この発明は、 糖鎖合成酵素および該酵素をコードする D NAに関するものであ る。 さらに詳しくは、 本発明は N—ァセチルガラクトサミン 2, 6シアル酸転移 酵素 &1 ( 2, 6-シァル酸転移酵素) および該酵素をコードする DNAに関す るものである。 該酵素は、 癌転移抑制およびウィルス感染抑止効果を有する薬剤 として、 あるいは薬剤にシアル酸を付加することにより生理作用を増加させるた めの試薬等として有用である。
また、 本発明は糖鎖合成酵素の製造方法に関するものである。 さらに詳しくは、 本発明はシァル酸転移酵素を微生物で発現させ、 シァル酸転移酵素を大量に取得 する方法に関するものである。 背景技術
シアル酸は、 例えば細胞 -細胞間伝達、 細胞基質相互作用、 細胞接着等の重要 な生理作用をつかさどる物質である。 発生、 分化、 及び癌遺伝子のトランスホー メーシヨン等の過程において調節をうけた、 種々の異なる細胞表面シアル酸が存 在することが知られている。
シアル酸は糖蛋白および糖脂質の炭化水素基の末端位置に存在しており、 翻訳 の後の過程で酵素的に CMP- Siaからこれらの部位に導入される。 例えば、 糖蛋白 には 3種の結合様式、 すなわち Siaひ 2, 6Gal, Siaa2, 3Gal及び Siaひ 2, 6GalNAc が共通に存在しており (Hakomori, S., Ann. Rev. Biochem. , 50, pp.733-764, 1981) ガングリオシドには高頻度に 2種の結合様式、 すなわち Sia 2. 3Gal及び Siaひ 2, 8Siaが存在している (Fishman, P. , and Brady, R. O. , Science , 194, PP. 906- 915, 1976) 。

Chem. , 267, pp.21004- 21010, 1992: 特表平 5-504678号公報; および Lee, Y. et al., Eur. J. Biochem, 216, 377 - 385, 1993)、 Gal S 1, 3(4)GlcNAcひ 2, 3-シアル酸転移酵素 (Gal 83(4)GlcNAc-ひ 3ST)をコードする cDNA(Wen, D. X et al., J. Biol. Chem. , 267, 21011-21019, 1992; 及び Kitagawa, H. et al. , Biochem. Biophys. Res. Commun. 194, 375)、 および GalySl,3GalNAc/Gal /Sl,4GlcNAc ひ 2, 3-シアル酸転移酵素をコードする cDNA (Sasaki, K. et al. , J. Biol. Chem. , 268, 22782-22787, 1993) もクロ一 ニングされている。
一方、 6&1^じひ2,6-シァル酸転移酵素にっぃては、 この酵素を分離した報告も あるが (Hakomori, S., Ann. Rev. Biochem., 50, 733-764, 1981) 、 物質として 同定できる程には精製されておらず、 反応特異性、 性質の安定性、 供給量に問題 があり、 実用に供することはできなかった。 また、 6&1^じひ2,6-シァル酸転移酵 素 (EC 2.4.99.3; GalNAc-ひ 6ST)をコードする cDNAはクローニングされていなレ、。 いままで構造が明らカ、にされた上記のシアル酸転移酵素は、 NH2-末端領域に位 置する疎水性セグメントを有しており、 この疎水性セグメントにより細 に経 膜的に固定される I I型の経膜蛋白である。 従って、 シアル酸転移酵素遺伝子を含 むベクターを哺乳類細胞にトランスフ ク卜して発現させると、 発現した酵素が 細胞膜に固定されてしまい、 細胞外に分泌されないという問題があった。 また、 哺乳類細胞を用いて発現させると、 細胞内酵素濃度が一定以上になると酵素の発 現が低下するという問題がある。
この様な問題を解決するために、 シアル酸転移酵素の活性ドメイン部分とシグ ナルぺプチド部分とを有する分泌型の融合蛋白を製造することもできる。 この方 法は、 シアル酸転移活性を保持しシアル酸転移酵素として作用する蛋白を細胞外 に分泌させるので、 細胞培養液からシアル酸転移酵素を容易に調製できるという 特徴がある。 しかしながら、 哺乳類の細胞を用いてシアル酸転移酵素を発現させ た場合には、 トランスフ クトした細胞が不安定であったり、 細胞培養操作が煩 雑になる場合がある。 また、 大量のシアル酸転移酵素を発現させるには長期にわ たって大量の細胞を培養する必要があり、 費用や設備の点で問題が生じる可能性 もあ o。
一方、 哺乳類細胞中に発現する酵素をコードする cDNA をクローニングし、 酵 素または酵素の可溶性蛋白をコードする遺伝子を含む組み替えべクタ一を製造し て、 該ベクターにより微生物を形質転換する方法は当業者に周知である。 この方 法により製造された形質転換体を培養することにより、 目的の活性を保持する酵 素あるいは可溶性酵素を微生物中で発現させて、 目的の酵素を大量に生産するこ とができる。
この方法は、 例えば、 形質転換された微生物を培養した後、 微生物をリゾチ一 ム等により溶菌して酵素を抽出する工程を含んでいる。 しかしながら、 短時間の うちに微生物の菌体内に大量の不溶性あるいは可溶性蛋白が発現するため、 菌体 内で蛋白が凝集して蛋白凝集物や沈殿物を形成することがあり、 このような蛋白 凝集物や沈殿物中から蛋白を抽出する必要があつた。
上記の蛋白凝集物や沈殿物中から蛋白を抽出するためには、 一般には尿素や塩 酸グァニジンが用いる方法が採用されている。 この方法では、 尿素などにより蛋 白を一旦変性させて (疎水性部分を露出させて) 可溶化したのち、 さらに脱変性
(renatuation) 処理を行うのが一般的である。 脱変性処理は透析によって尿素を 除去することにより行われる力 尿素の除去に際して、 それぞれの酵素に固有の 至適条件 (例えば、 PH、 塩濃度、 温度等) を選択しなければならないという問題 があり、 この条件の選択に多大な時間を要するのが現状である。 条件の設定を誤 つた場合には、 回収された酵素には活性がほとんど残存しなくなるので、 とりわ け脱変性処理の条件選択は重要である。
従って本発明は、 精製された GalNAc 2, 6-シアル酸転移酵素を提供することを 目的とするものである。 また本発明は、 GalNAcひ 2, 6-シアル酸転移酵素をコード する cDNAをクローニングし、 6&1^じひ2,6-シァル酸転移酵素をコ一ドする D NA 配列および該酵素のアミノ酸配列を提供することを目的としている。 また、 本発 明は、 上記の GalNAc 2, 6-シアル酸転移酵素の活性ドメインを含む細胞外分泌型 の蛋白を提供すること、 並びに上記蛋白を微生物中で大量に発現させる方法を提 供することを目的としている。 菌体内に発現するシアル酸転移酵素の凝集物から 該酵素を抽出して効率的に脱変性する方法を提供することも本発明のさらに別の . 目的である。 発明の開示
本発明者は上記の目的を達成すべく鋭意努力し、 ニヮトリ胚から GalNAcひ 2, 6- シアル酸転移酵素をコードする cDNAをクローニングし、 本発明を完成するに至つ た。 すなわち本発明は、 配列表の配列番号 1に示されるアミノ酸配列により特定 される6&1^( 2,6-シァル酸転移酵素 P-B1 を提供するものである。 また、 本発 明は、 上記の6&1 じひ2,6-シァル酸転移酵素 P-B1 のアミノ酸配列をコードする GalNAcひ 2, 6-シアル酸転移酵素遺伝子、 およびその一態様である配列表の配列番 号 1に示される核酸番号 1から 1698で示される塩基配列を有する GalNAcひ 2, 6 -シ アル酸転移酵素遺伝子を提供するものである。 さらに、 上記の6&1^じひ2,6-シァ ル酸転移酵素遺伝子を含む組み換えベクター、 およびその一態様であるプラスミ ド; I CEB- 3034、 上記の組み換えベクターにより形質転換された形質転換体、 並び に、 配列表の配列番号 1に示されるアミノ酸配列のアミノ酸番号 233 - 566 により 特定される Ga 1 NAc a 2, 6-シァル酸転移酵素活性ドメインも提供される。
GalNAcひ 2, 6-シアル酸転移酵素 P-B1 は、 蛋白に直接結合した N—ァセチルガ ラクトサミンの 3位の水酸基の置換の有無にかかわらずシアル酸を 6位に転移す る作用を有している。 このため NeuAc 2, 6GalNAc-蛋白の構造ができやすいが、 これによつてこの糖鎖の伸長は終結してしまう。 従って、 より長い糖鎖が望まし レ、場合には糖鎖が十分伸ぴ切つた後に初めてこの酵素を使うように糖鎖合成シス テムを設計する必要がある。 このような理由から、 3位の水酸基が置換されずに 蛋白質にひグリコシド結合している N—ァセチルガラクトサミンに対してはシァ ル酸の転移を行なわず、 一方、 3位の水酸基がガラクトースまたは還元末端側に ガラクトースを有する糖鎖で置換されている場合にのみ、 蛋白質にひグリコシド 結合している N—ァセチルガラクトサミンの 6位にシアル酸を転移させるシアル 酸転移酵素は極めて有用である。
そこで本発明者は、 ニヮトリ精巣から上記の特徴を有する GalNAcひ 2, 6-シアル 酸転移酵素をコードする cDNAをクローニングし、 配列表の配列番号 3に示される アミノ酸配列により特定される GalNAcひ 2, 6-シアル酸転移酵素 P- B3 に係る発明 を完成した。 すなわち本発明は、 上記の0&1^(; 2,6-シァル酸転移酵素 P- B3 の ァミノ酸配列をコードする GalNAcひ 2, 6-シアル酸転移酵素遺伝子、 およびその一 態様である配列表の配列番号 3に示される核酸番号 1から 1212で示される塩基配 列を有する GalNAc 2, 6-シアル酸転移酵素遺伝子を提供するものである。 さらに 上記の GalNAcひ 2, 6-シアル酸転移酵素遺伝子を含む組み換えべクタ一、 およびそ の一態様であるプラスミ ド I CEB3-T20、 ならびに上記組み換えベクターにより形 質転換された形質転換体が提供される。
さらに本発明者は、 上記の GalNAcひ 2, 6-シアル酸転移酵素の構造のうち活性に 係わる部分 (活性ドメイン) を含む細胞外分泌型の蛋白を提供することを目的と して研究を行った結果、 上記の GalNAc 2, 6-シアル酸転移酵素の活性ドメインで あるポリべプチド部分を特定することに成功し、 該ポリべプチド部分とシグナル ぺプチドとを含む細胞外分泌型の蛋白であって GalNAcひ 2, 6-シアル酸転移を触媒 する蛋白に係る発明を完成するに至った。 その一態様として、 配列表の配列番号 2に示されるアミノ酸配列により特定される蛋白 SB-690が提供される。 また、 上記蛋白をコードする遺伝子、 およびその一態様として配列表の配列番号 2に示 される核酸番号 1から 1065で示される塩基配列を有する遺伝子、 ならびに上記遺 伝子を含む組み換えベクター、 およびその一態様であるプラスミ ド pcDSB-690が 提供される。 さらに上記の組み換えベクターにより形質転換された形質転換体、 および上記形質転換体を培養し、 培養物から上記の蛋白を採取することを特徴と する上記蛋白の製造方法も提供される。
また本発明者は、 不溶性形態のマウス Gal /S l, 4GalNAc ひ 2, 6-シアル酸転移酵 素を大腸菌中で発現させた後、 該酵素を尿素で抽出して至適条件で脱変性するこ とにより、 高度に活性の回復した Gal ;S l, 4GalNAc ひ 2, 6-シアル酸転移酵素を製 造できることを見出し本発明を完成した。 すなわち本発明は、 シアル酸転移酵素 の製造方法であって、 以下の工程:(a) シアル酸転移酵素を微生物中で発現させ る工程;(b) 菌体内に蓄積するシアル酸転移酵素を含む蛋白凝集物または沈殿物 から該酵素を約 5〜9 の尿素により抽出する工程;(c) 上記工程 ) で得られ た抽出物を脱変性用組成物で希釈して約 1〜4 の尿素を含む 1次希釈物を得る 工程;(d) 上記工程 (c) で得られた 1次希釈物をさらに脱変性用組成物で希釈し て約 0. 5 〜2 M の尿素を含む 2次希釈物を得る工程;および (e) 上記工程 (d) で 得られた 2次希釈物から透析により尿素を除去して脱変性シアル酸転移酵素を得 る工程を含む方法を提供するものである。 図面の簡単な説明

本発明の GalNAc 2, 6-シアル酸転移酵素の最も好ましい例として GalNAcひ 2, 6- シアル酸転移酵素 P-B1 および P-B3が提供される。 以下、 本明細書において本 発明の酵素の一例として GalNAcひ 2, 6-シアル酸転移酵素 P- B1 および P- B3 につ いて詳細に説明するが、 本発明の GalNAcひ 2, 6-シアル酸転移酵素は GalNAcひ 2, 6 - シアル酸転移酵素 P- B1 または P-B3 に限定されることはなく、 本発明により初 めて明らかにされた GalNAcひ 2, 6-シアル酸転移酵素 P- B1及び 又は P- B3の活性ド メイン、 あるいはそのァミノ酸配列の一部を改変ないし修飾した GalNAc 2, 6-シ ァル酸転移酵素活性ドメインを有する GalNAc a 2, 6-シアル酸転移酵素は、 レ、ずれ も本発明の範囲に包含される。 このような活性ドメインの好ましい例としては、 例えば、 配列表の配列番号 1に開示したァミノ酸配列のァミノ酸番号 233-566 に より特定される GalNAc 2, 6-シアル酸転移酵素活性ドメインを挙げることができ る。
GalNAc 2, 6-シアル酸転移酵素 P-B1 および GalNAc ひ 2, 6-シアル酸転移酵素 P-B3をコードする cDNAの単離方法は、 それぞれ、 以下の実施例に詳細に説明され ている。 もっとも、 GalNAc 2, 6-シアル酸転移酵素 P-B1 または P- B3 をコード する cDNAの単離方法は上記方法に限定されることはなく、 当業者は下記の実施例 に記載された方法を参照しつつ、 あるいは、 この方法を適宜修飾ないし変更する ことにより、 容易に目的の cDNAを単離することができる。.また、 本発明の目的を 達成するためには、 配列表の配列番号 1ないし 3に記載された塩基配列を合成し て用いることも可能である。
配列表の配列番号 1に記載された GalNAcひ 2, 6-シアル酸転移酵素 P-B1をコード する DNA及び配列番号 3に記載された GalNAcひ 2, 6-シアル酸転移酵素 P-B3をコー ドする DNA は、 それぞれ本発明の好ましい態様であるが、 本発明の GalNAcひ2, 6 - シアル酸転移酵素 P-B1 をコードする DNA または GalNAcひ 2, 6-シアル酸転移酵素 P - B3をコードする DNA はこの特定の態様に限定されることはなく、 本発明により 明らかにされた GalNAcひ 2, 6-シアル酸転移酵素 P-B1 または GalNAcひ 2, 6-シアル 酸転移酵素 P- B3 のアミノ酸配列をコードする DNA は全て本発明の範囲に包含さ れる。 例えば、 配列表の配列番号 1に開示したアミノ酸配列の 233- 566 により特 定される GalNAcひ 2, 6-シアル酸転移酵素活性ドメインをコードする DNA は本発明 の好ましい態様である。 また、 配列表の配列番号 1に開示した核酸配列の核酸番 号 699- 1698により特定される DNA は本発明の特に好ましい態様である。
本発明の6&1^じひ2,6-シァル酸転移酵素、 例えば P- B1または P-B3は、 発現後に 細胞内に留まり、 細胞外に分泌されないことがある。 また、 細胞内濃度が一定以 上になると、 酵素の発現量が低下する可能性がある。 上記の GalNAcひ 2, 6-シアル 酸転移酵素 P- B1 または P-B3 の GalNAcひ 2, 6-シアル酸転移活性を有効に利用す るために、 本酵素の活性を維持し、 かつ発現時に細胞から分泌される可溶性形態 の蛋白を製造することができる。 このような蛋白として、 上記の GalNAcひ 2, 6-シ アル酸転移酵素 P- B1 または P-B3 の GalNAcひ 2, 6-シアル酸転移酵素活性に関与 泌型の蛋白であって GalNAc 2, 6-シアル酸転移を触媒する蛋白を挙げることがで さる。
これまでにクローニングされたシアル酸転移酵素は、 他のグリコシル—トラン スフ ラーゼと同様のドメイン構造を有している。 すなわち、 NH2 末端の短い細 胞質中尾部、 疎水性のシグナルアンカードメイン、 蛋白分解感受性を有するステ ム領域、 および C00H-末端の大きな活性ドメインを有する (Paulson, J. C. and Col ley, K. J. , J. Biol. Chem. , 264, 17615-17618, 1989) 。 本発明の GalNAcひ 2, 6-シアル酸転移酵素 P-B1 の経膜ドメインの位置を調べるために、 カイト及び ドゥーリ トル (Kyte, J. and Dool i tt le, R. F. , J. Mol. Biol. , 157, 105-132, 1982) の方法に従って作成した疎水性分布図を利用することができる。 また、 活 性ドメイン部分の推定には、 各種のフラグメントを導入した組換えプラスミ ドを 作成して利用することができる。 このような方法の一例は本明細書の実施例に詳 細に記載されている力 経膜ドメインの位置の確認や活性ドメイン部分の推定方 法は、 この方法に限定されることはない。 例えば、 6&1 じひ2,6-シァル酸転移酵素 P- B1 または P- B3 の活性ドメインで あるポリべプチド部分とシグナルべプチドとを含む細胞外分泌型の蛋白の製造の ためには、 例えばシグナルべプチドとして免疫グロプリンシグナルべプチド配列 を用い、 GalNAc 2, 6-シアル酸転移酵素 P- B1または P-B3の活性ドメインに対応す る配列を該シグナルぺプチドにインフレーム融合させればよい。 このような方法 として、 例えば、 ジョブリンらの方法 (Jobl ing, S. A. and Gehrke, し., Nature (Lond. ), 325, 622-625, 1987)を利用することができ、 本明細書の実施例には、 0&1^じひ2,6-シァル酸転移酵素 P-B1 について、 その具体的方法が詳細に説明さ れている。 もっとも、 シグナルペプチドの種類やシグナルペプチドと活性ドメィ ンの結合方法は上記方法に限定されることはなく、 当業者は、 GalNAc «2,6^ 7 ル酸転移酵素、 このましくは GalNAcひ 2, 6-シァル酸転移酵素?-81または P-B3 の 活性ドメインであるポリぺプチド部分を適宜選択することができるし、 それらを 利用可能な任意のシグナルぺプチドと適宜の方法により結合することにより細胞 外分泌型の蛋白を製造することができる。 このような蛋白の最も好ましい例とし て、 本発明の蛋白 SB-690を挙げることができる。
本発明の別の態様によれば、 シアル酸転移酵素の製造方法であって、 以下のェ 程:(a) シアル酸転移酵素を微生物中で発現させる工程; (b) 菌体内に蓄積する シアル酸転移酵素を含む蛋白凝集物または沈殿物から該酵素を約 5〜9 Mの尿素 により抽出する工程;(c) 工程 ) で得られた抽出物を脱変性用組成物で希釈し て約 1〜4 Mの尿素を含む 1次希釈物を得る工程;(d) 工程 (c) で得られた 1次 希釈物をさらに脱変性用組成物で希釈して約 0. 5〜2 の尿素を含む 2次希釈物 を得る工程;および (e) 工程 (d) で得られた 2次希釈物から透析により尿素を除 去して脱変性シァル酸転移酵素を得る工程を含む方法が提供される。 上記のとお り、 シアル酸転移酵素群は共通のドメイン構造を有しているので、 本発明の製造 方法は、 あらゆる型のシアル酸転移酵素の製造に適用することが可能である。 例 えば、 本発明の0&1^じ 2,6-シァル酸転移酵素ゃ Gal S l, 4GalNAc ひ 2, 6-シアル 酸転移酵素などは、 本発明の方法で好適に製造可能である。
上記発明の一態様は、 工程 (b) において約 8M の尿素を用い、 工程 (c) におい て約 2〜3 Mの尿素を含む 1次希釈物を得、 工程 (d) において約 1〜2 の尿素 を含む 2次希釈物を得、 さらに工程 (e) において 2次希釈物を 2価のカチオンの 存在下で透析する方法である。 また、 上記発明の別な態様は、 工程 ) において 約 8Mの尿素を用い、 工程 (c) において 1次希釈の後に 12時間以上 4°Cで放置 して約 2〜3 Mの尿素を含む 1次希釈物を得、 工程 (d) において 2次希釈の後に 48時間以上放置して約 1〜2 の尿素を含む 2次希釈物を得、 さらに工程 (e) に おいて 2次希釈物を 2価のカチオンの存在下で透析する方法である。 また、 上記 工程 (c) で用いる脱変性用組成物が 1〜2M尿素, 20 mM MOPS- NaOH, 0. 5 NaCl, 20 mM ラクトース, 0. 5 mM EDTA (pH 7. 0)を含み、 工程 (d) で用いる脱変性用組 成物が 20 mM MOPS-NaOH, 0. 5 NaCl, 20 mMラクトース, 0. 5 mM EDTA (pH 7. 0) を含む方法も好ましい方法である。
本発明のシアル酸転移酵素の製造方法の第一工程は、 シアル酸転移酵素を微生 物中で発現させる工程である。 このような目的のためには、 すでにクローニング されたシアル酸転移酵素遺伝子を利用することができる。 シアル酸転移酵素をコ —ドする cDNAとしては、 例えば、 本発明の6&^ じ 2,6-シァル酸転移酵素をコ一 ドする cDNAのほか、 Gal S L 4GlcNAc ひ 2, 6-シアル酸転移酵素 (Gal S4GlcNAc- ひ 6ST)をコードする cDNA (上記の Weinstein ら、 Grundmann ら、 Bastら、 および Hamamotoらの文献参照) 、 Gal S l, 3(4)GlcNAcひ 2, 3-シアル酸転移酵素 (Gal 3 (4)GlcNAc_ひ 3ST)をコードする cDNA (上記の Wenら、 および Ki tagawa らの文献 参照) 、 6&1 /81,36&1^じ &1 /31,4^じ^(:ひ2,3-シァル酸転移酵素をコ一ドする cDNA (上記 Sasaki らの文献参照) 、 Gal S l, 3GalNAc ひ 2, 3-シアル酸転移酵素 (Gal S 3GalNAc -ひ 3ST)をコードする cDNA (上記 Gi l lespieらの文献および特表平 5-504678号公報; ならびに Leeらの文献参照) などを用いることができる。 これ らの核酸配列中に存在するシ了ル酸転移酵素遺伝子を天然型酵素の発現のために そのまま利用してもよい。
また本発明では、 上記の天然型シアル酸転移酵素の他、 天然型シアル酸転移酵 素のポリぺプチド配列の一部が除去あるいは修飾された非天然型のシアル酸転移 酵素を微生物中で発現させてもよい。 例えば、 シアル酸転移酵素は NH2-末端領域 に位置する疎水性セグメント (経膜領域) を有しているので、 この疎水性セグメ ントを除去した可溶性形態のシアル酸転移酵素を微生物中で発現させることが好 ましい。 さらに、 疎水性セグメントおよびサイトゾル ·セグメントを除去してお くことも好ましい。
シアル酸転移酵素の発現用の組み替えベクターを製造するために、 例えば PCR 法によつて天然型シァル酸転移酵素遺伝子の全配列あるいはその一部の領域を選 択的に増幅することができる。 例えば、 開始コドンとクローニング 'サイトを有 し、 サイトゾル · ドメインと経膜ドメインを欠くシアル酸転移酵素遺伝子 (PCR断 片) を容易に製造することができる。 このようなシアル酸転移酵素遺伝子は開始 コドンとクローニング ·サイトを有しているので微生物発現用ベクターの導入に 好適であり、 天然型シアル酸転移酵素のポリぺプチド配列の一部が除去された非 天然型のシアル酸転移酵素をコードしているので、 微生物中で非天然型の可溶性 シアル酸転移酵素を発現させるので好ましい。 .
本発明の方法では、 シアル酸転移酵素の発現のために、 例えば大腸菌等の微生 物を用いることができる。 このような微生物を形質転換するために好適な微生物 発現用ベクターは、 当業者により適宜選択されうる。 例えば、 微生物として大腸 菌 JM109CDE3) 等を用いた場合には、 pET3b (Studier, F. W. et aし Method. Enzymol. , 185, pp. 60- 89, 1990)等の微生物発現用ベクターを用いることができ る。 上記のシアル酸転移酵素遺伝子を微生物発現用ベクターに導入する方法、 お よび微生物を上記の組み替えべクタ一により形質転換する方法は、 レ、ずれも当業 者に周知である。
形質転換体の培養は、 形質転換された微生物を培養する方法として当業者に周 知の方法により行うことができる。 目的のシアル酸転移酵素を微生物菌体内で効 率的に発現させるために、 例えば形質転換体の対数増殖期に T7-RNAポリメラ一ゼ を誘導することにより、 組み替え蛋白の産生を開始することができる。 このよう にして得られる形質転換体の菌体内には天然型シァル酸転移酵素あるいは非天然 型シアル酸転移酵素が大量に発現されており、 通常、 シアル酸転移酵素は蛋白凝 集物あるいは沈殿物を形成している。
本発明の方法の第二工程は、 菌体内に蓄積したシアル酸転移酵素を含む蛋白凝 集物または沈殿物からシアル酸転移酵素を 5〜10 Mの尿素により抽出する工程で ある。 シアル酸転移酵素を含む蛋白凝集物または沈殿物を菌体外に露出させて分 離するためには、 培養した形質転換体を例えばリゾチームおよび Triton X- 100等 で処理した後、 遠心処理により不溶性分画を集めればよい。 その後に、 沈殿をバ ッファー (例えば、 10 mM Tri s-HCL pH 7. 4) に蛋白濃度が l〜10mg/ml 程度と なるように懸濁し、 尿素による抽出操作を行う。
例えば、 上記の懸濁液に最終濃度が 5-10M、 好ましくは 8Mとなるように固形尿 素を加え、 沈殿を 15分間〜 2時間、 好ましくは 30分間にわたり 4〜25で、 好まし くは 10°Cで抽出する。 特定の理論に拘泥するわけではないが、 上記の抽出液に含 まれるシアル酸転移酵素の疎水性部分が尿素の作用によりが露出し、 その結果、 シアル酸転移酵素が可溶化し T蛋白凝集物または沈殿物から抽出される。
その後、 抽出液を例えば 12, 000 x gで 15分間遠心して沈殿物を除去するこ とにより、 変性されたシアル酸転移酵素を含む抽出液を得ることができる。 この 抽出的は、 一般に 0. 5 mg/ml程度の蛋白を含んでいる。 例えば、 抽出に 5. 7M尿 素を用いた場合には、 約 80%程度の蛋白を回収することができる。 また上記の抽 出に際して、 それぞれの最終濃度が 0. 3Mおよび 20 mMとなるように、 NaCl と Tris-HCl (pH 7. 4) を加えておくことが好ましい。 なお、 抽出操作の一例は以下 の実施例に詳細に説明されている。
上記の抽出物に含まれるシアル酸転移酵素は疎水性部分が露出しており、 高次 構造が損なわれているので、 本発明の方法では、 第三工程として上記抽出物に含 まれるシアル酸転移酵素の脱変性を行う。 本明細書において、 脱変性とは、 抽出 過程で損なわれた蛋白の高次構造を復元して酵素活性の一部または全部を回復さ せることをいう。 この工程は、 脱変性用組成物を用いて上記抽出物を段階的に希 釈し、 尿素濃度を徐々に低下させることにより効率的にシアル酸転移酵素の脱変 性を行うことを特徴としている。
脱変性工程は、 例えば、 上記の抽出物を脱変性用組成物で希釈して 1〜4 M の 尿素を含む 1次希釈物を得る工程; 1次希釈物をさらに脱変性用組成物で希釈し て 0. 5〜2 Mの尿素を含む 2次希釈物を得る工程;および 2次希釈物から透析に より尿素を除去して脱変性シアル酸転移酵素を得る工程を含むものである。
その好ましい態様は、 上記の抽出物を脱変性用組成物で希釈して 2〜3 Mの尿 素を含む 1次希釈物を得る工程; 1次希釈物をさらに脱変性用組成物で希釈して 1〜2 の尿素を含む 2次希釈物を得る工程;および 2次希釈物を 2価のカチォ ンの存在下で透析して尿素を除去し脱変性シアル酸転移酵素を得る工程を含む方 法である。 また、 さらに好ましい態様は、 上記抽出物を脱変性用組成物で希釈し た後に 12時間以上 4°Cで放置して 2〜3 Mの尿素を含む 1次希釈物を得る工程; 1次希釈物をさらに脱変性用組成物で希釈した後に 48時間以上放置して 1 〜2 M の尿素を含む 2次希釈物を得る工程;及び 2次希釈物を 2価のカチオンの存在下 で透析して尿素を除去し脱変性シアル酸転移酵素を得る工程を含む方法である。 脱変性用組成物としては、 例えば、 2 M尿素, 20 mM MOPS-NaOH (MOPS : 3- モ ルホリノプロパンスルホン酸) (pH 7. 0), 0. 5M NaCL 10 mMラクトース, 0. 5 mM EDTAや、 2 尿素, 20 mM Tris-HCl , 0. 3 NaCl, 20 mMラクトース, 0. 5 mM EDTA (pH 7. 4)を用いることができ、 さらに、 後者の組成物の各成分を、 例えば 20 mM Tri s-HCl (pH8. 0) ; 20 mM MOPS-NaOH (pH7. 0) ; 20 mM MES-NaOH (pH6. 0) ( ES: 3-モルホリノエタンスルホン酸) ; 0. 5 M NaCl; 0. 1 M NaCl ; または 1M尿素の ように変更した組成物を用いてもよい。 また、 尿素またはラクトースを含まない 組成物を用いてもよい。 これらのうち、 2 M尿素, 20 mM MOPS-NaOH, 0. 5M NaCl, 20 ラクト一ス, 0. 5 mM EDTA (pH 7. 0)を用いることが好ましレ、。 NaClの濃度 が 0. 1M を下回る場合、 あるいは pHが 9を上回る場合には、 脱変性効果が低下 するので好ましくない。 一般に、 脱変性用組成物を添加した後の塩濃度を 0. 3〜 0. 5、 pHを 6〜8 とすることが好ましい。
最初の希釈操作は、 上記の脱変性用組成物を用いて、 上記抽出物の最終蛋白濃 度が 0. 01 〜 0. 05 mg/ml 、 好ましくは約 0. 02 mg/ml となるような 1次希釈物 を得る操作である。 例えば、 上記抽出物を 10〜 40倍、 好ましくは約 20倍程 度に希釈すればよく、 尿素の濃度を 1〜4 M、 好ましくは上限を 3M、 あるいは下 限を 2M程度とする。 希釈操作は一般に 4°Cで行うことが好ましい。 この 1次希釈 混合物を、 好ましくは 4°Cで 12時間以上、 特に好ましくは約 12時間放置して、 脱変性を徐々に開始させる。
上記の 1次希釈物を、 好ましくは尿素を含まない同容量の脱変性用組成物で希 釈して、 尿素濃度を半分程度にする 2次希釈を行う。 この希釈操作により 2次希 釈物中の尿素濃度を 0. 5〜2 程度、 好ましくは上限を 2M、 あるいは下限を 1M程 度 (例えば 1〜2M) 、 特に好ましくは約 1. 2M程度にすべきである。 この 2次希 釈物を約 4°Cで 40時間〜 2週間、 好ましくは 48〜72時間、 特に好ましくは約 48時 間放置して脱変性を徐々に進行させる。
その後、 脱変性を完全に行うために、 上記の 2次希釈物を、 例えば尿素を含ま ない脱変性用組成物に対して透析し、 残存する尿素を完全に除去する。 透析操作 は、 例えば 4 で 48 時間程度行うことができ、 透析液としては、 上記の脱変性 用組成物の他、 シアル酸転移酵素が安定に保存される緩衝液等を用いることがで さる。
また、 上記の 1次希釈および 2次希釈工程、 並びに最終の透析工程を 2価の力 チオンの存在下で行うことにより、 さらに脱変性効果を高めることができる。 2 価のカチオンとしてはマグネシウムイオン、 マンガンイオン等を挙げることがで き、 これらは例えば 1〜10 mM程度、 好ましくは 5 mM程度の濃度で用いること ができる。 上記の透析工程を 2価のカチオンの存在下で行うことが特に好ましい。 なお、 最終の透析工程によって尿素を完全に除去する前にジチオスレィトールや メルカプトェタノ一ル等の還元剤を配合すると、 酵素活性が失われる場合がある が、 尿素を完全に除去することにより酵素は還元剤に対する抵抗性を回復し、 シ アル酸転移酵素活性を示すようになる。
以下、 本発明を実施例により具体的に説明するが、 本発明の範囲はこれらの実 施例に限定されることはない。
実施例
(A) 6&1^じひ2,6-シァル酸転移酵素 P- B1 の製造
GalNAcひ 2, 6-シアル酸転移酵素 P-B1 の cDNAクローンを得るために、 2個の縮 重ォリゴヌクレオチド (ST-107及び ST- 205) による PCR を、 铸型としてニヮトリ 胚 cDNAを用いて行った。 約 150bpの望ましいサイズのフラグメントを得た。 PCR 組換産物の中で、 CEB1と呼ばれるクローンは、 Ga 1 β 4G1 cNAc- 6STRL(180- 225残 基) 、 Gal 33(4)GlcNAc-ひ 3STRLC158- 203残基) 及び Gal β 3GalNAc- a 3STPS 144 - 189残基) の既知のシァリルモチーフとは異なる独特のアミノ酸配列を有してい た。 CEB1のシァリルモチーフである Ga 1 4G 1 cNAc- a 6STRL、 Gal 53(4)GlcNAc - ひ 3STRL及び Gal 83GalNAc - 3STPS と CEB1の相同性は、 それぞれ 56%、 58%及 び 60%であった。
CEB1からの cDNA ィンサートを用いての 6日齢ニヮトリ胚 cDNA ライブラリ一 のスクリーニングを行い、 いくつかの cDNA クローンを同定した。 そのうちクロ ーン I CEB- 3043は 2. 7 kb インサートを含んでいた (第 1図) 。 別のオーバーラ ッビングクローンを得るために、 ランダムプライムド cDNAライブラリ一を; I CEB - 3043の 5 ' -末端の 0. 8 kb EcoRI-Bgl l l フラグメントとハイブリダィゼーショ ンすることにより再スクリーニングを行った。 この cDNA ライブラリ一から 15個 のクローンが単離された。 そのうちのクローン; I CEBHADはクローン; I CEB - 3043の 5 ' -末端と 160 bp に渡ってオーバーラップする 220 bp インサートを含んで いた。
これらの 2個の cDNA を結合したものは、 ヌクレオチド 1699 の TGA停止コド ンで終わる 1. 7 kbのオープン · リーディング'フレームを含むものであった。 ポ リ(A) から 23ヌクレオチド上流にあるポリアデニル化シグナル (AATAAA)は 3 '末 端に存在している。 このオープン ' リーディング'フレームの翻訳により、 通常 の開始配列を持つヌクレオチド 1のメチォニン残基 (Kozak, M. , NatureCLond. ), 308, 241-246, 1984)で開始し、 分子量 64, 781 の 566アミノ酸から成る蛋白で ぁる本発明の6&1^じひ2,6-シァル酸転移酵素 P-B1 (実施例において、 単に P - B1 と呼ぶことがある) が発現する。 本発明の6&1^じひ2,6-シァル酸転移酵素をコ一 ドする遺伝子を含む cDNA、 本発明の GalNAc 2, 6-シアル酸転移酵素をコードする 遺伝子である; I CEB-3043の塩基配列、 および本発明の GalNAc 2, 6-シアル酸転移 酵素 P- B1 のァミノ酸配列を配列表の配列番号 1に示す。
ポリメラーゼ連鎖反応法 (PCR)
PCR は Gal y64GlcNAc—ひ 6STRL(Weinstein, J. et al., J. Biol. Chem. , 262, 17735-17743, 1987)、 Gal 4G1 cNAc- a 6STHP (Grundmann, U. et al. , Nucleic Acids Res. , 18, 667, 1990)、 及び Gal 53GalNAc— 3STPS (Gi l lespie, W. et al. , J. Biol. Chem. , 267, 21004-21010, 1992)中の保存領域に由来する縮重プ ライマー [5 'プライマー ST107 : TGGGCCTTGGI KA/OAGGTGTGCTGTTG及び 3 'プラ イマ一 ST205 : AGGCGAATGGTAGTTTTTG(A/T)GCCCACATC]を用いて行った。 cDNAを得 るために、 3日齢のニヮトリ胚からの poly(A)リツチ RNA (2 g)を、 ォリゴ- dT プライマ一 (フアルマシア社) ; dATP、 dCTP、 dGTP、 及び dTTPをそれぞれ 1 mM; 並びに 2 U/ z l の RNase阻害剤'(プロメガ社) と共に、 50// 1 の 10 mM Tris-HCl (pH8. 3)、 50 mM KCU 1. 5 mM MgCl 2及び 0. 001%ゼラチン中 0でで 10分間イン キュペートした後、 100 / U のモロニ一 ·ネズミ白血病ウィルス逆転写酵素 (BRL) を加えてさらに 42°Cで 60分間ィンキュベ一トした。
上記反応液を 94°Cで 3分間加熱した後、 0.2 / g の poly(A)リツチ RNAから製 造した cDNAを、 50 / 1 中に 10 mM Tris-HCl (pH8. 3), 50 mM KC1、 1. 25 mM gCh, 0. 001%ゼラチン、 dATP、 dCTP、 dGTP及び dTTPをそれぞれ 200 /zM、 Taq DNA ポリ メラ一ゼ (プロメガ社) を 2U、 並びに 40 pmole の各 PCRプライマーを含む混 合液中における PCR実験に用いた。 35サイクルの PCR増幅を行い、 各サイクルは 96°C, 45秒間の熱変性; 50° 60秒間のァニ一リング;及び 72°C、 60秒間の伸長 とした。 PCR生成物を 3%ァガロースゲルで展開し、 150bp に相当する DNAフラ グメントをゲルから溶出し (キアエックス 'キット、 キアゲン社) 、 平滑末端化 し、 キナーゼ処理した後、 PUC119の Smal部位の中にサブクロ一ニングし、 最終的 に配列決定した。
cDNAライブラリ一の作成
全 RNAをグァニジニゥ厶チオシァネート法、 及びそれに続く 5. 7 CsCl溶液中 における遠心によりニヮトリ胚(6日齢) から調製した(Sambrook, J. , Molecular Cloning: a Laboratory Man謹 1, 2nd ed. Cold Spring Harbor Laboratory, Cold Spring Harbor, N. Y. 1989)。 poly (A)リツチ RNA はォリゴテツクス- dT30 (宝酒造) で精製し、 オリゴ- dT プライマー及びランダムプライマーに; I ZAPI I (ストラタジーン社) 及び cDNA合成キット (フアルマシア社) を用いた cDNAラ ィブラリーの作成に使用した。
cDNAライブラリーのスクリーニング
増幅された cDNAライブラリ一 (lxlO6 プラーク) をニヮトリ胚 PCRフラグメン 卜でスクリーニングした。 プラークを移したフィルターを、 5 X SSC、 0.2% SDS、 5 Xデンハルド溶液及び 10 〃g/ml変性サケ精子 DNA 中で 3 2P-放射性ラベル化 DNA プローブと 65°Cで 12時間にわたりハイブリダィズさせた。 その後、 2X SSC、 0. \% SDS中で 65°C、 20分で二回洗浄した。 単離されたファージクローンからブラ スミ ドを得るために、 ファージミ ドレスキューを; I ZAPI I クローニングキットの 製造元 (ストラタジーン社) の使用説明書に従って行った。 cDNAインサートは、 Bluescriptプラスミ ドとして直接切り取った。 プラスミ ドの作成はサムブルック ら (Sambrook, J. et al. , Molecular Cloning : a Laboratory Manual)によって 述べられている分子クローニング標準法を用いた。
DNA配列分析
上記のィンサートの DNA配列を、 T7-DNAポリメラーゼの铸型として一本鎖 DNA を用いるジデォキシ鎖停止法 (Sanger, F. et al. , Proc. Natl. Acad. Sci. USA, 74, 5463-5467, 1977)により決定した。 シーゲンシング反応及び電気泳動はォ一 トリード DNA シーゲンシングキット及び DNA シーケンサー (フアルマシア社) を 用いて行った。 一本鎖 DNA は、 ヘルパーファージ R408 (ストラタジーン社) で重 複感染させた後に大腸菌 XL-Blue (ストラタジーン社) から調製した。 配列デ一 夕は PC/Gene (ティジンシステムテクノロジ一社) を用いたコンピューターで分 折した。
ノーザン及びサザンプロッティング
遺伝子の存在を確認するために、 ニヮトリゲノム DNAのサザン ·プロット分析 を行った。 i CEB-201 の EcoRI cDNA インサートとのハイブリダィゼ一シヨンを 行い、 EcoRI及び BamHIで切断した DNAでは単一のバンド、 Hindi I I 及び Sacl で切断した DNAでは 2バンドが得られた。 この単純なハイブリダイゼーションパ ターンによれば、 クローニングされた cDNAが単一コピー遺伝子であることが明 らカヽである。
また、 胚発生過程における転写パターンを調べるために、 ノーザンプロットハ イブリダィゼ一シヨンを行った。 6, 8, および 10日齢のニヮトリ胚由来 R NAを 分析したところ、 3. 0 および 2.2 Kb の 2種の R NAが検出された。 3. 0 Kb転写 が優勢であり、 胚発生の全期間において常に発現していた。 低レベルの 2. 2 Kb転 写が 6日齢の胚に検出されたが、 8および 10日齢の胚からは消失していた。 ニヮ トリの脳、 心臓、 肝臓、 胚、 腎臓、 及び精巣から得た 10// g のポリ Aリツチ RNA を用いて遺伝子発現を分析したところ、 極めて低レベルの 3. 0および 4. 0 Kbの転 写が精巣に検出されたが、 他の組織からはほとんど検出されなかった。 各実験の 詳細は以下のとおりである。
ノーザンブロット用に、 ニゥトリ胚からの変性 poly(A)リツチ RNA 5 pt g をホ ルムアルデヒドーァガロースゲル上でサイズ分画化した後、 ハイボンド N+ ナイ ロンメンブレン (アマシャム社) 上にプロッティングさせた。 サザンブロット用 には、 ニヮトリ胚から調製した 7. 5 / g のゲノム DNA を制限酵素 EcoRI 、 BamHI 、 Hindi 11 及び Saclで切断し、 0. 6% ァガロースゲル上でサイズ分画化した。 電気 泳動の後、 ゲルを 0, 5 N NaOH及び 1. 5 M NaCl 中で変性 (30分間) させた後、 0. 5 Tris-HCl (pH7. 5) 及び 1. 5 M NaCl 中で中性化し (30分間) 、 DNA をハイ ボンド N+ ナイロンメンブレン上に移した。 ノーザン及びサザンフィルターは両 方とも 50%ホルムアミ ド、 5 x SSC、 5 xデンハルド溶液、 0. 5% SDS及び 10〃g/ml 変性サケ精子 DNA 中で 37で、 1時間にわたりプレハイブリダィズさせた後に、 プ レハイブリダィゼ一シヨンと同じ条件で 12時間、 3 -放射性ラべル化0^ プロ一 ブとハイブリダィズさせた。 プローブとしては、 マルチプライムラべリングシス テム (アマシャム社) でラベルした; I CEB201である 0. 6kb EcoRI cDNA インサ一 トを用いた。 フィルターを 2 x SSC及び 0. 1% SDS中で 65°C;、 10分で二回洗浄し、 次いで 0. 2 x SSC及び 0. 1% SDS中で 65°C、 30分で二回洗浄した後、 -70 でで約一 日間、 コダック XAR フイルムに感光させた。
上記のとおり解明された本発明のシアル酸転移酵素 P- B1 のアミノ酸配列は、 従来公知のシアル酸転移酵素のァミノ酸配列には観察されない以下の特徴を示す るものである。
(i) 従来クローニングされたシアル酸転移酵素は全て I I型膜蛋白であり、 それら は他のグリコシル—トランスフェラ一ゼと同様のドメイン構造を有する。 すなわ ち、 NH2 末端の短い細胞質中尾部、 疎水性のシグナルアンカー.ドメイン、 蛋白分 解感受性を有するステム領域、 及び C00H-末端の大きな活性ドメインを有してい る。 一方、 本発明のシアル酸転移酵素 P-B1 は、 大きなステム領域 (または中間 領域) を有するものである。 -
(i i)本発明のシアル酸転移酵素 P-B1 は、 PEST領域 (233- 258残基) を有している 。 細胞内半減期が 2時間未満の蛋白のアミノ酸配列は、 プロリン、 グルタミン酸 、 セリン及びスレオニン残基が多い領域 (PESTと呼ばれる: Rogers, S. et al. , Science, 234, 364-368, 1986) を 1個以上含むことが知られている。 これらの PEST領域は、 一般にプラスの電荷を持つ数個のアミノ酸を含む集団を側面に擁し ている。 従来公知のシアル酸転移酵素はこの領域を有していない。
(i i i) 8アミノ酸 (SSSXVSTC)から成る領域が 2力所、 247-254残基及び 330- 337 残基に見出された。 他の蛋白のジーンバンクデータベースの検索からこの配列に 類似する配列は明らカ、にされなかった。
従来より公知のシアル酸転移酵素は、 驚くほど組織特異的発現を示すものであ つたが、 これは細胞型特異的糖鎖構造の存在に相関しているものと考えられてい る (Paulson, J. C. and Col ley, K. J., J. Biol. Chem. , 264, pp. 17615-17618, 1989) 。 ノーザン ·プロッティングの結果によれば、 シアル酸転移酵素 P-B1 の mRNAの発現パターンが変化することが明らかである。 シアル酸転移酵素 P- B1の遺 伝子から 3種の異なるサイズの mRNA (4. 0、 3. 0及び 2. 2 kb)が転写されること は、 それらが 0&1 6 1 , 461じ^(;- 2,6-シァル酸転移酵素 &1 ^ 401じ^じ-ひ63丁1^) 及び Gal /5 1, 3(4)GlcNAc a 2, 3-シアル酸転移酵素 (Gal 33(4) GlcNAc- ひ 3STRL) (Weinstein. J. et al. , J. Bi ol. Chem. , 262, 17735-17743, 1987; 及び Wen, D. X. et al. , J. Biol. Chem. , 267, 21011-21019, 1992)に観察されたような、 選択的スプライシングメカニズム及び選択的プロモーター利用メカニズムを通じ て発現されることを示唆している。 このことは、 シアル酸転移酵素 P- B1 に対し ては単一コピーの遺伝子しか存在しないことがサザン ·ハイブリダイゼ一ション により示されていることからも明らかである。
(B) 可溶性形態の蛋白 SB- 690 の製造
本発明の GalNAcひ 2. 6-シアル酸転移酵素 P-B1 の GalNAc 2, 6-シアル酸転移活 性を利用するため、 本酵素の活性を維持し、 かつ、 発現時に細胞から分泌される 可溶性形態の蛋白 SB-690 を製造した。
これまでにクローニングされたシアル酸転移酵素は、 他のグリコシルートラン スフヱラーゼと同様のドメイン構造を有している。 すなわち、 NH2 末端の短い細 胞質中尾部、 疎水性のシグナルアンカードメイン、 蛋白分解感受性を有するステ 厶(stem)領域、 及び C00H-末端の大きな活性ドメインを有する。 本発明の GalNAc a 2, 6-シアル酸転移酵素の経膜ドメインの位置を調べるために、 疎水性分布図を カイト及びドゥーリ トル (Kyte, J. and Dool i ttle, R. F. , J. ol. Biol. , 157, 105-132, 1982)の方法に従って翻訳された配列から作製した (第 2図) 。 この結 果、 本発明の GalNAc a2, 6-シアル酸転移酵素 P-B1 の疎水性経膜ドメインはアミ ノ酸番号 17から 37の 21ァミノ酸残基であることが示唆された。
上記のとおり、 GalNAcひ 2, 6-シアル酸転移酵素の疎水性シグナルアンカ一ドメ インはアミノ酸残基 17から 37に位置している。 铸型として pSB-BGLを含むインビ トロの翻訳 Z転写系では蛋白 (33 kDa)が合成されるにもかかわらず、 pcDSB-BGL でトランスフヱクシヨンした細胞からの培地は有意の活性を持たないことから、 233 から 269 残基が酵素活性に対して何らかの必須残基を含むことが明らかであ り、 活性ドメインは 233-566の周辺と推定された (第 3図) 。 これはクロ一ニン グされた他のシアル酸転移酵素のものに匹敵するサイズである。 上記の活性ドメ ィンを含む可溶性蛋白を作るために、 P- B1の推定活性ドメインに対応する配列を 免疫グロプリンシグナルぺプチド配列にインフレーム融合させた(Jobl ing, S. A. and Gehrke, L., Nature (Lond. ), 325, 622-625, 1987)。 以下に実験の詳細を示 す。
ベクタ一プラスミ ド PUGS を、. pBluescript SK(+) プラスミ ドの Pst l- Xholフ ラグメントを 117 bp の合成 DNA フラグメントで置換することにより作成した。 このフラグメントは、 5 '末端に合成 Pst l部位を持つ 43 bp のアルファルファモ ザイクウィルス(Jobl ing, S. A. and Gehrke. L., NatureCLond. ) 325, 622-625, 1987) の 5 ' - 非翻訳 (untranslated)リーダー配列を含んでおり、 この配列には 3 '末端に 17 bp の合成 EcoRI 、 Bgl l l 及び Xhol クロ一ニング部位を持つマウ ス免疫グロブリン M重鎖シグナルペプチド配列(57 bp) (Boersch-Supan, . E. et al., J. Exp. Med. 161, 1272-1292, 1985) が続いている。 このフラグメントの ヌクレオチド配列は 5 ' -CTGCAGGGTTTTTATTTTTAATTTTCTTTCAAATACTTCCACCATGAAA TTCAGCTGGGTCATGTTCTTCCTGATGGCAGTGGTTACAGGGGTCAATTCAGAATTCCAGATCTCGAG-3 ' しあ d o
本発明の GalNAc a2, 6-シアル酸転移酵素をコードする遺伝子である; I CEB- 3043 を EcoRVで部分切断し、 1. 8 kbのフラグメントを pBluescript SK(+)の EcoRV部 位にサブクローニングして、 pCEB-1800 を生産した。 このクローンは; I CEB- 3043 の 0. 8 kbの 3 ' -非翻訳領域を欠くものである。
0&1^じ 2,6-シァル酸転移酵素 P-B1 の活性ドメインを 5 ' - プライマ一とし てその中央部分に合成 EcoRI 部位を持つ 5 ' -AGGGCTGCTGAATTCACTGAGCCACAG-3 ' (ヌクレオチド 679-708) 、 および 3 'ユニバーサル M13シーゲンシングプライ マー、 さらに鐯型として pCEB-1800 を用いた PCR によって生産した。 この PCR産 物を EcoRI 及び Xholで切断した後、 上記べクタ一プラスミ ド pUGSの EcoRI/XhoI 部位に結合させてプラスミ ド pSB- 690 を得た。 このプラスミ ドには、 ;i CEB- 3034 遺伝子のぅち6&1^(;ひ2,6-シァル酸転移酵素 P- B1 の活性ドメインをコードする 領域が免疫グロプリンシグナル配列の 3 '末端にインフレーム融合された配列が 含まれるものである。 この融合フラグメントを Pst l及び Xholで pSB - 690から切り 取った後、 pcDSRひ発現ベクターの Pst l/Xhol部位中に挿入して pcDSB- 690を得 た。
対照として、 6&1^じ 2, 6-シァル酸転移酵素 P-B1 の活性ドメインの一部を欠 く蛋白である SB- BGLを以下のとおり製造した。 pCEB- 1800及び pUGSを Bgl l l で切 断し、 粘着末端を DNAポリメラーゼのクレノウ(Klenow)フラグメントを用いて充 塡した。 DNAポリメラーゼのクレノウフラグメントを熱変性させた (94°C、 20分 間) 後、 これらのプラスミ ドを Xholで切断した。 pCEB- 1800 からの 1. 0 kb フラ グメントをゲル精製し、 pUGSの平滑末端を持つ Bgl l l/Xhol部位中にサブクロー二 ングしてプラスミ ド pSB-BGL を得た。 pSB-BGLからの Pst l/Xholフラグメントを pcDSRひの発現べクタ一の Pst l/Xhol部位中でサブクローニングして pcDSB- BGL を得た。
上記の蛋白の発現は以下のように行った。 COS- 7細胞に DEAE-デキストラン法 (McCutchan, J. H. and Pagano, J. S. J. Nat l. Cancer Inst. 41, p. 351-357, 1968) を用いて 5 g のプラスミ ド DNA によりトランスフエクシヨンした。 48時 間のトランスフエクシヨンの後、 培地を回収し、 セントリコン 30フィルタ一 (ァ ミコン社) を用いて酵素アツセィ用に 10倍に濃縮した。 代謝ラベル用に、 COS細 胞 (60-圆培養皿) を Met- フリーの培地 (ダルベッコの改良イーグル培地及び 2%ゥシ胎児血清)(GIBC0)で洗浄した後、 同じ培地で 1時間ィンキュベートした。 細胞を 1. 5 ml の Met- フリーの培地中 10 MBq/皿のエクスプレス3 5 S プロティ ンラベリングミックス (デュポン一ニューイングランドヌクレア社) で 2時間、 パルスラベルした。 これらの細胞をその後 Met- フリーの培地で洗浄し、 ェクス プレスラベルなしの培地中で 5時間チェイスした。 分泌された蛋白を含む培地を 回収し、 10倍に濃縮した後、 SDS-ポリアクリルアミ ドゲル電気泳動、 次いでフル ォログラフィ一に掛けた。
発現した蛋白の酵素活性を以下のようにして測定した。 ァクセプ夕一としてォ リゴサッカライド及びグリコプロテインを用いたアツセィを、 総量が とな る 50 mM力コジル酸ナトリウムバッファ一 (pH 6.0) 、 50 M C P- [ ' C-]NeuAc (0. 9 Bq/pmol) 、 1 mg/ml ゥシ血清アルブミン、 2 mg/ml ァクセプター基質及び 1 PL \ 濃縮 COS細胞培地の存在下で行い、 30°Cで 2時間インキュベートした。 ィ ンキュペート時間終了時に、 アツセィ混合液 1〃1 をシリカゲル 60HPTLCプレー ト (メソレク社、 ドイツ) にのせた。 このプレートをエタノール: ピリジン: n -ブ 夕ノール:水:酢酸 (100: 10: 10:30:3) で展開し、 放射能活性を BAS2000 ラジ ォイメージアナライザー (富士写真フィルム製、 日本) で可視化して定量した。 原点に残った放射能はシアル化糖蛋白と見なした。
シアル化産物の同定は以下のように行った。 ァシァ口- BSMを pcDSB- 690 COS細 胞培地中で CMP- [ 1 4C]NeuAcで再シアル化して還元し、 /S—除去ォリゴサッカライ ドを製造した。 —除去はカールソンの方法 (Carlson, D. M. . J. Biol. Chem. , 243, 616-626, 1968) に従って行った。 ァシァロ-BSM (各 100/zg)を、 インキュ ベーシヨン時間が 12時間であることを除き、 上記と同じ条件下で pcDSB- 690 COS 細胞培地中で CMP-[ 1 4C- ]NeuAc でシアル化した。 反応を 0.5 M HC1中 1% リン夕 ングステン酸 500 1 を加えて停止し、 次いで 10, 000 xgで 5分間遠心した。 ペレットを同じリンタングステン酸溶液で一回洗浄し、 さらにメタノールで一回 洗浄した後、 0. 5 mlの 0. 05M NaOH及び 1M NaBH4 溶液中に溶解し、 45°Cで 30時間 ィンキュベートした。
インキュベーション時間の終了時に、 この溶液を酢酸で pH 6 に中和し、 凍結 乾燥した。 この乾燥産物を 50 ιι \ の水に溶解し、 水で平衡化及び溶出するセフ アデックス G-15 カラム (0.5 X 5 cm) 上のゲル濾過によって脱塩した。 放射能 活性のある分画はそれ以上精製せずに産物同定用の薄層クロマトグラフィ一に付 した。 異なる 2種類の展開溶媒を用い、 天然 BSMからの NeuAcひ 2, 6GalNAc-ォー ル及び Gl cNAc ^ 1, 3 [NeuAc a 2, 6] GalNAc-ォールと共泳動させた。 転移したシァリ ル残基の比率は 1 :0. 9 :0. 6であった。 シアル化 GalNAc- SerNAc を異なる 2種類の 展開溶媒系において NeuAc 2, 6GalNAc- SerNAcと共泳動させることにより、 本発 明の蛋白 SB-690が、 グリコプロテイン中の Ser又は Thr残基に直接結合してい る GalNAc に NeuAc a2, 6結合を形成することが示された。
pcDSB-690 でトランスフヱク小された細胞を培養した培地にはシアル酸転移酵 素活性が検出され、 pcDSB- 690により発現される本発明の蛋白 SB-690 がシアル 酸転移酵素活性を保持しつつ、 細胞外に分泌されていることが明らになった。一 方、 cDSB-BGLでトランスフエク卜された細胞を培養した培地からはシアル酸転移 酵素活性が検出されなかつた。
本発明の蛋白 SB- 690 の受容体特異性を pcDSB-690でトランスフヱクシヨンさ れた C0S-7細胞培養濃縮培地で調べた。 表 1に示すように、 ァシァロムチン、 フ エツイン及びアジア口フ ツインは良好な受容体として作用した。 注目すべきこ とに、 フヱツインはァシァロフヱツインよりも優れた受容体であることが示され た(Baubichon- Cortay, H. et al. , Carbohydr. Res. , 149, 209-223, 1986; お よび Brockhausen, I. et al. , Biochemistry, 29, 10206-10212, 1990) 。 他の グリコプロテイン、 オリゴサッ力ライド及びグリコリピッドは GalNAc- SerNAcを 除いて、 ァクセプターとして作用しなかった。 これらのデータは当ァクセプ夕一 部位がひ一グリコシド結合を通じてグリコプロテイン中の Ser又は Thr残基に直 接結合している GalNAc であることを示唆している。

-GalNAc 2 表中、 "0" は 1 pmole/hr/10^1 培地未満であることを示す。 従来クロ一ニングされたシァル酸転移酵素は Ga 1 -部分に対してのみァクセプ ター特異性を示すものであった。 一方、 本発明の GalNAc 2, 6-シアル酸転移酵素 P - B1および本発明の蛋白 SB-690は、 GalNAc-部分に対してァクセプ夕一特異性を 示すが Ga卜部分に対してはァクセプター特異性を示さないものである。 以下の 知見は、 本発明の GalNAcひ 2, 6-シアル酸転移酵素 P-B1 および本発明の蛋白 SB- 690力、 ひ 2, 6-結合を有する CMP-NeuAcを、 グリコプロテインの Thr/Serに 0- 結合している GalNAc-残基上に転移させる GalNAc 2, 6-シアル酸転移酵素活性 を有することを示すものである。
(i) COS細胞中の pcDSB- 690の発現により Thr/Ser残基に結合している GalNAc 部分のみに対するァクセプター特異性が明らかにされた。 一方、 試験した他の基 質に対しては検出可能な酵素活性は全く見られなかった (表 1) 。
(ii) ゥシ下顎腺ァシァロームチン及び GalNAc-SerNAc から得られたシアル化産 物はひ 2, 6結合を通じて GalNAc部分に結合しているシアル酸を有することが示さ れた。
2種類の型、 すなわちゥシ下顎腺型及び肝臓 (脳) 型の GalNAc-ひ 6STが報告 されており、 それらは異なるァグセプター特異性を持つ (Bergh, . E. et al., J. Biol. Chem. , 258, 7430-7436, 1983)。 前者の酵素は GalNAc、 Gal y31, 3GalNAc 及び NeuAcひ 2,3GalySl,3GalNAc に対して広範囲の特異性を持つが、 後者はグリ コプロテインの NeuAca2,3Gal Sl,3GalNAc部分に対してのみ特異性を持つ。 本 発明の GalNAcひ 2, 6-シアル酸転移酵素 P-B1 および本発明の蛋白 SB-690 のァク セプタ一特異性は、 前者の酵素と同様であることを示している。
ァシァロムチンのァクセプター部位の検討から NeuAcひ 2, 6GalNAc-Ser/Thr が 最も多い産物であることが示された。 し力、し、 ゥシ下顎腺ァシァロムチン中の糖 共役体の比率、 すなわち、 GalNAc- Ser/Thr、 GlcNAc^ 1, 3GalNAc-Ser/Thr および GalySl,3GalNAc-Ser/Thr がそれぞれ 65%、 25¾及び 5% に当たること(Tsuji, T. and Osawa, T., Carbohydr. Res., 151, pp.391- 402, 1986) を考慮すると、 本発明の GalNAc 2, 6-シアル酸転移酵素 P- B1 および本発明の蛋白 SB- 690 は、 以下のァクセブ夕一優先順位を有するものである: Gal Sl,3GalNAc- Ser/Thr > GlcNAc 3L3GalNAc-Ser/Thr > GalNAc - Ser/Thr。 —方、 ほとんど全ての放射能 活性が弱アルカリ処理によって解離し、 かつ、 フヱツインの方がァシァ口フェツ インよりも好ましい (表 1) という事実は、 ゥシ肝臓 (Bergh, M.E. et al., J. Biol. Chem. , 258, 7430-7436, 1983)、 およびラット脳 (Baubichon- Cortay, H. et al., Carbohydr. Res., 149, pp.209- 223, 1986) の GalNAc 2, 6-シアル酸転 移酵素について報告されているように、 NeuAcひ 2,3Gal>Sl,3GalNAc- Ser/Thr が Gal /81,3GalNAc- Ser/Thr よりも好ましい基質であることを示している。
GalNAc-SerNAc のシアル化はァシァロムチン上の対応する残基のシアル化より もはるかに遅い (表 1 ) 。 ブロックハウゼンらによれば (Brockhausen et al. , Biochemistry, 29, 10206-10212, 1990)、 最低 5アミノ酸の長さが有効なシンセ ターゼ活性にとって必要である。 本発明の GalNAcひ 2, 6-シアル酸転移酵素 P-B1 および本発明の蛋白 SB-690 も、 ペプチド部分に同様の直接作用を有するものと 思われる (表 1 ) 。
なお、 上記の製造例 (A) および (B) に用いた試薬等は以下の通りである。 フエ ツイン、 ァシァ口フェツイン、 ゥシ下顎腺厶チン、 ひ 1-酸グリコプロテイン、 ガ ラク トース S l, 4-N-ァセチルガラクトサミン、 CMP- NeuAc、 ラクト -N-テトラオ一 ス、 ベンジル -GalNAc 、 N-ァセチルラクトサミン、 及びトライ トン CF-54 はシグ マ社 (セント 'ルイス、 USA)から入手した。 CMP- [ 1 C]NeuAc(l lGBq/mmole) はァ マシャム社(U. K. )から入手したものである。 N-ァセチルガラクトサミン 1, 4-ガ ラクトースは梶本博士 (理化学研究所、 日本国埼玉県和光市) より寄贈されたも のを用いた。 2-ァセトアミ ドおよび 2-デォキシガラクトシルひ N-ァセチルセリン (GalNAc-SerNAc) は、 グランドラー及びシュミッ ト (Grimdler G. , and Schmidt R. R., Liebigs Ann. C em. , 1984, 1826-1847, 1984)の方法によって合成した。 NeuAcひ 2, 6-GalNAc- SerNAc は NeuAcひ 2, 6GalNAc- Ser (メクト社) からピリジン- 水中で無水酢酸でァセチル化によって製造した。 NeuAcひ 2, 6GalNAc-オール及び GlcNAC /51, 3 [NeuAc 2, 6]GalNAc -オールは辻および大沢(Tsuj i, T. and Osawa T., Carbohydr. Res. , 151. 391-402, 1986)の方法によってゥシ下顎腺厶チンか ら製造し、 270MHz 及び 1 3C NMRにより同定した(Savage, A. V. et al., Eur. J. Biochem. , 192, pp. 427-432, 1990 ; 及び Savage, A. V. et al. , Eur. J. Biochem. , 193, 837-843, 1990) 。 合成プライマーはァプライド .バイオシステ ム 394DNAシンセサイザーで合成した。 制限酵素 Smal、 EcoRI、 BamHk Hindi 11 、 Sacl、 X oK Bgl l l 及び Pst Iは宝酒造から入手した。
(C) GalNAcひ 2, 6-シアル酸転移酵素 P-B3 の製造
GalNAc 2, 6-シアル酸転移酵素の cDNAクローンを得るために、 2個の縮重ォ リゴヌクレオチド (ST-107及び ST- 205) による PCR を鐯型としてニヮトリ胚 cDNA を用いて行った。 約 150bpの望ましいサイズのフラグメントをァガロースゲル電 気泳動により精製した。 この PCR産物の塩基配列の決定を行ったところ、 Gal /S l, 4GlcNAc 2, 6-シアル酸転移酵素 (Kurosawa, N. , et al. , Eur. J. Biochem. , 219, 375-381, 1994) 、 及び GalNAcひ 2, 6-シアル酸転移酵素 P - Bl をコードする PCR産物の他に、 新規ァミノ酸配列をコ一ドする PCR産物として PCRB3が得られ た。 pCRB3 のシァリルモチーフと上記の各シアル酸転移酵素のシァリルモチーフ との一致度は、 65%-57%であった。
該遺伝子の完全なコ一ド領域を確認するために、 幼ニヮトリ精巣の cDNAライブ ラリーを pCRB3の cDNAインサートでスクリーニングした。 5 x lO5 個の別々のク ローンについてスクリ一二ングしたところ、 ィンサ一トサイズが 2. 05 kbである ポジティブクローン; I CEB3-T20が得られた。
この cDNAクローンのヌクレオチド配列は、 45.8 KDaの分子量を持つ 404ァミノ 酸をコードする 1212bpのオープンリーディングフレームを含んでいた。 このォ一 プンリーディングフレームは、 通常の翻訳開始配列 (Kozak, M. , Nature, 308, 241-246, 1984)を持つヌクレオチド 1位のメチォニンコドンで始まり、 ヌクレオ チド 1213位の TGA停止コドンで終了している。 また、 384 bpの 5 '非翻訳配列及 び 451 bp の 3 '非翻訳配列に挟まれている。 開始部位の 5 ' DNA配列は、 3個 のリーディングフレームの全てに停止コドンを含んでいる。 i CEB3-T20のヌクレ ォチド配列及び推定ァミノ酸配列を配列表の配列番号 1に示した。 このアミノ酸 配列を有する GalNAc 2, 6-シアル酸転移酵素を P-B3 と命名した。
上記の GalNAcひ 2, 6-シアル酸転移酵素 P-B3 (GalNAcひ 2, 6-シアル酸転移酵素 P- B1を ST6GalNAcAと呼ぶ場合、 本酵素を ST6GalNAcBと呼ぶこともある) は、 現在ま でにクローニングされている全てのシアル酸転移酵素に見られているように、 荷 電した残基に接する 17ァミノ酸の N-末端疎水性配列を含む I I型の経膜ドメインを 有している。 GalNAcひ 2, 6-シアル酸転移酵素 P-B3の一次配列を DNA及び蛋白デー 夕バンクの他のァミノ酸配列と比較したところ、 クローニングされている全ての シァル酸転移酵素と二領域で類似性を有することが明らかになつた。
GalNAc a2, 6-シアル酸転移酵素 P-B3 の中心部にある一方の領域 (シァリルモ チーフ L) は 45アミノ酸から成り 64-24%の配列一致度を示す。 他方、 C00H末端 部分にあるもう一つの領域 (シァリルモチーフ S、 333-355残基) は 78-43%の 一致度を示す。 Gal.NAc 2, 6-シアル酸転移酵素 P-B3の全体的ァミノ酸配列一致度 は、 それぞれ、 ニヮトリの Gal y81, 4GlcNAc ひ 2, 6-シアル酸転移酵素 (Kurosawa, N. , et aし Eur. J. Biochem. , 219, 375-381, 1994) に対して 10%、 ニヮトリ の Gal yS l,3GalNAc 2, 3-シアル酸転移酵素 (Kurosawa, N. et al., Biochem. Bi ophys Acta. , 発行中) に対して 13%、 及びニヮトリの ST6GalNAcA(22)に対し て 32%であった。 これらの結果は、 クローニングされた該遺伝子がシアル酸転移 酵素遺伝子フアミリーに属することを示すものである。
実験の詳細は以下のとおりである。
ポリメラーゼ連鎖反応法 (PCR)
PCR は Gai 4GlcNAc-ひ 6STRL(Weinstein, J. et aし J. Biol. Chem. , 262, 17735-17743, 1987)、 Gal yg 4G 1 cNAc- a 6STHP (Gr undmann, U. et al. , Nuclei c Acids Res. , 18, 667, 1990)、 及び Gal y53GalNAc-ひ 3STPS (Gi l lespi e, W. et al. , J. Bi ol . Chem. , 267, 21004-21010, 1992)中の保存領域に由来する縮重プ ライマ一 [5 'プライマー ST107 : TGGGCCTTGGI KA/OAGGTGTGCTGTTG及び 3 'ブラ イマ一 ST205 : AGGCGAATGGTAGTTTTTG(A/T)GCCCACATC]を用いて行った。 cDNAを得 るために、 3日齢のニヮトリ胚からの poly(A)リツチ RNA (2/z g)を、 オリゴ- dT プライマー (フアルマシア社); dATP、 dCTP、 dGTP、 及び dTTPをそれぞれ 1 m ; 並びに 2 U/ / 1 の RNaseP且害剤 (プロメガ社) と共に、 50〃1 の 10 mM Tri s- HC1 (pH8. 3)、 50 mM KC1、 1. 5 mM MgCl 2、 および 0. 001%ゼラチン中、 0 °Cで 10分間 インキュベートした後、 100 μ. \] のモロニ一 'ネズミ白血病ウィルス逆転写酵素 (BRL) を加えてさらに 42°Cで 60分間ィンキュペートした。
上記反応液を 94°Cで 3分間加熱した後、 0. 2〃g の poly(A)リツチ RNA から製 造した cDNAを、 50〃1中に 10 mM Tri s- HCKpH8. 3)、 50 mM KC1、 1. 25 mM MgCl 2、 0. 001%ゼラチン、 dATP、 dCTP、 dGTP、 及び dTTPをそれぞれ 200 zM、 Taq DNA ポ リメラーゼ (プロメガ社) を 2U、 並びに 40 pmole の各 PCRプライマ一を含む 混合液中における PCR実験に用いた。 35サイクルの PCR増幅を行い、 各サイクル は、 96°C、 45秒間の熱変性; 50° (:、 60秒間のァニ一リング;及び 72°C、 60秒間の 伸長とした。 PCR生成物を 3%ァガロースゲルで展開し、 150bp に相当する DNA フラグメントをゲルから溶出した (キアエツクス ·キット、 キアゲン社) 。 この フラグメントを平滑末端化してキナーゼ処理した後、 PUC119の Smal部位の中にサ ブクローニングし、 最終的に配列決定した。
cDNAライブラリーの作成 全 RNA をグァニジニゥムチオシァネート法: 及びそれに続く 5. 7 CsCl 溶液 中における遠心により、 ニヮトリ精巣から調製した (Sambrook, J., Molecular Cloning: a Laboratory Mannual, 2nd edi tion) 0 poly(A)リツチ匪 をオリゴ テックス- dT30 (宝酒造) で精製し、 オリゴ -dT プライマ一及びランダムプライ マーに; I ZAPI I (ストラタジーン社) 及び cDNA 合成キッ ト (フアルマシア社) を用いた cDNAライブラリーの作成に使用した。
cDNAラィブラリーのスクリ一二ング
増幅された cDNAライブラリー ( 1 X 106 プラーク) をニヮトリ胚 PCRフラグメ ントでスクリーニングした。 プラークを移したフィルターを 5 X SSC;、 0. 2% SDS, 5 xデンハルド溶液及び 10 ^ g/ml変性サケ精子 DNA 中で 3 2P- 放射性ラベル化 DNA プローブと 65°Cで 12時間にわたりハイブリダィズさせた。 その後、 2x SSC、 0. 1% SDS中で 65°C、 20分で二回洗浄した。 単離されたファージクローンからブラ スミ ドを得るために、 ファージミ ドレスキューを Ι ΖΑΡί Ι クローニングキッ トの 製造元 (ストラタジーン社) の使用説明書に従って行った。 cDNAインサートは B1 uescriptプラスミ ドとして直接切り取った。 プラスミ ドの作成はサムブルックら (Sambrook, J. et al. , Molecular Cloning: a Laboratory Manual, 2nd ed. )に よって述べられている分子クローニング標準法を用いた。
DNA配列分析
上記のィンサー卜の DNA配列は、 T7- DNAポリメラーゼの铸型として一本鎖 DNA を用いるジデォキシ鎖停止法 (Sanger, F. et al. , Proc. Natl. Acad. Sci. USA, 74, 5463-5467, 1977)により決定した。 シーゲンシング反応及び電気泳動はォー トリード DNA シーゲンシングキッ ト及び DNA シーケンサー (フアルマシア社) を 用いて行った。 一本鎖 DNA は、 ヘルパーファージ R408 (ストラタジーン社) で重 複感染させた後に大腸菌 XL-Blue (ストラタジーン社) から調製した。 配列デー 夕は PC/Gene (ティジンシステムテクノロジ一社) を用いたコンピューターで分 折した。
遺伝子の存在を確認するためにニヮトリゲノム DNAのサザン ·プロッ ト分析を 行った。 ニヮトリゲノム DNA に対する cDNAィンサート pCRB3のハイブリダイゼ一 シヨンでは、 EcoRI による切断からは単一のバンドが、 また BamHIによる切断か らはニ本のバンドが得られた。 この単純なハイブリダイゼ一ションパターンによ れば、 クロ一ニングされた cDNAが単一コピー遺伝子であることが明らかである。 厳密度の低い条件下で PCRB3プローブを用いたマウス及びサルからのゲノム DNA のサザンブロット分析では、 この遺伝子が種族間で保存されていることが示唆さ れた。 サザンブロット分析用には、 マウス脳、 COS- 7細胞及びニヮトリ精巣から 調製したゲノミック DNA を各 7. 5 n gずつ制限酵素で切断し、 0. 6%ァガロースゲ ル上でサイズ分画化したものを用いた。
また、 GalNAcひ 2, 6-シアル酸転移酵素 P- B3遺伝子の mRNAサイズ及び分布をノ一 ザンブロット分析により測定した。 3、 6、 8、 10及び 12日齢の胚からの RNA分 析により、 4. 5 kb及び 2. 2 kb の 2種の RNA の存在が明らかになった。 4. 5 kbの mRNAは調査した全ての胚ステージにおいて豊富に発現されたが、 成体組織には発 現されていなかった。 2. 2 kbの mRNA は初期の胚ステージにおいてはさほど豊富 に発現されなかったが、 胚ステージ終期及び成体組織においては豊富であった。
2. 2 kbというトランスクリプトのサイズは、 得られた cDNAクローン(A CEB3-T20) がフルレングスに近いものであったことを示すものである。 ノーザンブロット分 析用には、 ニヮトリ胚からの poly(A)リツチ RNA 5 ja g及びニヮトリ組織からの 全 RNA 10 / g をホルムアルデヒドーァガロースゲル上でサイズ分画化したものを 用いた。
従来公知のシアル酸転移酵素は、 驚くほど組織特異的発現を示すものであった 力 \ これは細胞型特異的炭化水素構造の存在に相関しているものと考えられてい る (Paulson, J. C. and Col ley, K. J. , J. Biol. Chem. . 264, pp. 17615— 17618, 1989) 。 上記ノーザン ·プロッティングの結果によれば、 シアル酸転移酵素 P-B3 の mRNA の発現パターンが変化することが明らかである。 胚期特異的な 4. 5Kbの mRNAの正確な構造は明らかではないが、 シアル酸転移酵素 P- B3 の遺伝子から 2 種の異なるサイズの mRNAが転写されることは、 それらが Gal 4GlcNAc-ひ 2, 6 - シアル酸転移酵素 (Gal β 4G1 cNAc- a 6STRL)および Gal 1, 3(4)GlcNAcひ 2, 3-シァ ル酸転移酵素 (Gal y53(4)GlcNAc- ひ 3STRし)(Weinstein, J. et al., J. Biol. Chem. , 262, 17735-17743, 1987; および Wen, D. X. et al. , J. Biol. Chem. , 267, 21011-21019, 1992) に観察されたような、 選択的スプライシングメカ二ズ ム及び選択的プロモータ一利用メカニズムを通じて発現されることを示唆してい る。 このことは、 シアル酸転移酵素 P- B3 に対しては単一コピーの遺伝子しか存 在しないことがサザン ·ハイプリダイゼ一シヨンにより示されていることからも 明らかである。
シアル酸転移酵素 P- B3 の全長をコードする 1. 3 kb DNA フラグメントを、 合 成オリゴヌクレオチドプライマー (5' - ACGGCGCTCGAGCCAACCCGGAGAGCAGCG-3' 及 び 5' -CGTTGCCTCGAGAGTCCTTGCAGTGGGACT-3':下線部は合成 Xhol 部位を示す) を 用いて増幅させた。 増幅させた DNA フラグメントを Xholで切断し、 発現ベクター pcDSRひの Xhol部位に挿入し (Takebe, Υ·, Mol. Cell. Biol. , 8, pp. 466 - 472, 1988) 、 組換えプラスミ ド PCDB3STを得た。 このプラスミ ドインサートの配列を 決定し、 ポリメラ一ゼ連鎖反応のエラーの可能性がないことを確認した。
上記の組換えプラスミ ド PCDB3ST 5〃g を用いて COS- 7細胞中に DEAE- デキス トラン法によりトランスフヱクトした (McCutchan, J. H. and Pagano, J. S. , J. Natl, Cancer Inst. , 41, 351-357, 1968)。
トランスフニクトの 48時間後に培養細胞(l x 107)を回収し、 リン酸緩衝食塩水 で洗浄した。 その後 20mM MnCl 2 及び 25 mM MES, pH 6, 0を含むバッファー 2 ml に細胞を再懸濁した。 細胞懸濁液を 30, 000 x g で 30分間遠心し、 得られた細胞 ペレッ トを 0. 5 ml の 1% トライトン X-100、 50 mM NaCK 5 mM MnCl2> 25 mM MES, pH 6. 0 に再懸濁した後、 ソニケ一ターにかけた。 30,000 xg で 30分間遠心 した後、 上清をセントリコン 30 フィルター (アミコン) で 10倍に濃縮して以下 のアツセィに用いた。
受容体としてグリコプロテイン、 オリゴサッカライド、 及びグリコリピドを用 い反応液として 0. 1 M力コジル酸ナトリウムバッファー(pH 6. 0)、 10 mM MgCl2 、 0. 5 % トライトン CF54 、 12 zM CMP-[ ! C]NeuAc (1. 5 kBq)、 1 rag/ml 受容体基 質及び COS 細胞ライセート (総容量 10〃1)を用いてアツセィを行った。 反 応液を 37°Cで 1時間ィンキュベ一トした後の反応混合液を、 グリコプロティンを 受容体に用いた場合は SDS- PAGE に付し、 オリゴサッカライド及びグリコリピド を受容体に用いた場合はエタノール /1-ブ夕ノール Zピリジン Z酢酸 Z水 (100: 10: 10:3:30) を溶媒系に用いる HPTLCプレート (メルク社、 ダームシュタツ小、 ドイツ) クロマトグラフィーに付した。 シアル化受容体は BAS2000 ラジオィメ一 ジアナライザー (富士写真フィルム製、 日本) で定量した。
シアル化産物の同定は以下のように行った。 還元オリゴサッカライドは、 カー ルソンの S—除去法 (Carlson, D. . , J. Biol. Chem. , 243, pp. 616-626, 1968) により、 再シアル化グリコプロテインから製造した。 PCDB3ST でトランスフエク トされた C0S-7細胞ライセート中でァシァ口 BSMを上記と同条件下で CMP- [ 1 4C] NeuAc を用いてシアル化した。 フェツインから遊離した放射能ラベルされたオリ ゴサッ力ライドを NDVシァリダーゼで切断し、 さらに生成物を同定するためそれ 以上の精製を行わずに薄層クロマトグラフィーに付した。 なお、 BSM から遊離し たオリゴサッカライドを標準として用いた。 ァシァ口 BSM及びアジア口フエツイ ンは、 それぞれ GalNAc 2, 6-シアル酸転移酵素 P-B1 及び Gal 1 , 3GalNAcひ 2, 3— シアル酸転移酵素 (Lee, Y. -C, , et al., Eur. J. Biochem. , 216, pp. 377-385, 1993) で [ 1 4C] _シアル化し、 オリゴサッカライドは8—除去により製造した。 生 成した [ 1 4C]NeuAcひ 2,6GalNAc-オール、 Gal S L 3( [ I 4C]NeuAc 2, 6)GalNAc- ォ —ル、 及び [ 1 4C]NeuAcひ 2, 3Gal β 1, 3GalNAc-オールは放射能ラベル化ス夕ンダ一 ドとして用いた。
フェツインを受容体として用いた場合には、 組換えベクター pcDB3STでトラン スフェクトされた C0S-7細胞のライセー卜によってのみシァリル化された。 発現 した6&1^じ 2,6-シァル酸転移酵素 P-B3 はフヱツイン及びァシァロフヱツイン に対して強い活性を示し、 ァシァ口 BSM に対しては弱い活性を示した力、 BSM又 は N-グリコシド結合ォリゴサッカライドのみを有する他のグリコプロテイン(例: ひ 1-酸グリコプロテイン、 オボ厶コイド、 ァシァ口- ひ 1 酸グリコプロテイン及 びァシアローオボムコィド) に対しては有意の活性を有していなかった (表 2)。 さらに、 ォリゴサッカライド又はグリコスフインゴリピドは本発明の GalNAc 2, 6-シアル酸転移酵素 P-B3に対する受容体としては機能しなかった。 該酵素によ つてフヱツイン中に取り込まれた [ 1 4C]NeuAc残基は、 N-グリカナーゼ又は NDVシ ァリダーゼによる処理に対して耐性を示した。 フ ツインから放出された放射能 ラベルされたオリゴサッ力ライドは NDVシァリダ一ゼによる処理の後、 Gal ^ 1, 3 (NeuAcひ 2, 6)GalNAcオールと共移動した。 これらの結果は、 シアル酸残基がフエ ツインの 0-グリコシド結合ォリゴサッカライドの GalNAc残基上のひ 2, 6-リンケ一 ジを通じて転移されたことを示している。 従って、 発現された酵素は GalNAcひ 2, 6-シアル酸転移活性を有していることが明らかである。 しかしながら、 本発明の 6&1^じ 2,6-シァル酸転移酵素 P-B3 に対するァシァ口 BSMの受容体としての機 能はフエツイン及びァシァロフヱツインと比べると遙かに不十分であった。 この 受容体基質特異性は、 ァシァ口 BSMがァシァロフヱツインよりも遙かに良好な受 容体として機能する GalNAc a2,6-シアル酸転移酵素 P-B1 の受容体基質特異性と は異なっている。
本発明の GalNAcひ 2, 6-シアル酸転移酵素 P- B3 の基質特異性を明らかにするた めに、 フェツインをシァリダ一ゼ (コレラ菌) 及びS—ガラクトシ夕一ゼ (ゥシ 精巣) で逐次処理し、 できたァシァロフヱツイン及びァガラクトーアジア口フエ ツインを受容体として使用した。 シァリダーゼ処理されたフエツインへの NeuAc - 残基の取り込みは天然フヱツインへの取り込みの 1. 5倍に増加した。 3個の 0— グリコシド結合型ォリゴサッカライドがフヱツインに含まれていることが知られ ており、 そのうちの 2個は NeuAcひ 2, 3Gal 5 1,3GalNAc であり、 残りの 1個は、 NeuAc a2, 3Gal;8 1, 3(NeuAc ひ 2, 6)GalNAcである(Spiro, R. G. and Bhoyroo, V. D. , J. Biol. Chem. , 249, 5704-5717, 1974) 。 従って、 天然フヱツインにおい ては 3個の 0—結合型ォリゴサッカライドのうちの 2個につレ、ては GalNAc残基が 受容体として機能しうるが、 一方、 ァシァ口フエツインにおいては全ての〇一結 合型ォリゴサッ力ライドの GalNAc残基が本発明の GalNAcひ 2, 6-シアル酸転移酵素 P - B3によってシアル化されうるものである。
また、 ァガラクト—ァシァ口フェツインは本発明の GalNAcひ 2, 6-シアル酸転移 酵素 P-B3 の受容体として機能することができず、 さらに、 該酵素と共にインキ ュペートされたアジア口 BSMから yS—除去により遊離されるオリゴサッ力ライド については Gal /S l, 3( [ 1 4C]NeuAcひ 2, 6)GalNAc -オールのみが検出され、 [ 1 4C] - NeuAc 2, 6- GalNAc - オールは検出されなかった。
以上により明らかにされた本発明の GalNAcひ 2, 6-シアル酸転移酵素 P- B3 の諸 特性をまとめて示せば、 以下のとおりである:
(1-i) 〇一グリコシド結合型 (NeuAcひ 2, 3) Gal β 1, 3GalNAc配列を含むフェツイ ン及びァシァロフヱツイン(Spiro, R.G. and Bhoyroo, V.D., J. Biol. Chem. , 249, 5704-5717, 1974) は良好な受容体として機能するが、 全炭化水素鎖の 5% しか Gal/Sl,3GalNAc配列を含まないァシァ口 BSM (Tsuji, T. and Osawa, T., Carbohydr. Res., 151, 391-402, 1986)は受容体としての機能がはるかに不完全 であ 。
(1- i i) Gal 1, 3GalNAcひ 1一 Βζがァシァロ GM1 (Gal β\, 3GalNAc/31, 4Gal y5L3Glc β 1 - Cer)及び GMlb(NeuAcひ 2, 3Gal β 1, 3GalNAc β 1, 4Gal β 1, 3Glc^ 1- Cer)と同様 に受容体として機能しなかつたことから、 このシアル酸転移酵素の活性に対して 蛋白部分は必須である。
(2) β- ガラクトシダ一ゼで処理したァシァ口フェツイン (ァガラクトーァシァ ロフヱツイン) に対して活性を示さない。
(3) ァシァ口 BSMの炭化水素鎖の約 60%は GalNAc-0-Ser/Thr である(Tsuji, T. and Osawa, T., Carbohydr. Res., 151, 391-402, 1986)にもかかわらず、 [14C] シアル化ァシァ口 BSMから遊離されたオリゴサッ力ライド中には Galy31,3([14C] NeuAc a 2, 6)GalNAc- オールしか検出されない。
これらの結果は、 グリコプロテインの Thr/Ser に Q—結合した GalNAc残基上に ひ 2, 6 リンケージを持つ CMP- NeuAc を転移させる触媒活性を有する本発明の酵素 の受容体基質としては、 0-グリコシド結合型オリゴサッ力ライドの Gal^UGal NAc配列が不可欠であるが、 一方、 ガラクト一ス残基に結合したひ 2, 3結合型シ アル酸残基は必須ではないことを明確に示している。 従って、 本発明により初め てクローニングされた酵素 P- B3 は新しいタイプの GalNAcひ 2, 6-シアル酸転移酵 素である。 分子中央に位置する 45アミノ酸範囲 (シァリルモチーフ L)から C00H- 末端までの(180- 404残基) GalNAcひ 2, 6-シアル酸転移酵素 P-B3 の一次配列は、 GalNAcひ 2, 6-シアル酸転移酵素 P- B1 の一次配列と高い配列一致度を示す (第 4 図: 48% の一致度) 。 これらの GalNAcひ 2, 6-シアル酸転移酵素に独特な保存領域 は、 それらのシアル酸をひ 2, 6-リンケージにより GalNAc部分に転移させる酵素作 用に相関するものであると思われる。 表 2 本発明の GalNAc 2, 6-シアル酸転移酵素 P- B3 の受容体基質特異性

0は 0.5 pmol/h より少ないことを示す。 なお、 上記の製造例 (C) に用いた試薬および試料等は以下のとおりである。 フ エツイン、 ァシァ口フェツイン、 ゥシ顎下腺ムチン、 ひ 1-酸グリコプロテイン、 ガラクト一ス 81,4-N-ァセチルガラクトサミン、 CMP-NeuAc 、 Gal S 1.3GalNAc ひ 1 - Bz、 GalNAcひ 1- Bz及びトライ トン CF- 54 はシグマ社 (セント 'ルイス、 USA) から入手した。 CMP-[1 C]NeuAc(llGBq/mraole) はアマシャム社 (U.K.)から入手し たものである。 2-ァセトアミ ド、 及び 2-デォキシガラクトシルひ N-ァセチルセリ ン(GalNAc- SerNAc) はグランドラ一及びシュミッ ト (Grundler G., and Schmidt R., Liebigs Ann. Chem. , 1984, 1826-1847, 1984)の方法によって合成した。 コレラ菌 (Vibrio cholerae) 由来の NDV-シァリダーゼ及びシァリダーゼは、 それ ぞれォックスフオード ' グリコシステムズ社 (U. K.:)及びべ一リンガー ·マンハイ ム社 (ドイツ) から購入した。 ゥシ精巣由来の8—ガラクトシダーゼはべ一リン ガー ·マンハイム社 (ドイツ) から入手した。 合成プライマーはアプライド ·バ ィォシステム 394 DNAシンセサイザーで合成した。 制限酵素類は宝酒造から入手 したものである。
(D) 微生物中で発現したシアル酸転移酵素の精製
プラスミ ドの構築
P C R法によりマウス由来の Gal ^ l, 4GlcNAc ひ 2, 6-シアル酸転移酵素 cDNA (Hamamoto, T. et al. , Bioorg. Medicin. Chem. , 1, 141-145, 1993) に開始コ ドンとクロ一ニング ·サイトを結合した。 センスプライマ一として Ndel サイト を含む 5' -TGGCATATGGGGAGCGACTATGAGGCTCT-3'、 アンチセンスプライマーとして BamHI サイトを含む 5' -ATGAGGATCCCTGGCTCAACAGCG- 3'を用いた。 得られた PCR断 片(1152bp)は、 開始コドンと酵素の 29番目のアミノ酸残基から C-末端までをコー ドする領域とを含んでおり、 サイトゾル · ドメイ と経膜ドメインを欠くもので ある。 この PCR断片を発現用ベクター pET3b (Studier, F. W. et al. , Method. Enzymol. , 185, 60-89, 1990) の Nde卜 BamHI 部位 (T7プロモータの下流に位置 する) に導入した。 得られた組み替えベクターを PET3-MBS と命名した。 なお、 上記 PCR断片の核酸配列を配列表の配列番号 4に示す。
酵素の発現
ベクター pET3- MBS をトランスフエクトした大腸菌 JM109(DE3) を 100〃g/ml のアンピシリンを含む 100 ml の LB培地を用いて 37°Cで培養した。 600 nmの光 学濃度が 0.2-0. 4に達した時点で、 2 m IPTG ('イソプロピル —D-チォガラク トビラノシド) を加えて T7 RNA ポリメラーゼを誘導し、 組み替え蛋白の産生を 開始した。 細胞質および経膜ドメインを欠く組み替え蛋白が菌体中に不溶性の封 入物の形態で蓄積した。 PET3-MBS によりトランスフヱクトされた大腸菌 JM109 (DE3) の生育速度は、 寒天培地上および液体培地中のいずれにおいても、 未トラ ンスフ クトの大腸菌 JM109(DE3) の生育速度と同様であった。 2時間培養した 後に菌体を集め (湿重量約 lg)、 10 mlの 20 mM Tris-HCl (pH 8. 0)に懸濁し、 リゾチーム(0. 1 mg/ml)と DNase 1 (0. 01 mg/ml)で 30分間処理した。 Tri ton X - 100 を最終濃度 1% となるように加え、 4 °Cで 15分間 12, 000 x g で遠心して不溶分 画を集めた。 沈殿を 3 mlの 10 mM Tris-HCKpH 7. 4)に懸濁し、 使用時まで -30°C で貯蔵した。 可溶化および脱変性
0.5 mlの上記懸濁液に 0.48 g の固形尿素、 60〃1 の 5M NaCK 20〃1 の 1M Tris-HCKpH 7.4)、 および水を加えて最終の容量を 1 mlとした (最終濃度: 8M 尿素; 0.3 M NaCl; 20 mM Tris-HCl, pH 7.4) 。 沈殿を 10 °Cで 30分間抽出し、 12, 000 xgで 15分間遠心した。 抽出された蛋白の大部分は 42 k ダルトンの分子 量であった。 抽出に 5.7 M尿素バッファーを用いた場合には、 80%の蛋白を回収 することができた。
8 M尿素を含む抽出液各 0.1 ml を脱変性用組成物 (標準組成物: 2M尿素, 0.5M NaCl, 10 m ラクトース, 0.5 mM EDTA, 及び 20 mM MOPS-NaOH, pH 7.0)各 1.9 mlで希釈し、 最終蛋白濃度を約 0.02 mg/ml とした。 溶液を 4でで 12時間 放置した後、 同容量の脱変性用組成物で希釈して尿素濃度を半分にし (約 1.2M;)、 さらに 4でで 48 時間放置した。 その後、 シアル酸転移酵素活性を測定し、 この 時点で脱変性用組成物の組成の影響を分析した (表 3) 。 さらに得られた酵素を 脱変性用組成物に対して 4°Cで 48 時間透析して残存する尿素と還元剤を除去し た。 得られた試料をセントリコン- 30 フィルター (アミコン社製) で約 20倍に濃 縮した。
シァル酸転移酵素のァッセィ
50 zM CMP-[14C-]NeuAc(0.9 Bq/pmol)をドナー基質とし、 5 mの Gal^l,4Glc NAc(N-ァセチルラクトサミン) を受容体基質として、 シアル酸転移酵素活性を測 定した。 反応液に 1 mg/ml ゥシ血清アルブミン、 1 〃1 の酵素溶液、 および 50mM 力コジル酸ナトリウム (pH 6.0) を加えて最終容量を 10 1 とし、 37°Cで 1 時 間インキュベートした。 その後、 試料をシリカゲル 60 HPTLC プレート (メルク 社、 ドイツ) にのせ、 エタノール: ピリジン: n-ブ夕ノール:酢酸:水(100: 10: 10:3:30)を展開溶媒として展開した o各プレート上の放射能活性を BAS2000ラジ ォイメージアナライザー (富士写真フイルム社、 日本) で定量した (Lee, Y.-C. et aし Eur. J. Biochem. , 216, 377-385, 1993) 。 酵素活性の 1ュニットは 1 分あたり 1 〃モルのシアル酸転移を触媒する量と定義した。 また、 分枝オリゴ糖 に関して受容体特異性を調べるために、 Ν-ァセチルラクトサミン型の 2分枝ピリ ジルァミノオリゴ糖を受容体基質として用い、 HPLCにより蛍光分析した。 8M尿素による抽出液を 4°Cでの希釈を行わずに直接透析に付した場合、 0.5M 未満の尿素濃度で析出した蛋白の酵素活性はほとんど回復しなかつた。 2回目の 希釈後 48時間における最適希釈条件を検討した結果を以下の表 3に示す。 表中、 標準組成物は 2M尿素, 20 mM Tris - HC1, 0.3M NaCL 20 m ラクトース, 0.5 mM EDTACpH 7.4)であり、 他の組成物は標準組成物と異なる部分のみを示してある。 表 3 Gal/SL4GlcNAc ひ 2, 6-シアル酸転移酵素の脱変性条件の影響

0 は標準組成物に比べて 5%未満であることを示す c 標準組成物において 0.5M NaCl (PH 7.0)とした場合に最大の脱変性効果が観ら れたので、 以降の実験ではこの組成物を用いて脱変性を行った。 この条件を用い て 3回の独立の脱変性を行ったところ、 回収された総活性は 0.4-0.8 mU/0.1 ml 抽出物であった。 この脱変性ステージの酵素は CMP- NeuAc及び N-ァセチルラクト サミンについてそれぞれ 0.14 mおよび 20 m の高い Km値を与えた。 また、 試 験を行った条件では還元剤 (DTT および - メルカプトエタノール) は酵素活性 を阻害した。 これは、 アツセィ混合物中に 0.1M濃度で持ち込まれた尿素の作用 によるものと思われる。 さらに、 2回目の希釈後 12時間にはほとんど活性が観 られず、 ポリべプチドの再折り畳み過程が試験温度においては非常に遅いことが 明らかである。 1 〃M および 1 mMの還元剤の存在下に 8M尿素抽出液を 20 容量 の 2M尿素, 20 0PS-Na0H(pH 7. 0), 0. 5 NaCL 20 m ラクト—ス, 0. 5 mM EDTAを含む脱変性用組成物で希釈し、 試料を 4°Cで 12時間放置した後に再度希釈 して尿素の濃度を半分にし、 さらに透析により尿素と還元剤を除去した場合、 還 元剤を使用しない場合とほぼ同じ活性を与えた。 結果を以下の表 4に示す。 表 4

また、 脱変性させたマウス Gal yS l, 4GlcNAc ひ 2, 6-シアル酸転移酵素の基質特 異性は、 それぞれ 2 mg/ml の基質を用いてアツセィを行った。 生成物は HPTLCに より分析を行った。 受容体としてォリゴ糖および糖蛋白を用いた場合にはエタノ ール: ピリジン: n-ブ夕ノール:酢酸:水 (100: 10: 10:3:30) を移動相として用 レ、、 糖脂質を受容体として用いた場合はクロ口ホルム: メタノール: 0. 5% CaCl2 (55:45:8) を移動相として用いた。 脱変性された酵素の基質特異性と動力学的パ ラメ一夕一は、 ラット肝から得られた酵素のものと同様であった。 結果を表 5お よび表 6に示す。 表 5

f濃度は末端ガラクト- ス残基についてのものである, Gal 81, 4GlcNAc 2, 6-シアル酸転移酵素は、 N-ァセチルラク トサミン型の 2 分枝糖蛋白の異なる枝分かれを区別することができる(Joziasse, D. H. et al. , J. Biol. C em. , 260, 714-719, 1985 ; および Van den Ei jnden D. H. et at., Biochem. Biophys. Res. Comm. , 92, 839-845, 1980)。 脱シァリル化した 2分枝 PA-ォリゴ糖を本発明の方法により脱変性した酵素によりシァリル化して HPLC分 析を行った。 アツセィは 10 pmolの受容体基質と 0. 1 mM C P-NeuAc を用いて最 終容量 5 1 として行った。 反応液を 37でで 1時間インキュベートした後、 冷 水 90 n \ を加えて反応を停止した。 シァリル化されたピリジルァミノオリゴ糖 を同定するため、 逆相カラム(Shimpack CLC-0DS, 0. 6 cmx l5 cm, 島津製作所) を用いて各反応混合物の HPLC分析を行った。 カラムを 70%溶媒 A (10 mM リン 酸ナトリゥ厶, pH 3. 8) および 30%溶媒 B (0. 5¾ n-フ"夕ノール, 10 mM リン酸ナ トリウム, pH 3. 8) で平衡化した後、 30分間で溶媒 Bが 60% になるような直線濃 度勾配を用いて 55 °C 1 ml/分で流出させた。 ピリジルァミノオリゴ糖を蛍光分 析 (励起波長 320 nm, 発光波長 400 nm)により検出したところ、 脱変性した酵 素は未変性の酵素と同様に Manひ 1, 6分技上のガラクトース残基よりも Manひ 1, 3 分技上のガラクトース残基に対する選択性が高いことが示された。
尿素を完全に除去することにより、 脱変性した酵素は還元剤に対する抵抗性を 回復した。 また、 2価カチオンを添加して脱変性を行うことにより 10倍以上の 活性が回復した。 いかなる特定の理論に拘泥するわけではないが、 尿素の存在下 に 0. 5 mM EDTA を含む透析液に対して長時間透析を行うと、 酵素の立体を正しく 保持するために酵素に強固に結合している 2価カチオンが失われるのかもしれな レ、。 1.2 M尿素を含む脱変性用組成物で脱変性した場合にも 2価のカチオンを添 加することにより活性が上昇した。 得られた結果を表 7に示す。 表中、 活性は試 薬無添加のものに対する相対値である。 また、 5 mM MnCl2を用いて測定した場合 には、 脱変性した酵素の比活性は 0. 15 U/mg蛋白であり、 ラット肝から得られた 酵素 (Weinstein, J. et al, J. Biol. Chem., 257, pp. 13835- 13844, 1982)の約 2であった。 酵素の総収量は 0. 1 U/10ひ ml 培養液であった。 表 7

以上、 本発明の方法を Gal/Sl,4GlcNAc ひ 2, 6-シアル酸転移酵素について実施 例により具体的に説明したが、 本発明の方法はこれらの実施例に限定されること はない。 上記のとおり、 シアル酸転移酵素類は他のグリコシル転移酵素とは異な り極めて保存性の高い領域 (シァリルモチーフ) を有していることが知られてお り(Livingston, B.D. and Paulson, J.C., J. Biol. Chera. , 268, 11504-11507, 1993) 、 シアル酸転移酵素はすべて類似の高次構造を形成しているものと考えら れている (Drickamer, , Glycobiology, 3, 2-3, 1993)。 従って、 以上の実施 例により明らかにされた Gal;81,4GlcNAc ひ 2, 6-シアル酸転移酵素の脱変性方法 が他のシアル酸転移酵素の脱変性方法に対しても同様の効果を奏することは、 当 業者は容易に理解されよう。 また当業者は、 本明細書に記載された本発明の方法 を修飾あるいは改変することにより、 Gal/31,4GlcNAc ひ 2, 6-シアル酸転移酵素 のみならず他のシアル酸転移酵素を脱変性するための最適な条件を適宜選択する ことができょう。 なお、 上記の例 (D) に用いた試薬および試料は以下のとおりである。 ラット肝 Gal S l, 4GlcNAc ひ 2, 6-シアル酸転移酵素、 フェツイン、 ァシァロフヱツイン、 ゥシ下顎腺ムチン、 ひ 1-酸糖蛋白、 ガラクトース y8 1, 3- N-ァセチルガラクトサミ ン、 ラクト N-テトラオース、 および N-ァセチルラクトサミンはシグマ社 (セン ト ·ルイス、 米国) から入手した。 尿素は和光純薬工業 (大阪、 日本) から入手 し、 溶液は使用直前に調製した。 CMP- [ 1 C] -NeuAc [ 11 GBq/mmole]はアマシャム 社 (英国) から入手したものである。 ゥシ下顎腺ァシァロムチンおよびァシァ口 ひ 1-酸糖蛋白は、 対応の糖蛋白を緩和に酸処理することにより製造した。 N-ァセ チルガラクトサミン S l,4-ガラクトースは梶本博士 (理化学研究所、 日本国埼玉 県和光市) により寄贈されたものを用い、 ピリジルァミノオリゴ糖 (PA- sugar 001, 021, 022, および 023) は宝酒造 (京都、 日本) から入手した。 蛋白濃度 はゥシ血清アルブミンを標準として BCA プロテイン 'アツセィ 'キット (ピア一 ス社) により決定した。 透析チューブ (20/32) はビスカース社のものを用いた。 産業上の利用可能性
本発明により提供される新規 GalNAcひ 2, 6-シアル酸転移酵素 P- B1および P- B3、 および該酵素の活性ドメインであるポリぺプチド部を有し細胞外に分泌される蛋 白は、 例えば、 蛋白にヒト型の糖鎖を導入する試薬、 あるいはヒトに特異的な糖 鎖を欠く遺伝性疾患の治療のための医薬として有用である。 また、 癌転移抑制、 ゥィルス感染防止、 炎症反応抑制を目的とする医薬としても用いることが可能で ある。 さらに、 本発明の方法は、 シアル酸転移酵素を微生物中で大量に発現させ た場合にも、 菌体内の凝集物あるいは沈殿物から高度に活性の回復した酵素を大 量に回収することができるので有用である。 配列表 配列番号: 1

Claims

請 求 の 範 囲
1. 配列表の配列番号 1に示されるァミノ酸配列により特定される GalNAcひ 2, 6- シアル酸転移酵素 P-B1 。
2. GalNAc 2, 6-シアル酸転移酵素 P-B1 のァミノ酸配列をコ一ドする GalNAcひ 2, 6-シアル酸転移酵素 P- B1 遺伝子。
3. 配列表の配列番号 1に示される核酸番号 1から 1698により特定される塩基配 列を有する請求の範囲第 2項に記載の GalNAc 2, 6-シアル酸転移酵素遺伝子。
4. 配列表の配列番号 3に示されるァミノ酸配列により特定される GalNAc 2, 6 - シアル酸転移酵素 P- B3 。
5. GalNAc 2, 6-シアル酸転移酵素 P- B3 のァミノ酸配列をコ一ドする GalNAcひ 2, 6-シアル酸転移酵素 P- B3遺伝子。
6. 配列表の配列番号 3に示される核酸番号 1から 1212で特定される塩基配列 を有する請求の範囲第 5項に記載の GalNAcひ 2, 6-シアル酸転移酵素 P-B3遺伝 子。
7. GalNAc 2, 6-シアル酸転移酵素 P-B1 のァミノ酸配列をコードする GalNAcひ 2. 6-シアル酸転移酵素遺伝子を含む組み換えベクター。
8. プラスミ ド; I CEB- 3034である請求の範囲第 7項に記載の組み換えベクター。
9. GalNAc 2, 6-シアル酸転移酵素 P- B3 のアミノ酸配列をコードする GalNAcひ 2, 6-シアル酸転移酵素遺伝子を含む組み換えベクター。
10. プラスミ ドス CEB3-T20 またはプラスミ ド pcDB3STである請求の範囲第 9項 に記載の組み換えベクター。
11. 0&1^( 2,6-シァル酸転移酵素 P-B1 のァミノ酸配列をコードする GalNAcひ 2, 6-シアル酸転移酵素遺伝子を含む組み換えベクターにより形質転換された形 質転換体。
12. GalNAcひ 2, 6-シアル酸転移酵素 P-B3 のァミノ酸配列をコードする GalNAcひ 2, 6-シアル酸転移酵素遺伝子を含む組み換えべクタ一により形質転換された形 質転換体。
13. GalNAcひ 2, 6-シアル酸転移酵素 P- B1 または P-B3 の活性ドメインであるポ リぺプチド部分とシグナルぺプチドとを含む細胞外分泌型の蛋白であって GalN Ac a 2, 6-シアル酸転移を触媒する蛋白。
14. 6&1^0 2, 6-シァル酸転移酵素 P-B1 の活性ドメインが配列表の配列番号 1 に示されるアミノ酸配列の 233-566 により特定されるポリべプチドである請求 の範囲第 13項記載の蛋白。
15. 配列表の配列番号 2に示されるアミノ酸配列により特定される請求の範囲第 13項に記載の蛋白 SB- 690 。
16. 0&1^ 2, 6-シァル酸転移酵素 P-B1 または P- B3 の活性ドメインであるポ リベプチド部分とシグナルべプチドとを含む細胞外分泌型の蛋白であって GalN Ac a 2, 6-シ了ル酸転移を触媒する蛋白をコードする遺伝子。
17. 配列表の配列番号 2に示される核酸番号 1から 1065で示される塩基配列を有 する請求の範囲第 16項に記載の遺伝子。
18. 0&1^(: 2, 6-シァル酸転移酵素?-81 または P- B3 の活性ドメインであるポ リベプチド部分とシグナルべプチドとを含む細胞外分泌型の蛋白であって GalN Ac 2, 6-シアル酸転移を触媒する蛋白をコードする遺伝子を含む組み換えべク 夕— 0
19. プラスミ ド pcDSB-690である請求の範囲第 18項に記載の組み換えベクター。
20. GalNAcひ 2, 6-シアル酸転移酵素 P- B1 または P- B3 の活性ドメインであるポ リベプチド部分とシグナルべプチドとを含む細胞外分泌型の蛋白であって GalN Acひ 2. 6-シアル酸転移を触媒する蛋白をコードする遺伝子を含む組み換えべク ターにより形質転換された形質転換体。
21. 0&1 じ 2, 6-シァル酸転移酵素 P-B1 または P- B3 の活性ドメインであるポ リベプチド部分とシグナルべプチドとを含む細胞外分泌型の蛋白であって GalN Ac 2, 6-シアル酸転移を触媒する蛋白をコ一ドする遺伝子を含む組み換えべク ターにより形質転換された形質転換体を培養し、 培養物から該蛋白を採取する ことを特徴とする、 GalNAcひ 2, 6-シアル酸転移酵素 P-B1 または P-B3 の活性 ドメインであるポリべプチド部分とシグナルべプチドとを含む細胞外分泌型の 蛋白であって Ga INAcひ 2, 6-シ了ル酸転移を触媒する蛋白の製造方法。
22. 0&1^じひ2, 6-シァル酸転移酵素。
23. 哺乳類動物由来の GalNAc 2, 6-シアル酸転移酵素。
24. シアル酸転移酵素の製造方法であって、 以下の工程:
(a) シアル酸転移酵素を微生物中で発現させる工程;
(b) 菌体内に蓄積するシァル酸転移酵素を含む蛋白凝集物または沈殿物から該 酵素を 5〜9Mの尿素により抽出する工程;
(c) 工程 (b) で得られた抽出物を脱変性用組成物で希釈して 1〜4 Mの尿素を 含む 1次希釈物を得る工程;
(d) 工程 (c) で得られた 1次希釈物をさらに脱変性用組成物で希釈して 0. 5〜 2 M の尿素を含む 2次希釈物を得る工程;および
(e) 工程 (d) で得られた 2次希釈物を 2価のカチオンの存在下で透析して尿素 を除去し脱変性シァル酸転移酵素を得る工程
を含む方法。
25. 工程 (c) で用いる脱変性用組成物が 1〜 2M尿素, 20 m MOPS-NaOH, 0. 5 NaCl, 20 mM ラクトース, 0. 5 mM EDTA (pH 7. 0)を含み、 工程 (d) で用いる脱 変性用組成物が 20 mM MOPS-NaOH, 0. 5M NaCl. 20 mMラクトース, 0. 5 mM EDT A (pH 7. 0)を含む、 請求の範囲第 24項に記載の方法。
26. シァル酸転移酵素が0&1^(;ひ2, 6-シァル酸転移酵素または 6&1 5 1, 461じ^じ a 2, 6-シアル酸転移酵素である請求の範囲第 24項に記載の方法。
27. シアル酸転移酵素の製造方法であって、 以下の工程:
(a) シアル酸転移酵素を微生物中で発現させる工程;
(b) 菌体内に蓄積するシアル酸転移酵素を含む蛋白凝集物または沈殿物から該 酵素を 8M の尿素により抽出する工程;
(c) 工程 ) で得られた抽出物を脱変性用組成物で希釈した後に 4°Cで 12時 間以上放置して 2〜3 の尿素を含む 1次希釈物を得る工程;
(d) 工程 (c) で得られた 1次希釈物をさらに脱変性用組成物で希釈した後に 4 8 時間以上放置して 1〜2 Mの尿素を含む 2次希釈物を得る工程;および
(e) 工程 (d) で得られた 2次希釈物を 2価のカチオンの存在下で透析して尿素 を除去し脱変性シアル酸転移酵素を得る工程
を含む方法。
28. 工程(c) で用いる脱変性用組成物が 1〜 2M尿素, 20 mM MOPS-NaOH, 0. 5M NaCL 20 m ラクトース, 0. 5 mM EDTA (pH 7. 0)を含み、 工程 (d) で用いる脱 変性用組成物が 20 mM MOPS-NaOH, 0. 5M NaCl, 20 m ラクト一ス, 0. 5 mM EDT A (pH 7. 0)を含む、 請求の範囲第 27項に記載の方法。
29. シアル酸転移酵素が GalNAcひ 2, 6-シアル酸転移酵素または Gal S 1, 4GlcNAc a 2, 6-シアル酸転移酵素である請求の範囲第 27項に記載の方法。
