Description
The present invention relates to the production of bioethanol from lignocellulosic biomass. The biomass contains cellulose, hemicellulose and lignin; cellulose is a source of raw materials for the production of bioethanol from high added value by-products. This biomass is a source of environmental pollution with harmful consequences on human health and the climate, the present invention relates to a process for pretreatment and hydrolysis of biomass for the production of bioethanol.
Bioethanol production is carried out in four stages: pretreatment; 1 hydrolysis; fermentation and ethanol recovery. Pretreatment aims to separate and make accessible the closely linked and crystallized constituents of the BLC: lignin, hemicellulose and lignin. By thermal, chemical and/or biological action, the structure of the lignin is destroyed or separated, the hemicellulose is more or less hydrolyzed, and the structure of the cellulose is modified.
Due to the structure of cellulose, its crystallinity and its association with lignin and hemicelluloses still present, even after the pretreatment step, its hydrolysis into fermentable monomers (glucose) is a difficult operation which can be carried out by enzymatic hydrolysis.
The resulting fermentable sugar solution is subjected to the action of microorganisms for fermentation. Bacteria, yeasts and fungi are capable of producing ethanol through the fermentation of sugars.
Ethanol recovery is done in two stages, the first stage of ethanol recovery is distillation. This separation operation based on the volatility of the alcohol allows to go from a 10°GL solution to a 94°GL (Gay Lusac degree) ethanol solution. However, ethanol is not the only volatile compound in the fermented must. It must be found in the gas phase with other gaseous compounds such as fatty acids, alcohols, ethers and water. The gas phase will be condensed and will require a second distillation. The second stage is rectification: it allows the separation of ethanol from other volatile compounds. These products will be classified into two categories according to their capacity to be distilled before and after ethanol: head products and tail products. It will therefore consist of a fractional distillation of the products contained in the condensate.
Lignocellulosic biomass, in particular sawdust, contains cellulose; in particular sawdust contains cellulose, hemicellulose and lignin. Cellulose is a glucose polymer which has great potential for the production of bioethanol. The use of sawdust for the production of bioethanol is limited due to its recalcitrant structure (cellulose trapped by lignin, preventing access by enzymes) and its high lignin content.
According to patents No. TNP/2018/000248, No. PCT/EP2017/081056, No. PCT/EP2009/050690 and PCT/EP2009/000920, the processes for obtaining bioethanol from certain lignocellulosic biomasses are known, in particular those which are less recalcitrant and which have a low lignin content (10% 0 20%). The most frequently used bioethanol production process takes place according to the following steps: pretreatment, which consists of separating the lignin from the cellulose and hemicellulose so that the cellulose and hemicellulose are accessible to enzymes; chemical or enzymatic hydrolysis which allows the long chain of glucose and xylose contained in the cellulose and hemicellulose to be cut; fermentation which allows alcohol to be obtained using yeasts, and distillation which allows bioethanol to be obtained.
Patent No. TNP/2018/000248 uses ordinary chemical pretreatment by combining an alkaline reagent (soda, 4% to 12% by weight of dry matter) and an acidic reagent (sulfuric acid, 30% to 60% of the total volume), followed by enzymatic hydrolysis and fermentation. Patent No. PCT/EP2017/081056 uses chemical pretreatment with formic acid for the production of bioethanol. Patent PCT/EP2009/000920 uses a diphospho sugar epimerase (NDPSE) enzyme in the production of bioethanol from lignocellulosic biomass.
When it comes to ethanol production from sawdust and other biomasses with high lignin content, ordinary pretreatment and combination of pretreatments in parallel is not sufficient. A combination of both pretreatment methods in series will allow delignification of biomasses with high lignin content (20% to 30%). This objective is essentially achieved according to the invention by a process for the production of bioethanol from lignocellulosic biomass. Such a process comprises key steps with reagents and solvents.
La quantité de réactif qu’on peut utiliser varie de 4% !The reagent used according to the invention can be sodium hydroxide, sulfuric acid, etc., and the solvent used according to the invention can be ethanol, methanol, propanol, acetone, hexane and other organic solvents. The amount of reagent that can be used varies from 4%!
Les quantités d’enzymes utilisées varient selon l’activité enzymatique et les levures utilisées entre 0,1% à 1% en poids du volume de fermentation.at 12% by weight, favorably from 5% to 10% of the quantity of dry matter. The quantity of solvent used according to the invention ranges from 30% to 60% by volume, favorably 40% to 50% by volume of the quantity of total volume of solvent used for the pretreatment. The quantities of enzymes used vary according to the enzymatic activity and the yeasts used between 0.1% to 1% by weight of the fermentation volume.
The pretreatment with an alkaline reagent takes place at a temperature of 160°C to 190°C, advantageously between 170°C and 190°C. In the case where the biomass has many particles larger than 2 millimeters, mechanical grinding is carried out in order to obtain particles smaller than 2 millimeters. The pretreatment with solvent takes place at a temperature varying between 170°C and 215°C, advantageously at 180°C and 200°C.
Due to the structure of cellulose, its crystallinity and its association with lignin and hemicelluloses still present, even after the pretreatment step, its hydrolysis into fermentable monomers (glucose) is a difficult operation.
Les souches 80 généralement utilisées pour la fermentation des hexoses sont Saccharomyces cerevisiae et Zymomonas mobilisThe resulting solution of fermentable sugars is subjected to the action of microorganisms for fermentation. Bacteria, yeasts and fungi are capable of producing ethanol through the fermentation of sugars. The strains generally used for the fermentation of hexoses are Saccharomyces cerevisiae and Zymomonas mobilis.
After fermentation, ethanol is found in a must containing mainly water and biomass degradation products, substances added during the process but also other molecules produced by microorganisms simultaneously 85 with the production of bioethanol, such as higher alcohols. The must must be freed from these different substances to obtain bioethanol by distillation.
