Sposób otrzymywania reaktywnych estrowych pochodnych (S)-(+)-kamptotecyny o wzorze ogólnym 2, gdzie: R = H lub CH3.A method for obtaining reactive ester derivatives of (S)-(+)-camptothecin of the general formula 2, where: R = H or CH3.
Description
Przedmiotem wynalazku jest sposób wytwarzania reaktywnych estrowych pochodnych (S)-(+)-kamptotecyny, mogących znaleźć zastosowanie w leczeniu nowotworów.The subject of the invention is a process for the preparation of reactive ester derivatives of (S) - (+) - camptothecin, which can be used in the treatment of neoplasms.
Wzór 1 przedstawia strukturę CPT wraz z numeracją atomów.(S) - (+) - Camptothecin (CPT) is one of the very promising substances with anti-cancer properties. It is a modified monoterpene indole alkaloid first isolated in 1966 from the wood and bark of Camptotheca acuminata (Nyssaceae), a tree native to China. The CPT molecule has the S configuration at the C20 carbon atom. This fragment of the molecule is responsible for its excellent anti-cancer properties. The R isomer is pharmacologically inactive [Chavan S.P. and Venkatraman M.S., Arkivoc 2005, 165-169]. Formula 1 shows the structure of CPT with the numbering of atoms.

CPT jest stosunkowo łatwo dostępnym naturalnym produktem o dobrze poznanym mechanizmie działania in vitro. Niska toksyczność wobec normalnych komórek w stanie spoczynku to zasługa mechanizmu działania ograniczającym się do fazy S cyklu komórkowego [Lee N-J. i inni, Polym. Int. 2003, 52, 13391345]. Jak wiadomo, działanie cytostatyków polega m.in. na zaburzeniu cyklu komórkowego i powodowaniu śmierci komórki lub zahamowaniu jej podziałów i rozwoju. Cytostatyki ze względu na mechanizm działania można podzielić na następujące grupy: leki alkilujące, inhibitory mitozy, inhibitory topoizomeraz, antybiotyki cytostatyczne i antymetabolity. Spośród leków należących do inhibitorów topoizomeraz rozróżniamy cytostatyki oddziaływujące na topoizomerazę I i topoizomerazę II [The Pharmacological Basic of Therapeutics, Goodman & Gilman 2011 (12th Edition), Janiec W., Kompendium Farmakologii, Wydawnictwo Lekarskie PZWL, Warszawa 2008]. W konsekwencji wydłużona inhibicja topoizomerazy I jest uznawana za jeden z najistotniejszych czynników wpływających na aktywność terapeutyczną CPT [Wu S-F. i inni, Molecules 2008,13,1361-1371],CPT is a relatively readily available natural product with a well-known mechanism of action in vitro. In vivo studies show that CPT has significant antitumor activity in mice with human forms of cancer: lung, uterus, breast, pancreas and stomach. Low toxicity to normal resting cells is due to an action mechanism confined to the S phase of the cell cycle [Lee N-J. and others, Polym. Int. 2003, 52, 13391345]. As is known, the action of cytostatics is, inter alia, by disturbing the cell cycle and causing cell death or inhibiting cell division and development. Due to the mechanism of action, cytostatics can be divided into the following groups: alkylating agents, mitosis inhibitors, topoisomerase inhibitors, cytostatic antibiotics and antimetabolites. Among drugs belonging to topoisomerase inhibitors, we distinguish cytostatics affecting topoisomerase I and topoisomerase II [The Pharmacological Basic of Therapeutics, Goodman & Gilman 2011 (12th Edition), Janiec W., Kompendium Farmakologii, Wydawnictwo Lekarskie PZWL, Warsaw 2008]. CPT and its CPT derivatives inhibit the action of topoisomerase I, which is required for DNA replication and RNA transcription in cells [Wu S-F. et al., Molecules 2008, 13, 1361-1371]. CPT stabilizes topoisomerase I-induced breakdown of DNA strands, thereby preventing DNA strand reassembly, leading to immediate cell apoptosis. As a consequence, prolonged inhibition of topoisomerase I is recognized as one of the most important factors influencing the therapeutic activity of CPT [Wu S-F. et al., Molecules 2008,13,1361-1371],
Moreover, CPT turns out to be also a weak inhibitor of HIV and equine anemia virus replication, while its derivative 9-nitrocamptothecin has an inhibitory effect on HIV-1 replication in freshly infected monocyte cells.

W celu zredukowania tych problemów podjęto badania nad syntezą i stosowaniem w terapii licznych analogów i pochodnych CPT. Np. topotekan półsyntetyczny analog CPT jest efektywny w leczeniu wieloogniskowej leukoencefalopatii [Li Y-Y i inni, Molecules 2010, 15, 138-148].A limitation in the use of CPT is its toxicity to various organs or systems, necessitating a dose reduction, and on the other hand, poor solubility of CPT in water and body fluids. Sometimes the low stability of the lactone ring in the CPT structure is also discussed, leading to its opening to form carboxyl and hydroxyl moieties. In order to reduce these problems, research has been undertaken on the synthesis and application of numerous CPT analogues and derivatives in therapy. The most common CPT analogs with hydrogen atoms at carbon numbers 7, 9, 10 and / or 11 are substituted for topotecan [US Patent No. 7,547,785], irinotecan [US Patent No. 7,683,170], exatekan [US Patent No. 5,834,476] and other derivatives substituted therein [US Pat. Nos. 8,685,997, 8,592,588, 7,687,487, 7,655,640] having improved water solubility, being less toxic, or having other therapeutic or diagnostic support characteristics. For example, topotecan, a semi-synthetic analogue of CPT, is effective in the treatment of multifocal leukoencephalopathy [Li Y-Y et al., Molecules 2010, 15, 138-148].
In addition, the patent literature describes CPT conjugates formed by an ester bond with the hydroxyl group at the carbon atom 20. An example may be compounds composed of a CPT molecule bound by an ester moiety with a linker to antibodies or their fragments, constituting a group controlling the conjugate towards diseased cells [Patents U.S. Patent No. 8,877,901.8 834,886,8617,558]. Additionally, there are described such CPT esters which also have substituents on carbon atoms number 7, 9, 10, 11 or / and 12 [US Patents Nos. 8 779 138, 6 855 720, 6 492 335, RE39 707] combining various properties or functionality, e.g. increased water-solubility [US Pat. chemotherapy [US Patent No. 8,563,537].
Otrzymano m.in. koniugaty CPT z glikolem polioksyetylenowym [Fleming A.B i inni, Bioconjugate Chem. 2004, 15, 1364-1375; Warnecke A. i Kratz F., Bioconjugate Chem. 2003, 14, 377-387; Greenwald R.B. i inni, Bioorg. Med. Chem. Lett. 2003, 11, 2635-2639], poli(kwasem glutaminowym) [DeVries P. i inni, J. Med. Chem. 2003, 46, 190-193; Singer J.W. i inni, J. Control. Release 2001, 74, 243-247], β-cyclodekstrynami [Cheng J. J. i inni, Bioconjugate Chem. 2003, 14, 1007-1017], poli[N-(2- hydroksypropylo)metakrylamidem] [Caiolfa V.R. i inni, J. Control. Release 2000, 65, 105-119], poliacetalami lub/i poliketalami [Patent USA nr 8 491 880], poli[(N-karboksybutylo)-L-aspartamidem] [Fan N. i inni, Colloids and Surf. B Biointerfaces 2010, 75, 543-549; Zhang W. i inni, Colloids and Surf. B Biointerfaces 2010, 81,297-303], poliamidoaminami [Fan H. i inni, Polymer 2010, 51, 5107-5114] poli(metakrylanem 2-fosforylocholinooksyetylu) [Chen X. i inni, Bioconjugate Chemistry 2009, 20, 2331-2341] czy z poliestrami [Parrish B. i Emrick T., Bioconjugate Chemistry 2007, 18, 263-267; Sobczak M. i inni, J. Macromol. Sci., Part A Pure Appl. Chem. 2014, 51, 254-262].A separate group of CPT conjugates are their combinations with polymers. The conjugation of CPT and a biodegradable or bioresorbable polymer gives i.a. the possibility of increasing its hydro- or lipophilicity, the possibility of extending the durability, programming the distribution, absorption and release kinetics. Despite numerous attempts, very few solutions of this type have been developed to date. The vast majority are conjugates with only 1 or 2 CPT molecules attached to the end (s) of the polymer chain per polymer chain. Received, among others CPT conjugates with polyoxyethylene glycol [Fleming A.B. et al., Bioconjugate Chem. 2004, 15, 1364-1375; Warnecke A. and Kratz F., Bioconjugate Chem. 2003, 14, 377-387; Greenwald R.B. et al., Bioorg. Macromol. Sci., Part A Pure Appl. Chem. 2014, 51, 254-262].
Celem wynalazku było opracowanie sposobu otrzymywania reaktywnych estrowych pochodnych CPT, które ze względu na grupy funkcyjne obecne w ich strukturze są monomerami zdolnymi do polimeryzacji rodnikowej, która jest jedną z najprostszych metod polimeryzacji i w wyniku reakcji (ko)polimeryzacji rodnikowej mogą tworzyć wielkocząsteczkowe koniugaty CPT, w których na jedną cząsteczkę polimeru przypada wiele cząsteczek CPT.The aim of the invention was to develop a method for the preparation of reactive ester CPT derivatives, which, due to the functional groups present in their structure, are monomers capable of radical polymerization, which is one of the simplest polymerization methods and, as a result of radical (co) polymerization reaction, can form macromolecular CPT conjugates, in of which there are many CPT molecules per polymer molecule.

Osad poddaje się krystalizacji z rozpuszczalnika krystalizacji po czym osad oddziela się i poddaje suszeniu w suszarce próżniowej w temperaturze od 20 do 40°C przez 10 do 96 godzin.R = H or CH 3 , consists in the fact that the excess of acrylic acid halide or methacrylic acid halide is subjected to a non-equilibrium esterification reaction with a CPT molecule having a tertiary hydroxyl group, carried out in a nonprotonic polar organic solvent which may contain in its structure, in addition to carbon, hydrogen and oxygen, also chlorine, bromine and nitrogen atoms incompatible with water and able to dissolve the main reaction product and all reactants except CPT. The main advantages of the described modifiers include their reactivity in radical polymerization processes. Radical (co) polymerization of CPT (meth) acrylate derivatives can follow the mechanism of classical (free) radical polymerization and the mechanisms of controlled polymerization (CRP), including atom transfer radical polymerization (ATRP). In this way, CPT molecules can be immobilized on polymer chains to form polymer drug conjugates in which there is significantly more than 1 CPT molecule per polymer chain. It thus enables the dosage of drug per gram of conjugate to be defined to a fairly large extent. Additionally, the ATRP method makes it possible to obtain block or branched CPT copolymers, in which the comonomers will determine many properties of the final conjugate.
