Abstract

Kovalent modifisering av vevs- og celleoverflater som hindrer celleadhesjon. Fremgangsmåten utføres under akseptable in vivo-betingelser og anvender en biokompatibel polymer som har en reaktiv gruppe bundet til minst en ende som reagerer med grupper til stede eller på vevs- og celleoverflater under vandig og milde betingelser, og som dermed bindes kovalent til polymeren på vevs- eller celleoverflaten. Fortrinnsvis reagerer den reaktive gruppe bundet til polymeren med aminer og hydroksyler til stede på vevs- og celleoverflater. I en foretrukken utførelsesform frembringer fremgangsmåten ifølge oppfinnelsen kovalent modifisering av vevs- og celleoverflater ved hjelp av PEG- diisocyanatløsning. Fremgangsmåten er spesielt nyttig for å svekke blodplate- og leukocyttavsetning i blodkar og dermed motvirke tromboser og restenoser, en alminnelig komplikasjon ved vaskulære prosedyrer inkludert PTCA og vaskulær kirurgi. Videre er fremgangsmåtene ifølge oppfinnelsen nyttige ved å redusere trombogenisitet av transplantater og redusere komplikasjonene ved vaskulær kirurgi ved å maskere vevesoverflateproteiner fra blodelementer. Fremgangsmåten reduserer immunogenisiteten av transplanterte vev og celler og reduserer dermed nødvendigheten av immunsuppressiv terapi.
