R ^ CISO _-^/N°2 OH LbU2 WZÓR 6 « WZÓR 7 Prac. Poligraf. UP-PRL naklad 120+18 Cena 45 zl PL PL PL PL PL PLThe subject of the invention is a method for preparing phenyl esters of 2-carboalkoxyamino-benzimidazolyl-5(6)-sulfonic acid with antiredness activity. 2-Carboalkoxy-amino-benzimidazolyl derivatives with alkyl, acyl, phenoxy and phenylthio radicals in the 5(6) position are known as antiredness agents (P. Actor et al., Nature 215, 321 (1967); German patent specifications DOS No. 2029637, DOS No. 2164690, DOS No. 236r348). The invention relates to a method for preparing anti-red-active phenyl esters of 2-carboalkoxyamino-benzimidazolyl-5(6)-sulfonic acid of formula 1, wherein R1 is an alkyl group with 1-4 carbon atoms, R2 and R3 each independently of one another are hydrogen, a hydroxyl group, an alkoxy group with 1-4 carbon atoms, a halogen, trifluoromethyl, an alkyl group with 1-4 carbon atoms, a carboalkoxy group with 1-4 carbon atoms in the alkoxy radical or CN. Alkyl radicals in the substituents R1, R2 and R3 include: methyl, ethyl, propyl, isopropyl, butyl, s.c.-butyl, t.c.-butyl. Alkoxy groups in the substituents R2 and R3 include: methoxy, ethoxy, propoxy, isopropoxy and butoxy. Halogen atoms in the substituents R2 and R3 include: fluorine, chlorine, bromine and iodine. Carbalkoxy groups in the substituents R2 and R3 include: carbomethoxy, carboethoxy, carbopropoxy or carbobutoxy. Particularly preferred are compounds of formula I in which R1 is methyl, R2 is hydrogen, and R3 is hydrogen, chlorine, bromine, trifluoromethyl, methyl, ethyl, methoxy or ethoxy, R3 being particularly preferably located in the 3-position of the phenyl ring. The method for the preparation of phenyl esters of 2-carboalkoxy-amino-benzimidazole-5(6)-sulfonic acid of the formula 1, wherein R1, R2' and R3 have the meaning given above, according to the invention consists in reacting an o-phenylene-diamine derivative of the formula 2, wherein R2 and R3 have the same meaning as in formula 1, with an N-dichloromethylene-carbamic acid ester of the formula 3, wherein R1 has the meaning given above, at a temperature of -10° to +40°C in the presence of a base.2 100 496 In order to carry out the process, 1 mole of the o-phenylene-diamine derivative of the formula 2 in the presence of 2 moles of a base is reacted with 1 mole of an N-dichloromethylene-carbamic acid ester of the formula 3. Hydroxides, carbonates and hydroxydes are used as bases. and alkali metal or alkaline earth metal bicarbonates or tertiary organic bases. Examples include sodium hydroxide, sodium bicarbonate, sodium carbonate, potassium carbonate, potassium bicarbonate, triethylamine, pyridine and methyl-substituted pyridines. N-Dichloromethylenecarbamic acid esters of formula 3 can be prepared according to German patent specification DOS No. 1932297 from the known dichloromethylenecarbamic acid chloride in the presence of an inert organic solvent such as ether, dioxane, tetrahydrofuran, benzene, toluene, at a temperature of 0°-40°C. Examples of N-dichloromethylenecarbamic acid esters of formula III include methyl, ethyl, isopropyl, propyl, n-butyl and tert.-butyl N-dichloromethylenecarbamic acid esters. The reaction temperature can be varied within a larger range. Generally, a range of -10° to 60°C is used, preferably 0-30°C. The o-phenylenediamine derivative of formula II used as a starting material is obtained by reduction of the corresponding aminonitro derivative of formula IV, wherein R2 and R3 have the same meanings as in formula I. The reduction can, for example, be carried out by hydrogenation in the presence of Raney nickel and a solvent such as methanol or dimethylformamide at a temperature of 20°C to 60°C, or by treatment with reducing agents such as sodium dithionite. The aminonitro derivatives of formula IV are obtained by reacting the corresponding chloronitro compounds of formula V, wherein R1 and R3 have the same meanings as in formula I, with ammonia in a suitable solvent such as dioxane or methanol at elevated temperature and pressure. The chloro-nitro compounds of formula 5 are in turn prepared by reacting 3-nitro-4-chloro-benzenesulfonic acid chloride of formula 7 with a phenol of formula 6, wherein R2 and R3 have the same meaning as in formula 1, in an inert solvent in the presence of a base such as triethylamine. The phenyl esters of 2-carboalkoxyamino-benzimidazolyl-5(6)-sulfonic acid according to the invention are valuable chemotherapeutic agents and are suitable for combating parasitic diseases in humans and animals, such as intestinal worms and liver flukes. They are particularly effective against a large number of intestinal worms, e.g., Haemonchus, Trichlostrongylus, Ostertagia, Strongyloides, Cooperia, Chabertia, Oesophagostomum, Hyostrongylus, Ankylostoma, Askaris, and Heterakis. Particularly outstanding is their activity against gastrointestinal strongyloides, which primarily attack ruminant animals. Infestation of animals by these parasites causes significant economic damage, therefore the compounds according to the invention are used in particular in veterinary medicines. The active substances of formula I are administered, depending on the situation, in doses of 0.5-50 mg per kg of body weight for 1-14 days. For oral administration, tablets, dragees, capsules, powders, granules or pastes are used, which contain the active substances together with known excipients and carriers such as starch, powder, cellulose, talc, magnesium stearate, sugar, gelatin, calcium carbonate, finely divided silicic acid, carboxymethylcellulose or similar substances. For parenteral administration, solutions are used, e.g., oil solutions prepared using sesame oil, castor oil, or synthetic triglycerides, optionally with the addition of tocopherol as an antioxidant and/or with the use of active substances at the interface, such as sorbitan fatty acid esters. Furthermore, aqueous suspensions are used, which are prepared using ethoxylated sorbitan fatty acid esters, optionally with the addition of thickening agents, such as polyethylene glycol or carboxymethylcellulose. The concentration of the active substances according to the invention in the preparations is preferably 2-20% by weight for use in animal medicine; in medicines for humans, the concentration of the active substances is preferably -80% by weight. To determine the effects of the compounds of the invention, chemotherapeutic studies were conducted on sheep lambs weighing approximately 30 kg that were experimentally infected with Haemonchus contortus or Trichostrongylus colubriformis larvae. The experimental animals were housed in tiled stalls that were thoroughly cleaned daily. After the pre-treatment period (the time between infection and sexual maturity of the parasites with the beginning of the secretion of eggs or larvae) the number per gram of feces was determined using the modified McMaster method according to Wetzel (Tierarztliche Umschau, 6, 209-210, 1951). Immediately after the treatment of sheep, usually 4-8 animals per active substance, at least 2 at a time, was carried out. The animals were given doses of the products of the method in the form of a suspension in 10 ml of 1% Tylose suspension each time. Each time, 7, 14 and 28 days after the treatment, the number of eggs per gram of feces was determined again according to the above-mentioned methods and their percentage decrease was calculated compared to the initial value before the treatment. The products obtained by the method according to the invention are perfectly effective not only when administered orally, but also act parenterally in doses down to 2 mg/kg. They thus significantly outperform comparable benzimidazole derivatives, in particular all known 5(6)-substituted 2-benzimidazole carbamates. Example 1. 26.4 g of 3,4-diaminobenzenesulfonic acid phenyl ester, 20.2 g of triethylamine and 300 ml of chloroform are mixed and a solution of 15.6 g of N-dichloromethylenecarbamic acid methyl ester in 50 ml of chloroform is slowly added with stirring at a maximum temperature of 20°C. Stirring is continued for another hour, the precipitate is filtered off and washed with additional chloroform. For purification, the crude product is dissolved in dioxane, filtered with charcoal and treated with water. After After filtering, washing and drying, 5 g of 2-carbomethoxy-amino-5-benzimidazole-sulfonic acid phenyl ester is obtained with a decomposition point of 242°C. 3,4-Diamino-benzenesulfonic acid phenyl ester is prepared by hydrogenating 27 g of 3-nitro-4-amino-benzenesulfonic acid phenyl ester in 300 ml of methyl glycol with Raney nickel at standard pressure and room temperature. The catalyst is filtered off and after concentration, 3,4-Diamino-benzenesulfonic acid phenyl ester is obtained as a crude product in the form of 25 g of dark oil, which can be used directly for ring closure. To prepare 3,4-Diamino-benzenesulfonic acid phenyl ester 3-Nitro-4-amino-benzenesulfonic acid. 54 g of 3-nitro-4-chloro-benzenesulfonic acid phenyl ester in 500 ml of dioxane was kept under 5 atm of gaseous ammonia pressure for 5 hours at 50°C and then the solvent was removed. The residue was treated with 200 ml of a mixture of equal parts of methanol and water, after which a solid precipitate formed after a short time and was filtered off. After repeated recrystallization from methanol and then benzene, 28 g of 3-nitro-4-amino-benzenesulfonic acid phenyl ester was obtained, melting at 104°C. 3-nitro-4-chloro-benzenesulfonic acid phenyl ester was prepared by mixing 51 g of 3-nitro-4-chloro-benzenesulfonic acid chloride with 18.8 g of phenol in 120 ml of acetone and, while cooling at an internal temperature not exceeding 10°C, add 28 ml of triethylamine dropwise. Stirring is continued for a few hours at room temperature and then water is added, whereupon an oil separates and is worked up over ether. After recrystallization from methanol, 54 g of 3-nitro-4-chloro-benzenesulfonic acid phenyl ester is obtained, melting point 71°C. The following is prepared analogously, using appropriately modified starting materials: Example II.
