Opis wynalazkuDescription of the invention
Przedmiotem wynalazku jest sposób mikrobiologicznego oczyszczania gruntów z węglowodorów, przeznaczony do remediacji gruntów metodą in situ i ex situ. The subject of the invention is a method of microbiological soil remediation from hydrocarbons, intended for soil remediation by in situ and ex situ methods.
A method of microbiological remediation of soil from petroleum products is known from the Polish patent description No. PL 180141, which uses native bacteria collected in soil samples to be cleaned, from which, after isolation and identification, pathogenic bacteria are eliminated, and bacteria with the highest activity in the decomposition of hydrocarbons contained in petroleum products, are passaged and introduced into the contaminated soil in a minimum amount of at least 10 5 cells per 1 g of dry soil mass. During the remediation process, the amount of introduced bacteria is kept above the minimum amount and the bacterial species developing in the soil is identified. The multiplied bacteria are introduced into the soil by sprinkling it with a suspension of bacteria or on a solid mineral carrier, especially in the form of expanded clay. In addition, before multiplication, the isolated bacteria are cultivated on a liquid mineral medium under aerobic conditions with the content of 0.5 to 2% of petroleum products, and the species of bacteria showing the highest biodegradation activity of hydrocarbons contained in these products are selected for remediation.
From the description of the Polish patent application No. P.326136, there is known a method of cleaning land contaminated with petroleum substances and an installation for cleaning land contaminated with oil derivatives. The method consists in placing the contaminated part of the soil in the tank, adding calcium compounds and nitrogen and phosphorus fertilizers in the ratio 1C: 0.1N: 0.03P and kept in the tank for 3 to 6 days. Then, the treated soil is transported to the second tank, where it is heated with hot steam to a temperature of 19 ° 24 ° C and a humidity of 14-18% by weight is maintained and it is oxygenated by blowing from below. Then, the bacterial strains are introduced into the soil in the form of a liquid vaccine, by sprinkling and possible injection into deeper parts of the soil, the bioremediation process in the second tank being carried out in the cycle of 6-8 weeks.
Znany jest z polskiego opisu patentowego nr PL 184204 sposób mikrobiologicznego oczyszczania gruntów zanieczyszczonych związkami organicznymi metodą in situ. There is known from the Polish patent description No. PL 184204 a method of microbiological treatment of soil contaminated with organic compounds by in situ method. This method consists in determining the average concentration of pollutants, biodegradability of pollutants, the oxygen demand of microorganisms in a laboratory, and isolating the microflora useful for the biodegradation of pollutants, and then improving the surface structure of the soil by plowing it, and then adding a nutrient solution to the contaminated soil. forms of mineral fertilizers used in agriculture in the amounts and proportions selected in the laboratory, containing K + , NH4 + , HPO4 2- ions at the appropriate concentration. To the ground through the lance, and seeded on the surface of the indigenous microorganisms introduced previously isolated from the ground, adapted to a concentration of soil contamination and propagated to the required amount of about 0.2 x 10 -3 m 3 of microorganisms per 1 m 3 soil, then incorporated system biowentylacji and the soil cleaning process begins, during which the polluted air is extracted and directed to a special biofilter, while the biodegradation process takes place while maintaining the temperature in the range of 0 - 40 ° C, preferably in the range of 20 - 27 ° C, soil in the range of 50 - 80%, acidity in the range of pH 5-8, preferably in the range of 6 - 7.5, which affects the speed of soil remediation.
W procesie namnażania mikroorganizmów, prowadzonym w bioreaktorze, zapewnia się im również odpowiednią zawartość azotu i fosforu poprzez wprowadzanie związków tych pierwiastków, w postaci NH4NO3, (NH4)2SO4, NH4H2PO4,The method of microbiological soil treatment according to the invention consists in that the remediation process is carried out in a closed water circuit, preferably continuously. According to the method, the contaminated soil is moistened to a level of at least 50% of the total water capacity, with water and / or leachate containing autochthonous microorganisms, especially capable of decomposing hydrocarbons, in an amount of at least 10 7 cells per 1 g of soil dry weight. The resulting hydrocarbon-containing leachate is introduced into a bioreactor containing microorganisms, introduced as a seed obtained from a contaminated soil sample. The inoculation is prepared through the isolation and selection of autochthonous microorganisms, which are characterized by particularly high activity in the degradation of hydrocarbon compounds, while eliminating pathogenic microorganisms and microorganisms producing toxic metabolites. The cultivated inoculum is introduced into the bioreactor at the beginning of the purification process, and subsequent batches of leachate from the remediated soil are the main source of organic carbon, which is a nutrient substrate for subsequent batches of bacterial suspension returned to soil remediation. In the process of multiplication of microorganisms, carried out in the bioreactor, they are also provided with the appropriate content of nitrogen and phosphorus by introducing compounds of these elements in the form of NH4NO3, (NH4) 2SO4, NH4H2PO4,
PL 207 237 B1 (NH4)2HPO4 NH4HCO3 i/lub (NH4)2CO3. W toku namnażania mikroorganizmów rozkłada się biologicznie produkty naftowe i oczyszcza się odcieki. Uzyskaną zawiesinę bakteryjną w postaci biomasy wraz z oczyszczonymi odciekami recyrkuluje się do gruntu, wzbogacając go w mikroorganizmy oraz uzupełniając zawartość wody.(NH4) 2HPO4 NH4HCO3 and / or (NH4) 2CO3. In the course of the multiplication of the microorganisms, the petroleum products are biologically decomposed and the leachate is purified. The obtained bacterial suspension in the form of biomass together with the purified leachate is recirculated to the soil, enriching it with microorganisms and replenishing the water content.
Związki te wprowadzane są do gruntu poprzez zraszanie po ich rozpuszczeniu w odcieku; dobierane są w proporcjach C:N:P=70120:8-30:0,5-2.The process of multiplication of microorganisms is carried out in a bioreactor, under aerobic conditions, under continuous aeration and at a constant temperature, preferably 28 ° C, with the pH value maintained in the range of 6 - 8.2. The bacterial suspension obtained in the bioreactor, rich in microorganisms capable of decomposing hydrocarbons, is introduced into the treated soil by spraying and / or injection or by a system of internal drainage. Preferably the process is carried out continuously using at least two bioreactors. Nitrogen and phosphorus compounds in the form of NH4NO3, (NH4) 2SO4, NH4H2PO4, (NH4) 2HPO4 NH4HCO3 and / or (NH4) 2CO3, selected depending on the pH of the treated soil, are used to enrich the soil with biogenic elements. These compounds are introduced into the soil by sprinkling after dissolving them in the leachate; are selected in the proportions C: N: P = 70 120: 8-30: 0.5-2.
W trakcie procesu remediacji prowadzi się kontrolę parametrów fizyko-chemicznych i mikrobiologicznych gruntu takich jak: pH, zawartość produktów naftowych, azot ogólny, azot amonowy i azotanowy, fosfor ogólny, ogólna liczebność mikroorganizmów, liczebność mikroorganizmów zdolnych do biodegradacji węglowodorów, aktywność enzymatyczna mikroorganizmów glebowych.During the remediation process, the soil physicochemical and microbiological parameters are controlled, such as: pH, content of petroleum products, total nitrogen, ammonium and nitrate nitrogen, total phosphorus, total number of microorganisms, number of microorganisms capable of biodegradation of hydrocarbons, enzymatic activity of soil microorganisms.
The process of decomposition of hydrocarbons takes place in the treated soil in accordance with the method, and is stimulated by the circulation of effluents, which at the entrance to the bioreactor are characterized by an increased content of hydrocarbons, and after leaving the bioreactor in the form of a bacterial suspension, they are a source of water and microorganisms decomposing pollutants in the soil.
The process according to the invention is suitable for the in situ or ex situ treatment of all soils contaminated with hydrocarbons, especially petroleum products such as crude oil fractions, heating oils, diesel engine oils, etc. According to the invention, in the ex situ method, a pile is formed on an insulated surface, and the leachate from it is collected by a drainage system and is collected in a collecting sump, while in the in situ method for the multiplication of microorganisms, recirculated contaminated groundwater is used.
An advantage of the invention is that it does not require the use of stripping towers or other apparatus to clean the effluents as they are returned to the bioreactor and used as a food substrate for the multiplication of microorganisms. The use of a closed water circuit significantly reduces the consumption of water necessary to maintain the moisture level of the cleaned soil. Another advantage is that the ex situ method uses a closed leachate cycle, which eliminates the risk of secondary contamination of the environment. The method according to the invention greatly reduces or even eliminates the consumption of food substrate preparations for the multiplication of microorganisms. The process of multiplication of microorganisms according to the invention is significantly shortened due to the use of the same hydrocarbons as the organic carbon source in the environment, the most active of which were the microorganisms from which the inoculum was generated, called the inoculum.
Sposób według wynalazku został objaśniony w przykładach wykonania. The method according to the invention has been explained in the working examples.

W trakcie procesuThe method of microbiological remediation of soil contaminated with petroleum products, carried out ex situ cyclically, consists in extracting a part of soil in the amount of 100 m 3 with a hydrocarbon concentration of 5300 mg per kg of dry matter, with an alkaline reaction and formed in the shape of a prism on the insulation foil . The heap is simultaneously aerated and moistened through sprinklers with leachate coming from the ground and a bioreactor with a capacity of 600 dm 3 up to 50% of the total water capacity, or supplemented with ground water. The effluents from the heap are directed to the bioreactor, into which the inoculum - inoculum of autochthonous microorganisms is also introduced. The inoculation is prepared from a contaminated soil sample through the isolation and selection of indigenous microorganisms characterized by particularly high activity in the degradation of hydrocarbon compounds. It is controlled whether these microorganisms do not produce toxic metabolites and whether they are pathogenic, which are eliminated. The grown seed is introduced into the bioreactor at the start of the purification process. The content of the bioreactor is enriched with nutrients in the form of NH4NO3, NH4H2PO4 compounds in the proportions C: N: P = 10: 10: 1 and the pH of the effluent is regulated to a neutral level. Microorganisms in the bioreactor are multiplied under aerobic conditions, continuously introducing air and maintaining a temperature of 28 ° C and a pH of 7. During the process
