Abstract

Transfeksjonspartikler for levering av nukleinsyre inn i høyere eukaryote celler in vitno og in vivo omfatter en eller flere nukleinsyremolekyler kondensert av organiske kationiske molekyler. Diskrete og stabile partikler blir oppnådd ved kompleksdannelse av nukleinsyremolekyler med identiske eller forskjellige organfske kationiske forløpermolekyler uten kryssbinding av nukleinsyre-molekylene, og kovalent kobling av forløpermolekylene til hverandre på nuklein-syretemplatet. For spesifikk cellulær målsøking kan partiklene bære målsøkende molekyler, f.eks. sukkere. Foretrukne kationiske forløpermolekyler er lipofile detergenter som er koblet for å danne lipider. Partiklene inneholder fortrinnsvis bare ett nukleinsyremolekyl som gjør dem nyttige for genterapi og for. levering av store DNA molekyler.
