Definitions
- Parvoviruses form a viral family that possesses certain common traits (Cotmore et al., Adv. Virus Res. 33: 91-174 (1987)). They are the smallest known DNA viruses. Its genome is formed by a linear DNA molecule of about 5.0 kb in length enclosed within a protein capsid, of icosahedral symmetry and about 25 nm in size. Two large open reading phases are detected in the viral genome. The phase on the left codes for the non-structural proteins involved in viral replication, while the phase on the right codes for the structural proteins that form the viral capsid, which is made up of the VP1, VP2 and VP3 proteins. The mRNA of both reading phases is polyadenylated and 3'-coterminal.
- the CPV capsid contains two proteins whose amino acid sequences overlap widely, VP1 (82-84 KDa) and VP2 (67-70 KDa) (Paradiso et al., J. Gen. Virol. 39, 800-807 (1982) ; Surleraux et al., Arch. Virol. 82, 233-240 (1984); J. Gen. Virol. 62, 113-125 (1982)).
- PPV causes reproductive failures in sows, causing death and fetal mummification, abortions and other reproductive disorders of pregnant sows (Joo & Johnson, Veterinary Bulletin 46, 653-660 (1976); Mengeling, J. Am. Vet. Med. Assoc 172, 1291-1294 (1978)).
- PPV contains a single stranded DNA molecule of approximately 5000 nucleotides (Mollitor et al., Virology 137, 241-254 (1984)). The complete PPV genome sequence has been described by our group (Ranz et al., J. Gen. Virol. 70, 2541-2553 (1989)).
- a further object of this invention is new synthetic peptides, from the amino terminal end of PPV VP2, capable of inducing neutralizing antibodies against PPV that can be used in obtaining immunogenic compositions and in formulating new vaccines. Synthetic capable of protecting pigs from infection caused by PPV. These synthetic peptides are based on the immunogenic properties of the amino terminus of the majority protein VP2 of PPV. Immunogenic compositions and vaccines incorporating said peptide are another additional object of this invention.
- FIGURES Figure 1 shows the results obtained by determining, by means of a virus-specific ELISA (enzyme immunoassay), the titre of anti-CPV antibodies present in serum of dogs immunized with the peptides of this invention (dogs 3029—- ⁇ — and 3030 - —-) and in a control dog (3025 — Q-)
- the invention provides new peptides useful for the preparation of synthetic vaccines suitable for the protection of animals against infections caused by autonomous parvoviruses, such as CPV, MEV, FPLV and PPV.
- the new vaccines contain at least one chemically synthesized peptide from the amino terminal region of the VP2 protein of CPV or the VP2 protein of PPV.
- terminal amino region in the sense used in this description, the region comprising at least the first 25 amino acids of the VP2 protein should be understood.
- N s 1 are capable of inducing neutralizing antibodies against MEV, so they can be used in the formulation of vaccines capable of protecting visions from infection caused by MEV. Indeed, it has been proven that mints vaccinated with a mixture of peptides 1L15 and 7L15 were protected against subsequent infection with virulent virus (Example 6). This experimental confirmation with MEV makes it possible to assume that the peptides included within the SEC. ID. N B 1 can induce protection in cats against FPLV due to the structural homology between CPV, MEV and FPLV, all of them belonging to the same subgroup.
- All peptides included in the SEC are included within this invention. ID. N B 2 or resulting from the substitution of amino acids of those shown in SEC.ID. N s 2 by other functionally equivalent or from the deletion or insertion of some of said amino acids as long as the resulting peptides are capable of inducing neutralizing antibodies against PPV. For practical reasons synthetic peptides of a length between 6 and 20 amino acids are preferred.
- the peptides of the invention should preferably be soluble to allow their coupling to suitable carrier proteins that enhance their immunogenic character, such as the KLH protein.
- suitable carrier proteins such as the KLH protein.
- the following steps were basically followed: a) Chemical synthesis of the peptides corresponding to the most immunogenic regions of PPV VP2 protein. b) Coupling of said peptides to a carrier protein. c) Selection of peptides that induced in rabbits an antibody response capable of neutralizing PPV. d) Immunization of pigs with the previously selected peptides.
- Example 2 17 solid phase peptides were synthesized using Fmoc chemistry, as previously mentioned. in relation to synthetic CPV peptides.
- the 17 synthesized peptides were coupled to the KLH carrier protein (Example 3) and used to immunize rabbits (Example 7).
- KLH carrier protein Example 3
- Example 7 the KLH carrier protein
- peptides named 1L15, 5L16, 6L15, 8L15, 10L16 and 13L15 were capable of inducing antibodies that neutralized PPV (Table 5). Based on these results, it can be said that these peptides can be used in the formulation of new synthetic vaccines in order to protect animals against infection caused by PPV.
- immunogenic compositions containing said peptides can be prepared in an immunogenic manner, whereby they can be used to formulate vaccines capable of protecting dogs, cats and minks from infection caused by CPV, FPLV and MEV respectively (for what they use peptides comprised within SEQ. ID. N B 1) and to formulate vaccines capable of protecting pigs against infection caused by PPV (for which will be used peptides comprised within SEQ. ID. N B two).
- Said immunogenic compositions may be prepared by coupling at least one of the peptides of this invention to a suitable multimeric carrier protein or structure.
- compositions comprising an immunogenic conjugate of a protein (KLH) and a peptide of this invention are provided.
- these compositions may contain an immunogenic complex obtained by crosslinking the peptide or an immunogenic recombinant protein containing one of the peptides of this invention.
- This invention also provides vaccines that are characterized in that they comprise one of the aforementioned immunogenic compositions in combination with at least one immunological adjuvant.
- Such vaccines can be prepared by suspending at least one of the peptides of the invention coupled to a suitable multimeric carrier protein or structure, in an immunologically acceptable diluent plus an adjuvant.
- a vaccine may contain a mixture of the peptides of this invention as immunogenic agents.
- immunologically acceptable diluent saline solutions with tris buffer or other similar saline solutions can be used.
- suspensions of alumina gels can be used alone or in combination with other adjuvants commonly used in the formulation of vaccines such as QuilA or related and of similar potency.

- each letter represents an amino acid sequence designated as code amino acid nomenclature in one - letter (Biochemistry Structural Louisot P., Ed AC, I th Edition (1977). , pp. 372-373); the amino terminal end is located on the left side of the sequence while the carboxy terminal end is located on the right side of the sequence; the peptide code reflects the position of the rest of the N-terminal end in the VP2 sequence
- the peptides were purified using high performance liquid chromatography (HPLC). Quality was analyzed on a Delta PaK 5 or C18-100A column (0.39 x 15 cm), with a 680 model gradient controller and a detector
- diode array model 991.
- the peptides were purified on a 2 x 25 cm Bischoff Prap 2025 C18 column in a Hewlett Packard HPLC equipment, model 1082B and with a reading at 225 nm.
- Gradients of methanol were used with an increase of methanol of 1% per min at a flow of 10 ml / min in water and 0.1% (v / v) of trifluoroacetic acid (TFA).

- the amino acid composition analysis was performed according to the Pico-tag (Waters) procedures.
- the length of the Peptides are variable, between 15 and 20 residues, not including the N-terminal cysteine that is used to perform the coupling. All of them were sufficiently soluble to allow their conjugation to KLH.
- the synthesis of these 17 peptides was carried out by the chemistry of the Fmoc, according to the general procedure previously mentioned, on Rink R resins (Example 1). The sequences were obtained from PPV protein VP2 (Ranz et al., Cited supra; Sakurai et al. Virus Res. 13, 79-86 (1989)). The sequences are acetylated at the N-terminal end and amidated at the C-terminal. Some peptides were purified according to the protocol described in Example 1.
- the coupling of purified peptides to the KLH carrier protein was performed according to standard procedures using m-maleimidobenzoyl-N-succinimide ester (MBS) as a coupling agent (Lerner et al. (1981) Proc. Nati. Acad Sci. USA 78, 3403-3407). The dialysis steps were done at 4 ° C and the other incubations at room temperature. To each mg of KLH 1 mg of peptide was added after binding of a "linker" to the protein and incubated overnight at room temperature.
