Abstract

Przedmiotem wynalazku jest sposób otrzymywania wstrzykiwalnego biokompatybilnego nośnika leków, komórek lub ich kombinacji, w postaci mikrorusztowań, charakteryzujący się tym, że obejmuje następujące etapy: a) przygotowanie roztworu zawierającego polimer oraz co najmniej jeden rozpuszczalnik; b) formowanie włókien o średnicy włókna wynoszącej od 50 nm do 10 µm na kolektorze płaskim w procesie elektroprzędzenia; c) laserowe cięcie warstwy włókniny uformowanej na kolektorze na pojedyncze oddzielne mikrorusztowania rusztowania o grubości od 0,01 do 0,2 mm oraz o kształcie walca o średnicy podstawy wynoszącej od 0,1 do 0,4 mm albo graniastosłupa o długości boku podstawy wynoszącym od 0,04 do 0,4 mm; d) oddzielanie oddzielnych mikrorusztowań od kolektora; e) chemiczne modyfikowanie mikrorusztowań poprzez ich zawieszenie w wodnym roztworze NaOH o stężeniu 0,001-1M przez 1 - 120 min albo fizyczne modyfikowanie mikrorusztowań poprzez ich zawieszenie w wodnym roztworze białka o stężeniu 0,001-0,5M  przez 2 - 120 min; f) odpłukanie nadmiaru NaOH albo białka z powierzchni mikrorusztowań. Kolejnym przedmiotem wynalazku jest kompozycja do podawania przez wstrzykiwanie zawierająca bufor oraz biokompatybilne wzajemnie nieagregujące in vitro nośniki leków, komórek lub ich kombinacji, charakteryzująca się tym, że nośniki stanowią mikrorusztowania z elektroprzędzonych włókien przygotowane sposobem według wynalazku, gdzie każde mikrorusztowanie stanowi pojedynczy nośnik o grubości od 0,005 do 0,2 mm oraz o kształcie walca o średnicy podstawy wynoszącej od 0,05 do 0,5 mm albo graniastosłupa o długości boku podstawy wynoszącym od 0,05 do 0,5 mm, którego powierzchnia jest zmodyfikowana chemicznie roztworem NaOH lub fizycznie roztworem białka, przy czym mikrorusztowania zawieszone w buforze tworzą zawiesinę nieagregujących mikrocząstek. Kolejnym przedmiotem wynalazku jest zastosowanie kompozycji według wynalazku w leczeniu uszkodzeń kości, chrząstek, krążków międzykręgowych.
