Abstract

Przedmiotem wynalazku jest sposób przygotowania roztworu chitozanu o masie cząsteczkowej nie mniejszej niż 10 kDa oraz o stopniu deacetylacji nie mniejszej niż 20%, charakteryzujący się tym, że przygotowuje się roztwór chitozanu mikrokrystalicznego a następnie uzupełnia wodą w ilości, w której stężenie chitozanu względem jego suchej masy w wodnym roztworze wynosi nie mniej niż 0,001% wagowego. Tak otrzymany roztwór miesza się w temperaturze nie niższej niż 0°C, korzystnie w warunkach ciśnienia atmosferycznego. W trakcie mieszania roztwór nasyca się kwasowymi tlenkami nieorganicznymi, korzystnie ditlenkiem węgla w postaci gazowej i/lub suchego lodu do momentu uzyskania klarowności roztworu oraz rozpuszczenia chitozanu. Przedmiotem wynalazku jest też kompozycja, charakteryzująca się tym, że otrzymana jest poprzez zmieszanie roztworu chitozanu w wodzie wytworzonego powyższym sposobem w ilości nie mniejszej niż 80% wagowych z co najmniej jednym związkiem z grupy związków biologicznie czynnych i/lub związków sieciujących i/lub plastyfikatorów, w ilości nie większej niż 20% wagowych. Kompozycja korzystnie ma postać aerozolu. Wynalazek obejmuje również sposób wytwarzania hydrożelowej membrany chitozanowej, charakteryzujący się tym, że roztwór chitozanu w wodzie, otrzymany sposobem jak wyżej wylewa się do formy, a następnie kondycjonuje się w temperaturze nie niższej niż 1°C, w warunkach ciśnienia korzystnie atmosferycznego, w czasie nie krótszym niż 12 h. Przed wylaniem do formy i kondycjonowaniem do roztworu chitozanu w wodzie dodaje się co najmniej jeden związek z grupy związków biologicznie czynnych i/lub związków sieciujących i/lub plastyfikatorów, w ilości nie większej niż 20% wagowych i miesza się do czasu uzyskania homogenności.The subject of the invention is a method of preparing a chitosan solution with a molecular weight of not less than 10 kDa and a degree of deacetylation of not less than 20%, characterized in that a solution of microcrystalline chitosan is prepared and then supplemented with water in an amount in which the concentration of chitosan relative to its dry mass in an aqueous solution is not less than 0.001% by weight. The solution thus obtained is stirred at a temperature not lower than 0°C, preferably under atmospheric pressure. During stirring, the solution is saturated with acidic inorganic oxides, preferably carbon dioxide in gaseous form and/or dry ice until the solution is clear and the chitosan is dissolved. The subject of the invention is also a composition, characterized in that it is obtained by mixing a solution of chitosan in water obtained by the above method in an amount of not less than 80% by weight with at least one compound from the group of biologically active compounds and/or cross-linking compounds and/or plasticizers, in an amount of not more than 20% by weight. The composition is preferably in the form of an aerosol. The invention also includes a method for producing a hydrogel chitosan membrane, characterized in that the solution of chitosan in water, obtained by the method as above, is poured into a mold and then conditioned at a temperature of not less than 1°C, under conditions of pressure, preferably atmospheric, for not less than 12 h. Before pouring into the mold and conditioning, at least one compound from the group of biologically active compounds and/or cross-linking compounds and/or plasticizers, in an amount of not more than 20% by weight, is added to the solution of chitosan in water and mixed until homogeneity is achieved.
