Description
Opis wynalazkuDescription of the invention
The subject of the invention is a method for obtaining a mixture based on fermented turmeric rhizome, a mixture based on fermented turmeric rhizome in powder form and its use.
The traditional method of processing turmeric rhizome (Curcumae longae rhizoma) involves heat treatment and drying. Turmeric has been a well-known herbal ingredient for centuries, rich not only in curcumin and essential oils but also in a range of B vitamins and fat-soluble vitamins. Excessive heat treatment can have a particularly negative impact on B vitamins and can significantly reduce turmeric's antioxidant properties. However, unheated rhizome in its native state has little nutritional value due to its distinctive bitter taste, poor absorption, and high levels of native fiber, which inhibits digestion.
Poza właściwościami barwiącymi i przyprawowymi kurkuma, a szczególnie zawarte w niej kurkuminoidy wykazują różne właściwości lecznicze potwierdzone badaniami farmakologicznymi przeprowadzonymi z udziałem różnych zwierząt. Ekstrakty z kurkumy wykazują działanie przeciwzapalne, przeciwgrzybicze oraz przeciwnowotworowe (Amalraj, A., Pius, A. Gopi, S. Biological activities of curcuminoids, other biomolecules from turmeric and their derivatives - A review. Journal of Traditional and Complementary Medicine 2017, 7, 205-233; Yamamoto, H.; Hanada, K.; Kawasaki, K.; Nishijima, M. Inhibitory effect on curcumin on mamma-lian phospholipase D activity. FEBS Lett. 1997, 417, 196-198; Apisariyakul, A.; Vanittanakom, N.; Buddhasukh, D. Antifungal activity of turmeric oil extracted from Curcuma longa (Zingiberaceae). J. Ethnopharmacol. 1995, 49, 163-169). Kurkuminoidy, żeby mogły oddziaływać na organizmy żywe (ludzie i zwierzęta) muszą zostać w pierwszej kolejności dostarczone do organizmu, a następnie ulec wchłonięciu z przewodu pokarmowego do krwiobiegu (Amalraj i in., 2017; Jager, R. Lowery, R.P. Calvanese, A.V. Joy, J.M. Purpura, M. Wilson, J.M. Comparative absorp-tion of curcumin formulations. Nutrition Journal 2014, 13: 11, 1-8). Dried turmeric rhizomes contain 3-5% essential oils and 0.02-6.0% curcuminoids, such as curcumin, demethoxycurcumin, and bisdemethoxycurcumin. These curcuminoids can be present in varying amounts and proportions, depending on biological and environmental factors related to the cultivation of the rhizome itself and, in the case of oleoresin or extracts, on the technology used to obtain them. In addition to its coloring and spice properties, turmeric, and particularly its curcuminoids, exhibit various medicinal properties, confirmed by pharmacological studies conducted on various animals. Turmeric extracts have anti-inflammatory, antifungal and anticancer properties (Amalraj, A., Pius, A. FEBS Lett. 1997, 196-198; Vanittanakom, N.; Antifungal activity of turmeric oil extracted from Curcuma longa (Zingiberaceae). 1995, 49, 163-169). To have an effect on living organisms (humans and animals), curcuminoids must first be delivered to the body and then absorbed from the gastrointestinal tract into the bloodstream (Amalraj et al., 2017; Jager, R. Lowery, R.P. Calvanese, A.V. Joy, J.M. Purpura, M. Wilson, J.M. Comparative absorption of curcumin formulations. Nutrition Journal 2014, 13: 11, 1-8). Concentrated curcuminoids, like powdered turmeric itself, have a very intense, characteristic taste and are bitter, which practically disqualifies their direct consumption. In this form, they can be either encapsulated or used as a food supplement.
Most existing microbial fermentation methods for turmeric are liquid fermentation methods and use large amounts of water. Therefore, the yield is low and the process is difficult to conduct because a liquid fermentation device is required, and drinking water and post-fermentation water treatment are problematic. However, liquid fermentation favors the growth of fermenting bacteria.
Znane są zgłoszenia patentowe dotyczące technologii związanej z kurkumą fermentowaną. Lactobacillus plantarium, Bacillus subtilis i inne bakterie kwasu mlekowego są dodawane do fermentacji kłącz kurkumy, a następnie podgrzewane i suszone w celu wytworzenia materiału spożywczego - np. japoński dokument patentowy JP2949411. There are patent applications for fermented turmeric technology. Lactobacillus plantarium, Bacillus subtilis, and other lactic acid bacteria are added to ferment turmeric rhizomes, which are then heated and dried to produce a food grade material, e.g., Japanese patent document JP2949411. A method for producing fermented turmeric is disclosed in Japanese patent document JP2994607, according to which powdered turmeric rhizome is mixed with yeast and fermented with the yeast, or starch and monascus are added to ferment the turmeric.
Furthermore, a method for removing the bitterness of turmeric by fermenting a mixture obtained by adding vegetable oil to turmeric and further mixing it with yeast at room temperature or by heating at a low temperature is known from patent document JP2873678. Patent document JP4183371 discloses a method for producing fermented turmeric, wherein lactic acid fermentation is carried out using a medium containing 5 to 30% by mass of turmeric rhizome or dry powder thereof after or while amylase and protease act on the dry powder, and functional fermented turmeric foods prepared by adding sugars and lactic acid bacteria extract to dry turmeric rhizome powder, adding yeast and lactic acid bacteria, fermenting and drying.
Patent application JP2010213609A describes a method for producing a fermented product having α-amylase inhibiting activity, characterized by fermenting a material comprising a dry powder of zedoary or medlar rhizomes using two or more types of lactic acid bacteria, wherein the lactic acid bacteria belong to the genus Lactobacillus, Streptococcus, or Tetragenococcus. In the fermentation process, carbohydrates such as glucose, sucrose, and molasses, and proteins may be added as assimilation agents for the fermenting bacteria.
After culturing each of the bacteria, the bacteria are premixed before being added to the medium and are preferably added in an amount of 1 to 10% by weight based on the weight of the dry powder content. Fermentation is preferably carried out at a temperature of 20 to 50°C, preferably 40°C, and the fermentation time can be appropriately selected according to the progress of the fermentation depending on conditions such as pH and the number of initial bacteria. After completion of the fermentation, it is preferable to dry it in an oven so that the moisture content is 10% by weight or less. The drying method can be thermal drying or freeze-drying. In the case of thermal drying, the product temperature is 100°C. After drying, sterilization is carried out by a known method, such as heating necessary to obtain a fermented product used as a food material or a raw material for extracts. The fermented turmeric product can be used as an active ingredient in a composition for preventing or treating diabetes, but it can also be subjected to other treatments, such as extraction processing.

Mieszanina na bazie fermentowanego kłącza kurkumy w postaci proszku charakteryzuje się według wynalazku tym, że zawiera proszek liofilizowanej fermentowanej kurkumy w ilości 90,86%, proszek colostrum bovinum w ilości 8,99%, krzemionkę krystaliczną w ilości 0,15%.The mixture based on fermented turmeric rhizome in powder form is characterized according to the invention in that it contains freeze-dried fermented turmeric powder in the amount of 90.86%, colostrum bovinum powder in the amount of 8.99%, crystalline silica in the amount of 0.15%.
The essence of the invention is also the use of the mixture obtained by the above-described method based on fermented turmeric rhizome in the form of powder, containing freeze-dried fermented turmeric powder in the amount of 90.86%, colostrum bovinum powder in the amount of 8.99%, crystalline silica in the amount of 0.15%, as a food additive.
The inventive solution, through the use of lactic acid fermentation with a colostrum bovinum starter and subsequent processing without the use of high temperatures, eliminates pathogenic bacteria, while also increasing the digestibility and bioavailability of the ingredients through the hydrolytic action of lactic acid. Due to the lack of naturally occurring lactic acid bacteria in the raw material, lactobacillus bacteria obtained from bovine colostrum were used. After appropriate processing, these bacteria constituted a starter culture that allowed for the regulation of the turmeric rhizome fermentation process.

Uzyskany w ten sposób koncentrat bakterii mlekowych stanowi płynną mieszaninę starterową bogatą w bakterie kwasu mlekowego, których miano osiąga wartość rzędu 107 jednostek tworzących kolonię na ml.A lactic acid bacteria starter was prepared using bovine colostrum (Colostrum bovinum). Frozen blocks of Colostrum bovinum were heated to 38°C, then subjected to fat centrifugation, and the skim milk was separated. The separated skim milk was microfiltered (400 L) on 0.14 μπι ceramic isoflux membranes using a transmembrane pressure not exceeding 80,000 Pa—in this case, 80,000 Pa. The resulting lactic acid bacteria concentrate constitutes a liquid starter mixture rich in lactic acid bacteria, whose titer reaches 107 colony-forming units per ml.

W pierwszym etapie płukania surowiec zanurzono w 10% roztworze kwasu cytrynowego, po 1 minucie odsączono materiał i przeniesiono do zbiornika z czystą wodą. Po opłukaniu w czystej wodzie materiał został odsączony. Następnie mokre kłącza kurkumy rozdrabniano za pomocą tzw. kutra wysokoobrotowego. Jest to urządzenie wysokoobrotowe zaopatrzone w nóż stalowy (9000 obr./min). Ruchoma misa urządzenia zapewnia pełną dezintegrację cząstek. Proces rozdrabniania trwa 3 minuty. The acceptance of raw turmeric rhizomes and their selection are important elements of the production process, as they determine their quality. If the raw material arriving at the plant has an off-flavor, leaks, or is of an inappropriate consistency (e.g., rhizomes that are too soft, have an inappropriate turgor pressure), visible signs of mold, visible signs of bacterial diseases, discoloration, stains, etc., this constitutes grounds for returning the raw material. It is impossible to obtain a good-quality fermentation medium from raw material infected with mold or damaged, where microbiological spoilage has begun. Selection allows for the rejection of turmeric rhizomes that show signs of spoilage. At this stage, an organoleptic assessment is conducted, including consistency, odor, and appearance. The acceptable amount of discarded material should not exceed 0.1%. Chemical peeling is performed in accordance with safety regulations, using goggles and a protective mask. This peeling method allows for the minimum weight loss of the peeled material, as only the outer layer of the peel is dissolved. It is performed in a NaOH solution at 80°C. Peeling takes 1 minute. After this time, the rhizomes are drained from the solution and transferred to clean water to rinse away any remaining NaOH. Rinsing is a necessary step in chemical peeling. In the first rinsing step, the raw material was immersed in a 10% citric acid solution. After 1 minute, the material was drained and transferred to a tank of clean water. After rinsing in clean water, the material was drained. Next, the wet turmeric rhizomes were ground using a high-speed cutter. This is a high-speed device equipped with a steel knife (9000 rpm). The device's moving bowl ensures complete particle disintegration. The grinding process takes 3 minutes. During the grinding process, the turmeric rhizome was combined with the remaining ingredients of the fermentation mixture according to the recipe presented in Table 2. The ingredients were added at the grinding stage to ensure that all the elements were well combined.

Wwyniku fermentacji zdecydowanie wzrosła zdolność do redukcji wolnego kationorodnika DPPH, co wskazuje na przeciwutleniające działanie fermentowanego produktu. Dodatkowo zawartość colostrum wzmocniła ten efekt. As a result of fermentation, the ability to reduce the free cation radical DPPH significantly increased, indicating the antioxidant activity of the fermented product. The presence of colostrum further enhanced this effect.
As a result of fermentation, the levels of vitamin C and vitamin B5 decreased. The fermentation process had no effect on the levels of curcumin, vitamin K2, and vitamin B7.
Produkt po fermentacji zawierający dodatek colostrum zawierał w znaczny sposób więcej: Witaminy E, witaminy B9, witaminy B2 oraz witaminy K1.The fermented product containing added colostrum contained significantly more: Vitamin E, vitamin B9, vitamin B2 and vitamin K1.
The fermentation process increased the bioavailability of the ingredients and improved the product's parameters. This relationship is observable when comparing the parameters of the fermented and unfermented product. The addition of colostrum bovinum allows for a much broader spectrum of biological effects.
