Description

Przedmiotem wynalazku jest sposób wytwarzania nowych amidów kwasu cynamonowego o ogólnym wzo¬ rze 1, w którym X oznacza atom chloru, bromu lub jodu a R oznacza atom wodoru lub rodnik alkilowy zawiera¬ jacy 1-3 atomy wegla. Zwiazki te maja cenne wlasciwosci farmakologiczne i nadaja sie do stosowania w lecznic¬ twie.Stwierdzono, ze amidy kwasu cynamonowego o ogólnym wzorze 1 wykazuja dzialanie przeciwdrgawkowe u ssaków, na co wskazuja wyniki badan farmakologicznych na myszach po podaniu im tych zwiazków. Badaniami tymi sa: test maksymalnego elektroszoku (MES) na myszach, metoda opisana przez Woodbury'egoi Davenporta w Arch. Int. Pharmacodyn. Ther 92, 97-107 (1952) oraz test napadu padaczkowego wywolanego metrazolem (MET) na myszach, metoda opisana przez Swinyarda, Browna iGoodmana w J.Pharmacol.Exp. Therap. 106, 319-330(1952).Wszystkie zwiazki o zdefiniowanym wyzej wzorze 1 sa zwiazkami nowymi z wyjatkiem zwiazku, w którym X oznacza atom chloru a R oznacza atom wodoru. Zwiazek ten znany jest w literaturze (Chem. Surowy produkt rekrystalizuje sie z roztworu etanolu i wody otrzymujac amid kwasu 3-bromocynamonowego o temperaturze topnienia 110°C.Przyklad XV. PL PL PL PL PL PL PL PL PLThe present invention relates to a method for preparing new cinnamic acid amides of the general formula I, wherein X is a chlorine, bromine, or iodine atom and R is a hydrogen atom or an alkyl radical containing 1-3 carbon atoms. These compounds have valuable pharmacological properties and are suitable for use in therapy. Cinnamic acid amides of the general formula I have been found to exhibit anticonvulsant activity in mammals, as indicated by the results of pharmacological studies on mice following administration of these compounds. These studies include the maximal electroshock (MES) test in mice, the method described by Woodbury and Davenport in Arch. Int. Pharmacodyn. Ther 92, 97-107 (1952) and the metrazol (MET)-induced seizure test in mice, the method described by Swinyard, Brown and Goodman in J. Pharmacol. Exp. Therap. 106, 319-330 (1952). All compounds of formula 1 as defined above are new compounds except the compound in which X is a chlorine atom and R is a hydrogen atom. This compound is known in the literature (Chem. Abs., 1970, 72, 21696) as an intermediate in the synthesis of its derivatives, but no data on its pharmacological activity have been provided. Compounds of general formula 1 having a trans configuration are preferred, wherein the compound in which R is an ethyl group and X is a bromine atom shows outstanding anticonvulsant activity in both of the above-mentioned pharmacological tests. Examples of amides of the general formula 1 are the following compounds: N-methylamide of 3-bromocinnamic acid, N-n-propylamide of 3-bromocinnamic acid, N-isopropylamide of 3-bromocinnamic acid, N-methylamide of 3-chlorocinnamic acid, N-ethylamide of 3-chlorocinnamic acid and N-n-propylamide of 3-chlorocinnamic acid. The new cinnamic acid amides of the formula 1 are prepared according to the invention by acylation of a compound of the formula R-NH-W, wherein R is a hydrogen atom or an alkyl radical containing 1-3 carbon atoms and W is a hydrogen atom or a leaving group, with a suitable acid of the general formula 2, wherein X has the meaning given above, or a reactive derivative thereof, e.g. an ester or thioester. alkyl, in which the alkyl radical contains, for example, 1 to 4 carbon atoms, an amide, an acid halide, e.g., an acid chloride, or an acid anhydride. Depending on the type of acylating agent, different reaction conditions are used, but usually the reactants are heated together at reflux temperature, preferably in an inert liquid medium, e.g., ether, benzene, or cyclohexane. The most convenient method of synthesis is the action of an acid chloride on the appropriate amine. Preferably, one molar equivalent of acid halide is used for two or more molar equivalents of amine, wherein the molar excess of amine can be replaced by another base, e.g., triethylamine, pyridine, dimethylaniline, potassium or sodium carbonate. The reactions are carried out in various polar or non-polar solvents, e.g. water, alkanols e.g. methanol, ethanol, etc., ether, dioxane, benzene, toluene, xylene, petroleum ether, cyclohexane, tetrahydrofuran, chloroform and carbon tetrachloride. The reactions can be carried out in a wide temperature range, e.g. from -10°C to the boiling point of the reaction mixture. In the case of the reaction of an amide of the formula R-NH-W with an acid anhydride, it is advantageous to add a catalytic amount of sulfuric acid. The leaving group W in the formula R-NH-W is, for example, the group -COH (formide), -CORi, where R a is an alkyl radical with 1 to 4 carbon atoms (amide), -CONH2 (urea) or -COORi, where Rt is an alkyl radical with 1 -4 carbon atoms (urethane). Acids of the general formula 2 are obtained by classical methods used in organic synthesis, such as the Perkin synthesis, the Reformatsky reaction or the Kooevenager condensation. Compounds of the general formula 1 can be used to treat or prevent convulsions in mammals, e.g. mice, dogs and cats, as well as in humans. In particular, they can be used to treat major and minor seizures, psychomotor epilepsy, and focal seizures, at doses ranging from 2 to 200 mg/kg of body weight per day. The optimal dose depends on the compound used, the patient's condition, and the route of administration. However, the recommended dose is 20 to 60 mg/kg, most preferably 30 to 50 mg/kg of body weight per day. It is preferable to administer the daily dose in three separate doses. A convenient form of administration is tablets containing from 100 to 500 mg of the active compound of formula I. When used in therapy, the compounds of formula I can be administered in chemically pure form, but it is more advantageous to administer them together with acceptable carriers as suitable pharmaceutical preparations. The carrier must be "acceptable" in the sense that it is tolerated by the other ingredients of the preparation and does not affect the recipient of the preparation. The carrier may be a solid or a liquid or a mixture of solid and liquid substances. It is advantageous to prepare the preparation in unit forms, e.g. tablets, capsules or sachets for oral administration or suppositories for rectal administration. Pharmaceutical preparations may also contain other pharmaceutically active substances. These preparations may be prepared by techniques generally known in pharmacy, which essentially consist of mixing the ingredients. Unit forms of preparation for oral, rectal or parenteral administration contain the active compound of formula 1 preferably in an amount of from 100 mg to 500 mg. Preparations for oral administration are prepared in the form of powders or granules containing the active substances and diluents, dispersing agents and surface-active agents. They can be used to produce medicines in the form of solutions in water or syrups; capsules or sachets in which the substances are in a dry state or in the form of an aqueous or non-aqueous suspension; tablets preferably prepared from granules of the active substance with a diluent by crushing with the addition of binders and lubricants, or in the form of a suspension in water, syrup, oil or water-oil emulsion, with flavorings, preservatives, dispersants, thickeners and emulsifiers added. The granules or tablets may be coated and marked.* For parenteral administration (by intramuscular or intraperitoneal injection), the active compounds are prepared in unit-dose or multi-dose containers in aqueous or non-aqueous injection solutions which may contain antioxidants, buffers, bacteriostatic agents and solutes rendering the active compound solution isotonic with blood; or in aqueous or non-aqueous suspensions which may also contain dispersing agents and thickening agents. Injectable solutions and suspensions can be prepared extemporaneously from sterile powders, granules, or tablets which may contain diluents, dispersing agents, surface-active agents, binders, and lubricants. In the above-mentioned MES test, trans-3-bromocinnamic acid N-ethylamide has an ED50 when administered orally to mice and rats of 80 mg/kg and 26 mg/kg, respectively. Anticonvulsant activity was determined in mice using the maximal electroshock (MES) test, performed according to the method described by Woodbury and Davenport, supra, using corneal electrodes and a Wahlquist Electroshock Stimulator Model E (manufactured by Wahlquist Instrument Co., Salt Lake City, Utah). Trans-3-bromocinnamic acid N-ethylamide was dispersed in a 0.5% aqueous methylcellulose solution by homogenization in a homogenizer mill, obtaining particles of more than 51% less than 5 microns and more than 87% less than 20 microns, preparing solutions of 5, 7.5, 10, 15, and 20 mg in 1 ml of methylcellulose solution. Male ICR Blue Spruce mice, weighing an average of 22 g, were divided into 6 groups of 10 mice each. The trans-3-bromocinnamic acid N-ethylamide suspension was administered to the mice orally one hour before testing by direct intragastric injection with a syringe. Control mice received the same volume of 0.57% aqueous methylcellulose solution at the same time. A 5 mA current was applied to the mouse cornea for 0.2 seconds, and the time from the electric shock to the hindlimb extensor muscle response (convulsive extension of the hindlimb) was measured, which is referred to as the hindlimb extension latency. Animals whose hindlimb muscles did not exhibit convulsions were considered protected (p). An increase in mean latency periods above the control is indicative of the anticonvulsant activity of the drug. The obtained results are presented in the table. Each group contained 10 mice. A different group of mice was used for each dose study. In the table, p indicates that the mice were protected from electric shock. The following examples, in which all temperatures are given in C, illustrate the process of the invention. Example 1. 3-Bromocinnamic acid N-isopropylamide 11.4 g of trans-bromocinnamic acid in 75 ml of dry benzene is heated to reflux, and then a mixture of 12 g of thionyl chloride and 50 ml of dry benzene is added in such portions as to maintain the reaction mixture at constant reflux. After complete addition of the thionyl chloride, the reaction mixture is heated for a further 2 hours, after which the solvent and excess thionyl chloride are evaporated under reduced pressure to give about 12.3 g of trans-3-bromocinnamic acid chloride. After dissolving this product in 150 ml of toluene, the resulting solution was added dropwise, with stirring, to a solution of 10 g of isopropylamine in 200 ml of ether. The reaction mixture was then stirred at room temperature for 1 hour and then heated under reflux for another hour. The solvent and excess amine were removed under reduced pressure. The crude residue was triturated with water, filtered, and recrystallized from a mixture of ethanol and water. Crystalline white trans-3-bromocinnamic acid N-isopropylamide was obtained, melting at 85-86°C. Elemental analysis, nuclear magnetic resonance spectra, and infrared spectra confirm the compound's structure. Example II. 3-Bromocinnamic Acid N-Ethylamide. A solution of 12.3 g of trans-3-bromocinnamic acid chloride in 150 ml of anhydrous toluene is slowly added, with stirring at room temperature, to a solution of 10 g of ethylamine in 100 ml of dry ether. The reaction mixture is then heated under reflux for one hour, after which the solvent and excess amine are removed under reduced pressure, and the residue is triturated with water, filtered, and recrystallized from ethanol/water. White crystalline trans-3-bromocinnamic acid N-ethylamide is obtained, melting at 89-90°C. Nuclear magnetic resonance spectra, infrared spectrum, and elemental analysis confirm the compound's structure. Example III: 3-Bromocinnamic acid N-ethylamide: 14.8 g of trans m-bromocinnamic acid, 173 ml of ethanol, and 0.4 ml of concentrated sulfuric acid were stirred and heated to reflux for 15 hours. Then approximately 150 ml of ethanol was distilled off, and the concentrated solution was poured into 140 ml of ice and water. The cold mixture was then made basic with 40% sodium hydroxide and extracted with methylene chloride (4 x 60 ml). The combined organic extracts were dried over anhydrous potassium carbonate, filtered, and the solvent was evaporated under reduced pressure. Trans-3-bromocinnamic acid ethyl ester was obtained as a partially solidifying oil. The infrared spectrum confirmed the compound's structure. 8.4 g of the ester, 6.7 g of ethylamine, 18 ml of methanol, and 1 g of 4A molecular sieves were stirred and heated under reflux for 30 minutes. The reaction mixture was then cooled to approximately 45°C, 0.6 g of sodium methoxide was added, and the mixture was heated under reflux for 1.5 hours. After cooling, the reaction mixture was acidified with 12 ml of concentrated hydrochloric acid, and the sieves were removed by filtration. Ice water was added to the filtrate to precipitate the trans-3-bromocinnamic acid N-ethylamide. After recrystallization from ethanol, a product melting at 89-90°C is obtained. Studies of trans-3-bromocinnamic acid N-ethylamide (Examples 2 and 3) using differential calorimetry and a heated-stage microscope (for melting point measurement) have demonstrated the presence of two physically different polymorphic crystalline forms, designated Form I and Form II. Form II melts at 86-87°C and can be converted by comminution into Form I, which melts at 88-90°C. Example 4: 3-Bromocinnamic acid N-methylamide A solution of 5 g of trans-3-bromocinnamic acid chloride in 100 ml of dry benzene is added, with stirring, to a solution of 3 g of methylamine in 200 ml of ether. After the addition was complete, a stream of gaseous methylamine was slowly passed through the reaction mixture at room temperature for 1 hour. The solvent and excess amine were then removed under reduced pressure, and the residue was triturated with water, filtered, and recrystallized from ethanol/water. Trans-3-bromocinnamic acid N-methylamide was obtained, melting at 147-147.5°C. The nuclear magnetic resonance spectrum, infrared spectrum, and elemental analysis confirmed the structure of the compound. Example 5: 3-Chlorocinnamic Acid N-Ethylamide: 4.4 g of trans-3-chlorocinnamic acid chloride in 75 ml of dry benzene was reacted with an excess of ethylamine in 75 ml of dry ether by the procedure given in Example 2. Trans 3-chlorocinnamic acid N-ethylamide is obtained, melting at 87-88°C. The nuclear magnetic resonance spectrum, infrared spectrum, and elemental analysis confirm the structure of the compound. Example 6. Trans 3-iodocinnamic acid N-ethylamide Trans m-iodocinnamic acid chloride is prepared from trans m-iodocinnamic acid by the usual procedure given in Example 1. 6.6 g of the acid chloride in 200 ml of dry toluene are added, with stirring at room temperature, to a solution of 3.0 g of ethylamine in 300 ml of dry ether, and the reaction mixture is heated under reflux for one hour and then continued as in Example 2. White crystals of trans-m-iodocinnamic acid N-ethylamide are obtained, melting at 116-117°C. Nuclear magnetic resonance spectra, infrared spectrum, and elemental analysis confirm the compound's structure. i101781 P r / y k l 9 d y VII '-.XI. Proceeding according to the method given in Example II, the following compounds are obtained:* compounds with trans configuration, in which X and R have the meanings given below in formula 1. In all cases, nuclear magnetic resonance spectra, infrared spectra and elemental analysis confirmed the structures of the compounds. By proceeding as described in Examples I and IV, respectively, using 3-iodocinnamic acid chloride, compounds with the trans configuration are obtained, in which X and R have the meanings given below in formula 1: Example XII XIII X J J R CH/CHj/j M Melting point 109-110 ' 1 134-135 In both cases, the infrared and ultraviolet spectra and elemental analysis confirmed the structure of the compounds obtained. The melting points were determined after recrystallization of the compounds from a mixture of ethanol and water. Example XIV 3-Bromocinnamic Acid Amide A solution of 2.7 g of 3-bromocinnamic acid chloride in 50 ml of dry toluene was added, with stirring at ambient temperature, to 150 ml of a saturated solution of ammonia in dry ether, ammonia gas being introduced into the ether solution throughout the addition of the acid chloride. The solution was added over 15 minutes, after which the reaction mixture was stirred at 20-22°C for a further 2 hours under an ammonia atmosphere. The solvent and excess ammonia were removed under reduced pressure, and the residue was thoroughly triturated with water. The crude product was recrystallized from a solution of ethanol and water to give 3-bromocinnamic acid amide, m.p. 110°C. Example 15 3-Bromocinnamic Acid N-Ethylamide A mixture of 4.54 g of 3-bromocinnamic acid and 2.5 g of N^-dimethylurea is heated under nitrogen in a bath at 260-270°C for 1 hour with stirring. The reaction mixture is cooled and triturated with 100 ml of 1-2% sodium carbonate solution. The crude residue is washed with water and recrystallized from aqueous ethanol to give 3-bromocinnamic acid N-ethylamide, melting at 88°C. PL PL PL PL PL PL PL PL PL
