Description

Przedmiotem wynalazku jest sposób przygotowania podloza do fermentacji kwasu mlekowego.Kwas mlekowy otrzymywany na drodze fermentacyjnej jest produktem enzymatycznego rozkladu heksoz. Bakterie mlekowe do rozwoju w srodowisku fermentacyjnym, potrzebuja w skladzie pozywki specyficznych substancji wzrostowych, miedzy innymi bialek i produktów ich rozpadu. Pan i wspólprac, stwierdzili (Ing. Eng. Chem. t. 32, nr 5 s. 709, 1940), ze bakterie Thermobacterium delbrucki potrzebuja dla normalnego wzrostu substancji bialkowych o niepel¬ nym stopniu hydrolizy. Dla najczesciej stosowanych w fermentacji mlekowej zródel substancji bialkowych jakimi sa kielki slodowe i zarodki zbozowe (pszenne i zytnie) w/w autorzy stwierdzili istnienie blizej nieokreslonego „termolabilnego czynnika wzrostowego", który zanikal po ogrzaniu pozywki do temperatury przekraczajacej 65°C dajac w konsekwencji wydluzony czas fermentacji i niepelne odfermentowanie wprowadzonego cukru. Spozywczy nr 19, s. 35,1961) moczenia kielków slodowych w 6-7% kwasie mlekowym i sporzadzania pozywki z tak przygotowanego namoku. Zabieg ten hamowal rozwój macierzysty mikroflory kielków w czasie okolo 48 godz., a wiec dluzszym niz okres lag-fazy po zaszczepieniu szczepem produkcyjnym. Kwas mlekowy neutralizowany byl wprowadzona dodatkowa iloscia CaCo3 przez zaszczepieniem szczepem produkcyjnym.2 133344 Zmaczynski i Ziobrowski zaproponowali (Zeszyty Naukowe WSE Wroclaw nr 18/1963) sposób przygotowania pozywki polegajacy na ekstrakcji zarodków lub kielków w temperaturze 50°C w ciagu 1 godziny, a nastepnie na rozdzieleniu czesci stalej i czesci plynnej. Czystosc mikrobiologiczna zachowana byla przez czas dluzszy niz 1 doba, a wiec znacznie dluzszy niz lag-faza fermentacji mlekowej (8-10 godzin).Sposób wedlug wynalazku polega na tym, ze zarodki zbozowe lub kielki slodowe maceruje sie z dodatkiem ok. 1% preparatu enzymatycznego pochodzacego z odcieku po hodowli wglebnej grzybni Penicillium roaueforti w czasie od 20 do 24 godz., w temp. 22-25°C. Z kolei przeprowadza sie termiczna pasteryzacje pozywki uzupelnionej w niezbedne ilosci zródel wegla, azotu i fosforu w temp. 100° w ciagu 15 min. Proces maceracji mozna prowadzic odrebnie tzn. poza kadzia fermentacyjna.Proponowany sposób przygotowania podloza do fermentacji kwasu mlekowego z czesciowo zhydrolizowanego surowca zbozowego umozliwia uzyskanie pozywki odznaczajacej sie czystoscia mikrobiologiczna w okresie lag-fazy, przy jednoczesnym zmniejszeniu ilosci surowca zbozowego uzywanego do fermentacji do 50%. Zmniejszenie iloscia surowca zbozowego mozliwe jest w wyniku czesciowej hydrolizy substancji bialkowych, co powoduje lepsze wykorzystanie bialek zawartych w tym surowcu.Przyklad I. Fermentacje prowadzono w fermentorze o objetosci 2500 cm3 w temp. 50° przy pH = 6,3 utrzymywanym w sposób ciagly przez automatyczny korektor pH, neutralizujac powsta¬ jacy kwas mlekowy 3n zawiesina Ca(OH)2. Szczepem produkcyjnym byl szczep „Z" Thermobacte- rium delbrucki z kolekcji TPChiS AE we Wroclawiu. Podloze fermentacyjne zawieralo: 250 gcukru (10% w stosunku do objetosci), 2,5 g (NH4)2 HP04 (0,1% w stosunku do objetosci) i 12,5 g zarodków zytnich. Ilosc zarodków(surowca zbozowego) zostala zmniejszona o polowe w stosunku do powszechnie przyjetej w praktyce przemyslowej. Pozywka byla przygotowana dwuetapowo.Etap I polegal na 24 h maceracji przewidzianej do fermentacji ilosci zarodków w 2500 cm3 wody wodociagowej z dodatkiem 1% obj. (25 ml) preparatu enzymatycznego z odcieku po hodowli grzybni Penicillium roauefortii, w temp. pokojowej 25°C przy ciaglym mieszaniu. Aktywnosc preteolityczna preparatu wynosila 2000JK/lg s.s. grzybni. Po procesie maceracji oznaczono przyrost azotu a aminowego stwierdzajac, ze wynosi on 22% w stosunku do wartosci poczatkowej.Etap II polegal na uzupelnieniu pozywki do pelnego skladu przez wprowadzenie przewidzia¬ nej ilosci cukru i fosforanu dwuamonowego i gotowaniu jej w temp. 100°C przez 15 min. Po zakonczeniu gotowania pozywke poprowadzono do fermentora i po ustaleniu temperatury fer¬ mentacji i zalozonego pH szczepiono 1% zaszczepem szczepu produkcyjnego. PLThe subject of the invention is a method of preparing a substrate for lactic acid fermentation. Lactic acid obtained by fermentation is a product of enzymatic decomposition of hexoses. To develop in a fermentation environment, lactic bacteria need specific growth substances, including proteins and their breakdown products. Mr. and co-workers stated (Ing. Eng. Chem. Vol. 32, No. 5, pp. 709, 1940) that the Thermobacterium delbrucki bacteria needed protein substances with an incomplete degree of hydrolysis for normal growth. For the sources of protein substances most often used in lactic fermentation, such as malt sprouts and cereal germs (wheat and rye), the above-mentioned authors found the existence of an undefined "thermolabile growth factor", which disappeared after heating the nutrient to a temperature exceeding 65 ° C, resulting in a prolonged fermentation time and incomplete fermentation of the introduced sugar A. Moreover, they contain numerous microflora causing microbiological contamination during the process of lactic fermentation. There is a known method (Przem. Spozywczy No. 19, p. 35, 1961) of soaking malt sprouts in 6-7% lactic acid and making nutrients from the soak prepared. This treatment inhibited the development of the native microflora of the sprouts for about 48 hours, ie longer than the lag-phase period after inoculation with the production strain. In turn, the thermal pasteurization of the nutrient medium supplemented with the necessary amounts of sources of carbon, nitrogen and phosphorus is carried out at 100 ° for 15 minutes. The maceration process can be carried out separately, i.e. outside the fermentation vat. The proposed method of preparing the substrate for the fermentation of lactic acid from partially hydrolyzed cereal raw material allows to obtain a nutrient with microbiological purity during the lag-phase, while reducing the amount of cereal raw material used for fermentation to 50%. The reduction of the amount of cereal raw material is possible as a result of partial hydrolysis of protein substances, which results in a better use of proteins contained in this material. Example I. Fermentations were carried out in a fermenter with a volume of 2500 cm3 at 50 ° at pH = 6.3 maintained continuously for automatic pH corrector, neutralizing the lactic acid formed by the 3N Ca (OH) 2 suspension.
