TARIFNAME FAVIPIRAVIR IÇEREN TABLET OLUSTURMA Teknik Alan Mevcut bulus farmasötik teknoloji alaninda , virüs enfeksiyonlarinin tedavisinde kullanilan Favipiravir içeren tablet olusturma ile ilgilidir. Önceki Teknik Favipiravir, (T-705; 6-floro-3-hidroksi-2-pirazinkarboksamid) Toyama Chemical Co Ltd. tarafindan RNA virüslerine karsi gelistirilen bir antiviral ilaçtir. Favipiravir, molekül sekli asagida verilmistir. COVID-19 (SARS-CoV2 Enfeksiyonu) virüsünün dünya üzerinde yayilmasindan sonra virüs enfeksiyonlarinin tedavisinde Favipiravir etkin maddeli ilaçlar önem kazanmistir. RNA virüslerine karsi gelistirilen Toyama Chemical firmasina ait EP240716681 bulusta Favipiravirin tablet kütlesinin% 50 ila 95'ini olusturdugu belirtilmistir. Yine günümüzde Favipiravir etkin maddesinin kullanildigi çesitli buluslar bulunmaktadir. CN111349049A nolu bulus favipiravirin bir sentez süreci ve bunun bir ara maddesi ile CN111592499A nolu bulus favipiravirin bir hazirlama yöntemi ile ilgilidir Bulusun amaci COVID-19 (SARS-CoVZ Enfeksiyonu) ve benzeri virüs enfeksiyonlarinin tedavisinde kullanilan Favipiravir etkin maddeli ilacin dissolüsyon profilini etkilemeden ( çözünme sorunu olusmadan ) Favipiravir konsantrasyonu düsürülmesi ve antiviral ilacin gelistirilmesidir. Bulusun detayli olarak açiklanmasi Mevcut bulusta, tablet agirligi arttirilarak Favipiravir konsantrasyonu düsürülmüstür ve bu konsantrasyon degisikliginin ilaç etkin maddesinin çözünme profilini dengeleyen optimum Krospovidon ve Povidon K30 miktarlari belirlenerek ürün gelistirilmistir. Deneme 12Kuru karisim uygulamasi. Favipiravir 47,06 Povidon K30 6,59 Hidroksi Propil selüloz, L-HPC 21 23,53 Krospovidon 19,06 Aerosil 200 1,88 Sodyum Stearil Fumarate 1,88 Sonuç: Toz akiskanligi kötü geldi ve tablet baski gerçeklestirilemedi. Deneme 2: Toz akisini iyilestirme amaçli kompaktör ile üretim yapildi. Favipiravir 47,06 Povidon K30 6,59 Hidroksi Propil selüloz, L-HPC 21 23,53 Krospovidon 19,06 Aerosil 200 1,88 Sodyum Stearil Fumarate 1,88 Sonuç: Toz akisi iyi olmamistir. Bu yüzden tablet baski yapilamamistir. Deneme 3: Toz akisini iyilestirmek için yas granülasyon çalismasi yapildi. Favipiravir 47,06 Povidon K30 6,59 Hidroksi Propil selüloz, L-HPC 21 23,53 Krospovidon 19,06 Aerosil 200 1,88 Sodyum Stearil Fumarate 1,88 Sonuç: Toz akisi iyilestirildi tablet baski gerçeklestirildi. Fakat çözünürlügü düsük gelmistir. Deneme 4: Çözünürlügü arttirmak. Favipiravir 47,06 Povidon K30 3,29 Hidroksi Propil selüloz, L-HPC 21 23,53 Krospovidon 22,35 Aerosil 200 1,88 Sodyum Stearil Fumarate 1,88 Sonuççözünürlük referans ürün ile yakin geldi TR TR TR TR TR TR
