Abstract
Wynalazek dotyczy genetycznie zakodowanego, wykorzystującego zjawisko FRET biosensora do monitorowania aktywności metaloproteazy macierzy zewnątrzkomórkowej 9 (MMP-9). MMP-9 jest zewnątrzkomórkową endopeptydazą zaangażowaną zarówno w procesy fizjologiczne i patologiczne. Zakotwiczony w błonie komórkowej, genetycznie zakodowany, wykorzystujący zjawisko FRET, biosensor pozwala na badanie aktywności proteolitycznej MMP-9 z wysoką rozdzielczością czasowo-przestrzenną bezpośrednio w miejscu aktywności MMP-9 na komórce. Funkcjonalność biosensora, zarówno in vitro i in vivo w żywych komórkach została wykazana przy pomocy analizy ratiometrycznej cięcia biosensora przez oczyszczoną auto-aktywującą zmutowaną MMP-9.
