble tilsatt, og blandingen ble kokt under tilbakeløps-kjøling i 18 timer, ble filtrert og inndampet under dannelse av den urene forbindelse 14 (skjema 5). Residuet ble oppløst i 200 ml vann, og en løsning av 6,0 g natriumborhydrid i 30 ml vann ble tilsatt i løpet av 3 minutter under omrøring. Reaksjonsblandingen ble holdt ved omgivelsestemperatur i
20 timer og ble deretter surgjort. (Dowex<®>50 W-X(2)), filtrert, inndampet med methanol flere ganger og deretter tørket (0,1 torr). Residuet (den urene forbindelse 15; skjema 5) ble suspendert i en blanding av eddiksyreanhydrid og pyridin
(1:1, 600 ml), omrørt ved romtemperatur i 24 timer og ble deretter inndampet sammen med toluen flere ganger til et tykt, halvfast residuum uten lukt av pyridin. Residuet ble fordelt mellom 0,5 1 ether og 0,2 1 vann. Vannfasen ble ekstrahert med 2 x 100 ml ether, og de kombinerte etherfaser ble vasket med 50 ml vann, tørket (natriumsulfat) og inndampet. Inndampning sammen med toluen og tørking (0,1 torr) ga et blekgult residuum (31,5 g, hovedsakelig forbindelse 16,
skjema 5). 320 ml eddiksyreanhydrid inneholdende 1% konsentrert svovelsyre ble tilsatt til det tørre residuum, og reaksjonsblandingen ble omrørt ved romtemperatur. Reaksjonen ble overvåket med TLC (Si02 ether/toluen, 3/1). Etter omrøring
i 50 minutter ble reaksjonsblandingen helt over i 1 liter ether, og etherløsningen ble deretter vasket med 150 ml isvann, 150 ml mettet natriumhydrogencarbonatløsning og 150 ml isvann, og ble tørket (natriumsulfat) og inndampet sammen med toluen og tørket (0,1 torr) under dannelse av 32 g av et amorft residuum.Krystallisering fra ethanol ga den angjeldende forbindelse (12,3 g, 33% fra di-galacturonsyre). Kromatografi (Si02, EtOAc/isooctan, 1/1) av modervæsken etterfulgt av krystallisering fra ethanol økte utbyttet til 55%.

Angjeldende forbindelse 5 (skjema 5 og eksempel I), ble fremstilt i ren form ved deacetylering av forbindelse 16 etter følgende prosedyre:
2,5 g av forbindelse 16 ble oppløst i 10 ml varm tetra-hydrofuran. 20 ml methanol ble tilsatt, og blandingen ble avkjølt til romtemperatur. 3 ml (0,1M) methanolisk natrium-methylat ble tilsatt, og reaksjonsblandingen ble omrørt i 24 timer, hvoretter forbindelsen ble utfelt som nåler. Reaksjonsblandingen ble filtrert, og bunnfallet ble vasket med methanol og tørket (0,1 torr) under dannelse av 1,08 g (79%) av forbindelse 15.

ble fremstilt ved kromatografisk rensning av rå-materialet dannet ved første reaksjonstrinn, slik som beskrevet i eksempel III, metode B.. Forholdet mellom a- og 3-isomerene var ca. 3:1 (bestemt ved NMR).

NaBT^ som reduksjonsmiddel. Innføring av slike tyngre isotoper på carbonatomer bundet i 5-stilling resulterer i analytiske fordeler med hensyn til tilsvarende produkt-forbindelser. Innføring av deuteriumatomer for å erstatte hydrogenatomer medfører således at den forstyrrelse som tilveiebringes ved nærvær av lactose i massespektrometiske målinger elimineres. På den annen side vil innføring av tritiumatomer på angitte carbonatomer bevirke at de resulter-ende forbindelser er påvisbare ved radioaktive målinger. Begge disse faktorer letter vesentlig påvisningen av forbindelsene når de anvendes i testsystemer eller ved terapeutisk eller diagnostisk anvendelse. ;Eksempel . 1. Fremgangsmåte for fremstilling av en forbindelse av formel (VI):
hvori n er et helt tall opp til 10, innbefattende 0, R er hydrogen eller en alkylgruppe, fortrinnsvis med fra 1 til 6 carbonatomer, og X er hydrogen, deuterium eller tritium,karakterisert vedat man reduserer en forbindelse av formel (\):
hvori n har den ovenfor angitte betydning og R er en alkylgruppe, fortrinnsvis med fra 1 til 6 carbonatomer, hvoretter eventuelt den dannede forbindelse av formel (VI) hvori R er en alkylgruppe, fortrinnsvis med fra 1 til 6 carbonatomer, hydrolyseres under dannelse av en forbindelse av formel (VI), hvori R er hydrogen.
2. Fremgangsmåte ifølge krav 1,karakterisert ved. at det innføres deuterium-eller tritiumatomer på de i 5-stillingen bundne substituent-carbonatomer ved å anvende NaBD^eller NaBT^som reduksjonsmiddel.
