Abstract

Ekspresjonsvektorene og fremgangsmåtene som benytter et E.coli-uttrykkingssystem for storskalaproduksjon av IL-21 er beskrevet. Vektorene benytter den IL-21-kodende sekvensen med spesifikke endringer i nukleotider for å optimalisere kodons og mRNA-sekundærstruktur for translatering i E.coli. Ved å benytte ekspresjonsvektorene ble IL-21-genet produsert i E.coli til et nivå på større enn I g/I i tilsatt ladningsfermentering. Også inkludert er OmpTmanglende E.coli-stamme fransformert med en IL-21 -ekspresj onsvektor.
