Abstract
Inventia se refera la un procedeu de obtinere a fosfatazei alcaline, enzima utilizata în stabilirea diagnosticului în cazul bolilor cu etiologie virala, bacteriana si parazitara cat si pentru identificarea si determinarea cantitativa a o serie de subsatante active biologic. Precipitatul obtinut de la purificarea fractiunilor II si III Chon din procedeul de obtinere a gamaglobulinei se dizolva în apa distilata se trateaza cu alcool butilic în proportie de 10% dupa îndepartarea proteinelor de balast fosfataza alcalina este precipitata cu sulfat de amoniu dupa care se îndeparteaza omauritatile prin absorbtie pe gel de hidroxid de aluminiu 2% în raport de 10 ml gel la 100 ml solutie ce contine fosfataza alcalina pe gel de aluminiu ce se adauga în proportie de 25 ml gel la 100 ml solutie proteica, dupa centrifugare se elueaza cu solutie de clorura de sodiu IM, solutie clorura de sodiu 2M, solutie clorura de sodiu 2M ce contine 2,5% acid glutamic, fosfataza alcalina desorbitade ultimile doua solutii se dializeaza în apa, se concentreaza în vid la o concentratie proteuca de 10mg/ml, se dializeaza apoi în solutie tampon fosfat alcalin, se filtreaza steril si se conditioneaza prin liofilizare sau stabilizare cu sulfat de amoniu sau glicerina.The invention relates to a process for obtaining alkaline phosphatase, the enzyme used in the diagnosis of diseases with viral, bacterial and parasitic etiology, and for the identification and quantitative determination of a series of biologically active sub-active substances.
