Abstract

Oppfinnelsen vedrører en fremgangsmåte for homogen immun- analyse av en ligand i en flytende testprøve som omfatter: a) inkubering av en blanding av (1) den flytende testprøve; (li) hvor liganden under bestemmelse har bare en epitop, et kovalent konjugat av liganden med et hvilket som helst mono- eller polyepitopmolekyl som har minst en epitop atskilt fra ligandens epitop; (2) et labilt enzym; (3) et første bispesifikt mono- klonalt antistoff som beskytter enzymets aktivitet ved å binde seg til en første epitop på enzymet og som er istand til å binde liganden under bestemmelse; og (4) et andre bispesifikt mono- klonalt antistoff som er istand til å binde nevnte enzym og en andre atskilt epitop på liganden under bestemmelse, eller en epitop av nevnte kovalente konjugat; hvorved det dannes et kvaternært immunkompleks; b) etter inkubering, å utsette blandin- gen for betingelser, hvorved et hvilket som helst enzym som ikke er tilstede i et slikt kvaternært immunkompleks blir inaktivert; og c) å bestemme mengden av påvisbart enzym, idet bestemmelsen relateres til mengden, dersom det er noen, av ligand i test- prøven.
