Abstract
Przedmiotem zgłoszenia jest sposób wytwarzania rusztowania komórkowego o dużej elastyczności, w którym prepoli(sebacynian glicerolu) o ciężarze cząsteczkowym Mn 1—20 kg/mol i stopniu estryfikacji 60-80%, rozpuszcza się w mieszaninie rozpuszczalników polarnych i niepolarnych, w stężeniu 10-65% wag. i miesza się do całkowitego rozpuszczenia polimeru, następnie do roztworu dodaje się 20-35% wag. alkoholu C1-C3 jako ciekłego porofora i miesza się do uzyskania jednorodnego roztworu, po czym roztwór wylewa się na podłoże z mieszaniny NaCl i NaHCO3 o rozmiarach ziaren <500 μm i zamraża do temperatury -10-(-20)°C, a następnie przeprowadza wodną kąpiel płuczącą przez 2-5 h, w temperaturze 0-5°C, w stanie zamrożenia skafoldu, zaś gotowy skafold suszy się metodą liofilizacji.The subject of the application is a method of producing a highly flexible cell scaffold, in which prepols (glycerol sebacate) with a molecular weight of Mn 1-20 kg / mol and a degree of esterification of 60-80% are dissolved in a mixture of polar and non-polar solvents at a concentration of 10-65 wt.% and stirred until the polymer is completely dissolved, then 20-35 wt.% is added to the solution. of C1-C3 alcohol as a liquid blowing agent and mixed until a homogeneous solution is obtained, then the solution is poured onto a substrate made of a mixture of NaCl and NaHCO3 with grain sizes <500 μm and frozen to -10 - (- 20) ° C, and then an aqueous rinsing bath for 2-5 h, at a temperature of 0-5 ° C, when the scaffold is frozen, and the finished scaffold is dried by lyophilization.
