Abstract
En fremgangsmåte for bestemmelse av identiteten av en nukleotidbase ved en spesifikk posisjon i en nukleinsyre av interesse. Identiteten av nukleo- tidet er basert på hvorvidt en primer kan utvides i nærvær av et spesifikt substratnukleotid eller ikke. Nukleo- tidbasen som skal bestemmes befinner seg ved 3'-enden umiddelbart nærligg- ende primerens 3'-ende-nukleotid. Primeren vil bli utvidet dersom substratnukleotidet er komplementært til nukleotidbasen i nukleinsyren av interesse, og primeren vil ikke utvides dersom det ikke foreligger noen slik komplementaritet.
