Description
Foreliggende oppfinnelse vedrører nye behensyre-estere med den følgende generelle formel:
hvori X er "N" eller "CH", ikke-toksiske syreaddisjonssalter derav, en metode for fremstilling av .disse esterne og tera-peutiske komposisjoner derav med forlenget virkning.
I de siste år er estere av neuroleptisk aktive fenotiaziner blitt foreslått og funnet anvendelige i preparater med forlenget virkning ved parenteral adminstrering. Esterne som er funnet mest anvendelige er enanten-,dekanon- og palmitinsyre-estere av flufenazin som hovedsaklig administreres som sterieleløsninger i vegetabilske oljer som injuseres intramuskulært. Den neuroleptiske effekten til slike løsninger kan vare opptil 15 dager.
Også alifatiske karbokaylsyreestere av noen meget sterke neuroleptiske fenotiaziner med et fluoratom i 7-stilling er blitt | foreslått og omfatter estere av alifatiske karboksylsyrer med opptil og til og med 17 karbonatomer, særlig dekanon- og pal.mi- . tinsyreestere.
Under fortsatt arbeid med sådanne estere i farmakologiske eksprimenter ble det funnet at dekanon- og palmitinsyreesterne av det meget sterkt nevrolytiske 2-trifluormetyl-7-fluor-10-(31 -(4-(2-hydroksyetyl)-1-piperidyl)propyl)fenotiazin og 2-trifluometyl-7-fluor-10-(3'-(4-(2-hydroksyetyl)-1-piperazinyl) propyl)fenotiazin administrert i ordinære doser som oljeløs-ninger ved injeksjon hadde tendens til å gi for høye konsen-trasjoner i begynnelsen som forårsaket en utpreget sedativ virkning den første uken.
Det er nå overraskende blitt funnet at behensyreesterne med formel I ikke bare er en mer forlenget virkning, men også for-årsaker mindre sedativ virkning hos forsøksdyrene ved injeksjon i form av oljeløsninger. Det synes også som andre bi-effekter som ekstrapyrimidale symptomer hovedsaklig unn-gås .
Forbindelsene med formel I kan fremstilles - ifølge metoden ved foreliggende oppfinnelse.- ved å omsette en forbindelse med følgende generelle formel:
hvori X er som ovenfor angitt med et reaktivt derivat av
behensyre som et syrehalid eller anhydridet, hvor<p>å forbindelsen med formel I som dannes ved reaksjonen isoleres som fri base eller som en ikke-toksisk syreaddisjonssalt derav.
Foresteringsprosessen ifølge oppfinnelsen utføres fortrinns-' vis i nærvær av et inert organisk løsningsmiddel som et keton, fortrinnsvis aceton eller en eter som dietyleter.
Det reaktive derivatet av behensyre er fortrinnsvis et syrehalid, spesielt syreklorid eller syreanhydrid.
De ikke-toksiske syreaddisjonsaltene av forbindelsene med formel I er fortrinnsvis salter av farmasøytisk fordragelige syrer som mineralsyrer, f.eks saltsyre, bromhydrogensyre, fosforsyre, svovelsyre og liknende, og organiske syrer som eddiksyre, vinsyre, maleinsyre, sitronsyre, metansulfonsyre og liknende.
De følgende eksempler illustrer metoden ved oppfinnelsen men skal ikke ansees som begrensende: EKSEMPEL 1
Behensyreesteren av 2-trifluormetyl-7-fluor-10-(31 -(4-(2-hydroksyetyl)-1-piperidinyl)pro<p>yl)fenotiazin.
24 g 2-trifluormetyl-7-fluor-10-(3'-(4-(2-hydroksyetyl)-1-piperidinyl)propyl)fenotiazin, HC1 ble overført til den tilsvarende frie base på vanlig måte. Basen ble oppløst i 200
ml tørr aceton, 40 g behensyreklorid ble tilsatt og blandingen kokt ved tilbakeløp på et vannbad i en time. Reaksjonsblandi-ngen ble inndampet i vakuum, resten behandlet med en blanding av 200 ml vann og 200 ml eter hvorpå eterfasen ble fraskilt. Eterfasen ble så rystet med en kald 10% vandig natriumkarbonat-løsning til alkalisk reaksjon. Eterfasen ble separert, tørket over vannfritt magnesiumsulfat og inndampet i vakuum. Resten ble krystallisert fra heksan og bestod av behensyreesteren av 2-trifluormety1-7-flour-10-(3'-(4-(2-hydroksyetyl)piperi-dyl)propyl)-fenotiazin, som smelter ved 57 - 58°C. Utbytte: 22.5 g.
EKSEMPEL 2
Behensyreesteren av 2-trifluormetyl-7-fluor-10-(3'-(4-(2-hydroksyetyl)-1-piperazinyl)propyl)fenotiazin.
Når eksempel 1 ble utført ved anvendelse av 32 g 2-trifluor-metyl-7-fluor-10-(31 -(41 -(2-hydroksyetyl)-1-piperazinyl)-propyl)fenotiazin, 2 HC1 istedet for 2-trifluormetyl-7-fluor-10-(3'-(4-(-hydroksyetyl)-1-piperidyl)propyl)fenotiazin, HC1 erholdtes behensyreesteren av 2-trifluormetyl-7.fluor-10-1(3'-(4-(2-hydroksyetyl)-1-1.piperazinyl)propyl)-fenotiazin som en hvit krystallinsk substans som smeltet ved 62 - 64°C.
Utbytte: 25 g.
Oppfinnelsen gir videre farmasøytiske komposisjoner med
forlenget virkning som har som aktiv bestanddel, en forbindelse , med formel I eller en av dens ikke-toksiske syreaddisjonssalter sammen med et'farmasøytisk bæremiddel eller fortynningsmiddel.
De kan administreres til dyr inklusive mennesklige skapninger både oralt, parenteralt og rektale og kan ha form av f.eks. sterile løsninger eller suspensjoner for injeksjon, tabletter, suppositorer, kapsler og siruper. Resultater av administrering til mennekselige skapninger av komposisjoner ifølge oppfin-neslen har vært meget lovende.
'Fortrinnsvis foreligger imidlertid komposisjonene i form av sterile løsninger eller suspensjoner for injeksjon, og en foretrukken utførelsesform av oppfinnelsen kan injeserbare løsninger fremstilles fra en ikke-toskisk injeserbart fett eller olje, f.eks. et vegetabilsk olje, sesamolje, olivenolje, arakisolje eller etyloleat, og de kan i tillegg inneholde delingsmidler, f.eks. aluminiumstearat for å forsinke absorp-sjonen i kroppen. Slike oljeløsninger har en sterkt forlenget virkning ved intramuskulær injeksjon og tilfredstillende neuroleptisk virkning er oppnådd ved en enkelt intramuskulær injeksjon av ca. 25-40 mg og en forbindelse med formel I opp-løst i en lett.vegetabilsk olje i opptil 2 til 6 uker.
Den foretrukne forbindelsen med formel I for bruk i de farma-søytiske komposisjoner ifølge oppfinnelsen er behensyreesteren av 2-trifluormetyl-7-fluor-10-(31 -(4-(2-hydroksyetyl)-1-piperidyl)propyl)fenotiazin i det følgende kalt Lu 13-112
for korthets skyld.
Det følgende eksempler illustrer de injeserbare oljeaktige løsniner ifølge foreliggende oppfinnelse:
Løsningene fylles for eksempel i ampuller som hver inne-;-.: holder 1 ml oppløsninge.
Den aktive bestanddelen med formel I kan også administreres
i form av en suspensjon av mikronisert aktiv bestanddel eller et salt derav i sterilt fysiplogisk saltvann.
Ethvert annet farmasøytisk hjelpemiddel kan brukes forutsatt at det er forenelig med den aktive bestanddel, og ytterligere komposisjoner og doseringsformer kan likne dem som for tiden brukes for neuroleptika. Også kombinasjoner av en forbindelse med formel I så vel som dets farmasøytisk ikke-toksiske syre-addis jonssalter med andre aktive bestanddeler, spesielt andre neuroleptika, tymoleptiske eller liknende faller innen rammen av foreliggende.^oppfinnelsen.
Ved prøving av behensyreesterne med formel I er det funnet fordelaktig å anvende den velkjente prøven som er basert på det faktum at sterke neuroleptika antagoniserer apomorfin indusert brekninger hos hunder. Forsøket er beskrevet av P.A.J. Janssen, C.J.E.Niemegeers og K.H.L. Schellehaus: "Is it possible to predict the clinical effects of neuro-leptic durgs (major tranquillisers) form animal data", del II, Arzneimittel Forschung, 15_, 1196-1206 , 1965 . For å be-dømme esterne av forbindelsene med formel I er en modifisert metode beskrevet av M. Nymakr et al. i Acta pharmacol. et toxicol, 1973, 3_3 , 363-376 .
Prøven kan beskrives kort som følger:
Apomorfinantagonisme hos hunder
Som dyr ble anvendt voksne renrasede Beagleshunder av begge kjønn. Grensedoseringen av apomorfin hydroklorid for induk-sjon av brekninger hos hundene er bestemt til å ligge på 0.025 mg pr. kg intravenøst. Etter denne dosen opptrer brekninger innen få minutter vetter injeksjonen. Fire hunder ble brukt for hver doseringsnivå av drogen som ble injesért subkutant bak i nakken. Ved forskjellige tidspunkter etter drogedamini-streringen ble hundene så tilført apomorfin ifølge et "opp og ned" skjema som brukte doseringsområdet 0.025 - 0.400 mg pr.
kg intravenøst geometrisk fordelt. Hvis således f.eks. en hund hastet opp etter 0.1 mg pr. kg fikk den neste hunden 0.05 mg pr. kg eller 0.2 mg pr. kg hvis den første hunden ikke kastet opp, og så videre. På denne måten var det mulig å bestemme hvilket nivå apomorfin hundene ble beskyttet en viss tid. Hundene ble foret en halv time før prøven for å sikre et lett oppkast.
Estere som enanton- og dekanonsyreesterne av sterke neuroleptika er tidligere blitt foreslått, og enantonsyresteren av flufenazin har faktisk funnet anvendelse i senere år i form N av oljeløsninger for injeksjon med "sustained release effect". - Det er nå funnet at Lu 13-112 synes å være mer fordelaktig enn den tilsvarende palmetinsyreesteren (Lu 13-113) og betraktelig mer fordelaktig enn denne enantonsyreesteren av flufenazin. Lu 13-112 (1.6 mg pr. kg s.c.) beskyttet hundene mot oppkast opptil 21 dager når en løsnining i lett vegetabilsk olje ble injesert uten forstyrrende sedativ virkning og ga en maksimal beskyttelse (12 x terskeldosen), mens Lu 13-113 beskyttet hundene opptil 16 dager og forårsaket betraktelig sedativ virkning den første uken. Flufenazin enantat beskyttet hundene opptil 14 dager og ga bare en maksimal beskyttelse på 3 x terskeldosen uten synderlig sedativ virkning.den første uken.
Claims (11)
1. En behensyreester med formelen
karakterisert ved at X er "N" eller "CH" og syreaddisjonssalter derav med farmasøytisk fordragelige syrer.
2. Behensyreester ifølge krav 1 med følgende formel karakterisert ved at X 23 "N" eller "CH".
3. Behensyreester ifølge krav 1 eller 2 karakterisert ved formelen
4. Behensyreester ifølge krav 1 eller 2 karakterisert ved formelen
5. Fremgangsmåte for fremstilling av benhensyreester med formelen
hvor X er "N" eller "CH", og syreaddisjonsalter derav med1 :1 farmasøytiske fordragelige syrer, karakterisert ved at en forbindelse med den følgende formel
hvor X er som ovenfor angitt omsettes med et reaktivt derivat av behensyre, hvorpå forbindelsen med formel I som dannes ved reaksjonen isoleres som fri base eller som et ikke-toksisk syreaddisjonssalt derav.
6. Fremgangsmåte ifølge krav 4 karakterisert ved at det som reaktivt derivat av behensyre anvendes
syreklorider derav.
7. Fremgangsmåte ifølge krav 5 eller 6 karakterisert ved at 2-trifluormetyl-7-fluor-10-(31 -(4-(2-hydroksyetyl)-1-piperidyl)propyl)-fenotiazin omsettes med behensyreklorid, hvorpå esteren som dannes isoleres i form av fri base eller et ikke-toksisk syreaddisjonsalt derav.
8. Fremgangsmåte ifølge krav 5 eller 6 karakterisert ved at 2-trifluormetyl-7-fluor-10-(3 <1-> (4-(2-hydroksyetyl)-1-piperazinyl)propyl)fenotiazin omsettes med behensyreklorid, hvorpå den dannede ester isoleres i form av fri base eller et ikke-toksisk syreaddisjonssalt derav.
9. Fremgangsmåte iføgle krav 4, 5, 6, 7 eller 8 karakterisert ved at den dannede ester isoleres i form av den fri base.
10. En farmasøytisk komposisjon karakterisert ved at den består av en hovedmengde farmasøytisk bæremiddel og en farmasøytisk effektiv dose av en forbindelse ifølge krav 1.
11. Komposisjon ifølge krav 10 karakterisert ved at den aktive bestanddel er behensyreesteren av 2-tri-fluormetyl-7-fluor-10-(3 <1> (-4-(2-hydroksyetyl)-1-piperidyl) propyl)fenotiazin.
