Abstract
Foreliggende oppfinnelse angår generelt nye fremgangsmåter for syntesen av cyklopropylacetylen som er en vesentlig reagens for den asymmetriske syntesen av (S)-6- kloro-4-cyklopropyletynyl-4-trifluorometyl-1,4-dihydro-2H-3,l-benzoksazin-2-on; en nyttig humant-immunsvikt-virus (HIV) reverstranskriptaseinhibitor. Ved denne fremgangsmåten kondenseres cyklopropankarboksaldehyd med malonsyre for å danne 3- cyklopropylakrylsyre; 3-cyklopropylakrylsyre halogeneres for å danne (E,Z)-l-halo-2- cyklopropyletylen; og (E,Z)-l-halo-2-cyklopropyletylen dehydrohalogeneres for å danne cyklopropylacetylen. Denne forbedringen tillater høy omdannelse av billige, lett tilgjengelige startmaterialer til cyklopropylacetylen, høye generelle utbytter og kan utføres i en industriell målestokk.
