Abstract

Denne oppfinnelsen vedrører den fullstendige cDNA-sekvens av Alternaria alternata-allergenene Alt a 45 og Alt a 12. Det er blitt mulig innenfor rammen av den molekylærbiologiske analyse av allergenene å identifisere Alt a 45 som en proteindisulfidisomerase. Dette protein er et 45 kD stort protein som 47% av pasientene reagerer på, og er således et viktig primært allergen. Alt a 12 som 8% av pasientene reagerer på ble identifisert som et ribosomalt protein som er av interesse fordi autoantistoffer mot ribosomale proteiner blir funnet i pasienter som lider av lupus. Det gjenstår å se hvorvidt det er en eventuell forbindelse mellom allergi mot muggsopp og autcimmuniske sykdommer. Ved anvendelse av denne rekombinante sekvens og datamaskinanalyse er det blitt mulig å identifisere svært potente B- og T- celieepitoper og å anvende peptidene avledet fra de rekombinante proteiner for diagnosen av allergi mot alternaria alternata muggsopper. Peptidene har også vist seg egnet både for in vitro og in vivo allergenspesifik stimulering av T-celler proliterasjon, interleukinfremstiiling, og for blokkering av T-cellcne som resulterer i en toleranse av de allergenspesifikke T-lymfocytter.
