DESCRIPTION DE L’INVENTIONDESCRIPTION OF THE INVENTION
The present invention describes the microbial biosynthesis of bioplastics (PHA) or the process of producing polyhydroxyalkanoates (PHA) by PHA-producing microorganisms (e.g. Cupriavidus necator) using apple juice from cashew nuts as a renewable carbon substrate. . Said invention identified and used the nutrient enriched carbon substrate for high purity PHA production.
The proliferation of plastics from petrochemicals is a major concern due to negative environmental and social impacts. The inappropriate disposal of plastics and the rapid depletion of natural resources in production are the main factors leading to investigations to find sustainable alternatives to petroleum-based plastics (Muhammadi et al., 2015).
Polyhydroxyalkanoates (PHA) are considered to be compatible alternatives to replace conventional petroleum-based plastics because of their biodegradability, biocompatibility and non-toxic nature. Additional features like recyclability and thermoplastic nature give them many applications in the packaging industry, medicine and food industry (Ivanov et al., 2015). PHAs belong to the family of bio-polyesters which are produced and accumulated by a variety of microorganisms as a source of carbon and energy in the presence of excess carbon and limiting nitrogen or phosphorus.
PHBs are the most widely studied form of PHA and can accumulate up to 80% of the dry weight of the cell. Besides PHB, copolymers containing 3-hydroxyvalerate (3HV) and 3-hydroxyhexanoate (3-HHx) were also identified in 1974. Today, around 150 different monomeric constituents of PHA and over 300 microbial species, mainly microbial species. bacteria have been identified as being able to accumulate PHA (Anjum et al., 2016).
Sur laThe chemical structure of PHAs is shown in Figure 1 which contains repeating units of 100,000 to 30,000. In the case of PHB, R = CH3 and x = 1, while in the case of PHV, R = C2H5 and x = 1 . On the
Based on the number of carbon atoms, PHA can have a short chain length (3 to 5 carbon atoms) or a medium chain length (6 to 14 carbon atoms). The difference is due to the specificity of the PHA synthase substrate, in the case of Alcaligenes eutropus, the PHA synthase can polymerize 3-hydroxyalkanes (3-HA) consisting of 3 to 5 carbon atoms, while the PHA synthase of Pseudomonas oleovorans can polymerize 3-HA of 6 to 14 carbon atoms (Pena et al., 2014).
L'étape finale catalysée par l'enzyme PHA synthase, qui catalyse la polymérisation du 3-hydroxybutryle -CoA pour former Poly-3-hydroxybutyrate (PHB) (Fig.2)The biochemical pathway of PHA production has been well studied and consists mainly of three-step reactions in the case of the notorious bacteria Cupriavidus necator (formerly Alcaligenes eutropus, Ralstonia eutropha) (Kessler et al., 2001). In the first phase, two molecules of acetoacetyl-CoA undergo condensation to form acetoacetyl-CoA using the enzyme 3kéthothiolase. The second step catalyzed by the NADPH-dependent enzyme acetoacetyl-CoA reductase, which acts on acetoacetyl-CoA and reduces it to form 3-hydroxybutryl-CoA. The final step catalyzed by the enzyme PHA synthase, which catalyzes the polymerization of 3-hydroxybutryl -CoA to form Poly-3-hydroxybutyrate (PHB) (Fig. 2)
European Patent No. 0,046,344 describes the production of PHB using glucose as a carbon source for A.eutrophus NCIB 11599. The most commonly used substrates for the production of bioplastic are pure carbon sources such as glucose, fructose , lactose and vegetable oils. The cost of these pure carbon sources is one of the most important factors that make the production of PHA uneconomical and accounts for around 50-60% of the total expenditure for the production of PHA (Volodina et al., 2016). Therefore, an alternative approach is to use renewable, economically feasible or low cost and readily available carbon substrates for microbial growth and efficient production of PHA.
Bien que les pommes de cajouAgricultural residues and wastes such as cashew apple are low cost substrates and are suitable for microbial cultivation, as a source of carbon, nitrogen and minerals. Cashew apples are considered agricultural waste and its nutritious juice may be suitable as an inexpensive substrate due to the presence of 10-12% glucose content, as well as fructose, sucrose, vitamins and amino acids. Most of the time, cashew apples rot on the field after the nut is removed. Although the cashew apples
Page 3 of 24 can be used for the production of juices, ice cream and other food products, the use of cashew apples as fruit is very limited due to the presence of astringent compounds such as polyphenols and tannins (0, 35%) (Fontes et al., 2013). In addition, there is a presence of a waxy layer around the cashew apples which makes them inconvenient for human consumption. Therefore, the underutilization of cashew apples, the presence of a high concentration of reducing sugars, their high availability and low cost make them a potential and economical substrate for bioplastic production.
West Africa, India, Vietnam, East Africa and Brazil are the main producers of cashews. World cashew production in 2016 was 738,861 metric tons, which leads to 7 million tons of cashew apples because it is nine to ten times the weight of the nuts. Due to its abundant availability and low cost, cashew apple juice has been well studied as a substrate for the fermentative production of bio-surfactants, bioethanol, organic acids and enzymes (Prommajak et al., 2014, Betiku et al., 2016). However, no invention or literature has been reported regarding its use as a carbon substrate for the production of bioplastics.
The aim of the invention is to produce bioplastic from a renewable substrate at low cost, rich in sugar and suitable for microbial culture.
According to the invention, this goal is achieved with the process of producing polyhydroxyalkanoates (PHA) or bioplastics with high purity and high concentration of biomass, from apple cashew juice. The production of high purity PHA avoids the use of expensive chemicals required for further extraction and purification of PHA.
This process involves determining an appropriate concentration of reducing sugar for microbial growth. To this end, he describes a batch batch feeding strategy using cashew apple juice as a carbon substrate and a variation of minerals (nitrogen and phosphorus) to improve the biomass concentration and the PHA yield. PHA-producing microorganisms (Cupriavidus necator) are inoculated into the culture media of cashew apple juice to produce a high concentration of PHA. Subsequently, to make the process of producing PHA (bioplastics) more economical, the extracellular and intracellular proteins released by the bacteria are recovered from the supernatant to be used as animal proteins.

d) Traitement thermique du bouillon fermenté après la production d'PHA et récupération de 77% des protéines à partir du surnageant après extraction par PHA.d) Heat treatment of the fermented broth after the production of PHA and recovery of 77% of the proteins from the supernatant after extraction with PHA.

This is the place to clarify that cashew apple juice is underused. Dans la présente invention, bien qu'il y ait présence de potassiumThe physicochemical characteristics of the concentrated cashew apple juice (the cashew apple juice was concentrated by evaporation) are shown in Table 1. The presence of monosaccharide sugars, ie glucose and fructose in the Cashew apple juice, is advantageous because it allows easy and rapid assimilation by the bacterial cells producing PHA, thus improving the yield of PHA. In the present invention, although there is presence of potassium
Therefore, based on the physicochemical characterization of cashew apple juice, there was a high concentration of 5 glucose and fructose, while minerals and nitrogen can be supplemented to enhance the growth of the microorganism and of PHA production (Table 1).

Page 7 of 24 cashew apple as a renewable carbon substrate. The microorganism used in the present invention is one of the best PHA accumulating bacteria, Cupriavidus necator. However, the method can be applied to other PHA producing microorganisms. In order for the PHA production process to be successful, two pre-inocula are required, but the number of pre-inocula may vary depending on the experimental scale. The first pre-inoculum must be prepared by propagation of PHA-producing bacteria (e.g. C. necator) in mineral medium containing (per liter of distilled water) 20 g of glucose, 6.0 g of disodium hydrogenphosphate dodecahydrate ( Na2HPO4.12H2O) 2.4g of dihydrogenphosphate phosphate (KH2PO4), 1.0g of ammonium chloride (NH4CI), 0.50g of magnesium sulfate heptahydrate (MgSO4,7H2O), iron (III) chloride ) hexahydrate (FeCl3.6H2O) and calcium chloride hexahydrate (CaCh, 6H2O). The first pre-inoculum is maintained at a specific temperature, agitation and for a specific period of time, according to the conditions optimized for the microorganism. The PHA-producing strain (C.necator) is maintained with stirring of 150 revolutions per minute (rev / min) at a temperature of 30 ° C. for a period of 24 hours, in order to obtain the actively growing cells. After this period, 10% of the pre-inoculum volume is transferred to a second pre-inoculum. The second pre-inoculum is prepared using the same carbon substrate, which is to be used in production media, i.e. cashew apple juice. The second pre-inoculum is usually prepared to fit the microbial cells into the new substrate and decrease the latency period in the final production media. Along with the carbon substrate, other minerals and nitrogen are also added in order to achieve good microbial growth and PHA production. Preferably, the second pre-inoculum is also maintained at 150 rpm at 30 ° C for 24 h. After 24 h, 10% by volume of the second pre-inoculum is added to the final PHA production media, which generally have the same composition as the second pre-inoculum.
The proper concentration of reducing sugars is necessary to achieve the maximum concentration of biomass or microbial cell growth and PHA production. The low concentration of carbon substrate can limit microbial growth, while a high concentration of reducing sugars or carbon can cause inhibition of the substrate. In the present invention, the concentration of reducing sugars varied between 5 and 35 g.L-1 and an appropriate concentration of reducing sugars was used for a subsequent process.
Page 8 sur 24Page 8 of 24
The process for producing PHA in the present invention can be carried out by batch or semi-continuous processing. The semi-batch feed fermentation process ensures the regulation of the appropriate nutrient feed in the fermenter to avoid both limitation and inhibition of the substrate. In the present invention, different metered feeding strategies such as one pulse, two pulse and step feed of cashew apple juice have been applied in order to achieve high biomass concentration and PHA production. Feeding during the process was based on the consumption of reducing sugars or carbon substrate. The temperature of the fermenter was maintained at 30 ° C by circulating water through the jacket of the fermenter, while the pH 6.8 was maintained using 4N sulfuric acid and 4N NaOH. Dissolved oxygen should normally be maintained at around 30% throughout the fermentation process.
L'excès de source d'azote peut modifier la voie métabolique de la cellule microbienne vers la production de biomasse au lieu de la production de PHA.The present invention also relates to the use of different concentrations of minerals and nitrogen in the feed solution during the feed batch fermentation process, to verify the effect on microbial growth and PHA production. The excess nitrogen source can alter the metabolic pathway of the microbial cell towards the production of biomass instead of the production of PHA.
The present invention also refers to the heat treatment of the microbial cells at 95-121 ° C for 30 minutes in order to kill the microbial cells so that they are not able to consume the PHA accumulated after the end of the fermentation. The heat treatment also helps to disrupt the cell structure by releasing the PHA to the extracellular medium.
The present invention also refers to the use of a supernatant for recovering proteins after extraction of PHA from a heat-treated fermented broth. The supernatant is both made up of extracellular and intracellular proteins released outside of microbial cells during heat treatment. Proteins can be extracted using heat treatment and further by addition of carboxy methylcellulose to accelerate the process of their precipitation.

PHA was measured using a standard extraction method for methanol and chloroform (Comeau et al., 1988) using benzoic acid as an internal standard.
Different feeding strategies have been used to maximize biomass concentration and PHA content. In both fermenters, the production media were supplemented with minerals. However, the feeding was carried out using only cashew apple juice without the addition of minerals.
The composition of the other mineral medium was identical to that of the first pre-inoculum medium. The second pre-inoculum was also maintained at 30 ° C. for 24 hours and then, the total pre-inoculum was transferred into 2.7 L of production medium in the 5 L fermenter. The composition of the production media comprises 20 g / L concentration of reducing sugars with minerals (g / L): 6.00 Na2HPO4,12H2O, 2.4 15 KH2PO4, 1.00 NH4CI, 0.50 MgSO4.7H 2 O, 0.01 FeCl3.6H 2 0, 0.01 CaCl2.6H 2 O. The pH of the production medium was maintained at 6.8. The same procedure was followed for the two fermenters. The temperature was maintained at 30 ° C by circulating water through the jacket. The fermentation pH was automatically controlled to 6.8 ± 0.1 by computer-controlled peristaltic pumps using 4N sulfuric acid and 4N NaOH. OD and pH were continuously monitored using a dissolved oxygen polarographic probe and a pH sensor (MettlerToledo, USA), respectively.
In the first fermenter with a pulse feeding strategy, the feed, i.e. reducing sugars were added only once during the whole fermentation, while in the second fermenter with a feeding strategy in two pulses, the feed, i.e. the reducing sugars were added twice during the fermentation. In both fermenters, approximately the same PHA content as well as the biomass concentration was achieved. The PHA content was very high, namely 90 ± 2%, while a low biomass concentration of 15 ± 0.3 g / L was obtained. The low concentration of biomass could result from the lack of nitrogen and mineral content in the feed solution as only the carbon source was provided in the feed.
The discontinuous feeding strategy is essential to control the substrate concentration at its optimum level and to achieve maximum cell or biomass concentration as well as high biomass productivity. In this study, cashew apple juice was supplemented with minerals and nitrogen content in the feed solution. However, the concentration of reducing sugars, minerals and nitrogen was varied for two fermenters.
Le pH moyen a été maintenu à 6,8 et le pré-inoculum a été maintenu à l'agitation de 150 tr/min pendant 24h à 30 °C.The study begins with the preparation of the first and second pre-inoculum. The first pre-inoculum was prepared by inoculating C.necator cells from the mineral media agar plates into the mineral medium broth containing (per liter of distilled water) 20 g of glucose, 6.0 g of hydrogen phosphate. of disodium dodecahydrate (Na2HPO4,12H2O), 2.4g of dihydrogen potassium phosphate (KH2PO4), 1.0 g of ammonium chloride (NH4CI), 0.50 g of magnesium sulfate heptahydrate (MgSO4, 7H2O) , iron (III) chloride hexahydrate (FeCh, 6H2O) and calcium chloride hexahydrate (CaCb, 6H2O). The solution of (FeCl3,6H2O) and (CaCh, 6H2O) was sterilized by filtration through a 0.2 µm polyethersulfone (PES) filter. Glucose, NH4Cl and (MgSO4, 7H2O) were autoclaved separately at 121 ° C for 15 min. And these solutions were mixed aseptically after cooling. Le DO et le pH ont été surveillés en permanence au moyen d'une sonde polarographique à oxygène dissous et d'un capteur de pH (Mettler10 Toledo, USA), respectivementPage 13 of 24 was also maintained at 30 ° C for 24 hours and then the total pre-inoculum was transferred to 2.7 L of production medium in a 5 L fermenter. The composition of the production media comprises 10 g / L concentration of reducing sugars with minerals (g / L): 6.00 (Na2HPO4, 12H2O), 2.4 KH2PO4.1.00 NH4CI, 0.50 (MgSO 4 , 7H 2 O), 0.01 (FeCb , 6H2O), 0.01 (CaCl 2 , 6H2O). The pH of the production medium was maintained at 6.8. The same procedure was followed for the two fermenters. The temperature was maintained at 30 ° C by circulating water through the jacket. The fermentation pH was automatically controlled at 6.8 ± 0.1 by computer-controlled peristaltic pumps using 4N sulfuric acid and 4N NaOH. OD and pH were continuously monitored using a dissolved oxygen polarographic probe and a pH sensor (Mettler10 Toledo, USA), respectively
In Fermenter A, the feed was carried out using a high concentration of minerals and nitrogen was used, while in the case of Fermentor B, a low concentration of minerals and nitrogen was used (Table 3 )

The samples from the two fermenters were taken after 12 hours to measure the biomass concentration (g / L), the concentration of reducing sugars (g / L) and the PHA content (% m / m). The concentration of reducing sugar during fermentation was analyzed using the DNS Method (3,5-dinitrosalicylic acid) (Miller, 1959). The biomass concentration was measured according to the standard method (APHA, 1998). PHA was measured using a standard methanol and chloroform extraction method (Comeau et al., 1988) using benzoic acid as an internal standard.
After the end of the fermentation, a maximum and very high biomass concentration of 70.11 g / L was obtained in Fermentor A with a high concentration of minerals and nitrogen, followed by 42.39 g / L of. concentration of biomass in fermenter B with a low concentration of minerals and nitrogen. However, the PHA content in the case of fermenter A, increased to 80% and then started to decrease and reached 66% at the end of the fermentation, while the PHA content in the case of fermenter B a increased until the end of fermentation and reached 89%. The decrease in the PHA content after a specific period of time is due to the accumulation of excess nitrogen which alters the metabolic pathway of the microorganism towards biomass growth instead of the accumulation of PHA.
A maximum PHA concentration of 46 g / L was obtained when feeding with a high concentration of minerals and nitrogen, while a concentration of 37.5 g / L of PHA was obtained when feeding with a low concentration of minerals and nitrogen. A maximum and very high PHA yield of 0.88 g of PHA / g of substrate consumed was obtained in the case of the fermenter, fed with a low concentration of minerals and nitrogen, while the yield of PHA of 0 54 g of PHA / g of substrate consumed was obtained in the case of a fermenter with a high concentration of minerals and nitrogen. The obtained PHA yield of 0.88 g of PHA / g of substrate consumed is the highest observed using apple juice from chopped nuts as carbon substrate compared to other reported carbon substrates such as residual glcerol, oil frond juice and sucrose (Mozumder et al., 2014; Cavalheiro et al. 2009; Ahmad et al., 2013).

Destruction of cell lysis that occurs during thermal deactivation and lysis of ΓΑΡΤΗ-containing cells also releases intracellular proteins into solution. The protein concentration, surprisingly, in the lysate soup was very high (32-36 g / L). Therefore, the supernatant collected after heat treatment during centrifugation was used for protein extraction. The pH of the mixture was lowered to 3.5 using sulfuric acid and the supernatant was heated for 20 minutes at 90 degrees celsius with constant stirring at 250 rpm. The 30% protein recovery was obtained after heat treatment. Subsequently, the broth was cooled to 50 degrees Celsius and the pH was adjusted to 5.5 using NaOH. Carboxy methylcellulose (CMC polymer) was added to speed up the process of protein precipitation.
L'ensemble du procédé a permis de récupérer 77% des protéines du surnageant2.4% (w / v) CMC was added to the broth and mixed well for 20 minutes. After 20 minutes of reaction, the supernatant was left for 8 h to allow the obtaining of protein precipitates by gravity. The entire process made it possible to recover 77% of the proteins from the supernatant
