Abstract
Przedmiotem zgłoszenia jest sposób różnicowania łagodnych i złośliwych guzków tarczycy w materiale z biopsji guza, sklasyfikowanej zgodnie z systemem Bethesda, który obejmuje etapy izolacji RNA, oraz określenie poziomu ekspresji zestawu genów, który obejmuje: AMOT, C3, CAMK2N1, CFI, DCSTAMP, DIO1, DUSP5, EGR1, EMP2, FAM20A, FAXC, FLRT3, HBG1, IGFBP6, IGSF1, ITGA2, KCNQ3, MET, METTL7B, MPZL2, NOD1, OLR1, PDE5A, PSD3, SDC4, SLC26A4, SLC26A7, SLC5A8, SLPI, TM4SF1, TPO, TUSC3. Normalizację ekspresji 32 genów tego zestawu prowadzi się pod endogenną kontrolą co najmniej czterech genów normalizujących spośród EIF3A (EIF3S10), HADHA, UBE2D2, EIF5, PPIE oraz RPLPO. Do różnicowania guzków tarczycy wykorzystuje się algorytm klasyfikatora, który nauczony jest na danych ekspresji genów skorelowanych z charakterem guzka tarczycy, zwłaszcza łagodnych guzków tarczycy i nowotworu złośliwego, w tym raka brodawkowatego i pęcherzykowego tarczycy.
