Przedmiotem wynalazku jest sposób wytwarzania kwasu 2,5-dwuketoglikonowego.Kwas 2,5-dwuketoglikonowy jest uzytecznym pólpro¬ duktem w syntezie witaminy C. Z tego wzgledu wytwarza sie go przy pomocy wielu róznych rodzajów bakterii takich 5 jak Acetobacter melanogenum, Acetobacter aurantium, Gluconoacetobacter rubiginosus, Gluconoacetobacter li- auifaciens i Pseudomonas sesami. Jednakze zastosowanie tych drobnoustrojów jest niepozadane z przemyslowego punktu widzenia z powodu powstawania duzych ilosci 10 brunatnych lub zólto-brunatnych pigmentów jako produk¬ tów ubocznych hodowli przez co zmniejsza sie czystosc wytwarzanego kwasu 2,5-dwuketoglikonowego.W opisie patentowym St. Zjedn. For this reason, it is produced by a variety of different types of bacteria, such as Acetobacter melanogenum, Acetobacter aurantium, Gluconoacetobacter rubiginosus, Gluconoacetobacter liauifaciens, and Pseudomonas sesami. However, the use of these microorganisms is undesirable from an industrial standpoint because of the formation of large amounts of brown or yellow-brown pigments as by-products of the culture, which reduces the purity of the produced 2,5-diketoglyconic acid. U.S. Patent No. 3,790,444 discloses a method for producing the acid. 2,5-Diketoglyconic acid without the formation of accompanying brown pigments by a new strain designated Acetobacter fragum in a culture containing glucose and a nitrogen source as a nutrient. This method uses an initial pH of 5.8 and after 32 hours, 2,5-Diketoglyconic acid is obtained with a yield of 87% calculated as glucose. The invention relates to an economical process for the production of 2,5-Diketoglyconic acid using the readily available Acetobacter cerinus strain. The preferred strains are IFO 3263 and 3266, which produce 2,5-diketoglyconic acid with a yield of over 95% (calculated as glucose). 2,5-diketoglyconic acid is used as an intermediate in the production of ascorbic acid. An aqueous solution of 2,5-diketoglyconic acid can be selectively reduced to obtain a mixture of 2-ketogulonate and 2-ketoglyconate, which can be converted to ascorbic and erythorbic acids. In the method of the invention, 2,5-diketoglyconic acid is readily obtained by bacterial action on glucose, using the known, readily available strain of Acetobacter cerinus. All commonly available strains of Acetobacter cerinus were tested. In these studies, the yield of obtained ketoacids was 50-95% (calculated as glucose). When Acetobacter cerinus, preferably IFO 3263 or 3266, is used, the only ketoacid produced is the desired 2,5-diketoglyconic acid and the process is carried out with a yield of over 95% (calculated as glucose). The commonly available Acetobacter cerinus strains are: IFO 3262 (ATCC 12303), 3263, 3264, 3265, 3266, 3267, 3268, 3269. These strains are known and registered in the ATCC catalogue, 11th edition, 1974 as ATCC 12303 and in the register kept by the Institute for Fermentation in Osaka as Gluconobacter cerinus IFO 3262 - IFO 3270 (the difference in the strain name is due to the different (nomenclature used by the Osaka Institute). The above-mentioned Acetobacter cerinus strains are cultivated in an environment in which glucose is the main carbon source. These microorganisms do not require expensive nitrogen sources such as peptone or meat extract. Usually, cultivation is carried out in a medium containing 2.5-20% glucose and a cheap nitrogen source such as urea and inorganic nitrogen sources such as ammonium salts, especially ammonium sulfate, ammonium nitrate or ammonium phosphate, and nicotinic acid is added to the medium as a growth indicator. The medium also contains mineral salts and a source of phosphorus. The glucose concentration in the medium is varied within the range of 2.5-20%, preferably 10-12%, in order to obtain 2,5-diketoglyconic acid in the most economical way. The fermentation temperature is 20-35°C, preferably 25-30°C, and especially about 28°C. The initial pH of the culture medium is 3.5-7.5, preferably 5-6. During the fermentation process, the pH value is maintained at about 5.5 by adding sodium hydroxide solution. Calcium carbonate can also be used to regulate the pH value. It is added to the culture after steam sterilization under increased pressure in the amount of about 30 g per 110 g of glucose. After inoculation, the fermentation medium is mixed with a mechanical stirrer at a speed of 1700 rpm and aerated at a speed of 0.5-1 volume of air (volume of broth) per minute. Using Acetobacfer cerinus IfO 3263 or 3266, the fermentation is carried out until the yield of 2,5-diketoglyconic acid is at least 90% (calculated as glucose) (36-40 hours). Paper chromatography has shown that the conversion of glucose to 2,5-diketoglyconic acid proceeds as follows: glucose ^*" acid 2-Ketoglyconic acid - 2,5-Diketoglyconic acid glucose - 5-Ketoglyconic acid - + 2,5-Diketoglyconic acid. Whatman No. 1 and No. 4 paper were used and the solvent system was methyl ethyl ketone:acetone:formic acid:water (80:6:2:12). The acid spots were identified by spraying with a 0.2% ethanolic solution of o-phenylenediamine containing 1% nitric acid and heating to approximately 70°C (5-Ketoglyconic acid - blue; 2-Ketoglyconic acid - yellow; 2,5-Diketoglyconic acid - green). High-pressure liquid chromatography can also be used for identification purposes. 2,5-Diketoglyconic acid can be separated and recovered from the final fermentation broth by any conventional method known in the art. The filtered fermentation broth can be treated with borohydride and the resulting mixture of 2-ketoglyconic and 2-ketogulonic acids hydrolyzed to yield ascorbic and erythorbic acids. Example I. The pH value of the cooled medium is 5.0. Acetobacter cerinus IFO 3263 cells derived from an agar slant are added to the flask as a medium (5 ml of 20 ml of sterile aqueous suspension) and then the flask is shaken on a rotary shaker at 28°C for 24 hours. Example II. Fermentation is continued until the yield of 2,5-diketoglyconic acid exceeds 95% (calculated as glucose). The resulting product is isolated by chromatography of the crude fermentation broth. Proceeding as described above, but using Acetobacter cerinus IFO 3266, 2,5-diketoglyconic acid is obtained in a yield exceeding 95% (calculated as glucose). Patent Claims 1.
