Abstract
Farmasøytisk sammensetning i en enhetsdoseringsform for peroral administrasjon til et menneske eller lavere dyr som har en gastrointestinaltrakt omfattende en tynntarm og en colon med en lumen derigjennom som har et innløp til colon fra tynntarmen, omfattende: a. en sikker og virksom mengde av et terapeutisk aktivt middel som er innlemmet i eller bestrøket på overflaten av en doseringsform valgt fra grup- pen omfattende et sfærisk substrat, et elliptisk substrat, en hard kapsel eller en komprimert tablett, med en største diameter fra 3-10 mm; og b. et enterisk polymer-belegnings- materiale; hvor doseringsformen har en glatt overflate som er fri for kanter eller skarpe kurver; det elliptiske sub- strat og den harde kapsel har et forhold mellom den lange og den korte diameter som ikke er større enn ca. 1,5; det terapeutiske middel frigis ved et punkt nær innløpet til, eller i, colon; det enteriske polymer-belegningsmateriale begynner å løses opp i vandig medium ved en pH-verdi mellom 5 og 6,3; og det enteriske polymer-belegningsmateriale har en beleggtykkelse på minst ca. 250 um.A pharmaceutical composition in a unit dosage form for oral administration to a human or lower animal having a gastrointestinal tract comprising a small intestine and a colon with a lumen therethrough having an inlet to the colon from the small intestine, comprising: a. A safe and effective amount of a therapeutically active agent incorporated into or coated on the surface of a dosage form selected from the group comprising a spherical substrate, an elliptical substrate, a hard capsule or a compressed tablet, having a maximum diameter of 3-10 mm; and b. an enteric polymeric coating material; wherein the dosage form has a smooth surface that is free of edges or sharp curves; the elliptical substrate and the hard capsule have a long to short diameter ratio of no greater than ca. 1.5; the therapeutic agent is released at a point near the inlet of, or in, the colon; the enteric polymer coating material begins to dissolve in aqueous medium at a pH between 5 and 6.3; and the enteric polymer coating material has a coating thickness of at least approx. 250 um.
