Abstract

Przedmiotem zgłoszenia jest preparat doglebowy (bionawóz) zawierający żywe mikroorganizmy zdolne do wiązania atmosferycznego jaki ma postać liofilizatu nowego szczepu Azotobacter chroococcum KBIMZ6. Zgłoszenie zawiera także sposób wytwarzania preparatu doglebowego (bionawozu), zawierającego żywe mikroorganizmy zdolne do wiązania atmosferycznego, w którym, w kolejnych krokach: 1. Z podłoża z uprawy soi izoluje się bakterie z rodzaju Azotobacter, prowadzi się hodowlę wgłębną, korzystnie kontroluje się zdolność uzyskanych mikroorganizmów do wiązania azotu atmosferycznego i pozyskuje biomasę komórkową; 2. Z pozyskanej biomasy izoluje się DNA, identyfikuje się poprzez analizę porównawczą sekwencji genów 16S rRNA izolat KBIMZ 6; 3. Z uzyskanego izolatu KBIMZ 6 hoduje się biomasę w hodowlach okresowych; 4. Równolegle z uzyskanego izolatu KBIMZ 6 hoduje się biomasę w hodowlach okresowych z zasilaniem; 5. Liofilizuje się biomasę z płynu pohodowlanego uzyskanego w hodowlach okresowych lub hodowlach okresowych z zasilaniem.The subject of the application is a soil preparation (biofertilizer) containing living microorganisms capable of atmospheric binding in the form of a lyophilisate of a new strain of Azotobacter chroococcum KBIMZ6. The application also includes a method for producing a soil preparation (biofertilizer) containing living microorganisms capable of atmospheric binding, in which, in the following steps: 1. Bacteria of the genus Azotobacter are isolated from a soybean cultivation substrate, a submerged culture is carried out, the ability of the obtained microorganisms to bind atmospheric nitrogen is preferably controlled, and the cellular biomass is obtained; 2. DNA is isolated from the obtained biomass, the KBIMZ 6 isolate is identified by comparative analysis of the 16S rRNA gene sequences; 3. The biomass is grown from the obtained KBIMZ 6 isolate in batch cultures; 4. In parallel, biomass is cultivated from the obtained KBIMZ 6 isolate in fed-batch cultures; 5. Biomass is freeze-dried from the post-culture fluid obtained in batch cultures or fed-batch cultures.
