Abstract
Multipleks overlapp-forlengelses RT-PCR fremskaffer en effektiv fremgangsmåte for linking av to eller flere nukleotidsekvenser kodende for områder eller subenheter av et heteromert protein i en enkel reaksjon. Spesielt blir koplingen av sekvenser som koder for variable regioner fra f eks. immunoglobuliner. T-celle reseptorer eller B-celle reseptorer forenklet ved fremgangsmåten ifølge foreliggende oppfinnelse. Dette tillater en mer effektiv måte for generering av biblioteker av sekvenser som koder for variable regioner. Derme muligheten for å utføre den multiplekse overlapp- forlengelses RT-PCR ved bruk av templat avledet fra en isolert enkeltcelle muliggjør genereringen av bibliotek av kognate par i et høygjennomløpsformat.
