Abstract
Amilaz konsantrasyonlarinin belirlenmesi için, asagidaki kademeleri içeren bir yöntem saglanmaktadir: (a) bilinmeyen bir amilaz konsantrasyonuna sahip olan bir numunenin bir bölümünün belli bir sicaklikta 20-200 mg/litre arasindaki bir konsantrasyonda nisasta içeren bir su ile karisabilen tamponlu nisasta çözeltisi içinde kuluçkalanmasi; (b) bu sekilde elde edilen karisimin belli bir süre ile tepkimeye birakilmasi; (c) bir sulu iyodin çözeltisinin bir bölümünün birinci numuneyi elde etmek için bu karisima eklenmesi; (d) numunenin baska bir bölümünün iyodin çözeltisinin ikinci bölümünü ve nisasta çözeltisinin ikinci bölümünü içeren bir karisima eklenmesi ile ikinci numunenin hazirlanmasi; (e) her iki numunenin optik yogunlugunun veya remisyonunun 300-800 nm arasindaki bir dalga boyunda ölçülmesi; ve (f) bir kalibrasyon egrisi kullanilarak numunedeki amilaz aktivitesinin hesaplanmasi; burada birinci ve ikinci numune esit numune ve nisasta çözeltisi konsantrasyonuna sahiptir.
