Abstract
Przedmiotem wynalazku jest sposób przygotowania immunoczujnika do wykrywania wirusa grypy, gdzie czyste złote elektrody płucze się etanolem i umieszcza się w 10 mM etanolowym roztworze 1,6-heksanoditiolu (HDT) na 20 h w temperaturze pokojowej. Następnie elektrody płucze się kolejno etanolem i wodą dejonizowaną oraz odwraca się dołem do góry i nakrapla 10 µl roztworu złota koloidalnego (GCP) na 18 h w +4°C, w kolejnym etapie elektrody płucze się 0.1 M buforem PBS (145 mM NaCl, 2.7 mM KCl, 8.4 mM NaH2PO4, 1.6 mM Na2HPO4) i nakrapla się 10 µl roztworu przeciwciał anty-M o stężeniu 28 µg/ml w 0.1 M buforze PBS na 20 h w +4°C. W ostatnim etapie na powierzchnie elektrod nakrapla się 10 µl roztworu 0.5% (w/v) BSA w 0.1 M PBS na 2 h w +4°C, tak przygotowane elektrody płucze się buforem PBS i przechowuje w temperaturze +4°C do momentu użycia (nie dłużej niż jeden dzień). Ponadto ujawniono zastosowanie immunoczujnika do wykrywania w roztworze białka M oraz wirusa grypy.The subject of the invention is a method for preparing an immunosensor for detecting influenza virus, where clean gold electrodes are rinsed with ethanol and placed in a 10 mM ethanol solution of 1,6-hexanedithiol (HDT) for 20 h at room temperature. Then, the electrodes are rinsed successively with ethanol and deionized water, turned upside down and 10 µl of colloidal gold solution (GCP) is dropped for 18 h at +4°C. In the next step, the electrodes are rinsed with 0.1 M PBS buffer (145 mM NaCl, 2.7 mM KCl, 8.4 mM NaH2PO4, 1.6 mM Na2HPO4) and 10 µl of anti-M antibody solution with a concentration of 28 µg/ml in 0.1 M PBS buffer is added dropwise for 20 h at +4°C. In the last stage, 10 µl of a solution of 0.5% (w/v) BSA in 0.1 M PBS is dropped onto the electrode surfaces for 2 h at +4°C, the electrodes prepared in this way are rinsed with PBS buffer and stored at +4°C until use ( no longer than one day). Furthermore, the use of an immunosensor for detecting M protein and influenza virus in a solution is disclosed.
