Abstract

W niniejszym zgłoszeniu opisane zostały cząsteczki wykazujące dwie specyficzności: pierwszą, wynikającą ze specyficznego wiązania cząsteczki do receptora indukującego internalizację i/lub ukierunkowaną degradację lizosomalną oraz drugą, wynikającą ze specyficznego wiązania do celu molekularnego, transbłonowego lub zewnątrzkomórkowego. Fragment cząsteczki determinujący wiązanie do receptora indukującego internalizację i/lub ukierunkowaną degradacje lizosomalną jest polipeptydem lub domeną białkową, które mogą być połączone wiązaniem peptydowym za N-końcem Fc łańcucha ciężkiego przeciwciała, C-końcem łańcucha ciężkiego przeciwciała lub wstawione w łańcuch polipeptydowy we fragmencie łańcucha ciężkiego lub/i łańcucha lekkiego wiążącym antygen. Fragment cząsteczki warunkujący drugą specyficzność może być łańcuchem polipeptydowym (np. przeciwciało, fragment przeciwciała, receptor, fragment receptora, antygen, fragment antygenu, białko fuzyjne) lub związkiem małocząsteczkowym. Obie części mogą być powiązane linkerem. Opisane cząsteczki znajdują zastosowanie do indukowania internalizacji i ukierunkowanej, lizosomalnej degradacji receptorów transbłonowych oraz białek zewnątrzkomórkowych. Ujawnione zostały także metody wytwarzania bispecyficznych cząsteczek oraz ich zastosowanie. Podane są sekwencje opracowanych peptydów specyficznie wiążących IGF2R, które jednocześnie nie wykazują powinowactwa do IGF1R. Przykłady wykonania obejmują zastosowanie ujawnianych polipeptydów jako części odpowiadających za indukowanie internalizacji i kierowanie do degradacji lizosomalnej w rekombinowanych, bi- lub multispecyficznych cząsteczkach.
