本発明は、 試料、 殊にヒ ト検体中のヒト · トロンボモジユリンの免疫 学的測定に関する。 更に詳しくは試料中に微量存在するヒト · トロンボ ジュリンを免疫学的に高感度で、 正確に且つ容易に測定する方法、 その ための試薬及びそのためのキッ卜に関する。
b. 従来技術
トロンボモジユリンは、 1981年にシ一 'ティー .エスモンらによ つて、 ゥサギ肺の血管内皮細胞からトロンビンによるプロテイン Cの活 性化を著しく促進するコファクター蛋白質として発見され [ T. Esmon, W. G. Owen: Proc. Natl. Acad. Sci. USA, 78, 2249〜 22
52 (1981) ] 、 翌年精製された [N. L. Esmon, f. G. Owen, C. T. Esmon: J. Biol. Chem. , 257, 859〜 864 (1982) ] 。 そ の後ヒト · トロンボモジュリンも肺および胎盤から精製され [S. Kuros awa, et al. : Thromb. Res. 3 _, 353〜 364 (1985) など] 、 ヒ ト胎盤からのトロンボモジユリンは、 分子量が還元剤の存在下 105, 000、 非存在下 75, 000である糖蛋白質であることが報告された。 トロンボモジュリンは、 動脈、 静脈、 毛細血管、 リンパ管の内皮細胞の 細胞膜上にある膜.翠白質で、 脳をのぞいたほとんどの臓器中に存在する が、 特に肺と胎盤に多く存在することが知られている。 'またその生理作 用は、 脈管の内皮細胞上でトロンビンの血液凝固促進作用を凝固阻害作 用へと転換する生理活性物質であることが知られている。 すなわちトロ ンボモジュリンは細胞膜上でトロンビンと 1 : 1の複合体を形成するこ とにより トロンビンはその凝固因子としての性質を失うとともにプロテ イン C活性促進作用を発現し、 この活性化プロテイン Cを介して血液凝 固因子中の活性型第 V因子、 活性型第 VI因子を不活性化することにより、 凝固阻害作用を示すことが知られている。 更にトロンボモジユリンは活 性型第 X因子に直接結合してその活性を阻害することによりプロトロン ビンからのトロンビン生成を抑制することも知られている。
以上述べた如き生理作用を有するトロンボモジュリンは、 血液凝固 · 線溶系の新たな調節因子として注目されている。 したがって、 溶液中の 微量なトロンボモジユリンを容易かつ正確に測定することは、 循環器系 の基礎医学、 臨床医学の研究において果たす役割は非常に大きい。
殊にヒト検体中に微量存在するヒト · トロンボモジユリンを高感度で 測定することは、 血管壁の障害を伴なう各種の疾患の病態をモニターで きる可能性を有している。
近時、 試料中、 殊にヒト検体中の微量成分を測定する方法として抗原 抗体反応を利用する免疫学的測定方法が用いられている。 この測定方法 を診断薬として医療現場での多数検体の測定などに用いる場合、 主な必 要条件として次の点が挙げられる。 すなわち検体中の微量成分を測定し える感度があること、 測定対象のみを測定する特異性があること、 再現 性よく測定値が得られること、 かつ測定操作が容易、 簡便などである。 上記した観点から従来知られているヒト · トロンボモジュリンの測定 手段を見る時必ずしも充分満足しえないのが現状である。 すなわち、 ェ ツチ ·ィシィらの放射免疫測定法(radio關 unoassay、 以下 R I Aと略) の報告(H. Ishii et al. : J. Clin. Invest. , 7 6 , 2 1 7 8 ( 1 9 8 5 ) , H. Ishii et al. , Blood 6J7 , 3 6 2 ( 1 9 8 6 ) ) がなされ ているが、 この報告では、 ヒ ト ' トロンボモジュリンに対するポリクロ ーナル抗体と1 2 5 I― トロンボモジユリンを用いた競争法が記載されて いるが、 その感度は 1〜2 . S ngZn^程度であり、 また測定時間は 1 9 時間以上必要とされることから、 この測定方法は微量のトロンボモジュ リンを正確にかつ簡便に測定するには不満足な方法であった。 また抗体 としてポリクローナル抗体のみを用いるため特異性の点でも危険性が考 えられる。
その後、 サンドイッチ法による免疫学的測定方法が報告された。 西岡 らは、 ヒ ト肺から精製したトロンボモジュリンを抗原として得たポリク ローナル抗体を用いた方法を報告している [西岡淳二ら、 第 1 0回日本 血栓止血学会要旨集、 5 1頁、 (1 9 8 7 ) ] 。 この方法では抗体とし てポリクローナル抗体のみを用いていることから前記方法と同様、 ポリ クローナル抗体の特異性の点で問題が予想される。 更に特開昭 6 4 - 4 5 3 9 8及び特開昭 6 4— 4 7 3 9 1号広報が開示されている。 ここで は 2種のモノクローナル抗体のみを用いたサンドィッ ト法が示されてい るが、 本発明者らの検討によれば、 必ずしもモノクローナル抗体のみを 使ったサンドィツチ法によるヒ ト · トロンボモジュリンの測定は実際の 医療現場で使用する測定方法としては充分満足すべきものではなかった。 すなわち、 試料中の微量なヒ ト · トロンボモジュリンを測定するに必要 な感度を短時間にえることが出来なかった。
c 発明が解決使用とする課題 そこで本発明者らは、 前述した従来技術の問題点を解決し、 診断薬と して必要条件を備えた免疫学的測定方法、 試薬及びキットを提供するた め研究を進めた。
本発明の第 1の目的は、 試料中、 殊にヒト検体中の微量のヒト · トロ ンボモジユリンを高感度で測定することができる免疫測定方法、 試薬及 びキットを提供することにある。
本発明の第 2の目的は、 試料中のヒト · トロンボモジユリンを実際の 医療、 殊に臨床の場において、 使用しうる簡便で且つ短時間で測定する ことができる免疫学的測定方法、 試薬及びキットを提供することにある。 本発明の他の目的は試料中のヒト · トロンボモジュリンを、 測定する 人や測定条件 (時間、 温度) の影響を受けるので、 また使用する抗体の 特異性に出来る限り影響されることなく、 安定して高感度で測定するこ とができる免疫学的測定方法、 試薬及びキットを提供することにある。 本発明のさらに他の目的は、 以下の説明から一層明らかとなるであろ う o
d . 課題を解決するための手段
かく して本発明者らの研究によれば、 前記本発明の目的及び利点は、 下記ヒ ト · トロンボモジユリンの免疫学的測定方法、 免疫学的測定試薬 及び免疫学的測定キットによって達成されることが見出された。
[ I ]ヒ ト ' トロンボモジュリンの免疫学的測定方法;
試料中のヒト · トロンボモジユリンをサンドイッチ法により免疫学的 に測定する方法において、 不溶性担体に固定化した抗体 (第 1抗体) と 標識化した抗体 (第 2抗体) は、 いずれか一方がヒト · トロンボモジュ リンを認識するポリクローナル抗体であり、 他方がヒト · トロンボモジ ユリンを認識するモノクローナル抗体であるヒ ト ' トロンボモジユリン の免疫学的測定方法。

以下本発明について更に詳細に説明する。 [A— 1 ] 抗原の調製及び精製
本発明に使用されるポリクローナル抗体およびモノクローナル抗体を 得るための抗原としてのヒ ト · トロンボモジユリンは、 原則としては天 然の材料から抽出した天然型のものが用いられるが、 天然型のヒ ト · ト ロンボモジユリンと同等の免疫学的性質をもつものであれば蛋白工学的 又は遺伝子工学的手法によって得られるものでもよい。 その例として、 ハプテンすなわち天然型のヒ ト · トロンボモジユリンの抗原決定部位を 含むペプチドであってもよい。 更に、 ヒ ト · トロンボモジュリンを蛋白 分解酵素、 例えばエラスターゼ、 トリプシンなどによって得られるフラ グメントであってもよい。
天然型のヒ ト · トロンボモジユリンを得るための材料としては、 例え ばヒ ト肺、 ヒ ト血管あるいは培養ヒ ト ·血管内皮細胞とその培養上清な どが挙げられるが入手が比較的容易な点から好ましくはヒ ト胎盤である。 分離 ·精製は、 通常用いられる蛋白分離技術、 例えば塩析、 抽出、 遠心 分離、 限外濾過、 各種のクロマトグラフィー等を組み合せて行うことが できるが、 前記したエス · クロサヮらの文献記載の方法は、 その 1例で のる。
かく して得られた精製ヒ ト · トロンボモジユリンを抗原として使用し、 以下に説明する方法によりポリクロ一ナル抗体及びモノクローナル抗体 を調製することができる。
[A - 2 ] ヒ ト · トロンボモジュリンを認識するポリクローナル抗体の 調製:
本発明において使用されるヒ ト · トロンボモジユリンを認識するポリ クローナル抗体は、 前記の如く して得られたヒ ト · トロンボモジユリン 或いはそのフラグメントを抗原として、 それ自体知られた方法に従って 得ることができる。
例えば 「日本生化学会編、 続生化学実験講座、 5卷、 1一 1 0頁、 東 京化学同人、 1 9 8 6」 記載の方法によって得られる。
免疫動物としては、 哺乳動物であれば特に限定されるものではないが、 例えばヒ ト · トロンボモジユリンを抗原とする場合、 山羊、 ゥサギ、 モ ルモッテ、 ラッ トあるいはマウスなどが好んで挙げられる。
免疫した後、 採血し、 抗血清を得る。 得られた抗血清を通常使用され る方法、 例えば塩析、 抽出、 遠心分離、 限外濾過、 種々のクロマトグラ フィ一などを組合せて精製されたポリクローナル抗体を得ることができ る
[A— 3 ] ヒ ト · トロンボモジュリンを認識するモノクローナル抗体の 調製;
本発明において使用されるヒ ト · トロンボモジユリンを認識するモノ クローナル抗体は、 前記ヒ ト · トロンボモジュリン或いはそのフラグメ ントを抗原として使用し、 それ自体知られたケーラーとミルシュタイン による細胞融合法 (G. KOkler and Milstein, Nature(London), 2 5 6, 4 9 5 - 4 9 7 ( 1 9 7 5 ) ) により作製されたハイプリ ドーマを培養 して分泌させ、 その培養液から分離することにより調製される。 すなわ ち、 ヒ ト ' トロンボモジュリンでマウスを免疫した後、 このマウスのリ ンパ球をマウス · ミエ口一マ細胞と融合させハイプリ ドーマを作製する。 このようにして得たハイプリ ドーマは、 融合された種々のリンパ球のそ れぞれに応じて種々のモノクローナル抗体を産生するので、 目的とする モノクローナル抗体を産生するハイブリ ドーマをクロ一ニングによって クローン化されたハイプリ ドーマとして単離する。 このクローン化ハイ プリ ドーマをィン · ビトロで培養してモノクローナル抗体を分泌させる。 この培養上清から抗ヒ ト · トロンボモジユリン 'モノクローナル抗体を 分離する。
5 本発明において、 モノクローナル抗体としては、 ヒ ト · トロンボモジ ユリンとトロンビンとの複合体形成に対する阻害作用を有し且つヒ ト · プロテイン Cの活性化に阻害作用を有するものを使用するのが好ましい。 このようなモノクローナル抗体は公知であり、 例えば、 I. Maruyama et al. : J. Biol. Chem., 2 6 0_, 1 5 4 3 2 ( 1 9 8 5 ) 記載のモノク口 i o ーナル抗体特開昭 6 4 - 4 5 3 9 8号公報記載の TM— A 7 3と表示さ れたモノクローナル抗体がある。
前記した阻害作用を有するモノクローナル抗体を使用することが好ま しい理由は、 試料中のヒ ト · トロンボモジユリンとトロンビンとの複合 体又はそのフラグメントを認識せず、 遊離のヒ ト · トロンボモジユリン
1 5 又はそのフラグメントを直接認識し測定できるからである。
[ B ] 第 1抗体及び第 2抗体の調製;
本発明においては、 上記ポリクローナル抗体、 モノクローナル抗体を 不溶性担体に固定化した抗体 (第 1抗体) 或いは標識化した抗体 (第 2 抗体) に用いるが、 その組合せは一方をポリクローナル抗体とし、 他方 2 0 をモノクローナル抗体として用いる。 この組合せにより、 ヒ ト ' トロン ボモジユリンの免疫学的測定方法においてポリクローナル抗体の持つ高 い親和性とモノクローナル抗体の持つ高い特異性を同時にに発現させる ことが可能である。 その結果、 感度、 特異性、 再現性に優れた簡便な免 疫学的測定方法を得ることができる。 しかしながら、 本発明者らの研究によれば、 固定化した抗体 (第 1抗 体) 力、'、 ヒ 卜 · トロンボモジュリンを認識するポリクロ一ナル抗体であ り、 標識化した抗体 (第 2抗体) がヒ ト · 卜ロンボモジユリンを認識す るモノクローナル抗体である組合せが、 特に好ましい結果が得られるこ とがわかった。
[ B— 1 ] 固定化した抗体の調製
不溶性 (固体) 担体に固定化された抗ヒ ト · トロンボモジユリン抗体 (第 1抗体) は以下のようにして得ることができる。
不溶性固体担体としては、 天然から得られる重合体とその誘導体、 合 成重合体とその誘導体を挙げることが出来る。 前者には、 多糖類とその 誘導体、 例えばセルロース、 セフアデックス、 セファロ一ス、 カルボキ シメチルセルロース、 ニトロセルロース、 酢酸セルロース、 デキストラ ンなど、 あるいはガラス、 シリカゲルなどの無機重合体などがある。 ま た後者にはビニル系重合体、 例えばポリスチレン、 ポリエチレン、 ポリ プロピレン、 A B S、 ポリフッ化ビニル、 ポリアミ ンーメチルビニルェ 一テル—マレイン酸共重合体、 エチレン—マレイン酸共重合体など;縮 合系重合体、 例えば 6—ナイロン、 6 , 6—ナイ口ンなどのポリアミ ド、 ポリエチレン ·テレフタレートなどのポリエステル、 ァミノ酸重合体な どがある。 また、 その形状は試験管、 マイクロタイタ一プレート、 ビー ズあるいはメンブレンなど特に限定されない。 かくなる不溶性固体担体 に固定される抗体としては、 前述の如き抗ヒ ト · トロンボモジユリン抗 体の抗体分子、 抗原結合能が失われないそのフラグメント例えば
F (ab' ) 2、 F ab F ab或いは F acbなど; あるいは抗原結合能が失われ ない抗体分子またそのフラグメン卜の誘導体である。 これらの抗体を不 溶性固体担体へ固定する方法は、 物理的吸着法、 例えばポリスチレンの 固体担体を抗ヒ ト, トロンボモジユリン抗体の溶液に浸漬する方法など; イオン結合法、 例えばイオン交換樹脂あるいはアミノ基、 カルボン酸基、 スルホン酸基、 リン酸基などのイオン化する官能基を有する固体担体を 用いる方法など;あるいは化学反応による共有結合法、 例えばカルボキ シ · クロライ ド法、 カルボジィミ ド法、 無水マレイン酸誘導体法、 イソ シァネート誘導体法、 臭化シアン活性化多糖法、 ジァゾ法、 活性エステ ル法、 架橋試薬による担体結合法 (架橋試薬としてグルタールアルデヒ ド、 へキサメチレンイソシァネート、 コハク酸イミ ド、 マレイミ ド化合 物など) など;更には、 ヒト · トロンボモジュリ ンに対しては結合能は ないが抗ヒト · トロンボモジユリン抗体に対し生物学的反応により結合 し得る物質を介して結合する方法、 例えばプロティン A結合固体担体を 用いる方法などである。
さらに本発明者らの研究によれば、 不溶性固体担体としてその表面が 鏡面化されたもの、 すなわち表面が極めて平滑なものを使用することに より、 非特異的吸着が低くなり、 ヒト · トロンボモジュリ ンの測定感度 が向上することがわかった。
従来、 高感度な測定のための不溶性固体担体としては、 むしろその表 面を研磨して粗くし、 表面積を大きくしたものが用いられてきた。 確か に表面積を大きくすると、 抗体の固定量を増加させるというメリットが ある力、 しかしながら非特異的吸着も大きくなるというデメリツトがぁ つた。 本発明者らの研究したところによれば、 ヒト · トロンボモジユリ ンの測定においては、 中心線平均粗さ (R a ) が 1 . 5 m以下である 鏡面化された固体担体は、 抗体の固定量は粗面化された固体担体とほぼ 同等であるが、 非特異的吸着は飛躍的に減少し、 ヒ ト · トロンボモジュ リンの高感度測定に遊離であることを見出した。 かかる鏡面化された不 溶性固体としては、 その材質形状は特に制限されているが例えばポリス チレンビーズ、 ガラスビーズがあげられる。
5 中心線平均粗さ (Ra) は、 粗さ曲線からその中心線の方向に測定長 さ^の部分を抜取り部分の中心線を X軸、 縦倍率の方向を Y軸とし、 あ らさ曲線を y = f (X) で表わしたとき、 次式で与えられる R aの値を ミクロン単位で表わした値を意味する。
10 Ra J f (x)/dx この中心線粗さ (R a) については、 J I S B 0601-198 2 (日本) く ANS I B 46.1— 1979 (US A) 及び R468 - 1966 (I SO) に説明されている。
なお以下の本発明の実施例では、 不溶性担体は東京精密 (株) 製の表 i s 面粗さ計サーフコム ®(Surfcom570 A) を用いて表面粗さを測定した。
[B-2] 標識化した抗体の調製:
本発明において標識化した抗ヒ ト · トロンボモジユリン抗体 (第 2抗 体) は、 以下のようにして得ることができる。
標識化すべき抗体としては、 抗体全分子のみならず、 抗原結合能が失 0 われないそのフラグメント例えば Fab'、 ?(3 )2又は 3(:1)フラグメン トなどを使用することができる。
かかるフラグメントは、 F(ab')2や Fab'の場合には、 上述のように して得られたポリクロ一ナル抗体またはモノクローナル抗体を公知の方 法、 たとえば該抗体をペプシンで分解して F (at )2フラグメントとなすか、 更に
F(ab')2フラグメントを還元処理することによって Fab'フラグメント となすことにより得られる(H. Nisonoff et al. , Arch. Biochem. Bio phys., 89, 230 (1960) : P. Parham. , J. Immunol. , 131, 2895 ( 1983 ) など) 。
標識化に使用される標識物質としては、 ラジオァイソ トープが良く知 られているが、 工業的な生産を考えると、 公害に結びつく可能性がある 点、 使用期限がある点、 また使用者の特殊訓練が必要な点で、 これらの 欠点がなくかつ増巾作用のある酵素が適している。
またかかる抗ヒ ト · トロンボモジユリン抗体と結合させる酵素として は、 例えばリゾチーム、 マレート ·デヒ ドロゲナーゼ、 グルコース一 6 —フォスフエ一ト ·デヒ ドロゲナーゼ、 パーォキシダーゼ、 グルコース · ォキシダーゼ、 アルカリフォスファターゼ、 ルシフェラ一ゼ、 3—ガラ ク トシダーゼ、 アルコール ·デヒ ドロゲナーゼ、 インベルターゼなどを 例示することが出来る。 これら酵素と前記抗ヒ ト · トロンボモジュリン 抗体との結合抗体の結合方法は、 グルタルアルデヒ ド法、 過ョーソ酸法、 マレイミ ド法など通常の方法に従うことが出来る。 例えばマレイミ ド化 された抗体又は抗体のフラグメン卜と SH化された酵素を溶液中で反応 することにより行うことができる。 抗体又は抗体のフラグメン卜のマレ イミ ド化は、 例えばサクシンィミジル 4— (N—マレイミ ドメチル) シ クロへキサンカーボネート (SMCC) 、 スルホサクシンィミジル 4— (N—マレイミ ドメチル) シクロへキサンカーボネート (スルホ SMC C) 、 サクシンィミジル一メタマレイミ ドベンゾエート (MB S) 、 サ クシンィミジル 6—マレイミ ドへキサノエ一ト (EMCS) などにより マレイミ ド化することができる。 酵素への SH基の導入は公知の方法(例 えば石川編 「酵素免疫測定法」 医学書院参照) に従うことができる。 例 えば酵素と S—ァセチルメルカプト無水コハク酸 (AMSA) 又は N— サクシンイミジル一 3— (2—ピリジルチオ) プロピオネート (SPD P) などと反応することにより得られる。
かく して得られた酵素と抗ヒ ト · トロンボモジユリン抗体との結合抗 体 (第 2抗体) 、 すなわち酵素標識化抗体における酵素の標識数は、 分 子量の測定や吸光度、 酵素活性などの測定などにより求めることができ る。 たとえば、 酵素がペルォキシダーゼの場合には、 抗体とペルォキシ ダーゼに由来する 28 Onmの吸光度と、 ペルォキシダーゼに由来する 4 03 nmの吸光度を測定することにより標識数を求めることができる(E. Ishikawa et al. , J. Immunoassay., _4, 209— 327, (1983)、 p 243参照) 。
すなわち 403nroでは抗体の吸光度はなく、 ペルォキシダーゼのみ由 来の吸光どである故に、 該吸光度からペルォキシダーゼの濃度を算出す る。 280nmでは抗体、 ペルォキシダーゼ両者由来の吸光度であること から、 抗体の濃度は測定された 28 Onmの吸光度から、 403nmの吸光 度によって算出されたペルォキシダーゼの濃度と 28 Onmでのペルォキ シダーゼの分子吸光係数から算出されるペルォキシダーゼの 28 Onmの 吸光度の寄与分を引いた値と、 28 Onでの抗体の分子吸光係数から算 出する。 かく して得られたペルォキシダーゼと抗体の濃度から抗体への ペルォキシダーゼの標識数を求めることが出来る。
かく して本発明の標識した抗ヒト · トロンボモジユリン抗体の標識数 を求め、 測定感度との相関を研究した結果、 抗体全分子、 Fab'、 ?(3 )2又は 3 フラグメントに酵素が、 抗体 1分子当り平均 1.0分 子以上結合のものであればよいが、 かなでも 1.5分子以上結合したも の (多標識化抗体) がより好ましい結果を生ずることが明らかとなった。 試料中のヒト · トロンボモジュリンを測定するに充分な測定感度を得る ためには、 該多標識化抗体は酵素が抗体全分子、 Fal 、 F(al )2又は Facbフラグメント 1分子当り平均 1.5分子以上結合したものがよく、 より好ましくは 2分子以上である。
かかる多標識化抗体は、 たとえば前記マレイミ ド化抗体又は抗体フラ グメントと SH化された酵素とを反応させることによって製造される。 該反応条件としては、 たとえば該抗体と該酵素とのモル比 1 : 1以上、 好ましくは 1 : 3以上、 反応温度 4〜40°C、 好ましくは 4〜30°C、 反応 pH5.5〜7.5、 反応時間 5〜48時間の条件に従うことができ るが、 本発明の多標識化抗体がえられれば前記反応条件に限定されるも のではない。 この様にして得られた多標識化抗体は例えば、 ゲルクロマ トグラフィーを用いて反応液より分離精製することができる。
かくして得られた本発明の酵素で標識した抗体は、 試料中の微量のヒ ト · トロンボモジュリンを測定するに必要な測定感度で正しく測定する ことが出来る。
[C] 免疫学的測定方法:
本発明の測定方法における試料、 殊にヒト検体としては、 通常の臨床 サンプル、 例えば血清あるいは血漿形態の血液、 関節液、 リンパ液、 胸 腺水、 腹水、 羊水、 細胞組織液、 骨髄液、 尿などのヒト · トロンボモジ ユリンを含有する液体であればいずれであってもよい。 好ましいのは血 清または血漿形態の血液である。 本発明の免疫学的測定方法においては、 前記第 1抗体及び第 2抗体を 組合せて使用し、 いわゆるサンドィツチ法により、 試料中のヒ ト · トロ ンボモジユリンを測定する。
サンドイッチ法としては大きく 1ステップサンドイッチ法と 2ステツ プサンドイッチ法とがある。 1ステップサンドイッチ法は、 測定しょう とするヒ ト · トロンボモジユリンを含む試料、 不溶性担体に固定化した 抗体 (第 1抗体) 及び標識化した抗体 (第 2抗体) を同一反応系で抗原 · 抗体反応を行なわせ、 固定化抗体 ·抗原 ·標識化抗体の複合体を形成せ しめ、 洗浄操作の後、 標識物質の量を測定する方法である。 この場合、 試料 (検体) と固定化抗体と標識化抗体とを同時に存在させて反応させ てもよいし、 先ず試料と固定化抗体とを反応させてから標識化抗体を添 加して反応させてもよい。 また、 試料と標識化抗体とを予め反応させて から固定化抗体を添加してもよい。 いずれにしても洗浄操作前に固定化 抗体,抗原 ·標識化抗体の複合体を形成しさえすればよい。 一方、 2ス テツプサンドイッチ法は、 試料と固定化抗体とを先ず反応して固定化抗 体 ·抗原の複合体を形成せしめ、 しかる後試料を除去、 洗浄してから標 識化抗体を添加して固定化抗体 ·抗原 ·標識化抗体の複合体を形成せし める。 次いで洗浄操作の後、 標識物質の量を測定する方法である。 ポ リクロ一ナル抗体とモノクローナル抗体を用いる本発明によれば、 1ス テツプサンドイッチ法あるいは 2ステップサンドイッチ法により試料中 における微量なヒ ト · トロンボモジユリンを高い感度、 特異性をもって 再現性よく簡便に測定することが出来る。
上記における免疫反応に用いられる溶媒としては、 反応に悪影響を与 えない通常の各種のものいずれであってもよい。 例えばリン酸緩衝液、 トリス ·塩酸緩衝液、 酢酸緩衝液などの p Hが 6. 0から 8 . 0程度のも のを用いるのが好ましい。
測定に際しての免疫反応温度条件は、 構成要素である蛋白質の性質を 変性させず、 かつ免疫反応を著しく抑制しない限り特に制限はないが、 一般には 5 0 °C以下、 好ましくは約 4 °C〜 4 5 °C程度の温度条件下に約 5分から 5時間程度、 好ましくは約 3 0分から 3時間、 を要して反応を 行えばよい。
[D ] 免疫学的測定キット :
本発明においては、 不溶性固体担体に固定化した抗ヒ ト · トロンボモ ジユリン抗体 (第 1抗体) 、 標識化した抗ヒト · トロンボモジユリン抗 体 (第 2抗体) に、 免疫学的測定に際し必要なその他のものを組み合せ て、 ヒト ' トロンボモジュリンの量を測定するためのキットを構成する ことができる。
本発明の免疫学的測定キットにおいて、 ( i ) 前記第 1抗体及び (ii) 第 2抗体と共に組合される他の試剤としては、 下記の (iii) 〜 (vi) のものが挙げられる。
(iii) 酵素で標識化した場合には、 酵素活性を測定するための基 質および反応停止剤
(iv) 希釈剤
( V ) 洗浄剤及び
(vi) 標準物質
以下これらについて説明するが、 これらは通常免疫学的測定に使用さ れるものであればよい。
酵素活性を測定するための基質および反応停止剤: 本発明の測定キットおよび測定方法において、 標識物質として酵素を 使用した場合に使用される前記基質および反応停止剤は、 標識物質とし ての酵素の種類に対応して、 免疫学的測定において通常知られているも のを使用することができる。 その例としては、 ペルォキシダーゼの基質 として 2, 2'—アジノ一ジ一 [3ェチルベンツチアゾリンスルフォン 酸] ジアンモニゥム塩 (ABTS) 、 オルトフェニレンジァミ ン (OP D) 、 3, 3', 5, 5'—テトラメチルベンヂジン (TMB) 等があり、 停止剤としては H2S04、 HC 1、 酢酸、 グリシン緩衝液 (pH 10. 3) 、 フッ化ナトリウム溶液等がある。
アル力リフォスファタ一ゼの基質としては 4一二トロフエニルフォス フェート、 4—メチルゥンベリフェリルフォスフェート、 NADP等が める。
—ガラストシダーゼの基質としては 2—二トロフエ二ルー^— D— ガラク トシド、 4—メチルゥンベリフェリル一 — D—ガラク トシド等 がある。 停止剤としては、 0.1M Na2C03などがある。
希釈剤:
本発明の測定キッ トおよび測定方法に使用される希釈剤としては、 免 疫学的測定において通常使用されるものであればよい。 該希釈剤として 免疫反応に悪影響を与えないものであればよく、 例えば、 リン酸緩衝液、 トリス塩酸緩衝液、 酢酸緩衝液などの pHが 6.0〜8.0の範囲のもの が主として使用される。
洗浄剤
本発明においては、 免疫反応測定において一般に使用される洗浄剤を そのまま使用することができる。 しかしながら本発明者らによれば、 前 記の如く第 1抗体及び第 2抗体を組合せると共に特定の界面活性剤がヒ ト · トロンボモジユリンの測定における免疫反応を抑制せず、 免疫反応 に関与しない物質や標識抗体の非特異的吸着のみを抑制する効果がある ことがわかった。
本発明においては、 このような界面活性剤を免疫活性剤を免疫反応液 中に存在させたり、 洗浄液中に存在させると高感度測定に有利である。 免疫反応が 2段反応の場合には、 いずれに存在させることもできるが、 2段目の反応に存在させるのが好ましい。
本発明に用いられる界面活性剤は両面界面活性剤及び Z又は H L B値 (Hydraphile Lipophile Balance) が 16以上の非イオン界面活性 剤である。
111^8が16未満の界面活性剤は、 非特異的吸着を抑制すると同時に 免疫反応を抑制してしまい、 好ましくない。
HLB値が 16以上の非イオン界面活性剤としては、 例えば、 ポリオ キシアルキレンアルキルァリールエーテル系、 ポリォキシアルキレンァ ルキルエーテル系、 ポリォキシアルキレン多価アルコール脂肪酸エステ ル系、 ポリォキシエチレンポリォキシプロピレンポリオール系などの H L B値が 16以上のものであればよく、 例えばトリ トン X— 305 (H LB17.3) 、 トリ トン X— 405 (HLB17.9) 、 ェマルゲン 9 50 (HLB 18.2) 、 ェマルゲン 985 (HLB 18.9) 、 Tween 20 (HLB 16.7) 、 ブル口ニック F 68 (HLB 29) 、 テトラ ニック 707 (HLB>20) などが挙げられる。
両性界面活性剤としては、 例えば、 カルボン酸塩型 (ベタイン型) 、 スルホン酸塩型 (スルホベタイン型) およびリン酸塩型のものなどが挙 げられる。 スルホベタイン型は発癌性が問題になることもあり、 その使 用は工業的には必ずしも好ましいとは言えない。 好ましくはべタイン型 両性界面活性剤であり、 一般式 RiRzRsNR [式中、 Riは C5〜C22 アルキル基、 R2および R3は C! Csアルキル基、 R4は COO©置換 基を有する C! Csアルキル基を表わす] で示されるベタイン型である。 更に好ましくは、 R2、 R3がそれぞれメチル基であり、 R4が CH2CO ΟΘである。 この種の好適な界面活性剤としては、 がラウリル基で ある 「アンヒ トール 24 B」 (商品名) が花王株式会社より販売されて いる。
これらの界面活性剤は単独で使用することもできるし、 二種以上の界 面活性剤を併用することもできる。
洗浄液等における界面活性剤の濃度は 1重量%以下であり、 好ましく は 0.0025〜1重量%である。
本発明においては、 0.1重量%以下の界面活性剤量でも洗浄効果に 優れ、 更に洗浄時の泡立ちが少くなり、 その結果泡立ちが激しくて、 泡 の除去が煩雑となったり、 その除去が不十分になって逆に洗浄としては 不十分になったりするといつた問題も少なくなるので、 好ましい。
これら界面活性剤の溶媒としては水、 生理食塩水、 あるいはリン酸緩 衝液のような緩衝液など、 測定に悪影響を及ぼさないものならいずれで あってもよい。 本発明の測定キッ トおよび測定方法において使用される標準物質は第
1抗体及び第 2抗体のそれぞれが認識する抗原決定部位をいずれも少な くとも 1つ以上有する蛋白質またはポリベプチドが用いられる。 その代表的なものの 1つは、 天然の材料から抽出した天然型のヒト · トロンボモジユリンまたはそのフラグメントである。
天然型のヒ ト · トロンボモジュリンを得るための材料としては、 例え ばヒト胎盤、 ヒト肺、 ヒト血管あるいは培養ヒト ·血管内皮細胞とその 培養上清などが挙げられるが入手が比較的容易な点から好ましくはヒ ト 胎盤である。 分離 ·精製は、 通常用いられる蛋白分離技術、 例えば塩析、 抽出、 遠心分離、 限外濾過、 各種のクロマトグラフィー等を組み合せて 行うことができる力、'、 前記したエス · クロサヮらの文献記載の方法は、 その 1例である。
また、 ヒ 卜 · トロンボモジユリンのフラグメントは、 前記の如く して 得られた天然型のヒト · トロンボモジュリンを蛋白分解酵素、 例えばェ ステラ一ゼ、 トリプシンなど或いは蛋白分解試薬、 例えば臭化シアンな どを用いて、 それ自体知られた方法によって得ることができる [例えば S. Kurosawa et al. , J . Biol. Chem. , 262, 2206 (1987) 参照] 。
さらに上記天然型のみならず、 蛋白工学的または遺伝子工学的手法に よつて得られた前記性質を有するポリぺプチドであつてもよい。
蛋白質の使用
本発明者らの研究によれば、 ヒ ト ' トロンボモジュリ ンの測定におい ては、 免疫反応溶液に分子量 1.6万〜 5.0万及び等電点 1.0-5.0 である蛋白質又はそれを含む混合物を存在せしめ、 これらの免疫反応溶 液における最終濃度が 0.005-0.8重量%となるように調整すると、 非特異的吸着が抑制され、 したがってバックグランドが著しく低くなり 高い測定感度が得られることがわかった。 かかる蛋白質又はそれを含む 混合物は、 本発明の免疫測定試薬或いはキッ卜に免疫反応溶液中に前記 所定の量となるように含有せしめることもできる。 かかる蛋白質として は、 例えばカゼイン、 ペプシン、 オボグリコプロテイン、 ォロソムコィ ドなどがあげられる。 分子量 1 . 6万以下の蛋白質を用いた場合特異的 吸着が上昇する結果を得ており、 また 5. 0万以上の分子量では免疫非 特異的反応の低減が不充分かつ特異的免疫反応の低下が見られることに より、 分子量を 1 . 6万〜 5. 0万と定めた。 また等電点に関しても 5. 0以上の蛋白質を添加した場合非特異的吸着が上昇し、 また等電点が 1 . 0より低い場合特異的反応が抑制されるために等電点を 1 . 0〜5 . 0と 疋めた。
また種々の濃度の該蛋白質溶液たとえばスキム · ミルクを用いて免疫 測定方法を行なったところ、 0. 0 0 5重量%未満で抗原が 0であるに もかかわらず非特異的反応が著しく増加し、 また、 1ヶ月間、 該蛋白質 たとえばスキムミルク溶液の冷蔵庫における保存試験を行なった結果、 0. 8重量%より大ではスキムミルク溶液が再溶解不能の沈殿を生じた ため、 その保存安定性の面から 0. 8重量%以下と定めた。 以上 2つの 事実を考慮し、 試薬の安定性を満足し、 かつ非特異的反応を効果的に減 ずるか該蛋白濃度は 0. 0 0 5〜0. 8重量 の範囲が適当である。 この ような混合物としては、 例えば主成分として前記タンパク質 1 0〜6 0 重量%、 好ましくは 2 0〜5 0重量%、 糖 (例えば乳糖) 3 0〜8 0重 量%、 好ましくは 4 0〜6 0重量%、 その他脂肪 (例えば 0 . 5〜2重 量%) 、 灰分 (例えば 5〜1 2重量 、 水分 (例えば 2〜8重量%) などを含むことができる。 このような混合物として典型的なのはスキム ミルクである。 スキムミルクはタンパク質としてカゼィンを含むもので あるが、 カゼインを単独で使用した場合に比べて、 スキムミルクは、 免 疫反応溶液中における分散性が良く、 非特異的反応を抑制する効果が高 く、 温度 4°Cにおける保存性が良い (沈澱が生じにくい) という特徴を 有する。 なお、 本発明に用いるスキムミルクとしては、 脱脂したミルク であれば、 何の由来の乳であっても良い。 一番典型的なもののひとつと しては、 市販されている Difco社製のスキムミルクである。
なお本発明において、 タンパク質の 「分子量」 は浸透圧法によって測 定した分子量を意味し、 具体的には高分子溶液と純溶媒と溶媒分子は自 由に透すが、 溶出高分子は透さない半透膜を境として接した際に両液の 浸透圧差が高分子の分子量のパラメータとなることを利用してタンパク 質の分子量を測定するもので、 本発明では 6.66 M尿素溶液を用いて 4°Cで測定した値である。 また 「等電点」 はタンパク質をその等電点に 従って分離するクロマトフォーカシング法によって測定した値をいい、 具体的には PBE94 (フアルマシア製) ゲルを充填したカラム
(O.5 cm0x45 cm) を用い溶出液 0.025Mイミダゾール塩酸 (pH7.4) で測定した値をいう。
前記蛋白質は、 免疫反応溶液中において、 前記濃度となるように使用 されるが、 キットの場合、 蛋白質は、 標準物質、 第 2抗体又は希釈剤の いずれかに少なくとも含有せしめておくのが好ましい。 含有量は免疫反 応溶液中において前記濃度となるようにして適宜決められるべきである。 本発明方法でのヒト · トロンボモジユリンの測定における、 免疫反応 溶液中に、 前記した分子量 16, 000〜50, 000及び等電点 1.0 〜5.0である蛋白質又はそれを含む混合物とともにァ一グロプリンま たはァーグロプリン誘導体を免疫反応溶液における最終濃度が 0.00 2 5〜0. 1重量%の濃度で存在せしめる時、 非特異的吸着の抑制すな わちバックグランドの低下が更に得られ、 該蛋白質単独で用いられる場 合に比較して更に高い測定感度が得られるので好ましい。 ァーグロプリ ンとしては、 ヒ トまたは哺乳動物、 例えば牛、 ゥサギ、 ャギ、 などの血 清から分離されたものが挙げられるが、 特にヒ ト ' ァーグロプリンが好 ましい。 また y —グロブリン誘導体としては、 化学的に修飾された前記 ァ一グロブリン、 例えば S—アルキル化 y —グロブリン、 S —スルホ化 ァ一グロブリンなどが挙げられる。 次にその添加量は、 0 . 0 0 2 5重 量%未満では γ —グロプリン又はその誘導体の添加によるバックダラン ドの低下は小さく、 実質的に十分なその添加効果は得られない。 また 0 . 1重量%ょり多く添加しても 0. 1重量%添加した時以上の効果は認め 難いことと、 y —グロプリンが比較的高価であるため製造コス卜の観点 から添加量の上限を 0. 1重量%が適当である。 以上の理由によって添 加量は前記の範囲が適当である。
e 発明の効果
かく して、 本発明によれば、 試料、 例えば臨床サンプルなどの極く微 量のヒ ト · トロンボモジュリンを高感度で精度よく、 しかも簡便な操作 で測定することができる。 しかも測定の操作は比較的短かい時間で実施 することができる。
殊に本発明によれば、 試料検体中にヒト ' トロンボモジュリンとトロ ンビンの複合体が存在していても、 その存在によって遊離のヒ ト · トロ ンボモジユリンの測定の精度は、 何等の影響を受けない。

f 実施例
以下実施例により本発明を更に具体的に説明する。
実施例 1
家兎の皮下に、 前記エス · クロサヮらの文献記載の方法に従ってヒト 胎盤から精製したヒト · トロンボモジユリン 5 0 0 /z gとフロイント完 全アジュバンドとのェマルジヨンを投与した。 1 4日後にヒト · トロン ボモジユリン 2 5 0 tz gを同様にして皮下投与した。 更に 1 4曰間隔で 2回同様に皮下投与し、 1 0日目に採血し、 血清とした後、 酵素免疫定 量法、 すなわちヒ ト · トロンボモジユリンを抗原とし第 2抗体にホース ラディッシュパーォキシダーゼ標識したャギ抗ゥサギ I gG抗体で、 ヒ ト · トロンボモジユリンに対する抗体価を測定し 3 0 0万倍であること を認識した。 その後、 全血を採取、 血清とし、 それをプロテイン A-セ ファローズ- 4 Bカラムで精製してヒト · トロンボモジユリンに対する ポリクローナル抗体を取得した。
実施例 2
1 . 抗ヒト · トロンボモジユリン 'モノクローナル抗体のマレイミ ド 化 ヒ ト · トロンボモジユリンのトロンビンとの複合体形成に対する阻害 活性を有し、 かつプロテイン Cの活性化を阻害するモノクローナル抗体 (特開昭 64— 45398号公報記載の TM- A 73) 830 gの PB S溶液に、 サクシンィミ ジル 4- (N-マレイミ ドメチル) シクロへキサ ンカーボネート (SMC C) の DM F溶液 (1

13.9 を 25°Cで滴下した。 30分間撹拌した後、 セフアデックス G- 25の力 ラム (lcrax45cm) で溶離液として 0.1 Mリン酸緩衝液を用いて、 ゲル濾過しマレイミ ド化されたモノクローナル抗体を得た。
2. ペルォキシダ一ゼのチオール基導入
ペルォキシダーゼ (東洋紡) 143 ragの 0.1 Mリン酸緩衝液 (pH 6. 0) 溶液 14.3 に S-ァセチルメルカブト無水コハク酸の DMF溶液 ( 60 mgXmi) 176.4 ζ を 25 °Cで撹拌下徐々に滴下し、 60分間 反応させた。 この溶液に 0.1Mトリス塩酸緩衝液 (pH7.0) 5.72 I ^と 0.1M EDTA溶液 (pH 7.0) 1.14m、 1Mヒ ドロキシル ァミ ン溶液 (pH7.0) 11.4 を加え 30°Cで 5分間撹拌した。 溶液を透析チューブに入れ、 0.1Mリン酸緩衝液 (pH6.0) -5mM EDTAを外液として 4 °Cで二夜透析し、 S H基の導入されたペルォ キシダーゼ溶液を得た。
3. マレイミ ド化モノクローナル抗体とチオール基を導入したペルォ キシダーゼの結合
マレイミ ド化モノクローナル抗体 830〃gとチオール基導入ペルォ キシダーゼ溶液 ( 4.05 mgZn^) 1.02 m とを混合し、 該混合液を限 外濾過によって約 600 に濃縮、 25°Cで 24時間反応させた。 反 応液を HPLC (カラム TSK Ge23000 SW) 、 溶離液 PBS (pH7.2) で分離しペルォキシダーゼ結合モノクローナル抗体を得た。 得られたペルォキシダーゼ結合モノクローナル抗体の抗体とペルォキ シダーゼのモル比は 1 : 4.6であった。
実施例 3
実施例 1に記載の抗ヒト · トロンボモジユリン 'ポリクローナル抗体 を、 0.1M炭酸緩衝液 (pH9.5) で 5 ^gZm の濃度に調整し、 鏡面 化されたポリスチレンビーズ (ィムノケミカル: ィムノビーズ、 Surfc om 57 OA (東京精密社製) で測定した表面中心線平均粗さ (Ra) : 1.3 zm) を浸し 4°Cで 17時間静置した。 ビーズを 1%B SA- TB S (pH7.4) で室温で 2時間静置した。 TB S (pH7.4) で再度 3 回洗浄し、 抗体固定ビーズ Aを得た。 TB S (pH7.4) 中に使用する まで 4 °Cで保存した。
上記抗体固定ビーズ Aと同様の方法によって実施例 2に記載の抗ヒ ト · ト口ンボモジユリン ·モノクローナル抗体 (TM-A 73) を固定した 抗体固定ビーズ Bを得た。
一方、 0.5%B SA-0.05%Tween20— 5mMCaC - T B S (pH7.4) の希釈緩衝液でヒト · トロンボモジユリンの 0、 0.62 5、 1.25、 2.5、 5.0、 10.0 ng/ 溶液を作成した。 この 0. 4πιδづっをポリプロピレン製プラスチック小試験管に入れ、 ヒ ト · トロ ンボモジュリン濃度 0〜1 Ong/ の希釈列 Αと Βを作成した。 この希 釈列 Aの各試験管に抗体固定ビーズ Aを 1づっ加え、 また希釈列 Bに抗 体固体ビーズ Bを 1つづつ加え、 それぞれを 37°Cで 90分間インキュ 一べ一シヨンした。
TB S (pH 7.4) に Tween20を 0.05%溶解した液 (TB S-T と略) で 3回洗浄した後、 希釈列 Aの各試験管には実施例 2で調製した ペルォキシダーゼ標識抗ヒト · トロンボモジュリン ·モノクロ一ナル抗 体を抗体濃度 1 gZn^に 0.5%B S A-0.05%Tween20- 5mM CaCi2-TB S (pH 7.4) で溶解した液を 0.4 mづっ加え、 また希 釈列 Bの各試験管には、 実施例 2と同様の方法で、 実施例 1に記載の抗 ヒ ト · トロンボモジユリン ·ポリクローナル抗体から調製したペルォキ シダーゼ標識抗ヒ ト · トロンボモジユリン ·ポリクローナル抗体を前記 と同様にして調製した抗体濃度 4 ^gZm の液を 0.4mづっ加え、 それ ぞれ 37°Cで 30分間ィンキューべーシヨンした。
TB S-Tで 5回洗浄した後、 発色剤として、 2.5mM H2O2-0. 025% 3, 3', 5, 5'テトラメチルベンジジンの溶液を 0.4π^づっ 加え、 37 °Cで 30分間インキユーべーシヨンし、 1規定硫酸 li ^を加 え発色反応を停止し、 45 Onmの吸光度を測定した。
その結果得た検量線を第 1図に示した。
比較として、 実施例 2の抗体ヒ ト ' トロンボモジュリン ·モノクロ一 ナル抗体とェピトープを異にするモノクローナル抗体を、 前記抗体固定 ビーズ A、 Bと同様にして鏡面化されたポリスチレンビーズに固定し抗 体固体ビーズ Cを得た。 該固定ビーズの外は前記抗体固定ビーズ Aを用 いた時と同様にして (希釈列 C) ヒ ト ' トロンボモジュリンに対する検 量線を得た。 その結果を比較例として第 1図に示した。
第 1図から明らかな如く、 2種のモノクローナル抗体の組み合せ (希 釈列 C) にくらベて、 本発明のポリクローナル抗体とモノクローナル抗 体の組み合せ (希釈列 A及び B) においては、 高感度の検量線が得られ ることが明らかである。 実施例 4
実施例 1に記載の抗ヒト · トロンボモジユリン ·ポリクローナル抗体 を、 0.1M炭酸緩衝液 (pH9.5) で 20 gZmの濃度に調整し、 下 記第 1表に示した各種中心線平均粗さの E I A用のポリスチレンビーズ を浸し、 4 °Cで 17時間静置した。 ビーズを 1%B S A-TB S (PH7. 4) で室温に 2時間静置した。 TBSで 3回洗浄し、 各種抗体固定ビー ズを得た。
—方、 0.5%B S A-0.2%スキム · ミルク— 0.015%ヒト—< グ ロブリン- 15mM CaCi2-TB S (pH 7.4 ) の希釈緩衝液でヒ ト ' トロンボモジュリンの 0、 17ngZmg溶液を作成し、 この 0.2 mづっ をポリプロピレン製プラスチック小試験管に入れた。 更に、 各試験管に 実施例 2で調製したペルォキシダーゼ標識抗ヒト · トロンボモジユリン · •モノクローナル抗体の抗体濃度 3 g/miの 0.5%B S A-5mM C aC -TBS (pH7.4) 溶液を加え、 上記抗体固定ビーズを 1ケづっ 入れて、 37°Cで 90分間インキユーべーシヨンした。 TBS-Tで 3 回洗浄した後、 発色剤として 2.5mM H2O2-0.025%3, 3', 3, 5にテトラメチルベンジジンの溶液を 0.4m_gづっ加え 37。Cで 30分 間ィンキューべーシヨンし、 1規定硫酸 1111£づつ加えて発色反応を停止 した。
この溶液の 45 Onmの吸光度を測定し、 各種抗体固体ビーズの S ZN 比 (ここで Nはヒト · トロンボモジュリン OngZmgにおける吸光度であ り、 Sはヒ ト ' トロンボモジュリン 17ngZm こおける吸光度である) 及び測定感度 (ヒト · トロンボモジユリン OngZi ^の変動係数を 10% とし、 これから求めた 0点における [吸光度 +3x標準偏差] 値がヒト · ト口ンボモジユリン濃度が

とを結ぶ直線と交わる 点におけるヒ ト · トロンボモジュリ ン濃度を測定感度と定義する) を算 出した。 その結果を第 1表に示す。
第 1表で明らかなごとく、 本発明の Raが 1.5 m以下の鏡面化され た不溶性担体の使用により優れた感度でヒト · トロンボモジユリン測定 が可能となった。

実施例 5
実施例 1に記載の抗ヒト · トロンボモジュリン 'ポリクロ一ナル抗体 を、 0.1M炭酸緩衝液 (PH9.5) に 5 zgZmの濃度に調整し、 鏡面 化されたポリスチレンビーズ (ィムノケミカル:ィムノビーズ、 Surfc om 57 OA (東京精密社製) で測定した表面中心線平均粗さ (Ra) : 1.3 ^m) を浸し 4°Cで 17時間静置した。 ビーズを TBS (pH7. 4 ) にて 3回洗浄後、 1%BSA-TBS (pH7.4) で室温で 2時間 静置した。 TBSで再度 3回洗浄し、 TB S中に使用するまで 4°Cで保 存した。
ヒト · トロンボモジユリンの 0、 10ng/m6溶液を、 0.5%B S A- 5mM CaC - TBS (pH 7.4) にスキム ' ミルクを最終濃度 0、 0.005、 0.012、 0.025、 0.05、 0.1、 0.2%含有させ た溶液で調製し、 その 0.4 nigづっをポリプロピレン製プラスチック小 試験管に入れた。 各チューブに前記抗ヒト · トロンボモジュリン固定ビ —ズを 1ケづっ加え、 37°Cで 90分間ィンキューべーシヨンした。
TB Sに Tween20を 0.05%溶解した液 (TBS- Tと略) で 2回 洗浄した後、 実施例 2で作成したペルォキシダーゼ標識抗ヒト · トロン ボモジユリン ·モノクローナル抗体の抗体濃度 1 /zgZn^に 0.5%B S A-5mM CaC -TBS (PH7.4) にスキムミルクを最終濃度 0、 0.005、 0.012、 0.025、 0.05、 0.1、 0.2%含有させ た溶液で調製し、 前記スキム, ミルク濃度と同じチューブにそれぞれ 0. 4111£づつ加え、 37°Cで 30分間ィンキューべーシヨンした。
TBS- Tで 3回洗浄した後、 発色剤として、 2.5mM H202 - 0. 025% 3, 3', 5, 5'テトラメチルベンジジンの溶液を 0.4ι ^づ つ加え、 37 °Cで 30分間インキユーべーシヨンし、 1規定硫酸 lmgを 加え発色反応を停止し、 45 Οηπιの吸光度を測定した。
その結果得た免疫反応溶液中のスキム · ミルク濃度と、 ヒト · トロン ボモジュリン 0、 1 OngZm の吸光度との関係を第 2図に示した。 図 から明らかな如く、 スキム · ミルクの添加は特異的反応に影響しないで 非特異的吸着を抑制することが明らかである。
実施例 6
実施例 5に記載と同様にして抗ヒト · トロンボモジユリン ·ポリクロ ーナル抗体固定ビーズを調製した。
—方、 0.5%BSA— 0.015%スキム · ミノレク -5mM CaCi-T B S (pH7.4) でヒト-ァグロプリンの下記第 2表に記載の濃度の溶 液を調製し、 これらを希釈緩衝液としてヒ ト · トロンボモジユリンの 0 ngZn^及び 14ngZni溶液を調製した。 各々 0.2mづっをポリプロピ レン製小試験管に加えた。 実施例 2で調製したペルォキシダーゼ標識抗 ヒ ト · トロンボモジユリン ·モノクロ一ナル抗体の抗体濃度 3 ytzg/nii の溶液を上記希釈緩衝液で調製し、 ヒ ト · yグロプリン濃度を対応させ て上記小試験管に 0.2 づっ加えた。
その後、 おのおのの試験管に上記抗体固体ビーズを 1ケづっ入れ、 3 7°Cで 90分間ィンキューべーシヨンした。
TB S-Tで 3回洗浄した後、 発色剤として 2.5mM H202- 0.0 25% 3, 3', 5, 5'-テトラメチルベンジジン溶液を 0.4 mづっ加 え、 37 °Cで 30分間インキユーべーシヨンし、 1規定硫酸 lm6を加え 発色反応を停止して 45 Onmの吸光度を測定した。
測定した吸光度から、 各ヒ ト · ァグロブリン濃度での SZN比及び測 定感度を実施例 4と同様にして算出した。 その結果を第 2表に示す。 第 2表の結果から、 スキム ' ミルクを含有する免疫反応溶液中に更に rグロプリンを 0.0025%以上添加することによってヒ ト · トロン ボモジュリンの測定感度を高めることが出来ることがわかる。

ヒ ト · トロンボモジユリンの OngZm£および 17ngZni濃度の 0.5 %B S A-0.2%スキム · ミノレク— 0.015%ヒ ト . yグロブリン— 5 m M CaCiz-5 OmM Tris/HC^-O.1M NaC (pH 7.4) 液 0.2n^と、 実施例 2で得られたペルォキシダ一ゼ標識抗ヒト · トロ ンボモジュリン 'モノクローナル抗体の抗体濃度が 3〃gZi^である 0. 5%B S A-5mM CaC 25 OmM Tris/H Ci— 0.1 M NaCi (pH7.4) 溶液 0.2 miをポリプロピレン製小試験管にとり、 次に実 施例 5に記載と同様にして調製した抗ヒ ト · トロンボモジユリン ·ポリ クローナル抗体固定ビーズを 1ケづっ上記各小試験管に入れ、 37でで 90分間ィンキューべーシヨンした。
各小試験管内の溶液を吸引除去した後、 第 3表に示した書類の界面活 性剤の 0.05%を含有する生理食塩液 3 で 3回洗浄した。 洗浄後、 発色剤として 2.5mM
H2O2-0.025% 3, 3', 5, 5' -テトラメチルベンジジン溶液を 0. 4 πιβづっ加え、 3 7 °Cで 3 0分間インキユーべーシヨンし、 1規定硫酸 1 m を加え発色反応を停止して 4 5 0 nmの吸光度を測定した。 この吸光 度から実施例 4と同様にして S ZN比を算出した。 その結果を第 3表に 示す。

実施例 8
洗浄液として第 4表に示した各濃度のテトロニック Ί 0 2の生理食塩 液を用い、 実施例 7と同様に操作して使用濃度の効果を調べた。 その結 果を第 4表に示す。 第 4 表

上記結果から明らかなように 0.0025〜1.0%の範囲で3 ^比 は大きく、 すなわち高感度にヒ ト · トロンボモジュリ ンを測定できる。 2%では泡立ちが激しく、 かつヒ ト · トロンボモジユリン濃度 17ng/ の吸光度が著るしく低下した。
実施例 9
実施例 5に記載と同様にて抗ヒ ト · トロンボモジユリン · ポリクロー ナル抗体固体ビーズを調製した。
一方、 0.5%B S A-0.2%スキム · ミルク 0.015%ヒ ト · Ί — グロブリンー 5mM CaC - 5 OmM TrisZHC ー 0.1M C& Na C£ (pH7.4) で段階希釈したヒ ト · トロンボモジユリン溶液 (0〜 33.3ng/ffl^) を調製し、 各 0.2m づっをポリプロピレン製小試験管 に加えた。 次に実施例 2で作成したペルォキシダーゼ標識抗ヒ ト · トロ ンボモジユリン ·モノクローナル抗体の抗体濃度 3 gZraの液を 0.5 %B S A - 5mM CaC 2— 50mM Tris/H C^- 0. 1 M NaC£ (p H7.4) で調製し、 上記小試験管の各々に 0.2 づっ加えた。 さらに これらの小試験管に上記抗体固定ビーズを 1ケづっ入れ、 37°Cで 90 分間ィンキューべーションした。
0.005 %テトロニック 702— 5mM CaC 一生理食塩液で 3回 洗浄した後、 発色剤として 2.5mM H202- 0.025%3, 3', 5, 5'-テトラメチルベンジジン溶液を 0.4mづっ加え、 37°Cで 30分 間ィンキューべ一シヨンした後、 1規定硫酸 1 を加え発色反応を停止 して 45 Onmの吸光度を測定した。 その結果得た検量線を第 3図に示し o
第 3図から明らかな如く、 バックグランドが低く高感度 (実施例 4の 方法により算出した測定感度:

Claims

請 求 の 範 囲
1 . 試料中のヒト · トロンボモジユリンをサンドィツチ法により免疫 学的に測定する方法において、 不溶性担体に固定化した抗体 (第 1抗体) と標識化した抗体 (第 2抗体) は、 いずれか一方がヒト · トロンボモジ ユリンを認識するポリクローナル抗体であり、 他方がヒト · トロンボモ ジユリンを認識するモノクローナル抗体であるヒト · トロンボモジユリ ンの免疫学的測定方法。
2. 該第 1抗体ヒト · トロンボモジユリンを認識するポリクローナル 抗体であり、 且つ該第 2抗体がヒト · トロンボモジユリンを認識するモ ノクローナル抗体である請求項 1記載の免疫学的測定方法。
3. 該モノクローナル抗体が、 ヒト · トロンボモジユリンとヒト · ト ロンボビンとの複合体形成に対する阻害作用を有し且つヒト ·プロティ ン Cの活性化に阻害作用を有するものである請求項 1または 2記載の免 疫学的測定方法。
4. 該不溶性担体が鏡面化された表面を有するものである請求項 1〜
3のいずれかに記載の免疫学的測定方法。
5. 免疫学的測定方法における免疫反応溶液中に分子量約 1 6 0 0 0 〜約 5 0, 0 0 0および等電点 1 . 0〜5. 0である蛋白質を存在せしめ、 免疫反応溶液における該蛋白質の最終濃度が 0. 0 0 5〜0 . 8重量%と なるように調製する請求項 1〜 4のいずれかに記載の免疫学的測定方法。
6. 免疫反応溶液中にさらに、 7—グロブリンまたは 7—グロブリン 誘導体を 0. 0 0 2 5〜0. 1重量%の濃度を存在せしめる請求項 5記載 の免疫学的測定方法。
7. 免疫学的測定方法における洗浄剤として、 両性界面活性剤および /または H L B値が 1 6以上の非イオン界面活性剤を 0. 0 0 2 5〜1 重量%の濃度で含有する洗浄剤を使用する請求項 1〜 6のいずれかに記 載の免疫学的測定方法。
8. 試料中のヒ ト · トロンボモジュリンを不溶性担体に固定化した第 5 1抗体及び標識化した第 2抗体を使用して免疫学的に測定方するための 試薬であって、 一方の抗体がヒ ト · トロンボモジユリンを認識するポリ クローナル抗体であり、 他方がヒ ト · トロンボモジユリンを認識するモ ノクローナル抗体であることを特徵とする免疫学的測定試薬。
9. 該第 1抗体ヒト · トロンボモジユリンを認識するポリクローナル 1 0 抗体であり、 且つ該第 2抗体がヒト · トロンボモジュリンを認識するモ ノクローナル抗体である請求項 8記載の免疫学的測定試薬。
1 0. 該モノクロ一ナル抗体が、 ヒ ト · トロンボモジユリンとヒ ト · トロンビンとの複合体形成に対する阻害作用を有し且つヒ ト ·プロティ ン Cの活性化に阻害作用を有するものである請求項 8または 9記載の免

1 1 . 該不溶性担体が鏡面化された表面を有するものである請求項 8 〜1 0のいずれかに記載の免疫学的測定試薬。
1 2. 試料中のヒト · トロンボモジュリンを免疫学的に測定するため のキッ 卜であって、
20 (i) 不溶性固体担体に固定化したヒト ' トロンボモジュリンを認識す る抗体 (第 1抗体) 、
(ii) 標識化したヒト · トロンボモジユリンを認識する抗体 (第 2抗 体) 、
但し、 上記第 1抗体及び第 2'抗体は、 いずれか一方がヒ ト · トロ ンボモジユリンを認識するポリクローナル抗体であり、 他方がヒト ' ト口ンボモジユリンを認識するモノクローナル抗体である。
(iii) 酵素で標識化した場合には、 酵素活性を測定^ ·るための基質お よび反応停止剤
(iv) 希釈剤
(V) 洗浄剤、 及び
(vi) 標準物質
よりなる免疫学的測定キッ ト。
1 3 . 該第 1抗体ヒト · トロンボモジユリンを認識するポリクローナ ル抗体であり、 且つ該第 2抗体がヒト ' トロンボモジュリンを認識する モノクローナル抗体である請求項 1 2記載の免疫学的測定キッ ト。
1 4. 該モノクローナル抗体が、 ヒ ト · トロンボモジユリンとヒト · トロンボビンとの複合体形成に対する阻害作用を有し且つヒト ·プロテ イン Cの活性化に阻害作用を有するものである請求項 1 2または 1 3記 載の免疫学的測定キッ ト。
1 5. 該不溶性担体が鏡面化された表面を有するものである請求項 1 2〜1 4のいずれかに記載の免疫学的測定キッ ト。
1 6. 該洗浄剤が、 両面界面活性剤及び Z又は H L B値が 1 6以上の 非イオン界面活性剤を 0. 0 0 2 5〜1重量%含有している請求項 1 2D 〜1 5項のいずれかに記載の免疫学的測定キッ ト。
1 7. 該第 2抗体、 該希釈剤または該標準物質に、 分子量約 1 6, 0 0 0〜約 5 0, 0 0 0及び等電点 1 . 0〜5 . 0である蛋白質を含有せし めた請求項 1 2〜1 6のいずれかに記載の免疫学的測定キッ ト。
1 8 . 該第 2抗体、 該希釈剤まだは該標準物質に、 y—グロブリンま たは 7—グロプリン誘導体を含有せしめた請求項 1 2〜1 7項のいずれ かに記載の免疫学的測定キッ ト。
