Description
Foreliggende oppfinnelse angår insekt- og middfrastøtende midler, anvendelsen av disse midler for bekjempelse av insekt og/eller midd samt acylerte a ,to-aminoalkoholderivater.
Midler som frastøter insekter og midd (repellenter), har den oppgave å fraholde skadelige eller belastende leddfotdyr fra berøring, så vel som stikking og suging eller biting på overflater som virker tillokkende for dem, eksempelvis huden hos dyr og mennesker, når disse på forhånd ble behandlet med slike midler.
Som repellenter er det allerede foreslått tallrike virksomme stoffer (se for eksempel K.H. Btlchel i "Chemie der Pflanzen-schutz- und Schådlingsbekåmfungsmittel"; utgiver: R. Wegler, bind 1, Springer Verlag Berlin, Heidelberg, New York, 1970, side 487 og følgende).
Spesielt kjent og i lengre tid i bruk er 3-metyl-benzosyredi-etylamid (DEET), dimetylftalat og 2-etyl-heksandiol-l,3, hvorav fremfor alt DEET i praksis har fått en betraktlig betydning, (se for eksempel R.K. Kocher, R.S. Dixit, CI. Somaya; "Indian J. Med. Res.", 62, 1 (1974)).
En betraktlig ulempe ved de kjente repellenter er deres delvis relativt korte (bare få timer) permanentvirkning.
En del av de ved nedenstående formel (I) definerte forbindelser er kjent.
Tilsvarende polyhydroksyaminer er kjent for eksempel fra "Chemical Abstracts", 83 (25): 205/791 u.
Videre er N-alkanoyl- og -alkenoyl-hydroksyalkylpiperidiner kjent fra US-PS 3 178 439. Ytterligere piperidiner er kjent fra "Tetrahedron Suppl.", 8/del 1 (1966) side 113-121.
En insekt- og middavvisende virkning av disse forbindelser er imidlertid tidligere ikke vært kjent.
Det er nu funnet at de delvis kjente acylerte a ,co-aminoalkoholderivater med formel I
der
X betyr hydrogen, COR11 eller R13,
R<1> betyr C1_7~alkyl eller Cg_7-alkenyl,
R2, R11 og R<13> er like eller forskjellige og betyr C1_6-alkyl,
R<3->r<8> er like eller forskjellige og betyr hydrogen eller C^^-alkyl, hvorved R<2> og R<3> sammen med atomene hvortil de er bundet kan danne en 5- eller 6-leddet monocyklisk
ring,
n er 1 og
m er 0,
har en sterk insekt- og middfrastøtende virkning (repellent-virkning).
Repellentvirkninger er betraktlig bedre enn den til de fra teknikkens stand kjente repellenter. De ifølge oppfinnelsen virksomme stoffer er således en verdifull berikning av teknikken.
Foreliggende oppfinnelse angår i henhold til dette insekt- og middfrastøtende midler som karakteriseres ved et innhold av minst et acylert ot ,o)-aminoalkoholderivat med den ovenfor angitte formel I.
Som antydet innledningsvis angår oppfinnelsen også anven-deisen av slike acylerte a ,a>-aminoalkoholderivater med formel I for bekjempelse av insekter og/eller midd.
Midlene ifølge oppfinnelsen inneholder et derivat med formel I, kan også inneholde ytterligere Insektbeskyttende midler. Her kommer på tale alle praktisk vanlige repellenter (se for eksempel K.H. Btlchel, supra).
I tilfelle av repellentkombinasjoner anvendes fortrinnsvis de acylerte ot.u-aminoalkoholer med den generelle formel I sammen med repellenter som karboksylsyreamider, 1,3-alkandioler og karboksylsyreestere. I detalj skal det nevnes: 3-metyl-benzosyredietylamid (DEET), 2-etyl-heksandiol-l,3 (Rutgers 612) og ftalsyredimetylester.
De i formel I angitte rester har fortrinnsvis følgende betydning: Det foretrekkes forbindelser, hvori R<1> betyr C1_7~alkyl eller C3_7-alkenyl, X betyr COR<11> eller R<13>, R<2> og R11 er like eller forskjellige og betyr C.j_6-alkyl, R3 til R<8> er like eller forskjellige og betyr hydrogen eller C-alkyl, R<13> betyr C^^-alkyl og n betyr 1 og m betyr 0.
Helt spesielt foretrukket anvendes forbindelsene med den generelle formel I som repellenter, der
m = 0 og
n 1,
R<1> betyr C^-alkyl,
R2, R1<1> og R1<3> er like eller forskjellige og betyr C1_&-alkyl,
R<3->r<8> betyr hydrogen, og
X betyr hydrogen, COR<11> eller R<13>, idet R11 og R<13> har
ovennevnte betydning.
Videre anvendes helt spesielt foretrukket forbindelser med den generelle formel I som repellenter, der
m = 0 og
n =1,
R<1> betyr C3_4-alkyl,
R<2> og R<3> danner sammen med atomene hvortil de er bundet en 6-leddet ring,
R 4-R 8 etyr hydrogen, og
X betyr hydrogen og R1<3>, idet R13 betyr Cj^-alkyl.
Forbindelsene med den generelle formel I er enten kjent eller kan fremstilles efter kjente metoder og fremgangsmåter (se for eksempel Cesare Ferri, "Reaktionen der organischen Synthese", Georg Thieme Verlag, Stuttgart, 1978, side 223 og side 450).
Man får følgelig forbindelsene med formel I, når de i og for seg kjente eller efter kjente fremgangsmåter fremstillbare a,(i)-aminoalkoholer (se for eksempel Cesare Ferri, supra, side 211 ff henholdsvis 496-497) med formel II
der R<2> til R<*0> og n og m har den under formel I angitte betydning, først omsettes med i og for seg kjente karboksylsyreklorider med formel III
idet
R<*> har den under formel I kjente betydning,
eventuelt i nærvær av en syreakseptor som for eksempel trietylamin eller kalsiumkarbonat og eventuelt under anvendelse av et fortynningsmiddel som for eksempel toluen,
CH2CI2, tetrahydrofuran eller acetonltril, ved temperaturer mellom -40 og 110'C.
For fremstillingen av forbindelser med den generelle formel I der X er forskjellig fra hydrogen, foregår da i et andre reaksjonstrinn, eventuelt efter isolering av mellomproduktet med fri OH-gruppe, den videre acylering/alkylering med i og for seg kjente karboksylsyreklorider med formel IV
for fremstilling av forbindelser med formel I med X = COR<11>;

eventuelt i nærvær av en syreakseptor som for eksempel trietylamin eller kaliumkarbonat eller en base som natriumhydrid, eventuelt under anvendelse av et fortynningsmiddel som for eksempel toluen, tetrahydrofuran eller acetonltril, ved temperaturer mellom 0 og 110°C. Opparbeidelsen foregår efter vanlige metoder, for eksempel ved ekstrahering av produktene med metylenklorid eller toluen fra den med vann fortynnede reaksjonsblanding, vasking av den organiske fase med vann, tørking og destillering, eller såkalt "pådestil-
En ytterligere rensing kan foregå ved kromatografi på kiselgel med for eksempel heksan:aceton = 7:3 som elueringsmiddel.
Til karakterisering av forbindelsene tjener brytningsindeks, smeltepunkt, Rf-verdi eller kokepunkt.
Oppfinnelsen vedrører også nye acylerte a.u-aminoalkoholderivater som karakteriseres ved formel

idet R11 og R<13> er like eller forskjellige, og betyr
C1_6-alkyl, og
R11 betyr C2_6-alkyl eller C3_6-<a>lkenyl.
Man får de acylerte a ,a)-aminoalkoholder ivater med formel

Opparbeidelsen av forbindelsen ifølge oppfinnelsen med formel Ia foregår på analog måte, som omtalt ovenfor ved fremstilling av forbindelsene med formel I.
Virkningen av repellentene med den generelle formel I varer 1enge.
De kan derfor med godt resultat anvendes til beskyttelse av skadelige eller belastende, sugende og bitende insekter og midd.
Til de sugende insekter hører i det vesentlige stikkmygg (for eksempel Åedes-, Culex- og Anopheles-typene), sommer-fuglmygg (Phlebotomen), nisser (Culicoides-typer), myldremygg (Simulium-typen), stikkfluer (for eksempel Stomoxys calci-trans ), Tsetsefluer (Clossina-typer), bremser (Tabanus-, Haematopota- og Chrysops-typer), stuefluer (for eksempel Musea domestica og Fannia canicularis), kjøttfluer (for eksempel Sarcophaga carnaria), Myiasis-frembringende fluer (for eksempel Lucilia cuprina, Chrysomyia chloropyga, Hypoderma bovis, Hypoderma lineatum, Dermatobia hominis, Oestrus ovis, Gasterophilus intestinalis, Cochliomyia hominovorax), vamser (for eksempel Cimex lectularius, Rhodnlus prolixus, Trlatoma infestans), lus (for eksempel Pedlculus humanus, Haemotopinus suls, Damallna oris), lusfluer (for eksempel Melaphagus orinus), flott (for eksempel Pulex irritans, Cthenocephalides canls, Xenopsylla cheopis) og sandflott (for eksempel Dermatophilus penetrans).
Til de bitende insekter hører i det vesentlige kakerlakker (for eksempel Blatella germanica, Periplaneta americana, Blatta orientalis, Supella superllectilium), biller (for eksempel Sitophilus granarius, Tenebrio molitor, Dermestes lardarius, Stegobium paniceum, Anobium puntactum, Hylotrupes bajulus), termitter (for eksempel Teticulitermes lucifugus) og maur (for eksempel Laslus niger).
Til midd hører flott (for eksempel Ornithodorus moubata, Ixodes ricinus, Boophilus microplus, Amblyomma hebreum) og midd i snevrere forstand (for eksempel Sarcoptus scabiei, Dermanyssus gallinae).
De virksomme stoffer som anvendes i midlene ifølge oppfinnelsen kan anvendes ufortynnet eller fortrinnsvis fortynnet og lar seg overføre til de for repellenter vanlige formuleringer. De lar seg anvende i alle i kosmetikken vanlige administreringsformer, for eksempel oppløsninger, emulsjoner, geler, salver, pastaer, kremer, pulvere, stifter, sprays eller aerosoler fra forstøvningsbokser.
For anvendelsen i det ikke-kosmetiske området lar det virksomme stoffet seg innarbeide for eksempel i granulater, oljeforstøvningsmidler eller "slow-release"-formuleringer. Tilberedningene fremstilles på kjent måte ved sammenblanding eller fortynning av de virksomme stoffer med oppløsnings-midlet (for eksempel xylen, klorbenzener, paraffiner, metanol, etanol, isopropanol, vann), bærere (for eksempel kaoliner, leirjord, talkum, kritt, høydispers kiselsyre, silikater), emulgeringsmidler (for eksempel polyoksyetylen-fettsyreester, polyoksyetylenfettalkoholetere, alkylsulfo-nater, arylsulfonater) og dispergeringsmidler (for eksempel ligninsulfittavler, metylcellulose).
De virksomme stoffer kan blandes med hverandre i formule-ringene eller også anvendes i blanding med andre kjente virksomme stoffer (for eksempel solbeskyttelsesmidler). Tilberedningene inneholder vanligvis mellom 0,1 og 95 vekt-# virksomt stoff, fortrinnsvis mellom 0,5 og 90$.
For beskyttelse mot blodsugende insekter eller midd påføres midlene ifølge oppfinnelsen med virksomme stoffer enten på den menneskelige eller dyriske hud, eller man behandler bekledningsstykker eller andre gjenstander med midlene.
Også som tilsetning av impregneringsmidler, for eksempel ved tekstilbaner, bekledningsstykker, forpakningsmateriale, samt som tilsetning til polerings-, pusse- og vindusrensemidler er de virksomme stoffer egnet.
De følgende eksempler på tilberedninger og anvendelse av de virksomme stoffer skal illustrere oppfinnelsen videre.
Eksempel 1
Et repellentmiddel i form av en lotion til anvendelse på huden fremstilles ved sammenblanding av 30 deler av et av de virksomme stoffer, 1,5 deler parfyme og 68,5 deler isopropanol. Isopropanol kan erstattes med etanol.
Eksempel 2
Et repellentmiddel i form av en aerosol til påsprøytning på huden fremstilles idet man formulerer en 50 #-ig stoff-oppløsning bestående av 30 deler av et av de virksomme stoffer, 1,5 deler parfyme og 68,5 deler isopropanol med 5056 Frigen 11/12 (= halogenert hydrogen som drivgass) som forstøvningsbokspreparat.
Eksempel 3
En annen forstøvningsdose er sammensatt av 40 #-ig stoff-oppløsning med 20 deler av en av de virksomme stoffer, 1 del parfyme, 79 deler isopropanol og 60# propanrbutan I forholdet 15:85.
Det ble fremstilt individuelle formuleringer tilsvarende eksemplene 1, 2 og 3 under anvendelse av følgende virksomme stoffer ifølge fremstillingseksemplene nr. 1, 2, 3, 8 og 15.
De følgende eksempler av biologisk virkning viser overlegen-heten av stoffene i forhold til teknikkens stand (dietyltoluamid = DEET):
Eksempel A
Repellentprøve på marsvin.
Prøvedyr: Åedes aegypti (Imagines)
Antall prøvedyr: ca. 5 000
Oppløsningsmiddel: etanol (99,8#)
3 vektdeler virksomt stoff opptas i 100 volumdeler oppløs-ningsmiddel .
Et marsvin, hvis rygg har blitt barbert i et område på 50 cm<2 >fikseres i et trangt bur (boks) således, at bare den barberte flate er tilgjengelig for myggen. Efter behandling av flaten med 0,4 ml virksomt stoffoppløsning stilles marsvinet efter fordamping av oppløsningsmidlet, samt boks, i et bur som måler 60 cm x 60 cm x 60 cm og som inneholder kun med sukkervann forede prøvedyr av begge kjønn.
I 10 minutter observerer man hvor mange mygg som stikker marsvinet. Prøven gjentas efter en time. Forsøket gjennom-føres i maksimalt 14 timer eller så lenge inntil virkningen opphører.
Ved denne prøve viser for eksempel følgende forbindelser fra fremstillingseksemplene overlegen virkning i forhold til teknikkens stand (dietyltoluamid = DEET):

Eksempel B
Repellentprøve på marsvin
Prøvedyr: Culex pipiens fatigans
Antall prøvedyr: ca. 1 000
Oppløsningsmiddel: etanol ( 99, 8%)
3 vektdeler virksomt stoff opptas i 100 volumdeler oppløs-ningsmiddel .
Et marsvin, hvis rygg er blitt barbert i et område på 50 cm<2 >fikseres i et lite bur (boks), således at bare den barberte flate av ryggen er tilgjengelig. Efter behandling av flaten med 0,4 ml virksomt stoffoppløsning, plasseres marsvinet efter fordampning av oppløsningsmidlet samt boks, i et bur av dimensjoner 60 cm x 60 cm x 60 cm, som inneholder kun med sukkervann forede prøvedyr av begge kjønn.
Det iakttas over 10 minutter hvor mange mygg som stikker marsvinet.
Derefter tas dette ut og prøven gjentas efter 1 time. Forsøket gjennomføres maksimalt i 10 timer eller inntil virkningen opphører.
Ved denne prøve viser for eksempel følgende forbindelser fra fremstillingseksemplene overlegen virkning i forhold til teknikkens stand (dietyltoluamid = DEET):

FREMSTILLINGSEKSEMPLER
Fremstilllngseksempel 1
N.O-bis-acetyl-N-butyl-aminopropanol-l,3
40 g (0,3 mol) N-butyl-aminopropanol-1,3 og 100 ml trietylamin (0,72 mol) oppløses 1 1 liter tetrahydrofuran og blandes ved 20° C med 50 ml acetylklorid (0,7 mol). Man oppvarmer 1 dag under tilbakeløp og frafiltrerer derefter det faste stoff. Man tilsetter metylenklorid og vasker den organiske fase vaskes med vann. Derefter tørkes den organiske fase med magnesiumsulfat, oppløsningsmidlet avdestilleres på rotasjonsfordamper og destilleres i kulerørovn (Kokepunktgf6 135-140°C). For ytterligere rensing kromatograferes stoffet over 1 kg kieselgel (elueringsmiddel: cykloheksan:aceton = 7:3).
Utbytte: 52,2 g = 81$ av det teoretiske.
Fremstilllngseksempel 2
l-pentanoyl-2-(2-hydroksyetyl)-piperidin
65 g (0,5 mol) 2-(2-hydroksyetyl)-piperidin og 90 ml trietylamin (0,64 mol) oppløses i 1 liter tetrahydrofuran og blandes med -20°C med 80 ml (0,67 mol) valeriansyreklorid. Man oppvarmer i 1 dag ved 20°C og fjerner derefter oppløsnings-middel best mulig på rotasjonsfordamper, tar opp med metylenklorid, vasker med IN NaOH-oppløsning, tørker den organiske fase og fjerner oppløsningsmidlet destillativt på rotasjonsfordamper. For å fjerne forurensninger i form av bis-acylerte forbindelser tas det hele opp i 200 ml etanol og oppvarmes i 1 time ved 50°C med 200 ml IN natronlut. Resten dampes igjen inn i en rotasjonsfordamper, ekstraheres med CH2CI2/H2O, den organiske fase tørkes, dampes nok en gang inn i rotasjonsfordamper og destilleres i kulerørovn (Kokepunktgf2 165°C).
Utbytte: 65,4 = 61% av det teoretiske.
Fremstilllngseksempel 3
l-pentanoyl-2-(2-metoksyetyl)-piperidin
32 g (0,15 mol) l-pentanoyl-2-(2-hydroksyetyl)-piperidin oppløses i 300 ml tetrahydrofuran og blandes ved 20°C med 5,9 g (0,197 mol) natriumhydrid (80#-ig i paraffin). Man oppvarmer i 4 timer under tilbakeløp, og tilsetter derefter 20 ml metylenjodid (0,32 mol).
Derefter oppvarmes i 8 timer under tilbakeløp, derefter blandes ved 20°C med 100 ml ammoniumkloridoppløsning, ekstraheres med metylenklorid, den organiske fase tørkes med magnesiumsulfat og innroteres. Efter kromatografisk rensing (elueringsmiddel CH2CI2:i-CsHyOH = 1:1; kiselgel) dampes resten inn i rotasjonsfordamper og destilleres i kulerørovn (kokepunkt0,2 : 160<S>C).
Utbytte: 27,7 g = 81% av det teoretiske.
Analogt til de ovennevnte fremstillingseksempler 1 til 3 ble det de nedenfor tabellarisk oppførte ytterligere fremstillingseksempler fremstilt.

Insekt- og middfrastøtende middel, karakterisert ved et innhold av minst et acylert a.o-amino-alkoholderivat med formel (I):

R<1> betyr C1_7~alkyl eller C3_7-alkenyl,
R2, R11 og R<13> er like eller forskjellige og betyr
alkyl,
R<3->R<8> er like eller forskjellige og betyr hydrogen eller
C^_^-alkyl, hvorved R<2> og R<3> sammen med atomene hvortil de er bundet kan danne en 5- eller 6-leddet monocyklisk ring.

Anvendelse av acylerte a,o)-aminoalkoholderivater med formel (I) for bekjempelse av insekter og/eller midd.
