明 細 謇 プラスミ ド 技術分野
本発明は、 遗伝子組換え技法において使用されるプラスミ ドベクタ一、 及び該 プラスミ ドベクターを用いた遣伝子の発現方法に関する。 さらに本発明は、 かか るブラスミ ドベクターを用いる目的遗伝子の単雜方法に関する。 また本発明は、 遺伝子工学試薬として有用な制限酵素及びその遺伝子、 更に詳細には A c c I I I 制限酵素及びその DN Aに関する。 背景技術
遺伝子組換え技法によって目的とする遗伝子の発現システムを構築する際には 、 用いる宿主の RNAボリメラーゼに認識されるプロモーター支配下に目的遺伝 子を配 Sすることにより、 該遺伝子の発現を制御することが行われる。 しかし、 宿主に対して有害なタンパク質をコードする遗伝子の場合には、 用いるプロモー ターの発現制御が厳密に行えないことに起因して該遺伝子産物の発現のためにプ ラスミ ドの構築自体が不可能になることがある。
そのような問題点を解決すベく開発された発現システムとしては大腸菌を宿主 とし、 大媵菌に感染するバクテリオファージ T 7の RNAポリメラ一ゼを利用 した P E T—システム (ノバジヱン社製) がある [ジャーナル ォブ モレキュ ラー バイオロジー (J. Mol. Biol. ) 第 189巻、 第 113 〜130頁 (1986) 、 ジーン (Gene) 、 第 56巻、 第 125 〜135頁 U987) ] 。 p E T—システムは 、 プロモーター認識の特異性及び転写活性の高い T 7 RNAボリメラーゼを大腸 菌内で発現させ、 この T 7 R NAボリメラーゼが発現べクタ一上の T 7プロモー 夕一下流に配 Sされた目的遗伝子を耘写し、 高発現されるシステムである。 目的 の遺伝子の転写は T 7 RNAボリメラーゼによってのみ起こるため、 該ボリメラ ーゼを生産しない宿主中では目的の遗伝子を発現させることなくプラスミ ドの構 築をすることが可能であり、 目的遺伝子を宿主の有する RNAボリメラーゼに認 識されるプロモーター支配下において発現システムを構築する場合とは異なりブ ラスミ ドの構築自体が不可能になることはない。
しかし、 T 7 RNAボリメラーゼ遗伝子は、 λ—ファージベクター上にクロー ニングされて発現用の宿主に溶原化されているために宿主を自由に選ぶことがで きず、 宿主を変更しょうとする場合には煩雑な操作が必要となる。 さらに、 宿主 中の Τ 7 RNAボリメラーゼの発現があまり厳密に制御されていないため、 非誘 等状想の宿主内でも Τ 7 RNAボリメラーゼが発現されており、 その結果、 発現 ベクター上の Τ 7プロモータ一下流に配 された目的遗伝子の発現が非誘導状態 においても起こる。 このような目的遣伝子の非誘導状 «8における発現を低下させ るために、 Τ 7 RNAボリメラーゼの阻害物質である Τ 7リゾチームを用いて Τ 7 RNAボリメラーゼの活性を阻害したり [ジャーナル ォブ モレキュラー バイオロジー ( J. Mol. Biol. ) 第 219巻、 第 37〜44頁 (1991) ]、 T 7プロモ 一ター下流にラクトースオペレーターを配置して T 7 RNAボリメラーゼの T 7 プロモーターへのアクセスを防いだりしている [ジャーナル ォブ モレキユラ 一 バイオロジー ( J. Mol. Biol. ) 、 第 219巻、 第 45〜59頁 (1991) ]。
しかし、 これらの措置も転写活性の高い T 7 RNAボリメラーゼに対しては十 分でなく、 非誘導状 ISの T 7 RN Aボリメラーゼの活性を完全に阻止することは できない。 そのため、 目的の遗伝子産物が宿主に対して致死的な場合、 ブラスミ ドの構築はできてもその遺伝子を発現させるための形 »転換体を作製できないこ とがある。 つまり、 p E T—システムには宿主を自由に変更できないこと、 およ び T 7 RNAボリメラーゼの発現制御が正確でないこと、 以上 2つの解決すべき 問題点がある。
—方、 パクテリオファージ T 7に性質が類似したパクテリオファージとして 、 サルモネラ属钿菌 (Salmonella typhimuriuin) に感染するバクテリオファージ S P 6がある [サイエンス (Science ) 、 第 133巻、 第 2069〜2070頁 (1961) ] 。 バクテリオファージ S P 6が生産する RNAボリメラーゼは、 分子量が約 1 0 万のシングルべプチドであり、 プロモーター認識の特異性が強く転写活性も高い ため試験管内 R NA合成によく用いられている [ジャーナル ォブ バイオロジ カル ケミストリー ( Biol. Chem. ) 第 257巻、 第 5772〜5778頁 (1982) 、 ジ ヤーナル ォブ バイオロジカル ケミストリー (J. Biol. Chem. ) 、 第 257巻 、 第 5779〜5788頁 (1982) ] 。 さらに、 S P 6 RN Aボリメラーゼ遗伝子はすで にクローニングされ、 大腸菌内で大量発現されている [ヌクレイック ァシッズ リサーチ (Nucl. Acids Res. ) 第 巻、 第 2653〜2664頁 (1987) ] 。
宿主に対しその発現産物が致死的に働く遺伝子の例として、 制限酵素 (Restri ction Endonuc lease) 遺伝子がある。 本来制 酵素は、 産生している微生物の体 內に侵入したファージ等の外来 D N Aを切断することによる自己防御に利用され る。 一方、 制限酵素を産生する微生物は、 大抵の場合、 制限酵素と同じ塩基 K列 を認識する修飾酵素を産生し、 制限酵素による切断から自己の DNAを防御して いる。 すなわち、 修飾酵素は S»する塩基配列中の塩基の一つ以上にメチル基を 添加して DN Aを修飾し、 同じ配列を β織する制限酵素の結合、 或いは該酵素に よる D NA切断を不可能にする。 この機構を制限修飾系と呼び、 構成する制限酵 素及び修飾酵素の遗伝子対を制限修飾系遺伝子と呼ぶ。 従って、 制限修飾系遗伝 子の修飾醉素遺伝子が欠損した微生物中で制限酵素遺伝子が発現すると、 激生物 自身の DNAが切断され細胞は死に到る。 実際、 b o I制 IS修飾系遺伝子には 二種類の修飾酵素遺伝子が存在しているが、 このうちの一方の修飾酵素遺伝子の みを共存させることによる不完全なメチル化では宿主 D N Aの修飾が不完全とな り、 制限酵素遺伝子のクローニングが不可能であることが報告されている [ヌク レイツク ァシッズ リサーチ (Nucl. Acids Res. ) 第 2 1巻、 第 2 3 0 9〜2 3 1 3頁 U 9 9 3 ) ] 0 また、 制限修飾系遺伝子を保持する細胞より制 IS修飾系遺伝子の脱落が起こる と、細胞内に存在する修飾活性が不足してゲノム DNAのメチル化が不完全にな り、 細胞内に残存するごく少量の制限酵素によって自分自身のゲノム DNAが切 断されて致死的となることが明らかにされている [サイエンス (Science ) 第 2 6 7巻、 第 8 9 7〜8 9 9頁 ( 1 9 9 5 ) ] 。 このように制 K修飾系を構成する 修飾酵素非共存下では、 制 R酵素は細胞にとつて めて有害なタンパク質であり 、 その遗伝子を単独でクローニングし、 さらに発現させることはこれまでの技術 では不可能であった。
ここで制限酵素について触れると、 その酵素学的性質により制限酵素は I型、 I I型、 I I I 型に分類する事ができる。 その中でも、 特定の DNA堪基配列を認識 し、 その配列の內部戒いは近傍の特定の位置で切断する II型制限酵素は、 遠伝子 工学の分野で広く使用され、 これまでに種々の特異性を有する制限酵素が搛々な 微生物から単離されている [ヌクレイック ァシッズ リサーチ (Nucl. Acids Res. ) 第 24巻、 第 223〜235 頁 (1996) ] 。 以下、 本明钿害では I I型制限酵素を 「制限酵素」 と記載する。 しかし、 微生物によっては、 産生している制限酵素量 が少なかったり、 複数の制限酵素を産生している場合がある。 例えば、 制 K酵素 A c c【I I は通商産業省工業技術院微生物工業技術研究所 〔あて名: 日本国茨城 県筑波郡谷田部町柬 1丁目 1番 3 (郵便番号 3 0 5 ) 〕 に、 昭和 6 0年 1 1月 9 日 (原寄託日) より F E RM B P - 9 3 5として寄託されているァチネトバク 夕一 ·力ノレ: 3了セティカス (Acinetobacter caicoaceticus ) (以下 A c c ¾と 略する) により生産されるが、 該酵素の発現 iは低く、 また本菌は A c c I及び A c c II制限酵素を同時に産生している。 従って、 高純度、 安価な試薬として制 限酵素 A c c I I I を本菌を用い提供するには 度な製造技術を要した。 高純度で 安価な試薬として制限醉素を提供する場合、 目的の制限酵素遺伝子を単餱し、 目 的制限酵素のみを大量生産する遺伝子工学的生産は有効な手段である。 この目的 を達成するため、 従来制限酵素遗伝子を単餱する幾つかの方法が報告されている まず制限酵素を産生している徼生物のゲノム D N Aを適当な制限酵素により切 断し、 その断片を適当なプラスミ ドベクターに挿入し、 制限酵素遗伝子が発現す るクローンを選択する 「ショッ トガン』 方法が挙げられる。 目的クローンのスク リーニング法として、 例えば、 宿主に制限修飾系遺伝子が導入されると、 宿主が 自已防衛機能を獲得することを利用し、 ファージの感染に対する耐性を指標にし
、 制限修飾系遗伝子を単離する方法 [Ps t I :プロシーディングス ォブ ザ ナショナル アカデミー ォブ サイエンス ォブ ザ USA (Proc.Natl. Acad. Sci. )第 78巻、 第 1503〜 1 507頁 (1 981) 〕 がある。 しかし 、 本方法では、 制限修飾系遺伝子のサイズが単雜可能な範囲に収まる必要があり 、 さらに単鎗された制限修飾系遗伝子の発現が、 宿主を選択的に生存させうるに 十分なファージ耐性を示す必要がある。 一方、 制限修飾系遗伝子の一般的な特徴 として、 制限酵素遺伝子と修飾酵素遺伝子がゲノ厶上で近接していることが挙げ られ、 実際、 これまでに得られた制限修飾系遺伝子の多くで確認されている [ヌ クレイック ァシッズ リサーチ (Nucl. Acids Res.)第 1 9巻、 第 2539〜 2566頁 ( 1 991 ) ]。 そこで、 上記特徴を利用し、 修飾酵素遠伝子の発現 を指標とし制限修飾系遗伝子をスクリーニングする方法 [特開昭 63 - 8798 2、 ヌクレイック ァシッズ リサーチ (Nucleic. Acids Res.)第 1 9卷、 第 18 31〜 1 835頁 (1 991) ] がある。 しかし本法は、 修飾酵素遺伝子に制限 酵素遺伝子が近接していない場合、 制 IS酵素遺伝子を得ることはできない。
さらに上記の Γショットガン」 方法には、 根本的な問題がある。 それは、 ゲノ ム D N A供給源となる生物と宿主との面の転写翻訳機構の違 、である。 例えば制 限修飾系遗伝子に付随するプロモータ一及びリポゾ一ム結合部位が宿主内でうま く機能せず、 十分な遺伝子発現が起こらない場合には、 たとえ目的の遗伝子を含 有する形質転換体を得られていても、 その形質転換体を選択する事に多大な労力 を要する。 この点を回避するには、 例えば、 制限酵素タンパク質のアミノ酸配列 を解析し、 そのデータを基に PCRによる DNA增幅法により、 制限酵素産生菌 のゲノ厶 DN Aより制限酵素遺伝子を得、 既知のタンバク K発現システムを利用 する方法がある [特開平 6— 277070 ] 。 しかし、 従来のタンパク質発現シ ステムでは、 制限酵素の存在が宿主に対し致死的となるため、 何らかの方法、 例 えば該酵素と制限修飾系を構成する修飾酵素を共存させ、 宿主を保護する必要が める。
前述の制限酵素遺伝子の単離方法はいずれも、 該遗伝子とともに制限修飾系遠 伝子を構成する修飾酵素遺伝子を同時に単離することが必要であるが、 制限酵素 遗伝子単独で単離する方法も報告されている [ヌクレイック ァシッズ リサ一 チ (Nucleic. Acids. Res.)第 22巻、 第 2399〜2403頁 (1 994) ] 。 しかし、 この方法で例示されているのは、 酵素活性至適温度が 70で付近の制限 酵素をコードする遗伝子の単離であり、 宿主培養温度近辺で比活性の高い制限酵 素をコードする遗伝子を単離する場合には、 やはり修飾酵素遺伝子を共存させて いる。
また例外的に、 制 酵素 Dpn Iの様に DNA認識配列中の特定の核酸塩基が メチル化による修飾を受けていないと、 切断活性を示さなレ、制限酵素が存在する 。 このような性質を有する制 IS酵素遺伝子は、 宿主菌を選択することにより、 例 外的に、 他の特定の遺伝子非共存下でも単雛可能だと考えられる。 実際、 II型制 限酵素ではないが、 メチル化を受けた核酸塩基を含む特定の DN A塩基配列を認 識し、 DNA切断活性を有する Mr rタンパク質をコードした mr r遺伝子が単 離されている [ジャーナル ォブ パクテリォロジ一(J.Bacteriol.)第 1 73巻 、 第 5207〜5219頁 (1991) ]。
上述したように、 従来の技術により制限酵素途伝子の単離を行うには、 特殊な 場合を除き、 該遺伝子と制 K修飾系遺伝子を構成する修飾酵素遺伝子を同時に発 現させる必要があった。 発明の開示
従って、 本発明の第 1の目的は、 その遗伝子産物が宿主にとって致死的、 ある いは有害であるため、 従来技術では単雜が困難であったり、 発現システムの構築 が困難であつた遗伝子であっても単離することができ、 さらに宿主に導入して効 率よく該タンパク を発現させることのできるプラスミ ドベクターを提供するこ とにある。 本発明の第 2の目的は、 このブラスミ ドベクターを用いた目的遗伝子 の発現方法を提供することにある。 本発明の第 3の目的は、 このブラスミ ドべク 夕一を利用することによる、 目的遺伝子の単離方法、 特に制限修飾系を構成する 修飾酵素遺伝子の共存を必要としない、 制限酵素遺伝子の単離方法を提供するこ とにある。 本発明の第 4の目的は、 A c c i n 制限酵素活性を有するボリぺブチ ドを提供することにある。 さらに本発明の第 5の目的は A c c【1〖 制限酵素活性 を有するボリペプチドをコードする DN Aを提供することにある。
本発明者らは p E T—システムの問题点を解決するため、 発現制御が正確な新 しい発現システムとして、 まず S P 6 R NAポリメラーゼを用いた発現システム の構築を行い、 該システムの評価を試みた。
該発現システムは、 S P 6 RN Aボリメラーゼ遗伝子を iBini Fプラスミ ドに祖 み込んだシステムプラスミ ドを利用して構築されているため、 目的の遗伝子を S P 6プロモーター下流にクローニングした発現ベクターとこのシステムプラスミ ドを同時に宿主に導入することにより、 様々な大 I»菌で目的遗伝子の発現システ ムを構築することができる。 さらに、 1 a cプロモーターとアンチセンス技術を 利用してシステムブラスミ ド上の S P 6 R NAボリメラーゼ遺伝子の厳密な発現 制御を可能にした。 3—ガラクトシダーゼ遺伝子をレボーター遺伝子に用いて本 発現システムを評価した結果、 このシステムにおいて S P 6 R NAポリメラーゼ 遺伝子の発現は正確に制御されており、 非誘導状態で S P 6 RNAボリメラーゼ がほとんど発現していないこと、 および誘導後には目的遗伝子の高発現に必要十 分な S P 6 RNAボリメラーゼが発現し、 目的遺伝子が高発現することが明らか になった。
しかしながら大腸菌を宿主とした場合には、 S P 6プロモーターは大腸菌の R NAボリメラーゼによって非常に弱いながら δ織されるために、 その下流にある 目的遺伝子は非誘導状懋においてもごく少量発現することが明らかになつた。 こ のため、 遺伝子産物が大瞄菌に対して非常に有害で致死的に働く場合には発現シ ステムの構築は不可能となり、 本発明の目的を充分に達成することはできないこ とが判明した。
そこで、 本発明者らはさらに 意研究した結果、 目的 ¾伝子の発現を 2種類の 制御方法、 すなわち該遺伝子のコピー数の制御、 およびプロモーターを介した転 写制御とを組み合わせてコントロールする新規なシステムを採用することにより 、 意外にも (i)非誘導状! 18では目的遺伝子の発現を検出できないレベルに抑える ことが可能であり、 (i i)発現誘導を行つた場合には目的遗伝子を含有するブラス ミ ドのコピー数を上昇させると共に RNAボリメラーゼを発現させ、 該 RNAボ リメラーゼの認識するプロモーター下流に配置された目的遺伝子が転写、 翻訳さ れることを見いだした。 即ち、 本発明のブラスミ ドベクターは、 上記課題を解決 することができる、 遺伝子工学的分野において画期的な性 を有するプラスミ ド ベクターであり、 本発明者らはさらに該ベクターを利用した目的遗伝子の発現方 法を完成するに至った。
更に、 上記プラスミ ドベクターを用いて、 宿主に対しその発現産物が致死的に 働く遺伝子を単難する方法、 特に、 従来技術の欠点を克服した制 K酵素遠伝子の 単離方法を完成するに至つた。
また上記の方法を利用することにより、 従来得られていなかった A c c III 制 限酵素をコードする DNAを A c c lll 修 «ί酵素を共存させることなく単離する ことができ、 更に A c c n i 制限酵素を大量発現させることが確認できた。 即ち、 本発明の要旨は、
( 1 ) 宿主が本来有しない RNAボリメラーゼによって認織される、 目的遗伝 子の発現を制御するプロモータ一配列と、 外来因子による誘等によってコビー数 を增加させる複製起点とを有することを特徼とするプラスミ ドベクター、
(2) プロモーター配列がパクテリオファージ由来の RNAボリメラーゼによ つて認識されるものである前記 (1) に記載のブラスミ ドベクター、
(3) プロモーター配列が SP 6ファージ由来の RNAボリメラーゼによって 認識されるものである前記 (2) に記載のプラスミ ドベクター、
(4) プロモーター配列が配列表の配列番号 30で示される塩基配列を含むも のである前記 (3) に記載のプラスミ ドベクター、
(5) 複製起点がプロモーターの制御下にある、 前記 (1)〜(4) のいずれ か 1項に記載のブラスミ ドベクター、
(6) 複製起点が 1 a cプロモータの制御下にある、 前記 (1)〜(5) のい ずれか 1項に記載のブラスミ ドベクター、
(7) 選択マーカーとして薬荊耐性遺伝子を有する、 前記 (1)〜(6) のい ずれか 1項に記載のプラスミ ドベクター、
(8) PACE 60 K PACE 61 1、 p ACE 70 K PACE702か ら選択される前記 (7) に記載のブラスミ ドベクター、

本発明の第 1の発明はプラスミ ドベクターに関する。 すなわち、 宿主が本来有 しない RNAボリメラーゼによって認»される、 目的遗伝子の発現を制御するブ 口モーター配列と、 外来因子による誘導によってコビー数を增加させる複製起点 とを有することを特徴とするプラスミ ドベクターに関する。 該プラスミ ドベクタ 一が有する目的遺伝子の発現を制御するプロモーター配列としては、 特に限定さ れるものではないが、 ある特定の RNAボリメラーゼによって認識されるプロモ 一ター配列、 例えば T 7、 T 3、 S P 6等のパクテリオファージ由来の RNAボ リメラーゼによって認織されるプロモーター配列が使用でき、 とりわけ S P 6フ ァージ由来の RN Aボリメラーゼによって認戮されるプロモーター配列が好まし い。 S P 6ファージ由来の RNAボリメラーゼによって認識されるプロモーター 、 すなわち S P 6プロモーターの最小領域の塩基配列を配列表の配列番号 3 0に 示す。 さらに、 該プラスミ ドベクターは選択マーカーとして使用される薬剤耐性 遗伝子を有することができる。
また、 本発明のブラスミ ドベクターのコピー数の制御、 すなわち該ブラスミ ド ベクターの複製起点の機能の制御を行う外来因子としては、 特に限定されるもの ではないが、 例えばラクトース又はイソプロビル一 ;8— D—チォガラクトシド ( I P T G) のようなその構造類似物の添加、 ガラクトース又はその構造類似物の 添加、 ァラビノース又はその構造類似物の添加等の種々の化学物質の添加或いは トリプトファン囊度の低滅、 形質転換体の培養温度の謂螯等を挙げることができ る。 これらの因子により制御可能なプロモーターの支配下に複製起点をおくこと によってコピー数の制御が可能となる。 この目的に使用可能なプロモーターとし ては上記の目的を達成できるものであれば特に制限はなく、 たとえば宿主が大腸 菌である場合には l a c、 t r p、 t a c、 g a l、 a r a、 P L 等のプロモー ターを使用できる。 またこれらのプロモーターは RNAボリメラーゼ遺伝子の発 現誘導にも使用することが可能である。
本発明のブラスミ ドベクターの複製起点は、 特に限定されるものではないが、 例えばランナウエイプラスミ ドの複製起点であり、 複製起点が 1 a cプロモータ の制御下にあるプラスミ ドベクターが例示される。 この場合には、 コピー数の制 御はラクトース及びその構造類似物の添加、 特に好ましくはィソブロビル一 β― D—チォガラクトシド ( I P T G) の添加によって達成される。
本発明の第 2の発明は、 本発明のブラスミ ドベクターに発現させようとする目 的遺伝子を組み込んだプラスミ ドベクターと、 該プラスミ ドベクター上のプロモ
—ター配列を認識する RNAボリメラーゼ遺伝子とを宿主に導入し、 外来因子に より該ブラスミ ドベクターのコピー数の堆加を誘導すると共に該 RN Aボリメラ —ゼの発現の誘導を行って目的遗伝子の転写、 翻 IRを行うことを特徴とする、 目 的遣伝子の発現方法に関する。 ブラスミ ドベクター上のプロモーター配列を認識 する RNAボリメラーゼ遺伝子としては、 特に限定されるものではないが、 例え ば前記のようなパクテリオファージ由来の R N Aボリメラーゼ遺伝子を挙げるこ とができ、 とりわけ S P 6ファージ由来の RNAボリメラーゼ¾伝子が好適であ る。 また、 かかる RNAボリメラーゼの発現の、 又はブラスミ ドベクターのコピ 一数の増加の誘導を行う外来因子としては、 特に限定されるものではないが、 例 えばラクトース叉はイソプロビル一 /9一 D—チォガラクトシド ( I P T G) のよ うなその構造類似物の添加、 ガラクトース又はその構造類似物の添加、 ァラビノ ース又はその構造類似物の添加等の種々の化学物質の添加或いはトリブトファン 濃度の低減、 形質転換体の培養温度の調整等を挙げることができる。
プラスミ ドベクターのコピー数の増加の誘導と R NAポリメラーゼの発現の誘 導とは、 それぞれ別の外来因子の作用により行うことも、 あるいは同一の外来因 子の作用により行ってもよい。 また、 この場合 RNAボリメラーゼをコードする 遗伝子は、 宿主の染色体上に組み込まれていても、 あるいは本発明のプラスミ ド ベクターとは異なる別のブラスミ ドベクターやファージベクターによって宿主に 導入されていてもかまわない。 後者の場合には目的に応じて宿主を変更すること が容易に行える。 さらに、 この場合には RNAボリメラーゼ遺伝子を有するべク ターの宿主への導入は、 本発明のプラスミ ドベクターより先に行われても、 ある いは後で行われてもかまわない。
本発明における目的遗伝子は、 特に 15定されるものではないが、 宿主に対し致 死的となる、 あるいは有害なタンパク質をコードするものである場合にその意義 が発揮される。 このようなタンパク質としては、 例えば前記のような制限酵素、 その他の核酸分解醇素、 核酸結合性のタンパク質、 タンパク質分解酵素などが挙 げられる。
本発明は、 前記のように目的 *伝子の発現を 2種類の制御方法、 すなわち該遗 伝子のコピー数の制御、 およびプロモーターを介した転写制御とを組み合わせて コントロールするシステムを提供する。 本発明におけるプラスミ ド上にクロー二 ングされた目的遗伝子の発現の制御は、 従来のものと異なり極めて厳密であり、 非誘等状懋ではその発現を検出できないレベルに抑えることか可能である。 発現 誘導を行いブラスミ ドのコビー数、 すなわち目的遺伝子のコピー数を上界させる と共に RN Aボリメラーゼを発現させると、 該 RNAボリメラーゼの作用によつ てプロモーターに制御されて目的遺伝子が転写、 翻訳される。
従って、 本発明のブラスミ ドベクターを用いることによって、 従来技術では困 難であつた宿主に対して有害な、 あるいは致死的な遗伝子を含有する形質転換体 を作製し、 さらに該遺伝子の発現を行うことが可能となる。
例えば、 本発明のプラスミ ドベクターを使用し、 N s p7524lI I 制限酵素をコ 一ドする遺伝子のクローニングを試みたところ、 対応する修飾酵素遺伝子を共存 させることなく大腸菌に保持させることができた。 さらに、 得られた形質転換体 に RNAボリメラーゼ遺伝子を含有するシステムブラスミ ドを導入して発現誘導 を行うことによりその菌体内に N s p 75241 I I 制限酵素を発現させることも可能 であつた。
また、 本発明のプラスミ ドベクターを遺伝子ライプラリー作製のためのクロー ニングベクターとして用いることにより、 従来の方法を用いた場合には取得でき なかった遺伝子を単離し、 さらにその発現産物の活性を確認することを 1つの宿 主で行うことが可能となる。 もちろん本発明のブラスミ ドベクターの利用は宿主 に対して有害な産物をコ一ドする遗伝子のクローニングに限られるものではなく 、 一般的なブラスミ ドベクターとしても利用可能である。
本発明に係る RNAボリメラーゼ遗伝子を宿主に導入する方法として、 例えば 、 プラスミ ド pFSP 6、 ファージ Ml 3 s p 6を用いることができる。 プラス ミ ド pFSP 6の詳細な構築方法は参考例 (2) に記載されている。 該プラスミ ドで形質転換された大腸菌 HB 101は Escherichia coli HB 10 t/pFS P 6と命名、 表示され、 平成 7年 12月 22日 (原寄託日) より通商産業省工業 技術院生命工学工業技術研究所 〔あて名 ·. 日本国茨城県つくば市東 1丁目 1番 3 号 (郵便番号 305 ) 〕 に受託番号 FERM BP- 5742として寄託されて いる。 ファージ Ml 3 s p 6の構築方法は実施例 2 (6) に記載されている。
以下に、 プラスミ ド pFSP 6を用いた場合を例に挙げて、 本発明について更 に詳しく説明する。
まず、 上記のシステムプラスミ ド pFSP 6を利用して目的遺伝子の発現を行 うことができるブラスミ ドとして、 SP 6 RNAボリメラーゼによって認識され るプロモータ一配列と、 レポ一ター遺伝子としての^一ガラクトシダーゼ逮伝子 とを有するマルチコピーのモデル発現プラスミ ドであるプラスミ ド pMSP6L 、 PMSP6F (第 1図) 、 pMSP60 (第 2図) を構築し、 これらのモデル 発現ブラスミ ドを用いて行った試験結果について説明する。
これらのモデル発現ブラスミ ドの詳細な構築方法は参考例 (3) に記載されて いる。 これらのプラスミ ドには SP 6プロモーター配列として SP 6プロモータ 一の最小領域 (PMSP6L) , ネイティブな SP 6プロモーター領域 (p S P 6 F)、 又は SP 6プロモーターの最小領域と 1 a cオペレーター領域 (pM SP 60) がそれぞれ導入されている。
プラスミ ド pMSP 6 L又は pMSP 6 Fのみを表 1に示す大腸菌に導入した 場合には、 表 2に示されるように宿主大睡菌が SP 6RNAボリメラーゼを発現 しないにもかかわらず8—ガラクトシダーゼ活性が認められ、 その量は SP 6ブ 口モーターを有していないプラスミ ド pMS 434を導入したものよりも高い。 すなわち、 SP 6プロモーターが大腸菌由来の RNAボリメラーゼによって認識 され、 その下流の ;3—ガラクトシダーゼ遗伝子が転写、 翻訳されていると考えら れる。 また、 大腸菌 MR i 80は p c n B遺伝子に変異を有し、 上記のモデル発 現ブラスミ ドのような Co 1 E 1由来の複製起点を持つブラスミ ドのコピー数が 親株である MR i 7に比べて 1/6〜1Z1 3に低下する株であるが、 これを宿 主に用いた場合には 9一ガラクトシダーゼ活性も MR i 7の 1Z7〜1Z1 4に 滅少している。 このことから、 ^一ガラクトシダーゼ ¾伝子の発現のレベルと導 入されたブラスミ ドのコピー数との閎には相関があることが示される。 表 1

MRi80 17 27 4 O 97/34006 次に、 同じモデル発現プラスミ ドを上記のシステムブラスミ ド pFSP 6とと もに大腸菌に導入したうえ、 SP 6 RNAボリメラーゼ遗伝子の発現を誘導しな い条件下で ーガラクトシダーゼ活性を謂べてみると、 表 3に示されるようにシ ステムプラスミ ドを有さない大腸菌の場合と同程度の8—ガラクトシダ一ゼ活性 しか認められない。 このことは非誘導状 «8においてはブラスミ ド PFSP 6上の SP 6 RNAボリメラーゼ遗伝子の発現がほぼ完全に抑えられていることを示し ている。 一方、 イソプロピル一^一 D—チォガラクトシド ( I PTG) を添加し て SP 6 RN Aボリメラーゼ遗伝子の発現を誘導した場合には; δ—ガラクトシダ ーゼ活性は 1 8〜32倍上昇しており、 SP 6プロモーターを介して 3—ガラク トシダーゼ遺伝子が発現可能であることを示している。 特に大腸菌 MR i 80を 宿主とした場合には、 非誘導状想の 9一ガラクトシダーゼ活性は極めて低いレべ ルにおさえられているが、 誘導 4時間後には非誘導状態の 25〜32倍に上昇し ている。 表 3

さらに SP 6プロモーター下流に 1 a cオペレーター配列を挿入されているブ ラスミ ド PMSP 60では、 他のモデル発現プラスミ ドに比べて非誘導条件下で の /9一ガラクトシダーゼ活性が低下しているが、 その発現を完全におさえること はできない。 以上の結果は、 マルチコピーのブラスミ ドを使用した発現システム では目的遗伝子の発現を完全におさえることが難しく、 また、 この問題を解決す るにはシステムプラスミ ド上の RNAボリメラーゼ遗伝子の発現制御よりも目的 遗伝子を含むブラスミ ドのコビー数による制御の方が効果があることを示唆して いる。
そこで、 本発明者らはこれらの綰果をもとに新たな発現べク夕一の開発を行 、 、 本発明のプラスミ ドベクターを見出すに到った。
即ち、 本発明者らは非誘導伏想での発現レベルを下げ、 誘導後の発現量を確保 することができるランナウエイプラスミ ドベクターを構築した。 まず、 システム ブラスミ ドでは SP 6RNAボリメラーゼの発現が I PTGによって誘導される ことを考慮して、 I PTGの誘導によりコピー数を增加させることのできるラン ナウエイ複製起点を有するブラスミ ドを第 3図に示す操作に従って構築した。 ま ず、 プラスミ ドベクター pUC 1 9 (宝酒造社製) の Nd e Iサイトに配列表の 配列番号: 3および配列番号: 4にその埴基配列を示した 2つの DNA縝から作 製される 106Nd e I DNA断片を導入して得られるブラスミ ド pUC 106 Aより、 l a cプロモーター、 およびオペレーター領域を含む Pvu II断片を除 いたプラスミ ド pUC 106 Ad POを構築する。
次に該ブラスミ ドを铸型とし、 該ブラスミ ドの複製起点である RNAII領域近 傍に設定された RNAIIAプライマー (配列表の配列番号: 5に RNAIIAブラ イマ一の塩基配列を示す) と 106Nd e I配列末端部分に設定された 1870ブラ イマ一 (配列表の配列番号: 6に 1870プライマーの ¾基 B2列を示す) を用いた P CRによって得られる RNA1870断片をプラスミ ド pSTV28 (宝酒造社製) の Xb a Iサイトに揷入したプラスミ ド pHS 2870を得ることができる。 構 築されたブラスミ ド pHS 2870を第 4図に示す。 通常、 大腸菌 1細胞あたり に該プラスミ ドは 30コピー程度存在するが、 I PTGの添加によりそのコピー 数は数百に堆加する。 次に該プラスミ ドよりプラスミ ド PSTV28由来の P 1 5 A複製起点を含む Ac c I -Ns p I断片を除き (プラスミ ド pCRS 01)、 さらに不要の制限 酵素サイトを含む E c oR I -Xba I断片を除去した後 (ブラスミ ド pCRS 02)、 ブラスミ ド pMJR 1560 [ジーン (Gene)第 51巻、 第 225〜 267頁 (1987) ] 由来のラクトース · レブレッサー ( 1 a c Iq )遺伝子を含む DNA 断片を導入したプラスミ ド pCRS 04を構築することができる。 第 5図にブラ スミ ド pHS 2870からプラスミ ド pCRS 04を構築する ¾程を示す。 第 6 図にこうして構築されたブラスミ ド pCRS 04を示す。 該プラスミ ドは I PT Gで誘導されるランナウエイプラスミ ドであり、 通常は大腸菌 1細胞あたりに 1 〜2コピー存在するが、 I PTGの添加によりそのコピー数は数百に墦加する。 該プラスミ ド上に挿入される目的遺伝子の発現を制御するために、 該プラスミ ドの Nh e Iサイトに S P 6プロモーター配列と 1 a cオペレーター配列とを含 む 2本鎖オリゴヌクレオチド、 PSP, -0, .c EXリンカ一を導入し、 ブラスミ ド PACE 60 1を構築することができる。 PSPe -Ο, .ο EXリンカ一は配列 表の配列番号: 7および配列番号: 8にその塩基配列を示した 2つの DNA鎮か ら作製される。 該プラスミ ド pACE 601は選択マーカーとしてクロラムフエ 二コール耐性遺伝子を有するランナウエイブラスミ ドベクターである。
また、 上記のプラスミ ド pCRS 04の Nh e Iサイ卜にプラスミ ドベクター PUC 1 1 8 (宝酒造社製) 由来の /8—ラクタマーゼ遗伝子を含む BspHI断 片を導入した後 (ブラスミ ド pCRS 70)、 さらにクロラムフエ二コール耐性 遗伝子を含む Nc 0 I - Bs aA I断片を除いて上記の P„, -0,.c EXリン カーに置換することにより、 該リンカ一の挿入方向の異なるプラスミ ド pACE 701および pACE 702を構築することができる。 これらのプラスミ ドは選 択マーカーとしてアンビシリン耐性遺伝子を有するランナウエイブラスミ ドべク ターである。 こうして得られたブラスミ ド pACE 601、 PACE 701および pACE 702を使用した場合の目的遺伝子の発現制御の能力は上記同様に 一ガラクト シダーゼ遗伝子をレボーター遗伝子として謂べることができる。 プラスミ ド pM S 434 [ジーン (Gene)第 57巻、 第 89〜99頁 (1987) ) ] を铸型とし、 ブライ マー t r pA— N— Nc 0 I、 およびプライマー 1 a cZ— C一 Nc o Iを用い た P C Rによって增幅される S—ガラクトシダーゼ遗伝子を含む DN A断片を、 上記の 3つのブラスミ ドそれぞれの SP 6プロモーター下流に存在する Nc 0 I サイトに導入したモデル発現プラスミ ドを構築することができる。 プライマー t r pA-N-Nc o およびブライマー l a cZ— C一 Nco lの塩基配列を それぞれ配列表の配列番号: 9および配列番号: 1 0に示す。 こうして得られた プラスミ ドはそれぞれブラスミ ド pACE 601 Z、 pACE 701 Zおよび p ACE 702 Zと命名されている。 これらのモデル発現プラスミ ドで形質転換さ れた大腸菌では 一ガラクトシダーゼ活性は全く検出されず、 ーガラクトシダ 一ゼ遺伝子の発現が極めて厳格に制御されていることが示される。
本発明のプラスミ ドを用いて、 宿主に対しその発現産物が致死的に働く遺伝子 を単雜し、 さらに該遺伝子を発現させることが可能なことを Ns P7524 III制限 酵素遗伝子を用いて確かめることができる。 ブラスミ ド PBRN3は Ns p7524
ΠΙ制限修飾系遺伝子を含有しており、 該ブラスミ ドで形質転換された大腸菌 M C 1 061は Escherichia coli MC 1061 /p BRN 3と命名、 表示され、 平成 7年 9月 28日 (原寄託日) より通商産業省工業技術院生命工学工業技術研 究所 (あて名: 日本国茨城県つくば市東 1丁目 1番 3号 (郵便番号 305 ) 〕 に 寄託番号 FERM BP- 5741として寄託されている。 該形 »転換体より調 製したブラスミ ド pBRN3を铸型とし、 ブライマー L一 ORF、 およびブライ マー Ns pR— ORF 3を対とした PCRによる DNA堆幅反応により、 Ns p 7524 III制限酵素遺伝子のみを含む DNA断片を取得することができる。 配列表 の配列番号: 1 1および配列番号: 12にブライマー L一 ORF、 およびプライ マー Ns pR— ORF 3の塩基配列を示す。 得られた DN A断片を上記のプラス ミ ド PACE601の SP 6プロモーター下流の Nc 0 Iサイトに揷入して得ら れるプラスミ ド pACE 601— Ns p IIIを大腸菌 HB 101に導入すること により形質転換体 Escherichia coli HB 101/pACE 601- Ns p III を得ることができる。 該形質転換体は Ns p 7524 〖II修飾酵素遗伝子が共存しな いにもかかわらず Ns p 7524【II制限酵素遗伝子を、 宿主に安定に保持させるこ とが可能である。
さらに該形質転換体に上記のシステムプラスミ ド p F S P 6を導入した形質転 換体 Escherichia coli HB 101 Zp F S P 6 Zp AC E 601 - N s p III を作製することができる。 該形質転換体を培養し、 その適当な時期に I PTGを 添加して発現誘導を行うことにより培養物中で Ns p 7524 ΙΠ制限酵素が産生可 能なことを確認できる。
本発明の第 3の発明は、 上記プラスミ ドベクターを用いることを特徵とする、 目的遺伝子の単離方法を提供する。 本発明の単糠方法は、 宿主に対しその発現産 物が致死的あるいは有害な遺伝子、 特に制限酵素遺伝子の単雜に好適に適用され る。 本法を用いることにより、 これまで単離されていない制限酵素遗伝子、 例え ば A c c III制限酵素遗伝子を、 A c c IU修飾酵素遺伝子非存在下でも単離す ることが可能となる。 制限酵素遼伝子の単離は、 目的酵素産生菌のゲノム DN A を適当な制限酵素により断片化し、 得られた DN A断片を直接、 例えばブラスミ ド PACE 61 1に組み込むことによる、 いわゆる Γショットガン方法」 のほか 、 例えば制限酵素産生菌のゲノム DNAのカセットライブラリーを利用し実施す ることも可能である。 カセットライプラリーを铸型とし、 DNA增轘法により遗 伝子を獲得することにより、 宿主と転写機構の異なる徼生物より瞬製した遺伝子 も発現させることが可能になる。 以下、 カセットライブラリーを利用した制限酵 素遺伝子の単離方法について、 Ac c III制限酵素を例として述べる。
Ac cIII制限酵素産生菌、 すなわち Ac c菌のゲノム DNAのカセッ トライ ブラリーは、 以下のように作製できる。 即ち、 Ac c菌を培養して得られる菌体 よりゲノム DNAを抽出し、 適当な制 K酵素で消化した後、 得られた DNA断片 と相補的な突出末蜴を持つカセットとを連結する。 ゲノム DN A切断に使用する 制限酵素の種類を変え、 同様のカセットライブラリーを数種作製する。 これらを 合わせて Ac cゲノムカセットライブラリーと呼ぶことにした。 次に、 Ac c菌 より目的制限酵素タンパク質を精製し、 そのアミノ酸配列の一部を決定し、 この 配列に基づいてプライマーを合成する。 このプライマーとカセットプライマーを 用いて、 個々のカセットライブラリーを練型とした PCRによる DNA增幅反応 を行い、 Ac cll〖 制 IS酵素遺伝子断片を含む DNA断片を取得する。 得られた DNA断片の塩基配列を、 例えば、 PCR産物のダイレクトシークェンシングに より決定し、 Ac c III制限酵素遺伝子全長の塩基配列を決定する。
該塩基配列より Ac cIII制限酵素遺伝子全長を増幅することが可能なプライ マーを設計し、 Ac c菌のゲノム DNAを铸型とし、 PCRによる DNA増幅反 応により Ac cIU制限酵素遗伝子全長を含む DNA断片を取得する。 得られた DNA断片をプラスミ ド PACE 61 1の S P 6プロモーターの下流にコドンフ レームが合うように挿入して得られる組み換えブラスミ ドを、 例えば、 大腸菌 J Ml 09に導入することにより形質転換体を得る。 Ac c III制限酵素遺伝子が 発現可能な状 1»にある形質転換体の選択は、 各形質転換体を培養し、 更に本発明 の遺伝子発現方法によって遗伝子発現誘導を行い、 得られた各塔養物中の Ac c III制 酵素活性を確 Sするほか、 例えば、 各形質転換体に保持されているブラ スミ ド DN Aの制限酵素地図を作成することにより実施することができる。
実際、 このようにして得られた Ac clll制 IS酵素遺伝子をプラスミ ド pAC E 61 1に挿入し、 得られたプラスミ ドは pCRA 1 9と命名されている。 第 7 図にその制限酵素地図を示す。 図中、 太い実糠が Ac cIII制限酵素遺伝子を含 む DNA断片である。 プラスミ ド pCRA 19を導入した大腸菌 JM109は E scherichia coli J 109 I pCRA19と命名され、 平成 8年 5月 28日 (原寄託日) により通商産業省工業技術院生命工学工業技術研究所 〔あて名:日本国茨城県つ くば市東〖丁目 1番 3号 (郵便番号 305) 〕 に寄託番号 F E RM BP-57 43として寄託されている。 該形質転換体は Ac cin修飾酵素遺伝子が共存し ないにもかかわらず、 制 IS酵素 ¾伝子を安定に保持している。 上記の方法により
、 制限修飾系遺伝子を構成する修飾酵素遺伝子の共存を必要とせず、 制 β酵素遗 伝子を単離することができる。
本発明の第 4の発明は Ac cIII制限酵素活性を有するボリペプチドを提供す る。
本明細書において、 Ac clII制限酵素活性を有するボリペプチドの総称とし て A c c ΠΙ制限酵素なる用語を使用する場合がある。 本発明の A c cIII制眼 酵素は、 例えば配列表の配列番号: 1に記載のァミノ酸配列からなる。
また、 本発明の Ac clII制限酵素は、 前記のものに展定されることはなく、 配列表の配列番号: 1に記載のァミノ酸配列の全部または一部を含むボリぺブチ ドであって、 Ac clil制 酵素活性を有するボリペプチドをも含むものであり 、 さらに配列表の配列番号: 1に記載のアミノ酸配列、 又はその一部のアミノ酸 配列において、 1もしくは複数のアミノ酸残基が欠失、 付加、 揷入又は S換の少 なくとも一つがされており、 かつ Ac cll〖 制 16酵素活性を有するボリペプチド も本発明の範囲内である。
一般に天然に存在するタンパク質にはそれをコードする遺伝子の多形や変異の 他、 生成後のタンパク質の生体内および精製中の修飾反応などによってそのァミ ノ酸配列中にアミノ酸残基の欠失、 挿入、 付加、 置換等の変異が起こりうるが、 それにもかかわらず変異を有しないタンパク質と実質的に同等の生理学的活性、 生物学的活性を示すものがあることが知られている。 このように構造的に差異が あってもその機能や活性については大きな違いが認められないものは、 本発明の 範囲内である。 人為的にタンパク質のァミノ酸配列に上記のような変異を導入し た場合も同様であり、 この場合にはさらに多種多様の変異体を作製することが可 能である。 たとえば、 大揚菌で発現されたタンパク質の N末蜴に存在するメチォ ニン残基は多くの場合メチォニンァミノべプチダ一ゼの作用により除去されると されているが、 タンパク質の種類によってはその除去が完全には行われず、 メチ ォニン残基を持つもの、 持たないものの両方が生成される。 しかしこのメチォ二 ン残基の有無はタンパク質の活性には影響を与えない場合が多い。 また、 ヒトイ ン夕ーロイキン 2 (I L— 2)のアミノ酸配列中のあるシスティン残基をセリン に置換したボリぺプチドがィンターロイキン 2活性を保持することが知られてい る [サイエンス (Science)、 第 224巻、 1431頁 (1984) ]。
さらに、 遗伝子工学的に夕ンパク質の生産を行う I»には ¾合夕ンパク質として 発現させることがしばしば行われる。 たとえば目的タンパク質の発現量を増加さ せるために N末端に他のタンパク質由来の N末墦ペプチド鎖を付加したり、 目的 タンパク質の N末端、 あるいは C末端に適当なぺプチド鎖を付加して発現させ、 このべプチド鎖に親和性を持つ担体を使用することにより目的タンパク質の精製 を容易にすることなどが行われている。 したがって本発明の Ac c III制限酵素 とは一部異なつたアミノ酸配列を有するボリペプチドであっても、 それが本発明 の Ac cill制限酵素と本質的に同等の活性を示す限りにおいて、 本発明の範囲 内に属するものである。
Ac c【I〖 制限酵素を得る方法としては、 例えば、 Ac cin制眼醉素遗伝子 を含有する、 上記形質転換体 Escherichia coli JM109 I pCRA19を塔養し、 その 適当な時期に誘導剤の I PTGを添加し PCRA19コピー数を堆加させた後、 ファ ージ Ml 3 s p 6を感染させることにより、 その目的を達成することができる。 形質転換体の培養物から Ac cll〖 制限酵素を採取するにあたっては、 たとえ ば培養物より菌体を集菌後、 超音波破砕、 超遠心分離等により酵素を抽出し、 つ いで除核酸法、 塩析法、 ァフィ二テイク口マトグラフィ一法、 ゲル 法、 ィォ ン交換クロマトグラフィー法等を組み合わせて精製すればよい。 この精製におい て出発原料となる上記培養物は、 Ac c I、 Ac c IIといった他の制限酵素を含 まないため、 従来の精製方法と比べて容易に必要な純度の酵素標品を得ることが できる。
本発明の第 5の発明は Ac cIU制 酵素活性を有するボリペプチドをコード する DNAを提供する。
本発明の Ac cll〖 制 IS酵素活性を有するボリペプチドをコードする DNAは 、 配列表の配列番号: 1に記載のアミノ酸配列をコードする DNAからなるもの であり、 例えば配列表の配列番号: 2に記載の塩基配列からなる DNAが挙げら れるが、 これらに限定されるものではない。 即ち、 ①配列表の配列番号: 1に記 載のアミノ酸配列の全部または一部を含むボリペプチドであって、 Ac clII制 限酵素活性を有するボリペプチドをコードする DNA、 ②配列表の配列番号: 2 で示される DNAの全部、 又はその一部を含む DNAであって、 該 DNAの発現 産物が A c c Π〖 制限酵素活性を示す DNA、 ⑨配列表の配列番号: 1に記載の ァミノ酸配列又はその一部のァミノ酸配列において、 1艇もしくは複数のァミノ 酸残基が欠失、 付加、 揷入もしくは置換の少なくとも一つがされており、 かつ A c cIII制限酵素活性を有するボリべプチドをコードする DNA、 ④上記①〜③ に記載の DN Aにハイプリダイズ可能な、 Ac cill制限酵素活性を有するボリ ペプチドをコードする DNA、 等も本発明の範囲内である。
また、 上記で得られた DN Aをプローブとして厳密な条件下でハイプリダイゼ ーシヨンを行えば、 得られた DNA (配列表の配列番号: 2) と配列は少し異な るが、 同様な酵素活性を持つボリペプチドをコードする類似の D N Aを得ること ができ、 これも本発明の範囲内のものである。
かかる厳密な条件下とは、 例えば、 DNAを固定したナイロン膜を、 6XSS C (1 XSSCは塩化ナトリウム 8. 76g、 クェン酸ナトリウム 4. 4 lgを 1リットルの水に溶かしたもの) 、 1 % SDS、 100 g /mlサケ精子 DN A、 0. 1 % ゥシ血清アルブミン、 0. 1% ボリビュルピロリ ドン、 0. 1 % フイコールを含む溶液中で 6 5でにて 2 0時間プローブとともに保温してハ イブリダイゼーシヨンを行うことをいう。
A c c I I I 制限酵素をコードする類似の DNAをハイブリダィゼーシヨンによ り得る方法としては、 たとえば以下の方法があげられる。
まず、 適当な遺伝子源から得た DN Aを常法に従ってプラスミ ドゃファージべ クタ一に接練して DN Aライブラリーを作製する。 このライブラリーを適当な宿 主に導入して得られる形質転換体をブレート上で培養し、 生育したコロニーまた はプラークをニトロセルロースやナイロンの膜に移し取り、 変性処理の後に DN Aを膜に固定する。 この膜をあらかじめ" P等で標織したプローブ (使用するブ ローブとしては、 配列表の配列番号: 1に記載したアミノ酸配列の全部または一 部をコードするボリヌクレオチドであればよく、 たとえば配列表の配列番号: 2 に記載の塩基配列の全部または一部からなるまたはそれを含むポリヌクレオチド を使用することができる) を含む上記の組成の溶液中、 上記の条件で保温し、 ハ イブリダィゼーシヨンを行う。 ハイプリダイゼーシヨン終了後、 非特異的に吸着 したプローブを洗い流し、 オートラジオグラフィ一等によりプ o—ブとハイプリ ッドを形成したクローンを同定する。 この操作をハイプリッド形成クローンを単 離できるまで揉り返す。 こうして得られたクローンの中には、 目的の酵素活性を 有するポリべプチドをコ一ドする DNAが保持されている。
得られた DNAは、 たとえば次のようにしてその塩基配列を決定し、 それが目 的の酵素タンパク質をコードする DNAであるかを確認する。
塩基配列の決定は、 ブラスミ ドベクターを用いて作成された形質転換体の場合 、 宿主がェシヱリヒア ·コリであれば試験管等で培養を行い、 常法に従ってブラ スミ ドを調製する。 得られたプラスミ ドをそのまま錄型とするか、 あるいは揷入 断片を取り出して M l 3ファージベクター等にサブクローニングした後に、 ジデ ォキシ法により埴基配列を決定する。 ファージベクタ一で作成された形質転換体 の場合も基本的に同様な操作により塩基配列を決定することができる。 これら培養から塩基配列決定までの基本的な実験法については、 例えば、 ティ

得られた DNAが目的の A c c II I 制! ¾酵素をコードする類似の D NAである かどうかの確認は、 決定された塩基配列を配列表の配列番号: 2に記載した塩基 配列と比較して、 あるいは決定された塩基配列より推定されるァミノ酸配列を配 列表の配列番号: 1に記載したァミノ酸配列と比較して行うことができる。 得られた D N Aが目的の制 酵素をコードする領域の全てを含まない場合には 、 得られた DN Aの塩基配列をもとにしてプライマ一を合成し、 これを用いた P C Rによって足りない領域を堆幅したり、 あるいは得られた DN Aの断片をプロ —ブとして DN Aライブラリーのスクリーニングを繰り返すことにより、 全コー ド領域を得ることができる。
こうして得られた A c c Π〖 制 IS酵素をコードする類似の DNAを含有する形 質転換体を作成して該 DNAにコードされる酵素タンパク質を発現させ、 さらに 発現された酵素タンパク質を精製することができる。 形質転換体の作成、 酵素夕 ンパク質の発現、 精製はいずれも本発明のブラスミ ドを使用することにより、 実 施することができる。 こうして得られた酵素タンパク質は A c c II I 制限酵素活 性を保持している。
なお、 従来得られていない A c c III 修飾酵素及び該酵素をコードする DNA も、 上記の A c c【n 制限酵素をコードする DNAを利用することにより得るこ とができる。 例えば、 多くの場合に制 K酵素遺伝子と修飾酵素遺伝子とが接近し て存在することを利用し、 制限酵素逮伝子近傍のタンパク質をコードする遺伝子 領域を、 カセットライプラリーを鍔型とした DNA增幅反応により得、 適当な発 現ベクターに組み込み、 遺伝子発現を行い、 A c c II I 修飾醉素活性を適当な方 法で確認することにより実施できる。 A c c l【【 修飾酵素活性は、 例えば該形質 転換体より脚製した DNAが、 Ac cIIl制限酵素の切断活性に対する耐性を示 すことにより、 確認することもできる。 なお、 上記遗伝子領域の塩基配列を决定 し、 既知の制限修飾系の修飾酵素遗伝子簡の保存領域 [ジャーナル ォブ モレ キュラー バイオロジー(J.Mol.BioL)第 206巻 第 305〜321頁 (1 9 89) ] とのホモロジ一が確 Sできれば、 遺伝子発現を行う前に、 ある程度修飾 酵素遺伝子であるとの予測を立てることができる。
このようにして、 Ac c菌のゲノム DNAのカセットライブラリーを利用する ことにより、 Ac c III修飾酵素及び該醉素をコードする DNAを得ることがで きる。 配列表の配列番号: 13にアミノ酸配列を、 配列番号: 14に塩基配列を 示す。
また、 本発明における Ac cIII修飾酵素は前記のものに限定されることはな く、 Ac c III制 K酵素の項で述べたように、 配列表の配列番号: 13に記載の アミノ酸配列の全部または一部を含むボリペプチドであって、 Ac cU〖 修飾酵 素活性を有するボリペプチドをも含むものであり、 さらに配列表の配列番号: 1 3に記載のアミノ酸配列、 又はその一部のアミノ酸配列において 1もしくは複数 のアミノ酸残基が欠失、 付加、 揷入又は置換の少なくとも一つがされており、 か つ Ac c III修飾酵素活性を有するボリペプチドも本発明の範囲内である。
本発明における Ac cin修飾醉素をコードする DNAは、 配列表の配列番号 : 13に記載のアミノ酸配列をコードする DNAからなるものであり、 例えば配 列表の配列番号: 14に記載の塩基配列からなる DNAが挙げられるが、 これら に限定されるものではない。 すなわち、 ①配列表の E列番号: 13に記載のアミ ノ酸配列の全部または一部を含むボリペプチドであって、 Ac cIIl修飾醉素活 性を有するボリペプチドをコードする DNA、 ②配列表の配列番号: 14で示さ れる DNAの全部、 又はその一部を含む DNAであって、 該 DNAの発現産物が Ac c III修飾酵素活性を示す DNA、 ③配列表の配列番号: 13に記載のアミ ノ酸配列又はその一部のアミノ酸配列において、 1もしくは複数のアミノ酸が欠 失、 付加、 挿入もしくは置換の少なくとも一つがされており、 かつ Ac clil修 飾酵素活性を有するボリペプチドをコードする DNA、 ④上記①〜③に記載の D N Aにハイブリダィズ可能な、 Ac clil修飾酵素活性を有するボリペプチドを コードする DNA、 なども本発明の範囲内である。 以下に参考例および実施例をあげて本発明をさらに詳細に説明するが、 本発明 は以下の実施例のみに限定されるものではない。 また、 本明細書に記載の操作の うち、 プラスミ ドの謂製、 制限酵素消化などの基本的な操作については 1989 年、 コールド スプリング ハーバー ラボラトリー発行、 T. マニアテイス ( T. Maniatis ) ら纏集、 モレキュラー クローニング:ァ ラボラトリー マ二 ユアル第 2版 (Molecular Cloning : A Laboratory Manual 2nd ed. ) に記載の 方法によった。 参考例
( 1 ) 培地及び ¾養条件
大腸菌は LB培地 (トリプトン 1 %、 酵母エキス 0. 5%、 NaC l O. 5% 、 PH7. 0) を用いて 37 'Cで好気的に培養した。 抗生物質は大腸菌が保持し ているブラスミ ドに合わせて以下の S度で培地に添加した。 pFSP6 : 25 g mlのカナマイシン、 p XX 325 : 20 g Zmlのアンピシリン、 アンピシ リン耐性 C o 1 E 1タイプブラスミ ド: 50〃g Zmlのアンピシリン、 アンピシ リン耐性 pUC夕イブプラスミ ド: 100 tig Zffllのアンピシリン。 発現誘導実 験では、 定常期まで培養した培養液を新觯な培地に 1 %植菌した後、 37でで好 気的に培養し、 OD,0。 =0. 6 (6 108 cell/ml) に達した時点でイソプ 口ビル- ー D—チォガラクトシド (I PTG)を最終濃度が 0, 2mMになるよ うに添加し、 さらに培養を統けた。 (2) システムプラスミ ド pFSP 6の構築
システムプラスミ ド pFSP 6は大腸菌 JM109を宿主とし、 第 8図に示し た手順で構築した。 プラスミ ド pSP 6— 2 [ヌクレイック 了シッズ リサ一 チ ( Nucl. Acids Res. )第 巻、 第 2653〜2664頁 (1987) ] を H i n dill ( 宝酒造社製) で消化し、 末蜴を平滑化した後、 さらに BamHI (宝酒造社製) 消化して得られる SP 6 RN Aボリメラーゼ遗伝子を含む約 2. 81(1)の0 断 片を BamHIおよび Hi ncll (宝酒造社製) で消化したブラスミ ドベクター PUC 18 (宝酒造社製) と混合してライゲーシヨンを行い、 プラスミ ド pUC SPを構築した。 次にブラスミ ド pMJR 1560 [ジーン (Gene)第 51巻、 第 225〜267頁 (1987) ] を Kpn Iで消化し、 末 «を平滑化した後、 更に P s t
I (宝酒造社製) 消化して得られる 1 a c I q遺伝子を含む約 1. 3kbの DNA 断片を単離した。 上記のプラスミ ド pUCSPを H i n dill で消化し、 末端を 平滑化した後、 さらに Ps t lで消化し、 これと上記の約 1. 3kbの DNA断 片とを混合してライゲーシヨンを行い、 プラスミ ド pUCSP 1 a cを構築した ο
さらに pUCKm [ジャーナル ォブ モレキュラー バイオロジー ( J. o 1. Biol.)、 第 147巻、 第 217〜226頁 (1981) ] を P s t I消化して得られる 、 カナマイシン耐性遺伝子を含む約 1. 5kbの DNA断片を上記のプラスミ ド p UCSP l acの Ps t lサイトに導入してブラスミ ド pUCSP 1 a c Kmを 構築した。 該ブラスミ ドを A a t II (東洋钫社製) 消化した後に Ns p I (宝酒 造社製) で部分消化を行って約 6. 5kbの DNA断片を単離し、 さらにその両末 端を平滑化した。
一方、 プラスミ ド PXX325 [プロシーディングス ォブ ザ ナショナル アカデミー ォブ サイエンス ォブ ザ USA (Proc. Natl. Acad. Sci. USA)、 第 80巻、 第 4784〜4788頁 (1983) ] を H i n dill で消化し、 末端を 平滑化した後、 さらに S a 1 I消化して mini Fプラスミ ドの複製起点を含む約 6 . 8kbの DNA断片を単離し、 これを上記の約 6. 5kbの DNA断片と混合して ライゲ一シヨンを行い、 プラスミ ド pFSP 6を構築した。 また、 該ブラスミ ド で大腸菌 HB 101を形質転換し、 Escherichia coli HB 101/pFSP 6 を作製した。
(3)マルチコピーのモデル発現プラスミ ドの構築
S P 6プロモーターの最小領域を含む 2本鎖ォリゴヌクレオチドである PSPe リンカ一を配列表の配列番号: 15および配列番号: 16に塩基配列を示した 2 つの DNA鎖から作製し、 これと Xho l (宝酒造社製) および H i n dlU で 消化したプラスミ ド pMS 434 [ジーン (Gene)第 57巻、 第 89〜99頁 (1987) ] を混合してライゲーシヨンを行い、 該ブラスミ ド上の /8—ガラクトシダーゼ遗 伝子上流に上記のプロモーター配列が挿入されたモデル発現プラスミ ド pMSP 6 Lを構築した (第 1図) 。
一方、 PSP64 [ヌクレイック ァシッズ リサーチ ( Nucl. Acids Res. ) 第 12巻、 第 7035〜7056頁 (1984) ] を A c ellおよび Hi n dill 消化して得 られるネイティブな S P 6プロモーター配列を含む DNA断片を上記のプラスミ ド PMS 434の Xho I- Hi ndlllサイト閟に挿入することによりモデル 発現ブラスミ ド p MS P 6 Fを構築した (第 1図) 。 さらに SP 6プロモーター の *小領域と 1 a cオペレーター領域とを含む ΡβΡβ -0,.« リンカ一を配列表 の配列番号: 17及び配列番号: 18に塩基配列を示した 2つの DNA鎖から作 製し、 これを上記のプラスミ ド pMS 434の Xho I—Hi n dill サイト面 に挿入することによりモデル発現プラスミ ド pMSP60を構築した (第 2図)
(4) SP 6プロモーターの大腸菌の RN Aボリメラーゼに対するプロモータ 参考例 (3) で構築されたプラスミ ド pMSP 6 L、 pMSP 6 F、 および対 照として SP 6プロモーター配列を含まないブラスミ ド pMS 434を表 1に示 す大腸菌に導入し、 得られた形質転換体を参考例 (1) に示した方法で培養後、 それぞれの菌体の培養液を対数增¾期に採取し、 採取した培養液の一部で OD«e 0 を測定し、 残りは予め冷やしておいた試験管に移して最終濃度が 1 0 OmgZml になるようにクロラムフエ二コールを添加した。 この培養液を試料として 1 97 2年、 コールド スプリング ハーバー ラボラトリー発行、 J. H. ミラー ( J.H.Miller) 著、 ェクスペリメンッ イン モレキュラー ジエネテイクス (Ex periments in Molecular Genetics ) に記載の方法でS—ガラクトシダーゼ活性 の測定を行 o
表 2に示されるように、 SP 6プロモーターを含む配列が挿入されたモデル発 現ブラスミ ド pMSP6L、 あるいは PMSP6Fを導入された大腸菌 MC4 1 00は、 揷入されたプロモーター配列に M係なく、 同レベルの低い i8—ガラクト シダーゼ活性を示した。 しかし、 SP 6プロモーターを含まないプラスミ ド pM S 434を導入されたものではこれよりも低い酵素活性しか認められなかった。 さらに大腸菌 MR i 7、 MR i 80を宿主として検射した結果、 プラスミ ドのコ ピー数が滅少する MR i 80では^一ガラクトシダーゼ活性も低下した。
(5) システムブラスミ ド pFSP 6の評価
参考例 (4) で作製した各形質転換体にシステムプラスミ ド pFSP6を導入 したうえ参考例 (1) に示した方法で培養し、 I PTG添加前後の非誘導状 »と 誘導状想の ^一ガラクトシダーゼ活性を測定してその結果を表 3に示した。 各形 質転換体とも非誘導状想における 8—ガラクトシダーゼ活性は表 2に示されたブ ラスミ ド pFSP 6を導入されていない場合と同程度の值であった。
一方、 I PTG添加による誘導を行うと、 プラスミ ド pMSP6L、 および p MSP 6 Fを導入されたものでは非誘導状懇と比較して 1 8〜32倍の^ーガラ クトシダーゼ活性が認められたのに対し、 プラスミ ド pMS 4 34を導入された ものでは活性の上昇は見られなかった。
(6) l a cオペレー夕一配列の発現制御への効果
ブラスミ ド pFSP 6を導入された大腸菌 MC 4 1 00にプラスミ ド pMSP 6 L、 pMSP 6 F、 pMSP 60、 および pMS 434をそれぞれ導入して得 られる形質転換体を参考例 ( 1) に示した方法で培養し、 非誘導状態における; S 一ガラクトシダーゼ活性を測定した。 その結果を表 4に示す。 1 a cオペレータ —配列を含むブラスミ ド pMSP 60を導入されたものではブラスミ ド pMSP 6 L、 pMSP 6 Fに比べて ;8—ガラクトシダーゼ活性が低くおさえられている が、 なおわずかな活性の発現が見られた。 つまり、 1 a cオペレーター配列の導 入だけでは完全に非誘導状態での発現を止めることはできなかつた。 表 4

実施例 1
( 1 ) 培地及び培養条件
大腸菌は LB培地(トリプトン 1 %、 酵母エキス 0. 5%、 NaC 1 0. 、 pH7. 0) を用いて 37 'Cで好気的に培養した。 抗生物質は大腸菌が保持し ているブラスミ ドに合わせて以下の濃度で培地に添加した。 pFSP 6 : 25 g Zmlのカナマイシン、 アンピシリン耐性 Co 1 E 1タイプブラスミ ド : 50 g Zmlのアンビシリ ン、 アンピシリ ン耐性 pUCタイププラスミ ド: 1 0 0 zg Zmlのアンビシリン、 クロラムフエ二コール耐性ランナウエイプラスミ ド: 30 fig Zmlのクロラムフエ二コール、 アンピシリン耐性ランナウエイプラスミ ド: 30 figノ mlのアンビシリン。 発現誘導実験では、 定常期まで埤養した培養液を 新鲜な培地に 1 %楂菌した後 37でで好気的に培養し、 OD,。0=0. 6 (6 X 1 0, cell/mi) に達した時点でイソプロピル一 9一 D—チォガラクトシド ( I P TG)を最終濃度が 0. 2 π*Ιになるように添加し、 さらに培養を続けた。
(2) ランナウエイブラスミ ド pHS 2870の構築
発現プラスミ ド構築の基になったランナウエイプラスミ ド pHS 2870の構 築を第 3図に示す操作で行った。 配列表の配列番号: 3および配列番号: 4に塩 基配列を示した 2つの DNA鎖から作製された 106Nde IDNA断片を Nd e I消化したプラスミ ドベクター pUC 19と混合してライゲージヨンを行った 。 こうして得られたプラスミ ド pUC 106 Aを P vuH消化し、 セルフライゲ ーシヨンを行ってプラスミ ド pUC 106 AdPOを得た。 次いで、 このプラス ミ ド pUC 106 AdPOを練型とし、 RNAIIAブライマー (配列表の配列番 号: 5に RNAIIAブライマーの塩基配列を示す) と 1870プライマー (配列表の 配列番号: 6に 1870ブライマーの塩基配列を示す) を用いた PCRによって得ら れる增幅 DNA断片を Xb a I消化した後、 Xba I消化したプラスミ ド pST V28と混合してライゲーシヨンを行なってプラスミ ド pHS 2870を得た。 第 4図にプラスミ ド pHS 2870の構成を示す。
(3) ランナウエイブラスミ ド pCRS 04の構築
上記のブラスミ ド pHS 2870を Ac c I (宝酒造社製) および Ns p Iで 消化した後、 末缁を平滑化してセルフライゲーシヨンして P 15A複製起点を除 いたブラスミ ド pCRS 01を得た。 次に該プラスミ ドを E c oR I (宝酒造社 製) および Xba Iで消化した後、 上記同様に末端を平滑化してセルフライゲー シヨンを行いブラスミ ド pCRS 02を構築した。 さらにブラスミ ド pMJR 1 560を1< 11 1 (宝酒造社製) および Ps t Iで消化して得られる約 2kb の DNA断片の末端を平滑化した後、 上記のブラスミ ド PCRS02を NspV および Vsp I (ともに宝酒造社製) で消化した後に末端を平滑化したものと混 合してライゲーシヨンを行ない、 ブラスミ ド pCRS 04を得た。 ブラスミ ド p CRS 04構築の ¾程を第 5図に、 またブラスミ ド pCRS 04の構成を第 6図 に示す。
(4) ランナウエイ発現ベクター pACE 601の構築
Nhe I (宝酒造社製) 消化した後に末端を平滑化した上記のプラスミ ド pC RS 04と、 配列表の配列番号: 7および配列番号: 8に塩基配列を示した 2つ の DNA鎖から作製された PSPe -Ο,.ο EXリンカ一とを混合してライゲ一シ ョンを行ない、 プラスミ ド pACE 601を構築した。 第 9図にプラスミ ド pA CE601の構築の過程を示す。 なお、 これらのブラスミ ドの構築は大腸菌 JM 1 09を宿主として行った。
(5) ランナウエイ発現ベクター pACE 61 1の構築
ランナウエイ発現ベクター pACE 601を X ho i— Hi nd【IIで消化し 、 そのサイトに配列番号: 15、 配列番号: 16に記載の合成オリゴ DN Aから なる リンカ一を挿入して、 ランナウエイ発現ベクター pACE 61 1の構 築を行った。 第 1 0図に pACE 61 1の構成を示す。 また、 第 1 1図にプラス ミ ド PACE 81 1の構築の過程を示す。
(6) ランナウエイ発現ベクター pACE 701および pACE 702の構築 ブラスミ ドベクター pUC l 18 (宝酒造社製) を BspH I (NEB社製) で消化して得られる約 lkbの DNA断片の末端を平滑化し、 Nhe I消化した後 に末端を平滑化した上記のブラスミ ド pCRS 04と混合してライゲーシヨンを 行ない、 プラスミ ド pCRS 70を構築した。 第 1 2図にプラスミ ド pCRS 7 0の構築の «程を示す。 次に該ブラスミ ドを Nc o I (宝酒造社製) 、 および B s a A I (NEB社製) で消化した後に末端を平滑化したうえ、 上記の PSP, - Ot.c EXリンカ一と混合してライゲーシヨンを行ない、 該リンカ一の挿入方向 の異なる 2種のプラスミ ド PACE70 1、 および pACE 702を構築した。 第 1 3図にブラスミ ド PACE 701、 および pACE 702の構築の過程を示 す。 なお、 これらのプラスミ ドの構築は大腸菌 JM1 09を宿主として行った。
(7) モデルランナウエイ発現プラスミ ドの構築
上記のブラスミ ド p MS 434を铸型とし、 ブライマー t r pA— N— Nc o I、 およびブライマ一 1 a c Z— C一 Nc o Iを用いた PCRを行い、 3—ガラ クトシダーゼ遗伝子を含む DN A断片を得た。 ブライマー t r pA-N-Nc o I、 およびブライマー 1 acZ - C一 Nc o Iの塩基配列をそれぞれ配列表の K 列番号: 9および配列番号: 10に示す。 該断片を Nc 0 Iで消化した後、 上記 のブラスミ ド pACE 60 1、 pACE701、 および PACE702を Nc o Iで消化したものそれぞれと混合してライゲージヨンを行ない、 PSP, -0,.e 配列下流に; S—ガラクトシダーゼ遗伝子が導入されたモデルランナウエイ発現ブ ラスミ ド pACE601 Z、 PACE701 Z、 および PACE702 Zを得た o なお、 これらのブラスミ ドの構築は大陽菌 JM1 09を宿主として行った。
(8) ランナウエイ発現プラスミ ドの非誘導状 «8での発現レベルの評価 上記のシステムブラスミ ド PFSP6を導入された大腸菌 MC41 00 (以下
MC 4 100/pFSP 6と略す) に上記のモデルランナウエイ発現ブラスミ ド PACE 60 1 Z PACE 701 Z. および p ACE 702 Zを導入して得ら れる形質転換体 MC 4 100ZpFSP6ZpACE601 Z、 MC 4100/ pFSP6ZpACE701 Z、 および MC4100 /p F S P 6 /p A C E 7 02 Zを実施例 ( 1 ) に示した条件で培養し、 対数增¾期に採取した培養液につ いて参考例 (4) に示した方法で9一ガラクトシダーゼ活性を測定した。 その結 果、 いずれの形質転換体においても 6—ガラクトシダーゼ活性は検出限界以下で あり、 参考例 (4) に示したマルチコピープラスミ ドを用いた場合に比べて顕著 な発現抑制効果が見られた。
(9) Nsp7524【【I制限酵素遺伝子発現プラスミ ドの構築
Nsp7524UI制限修飾系遗伝子を含有するプラスミ ド pBRN3を Escheri chia coli C 106 l/pBRN3 (FERM BP- 5741 ) より調製し 、 これを铸型としてプライマー L一 ORF、 およびブライマー Ns pR— ORF 3を用いた PCRを行い、 Nsp7524Il〖 制限,素遺伝子のみを含む約 1 kbの D NA断片を得た。 配列表の配列番号: 1 1および配列番号: 12にプライマー L 一 ORF、 およびブライマー Ns pR - ORF 3の塩基配列を示す。 該断片を N c 0 Iで消化した後、 上記のプラスミ ド PACE601を Nco lで消化したも のと混合してライゲーシヨンを行ない、 PSPe -0,.c 配列下流に Ns ρ7524Π I制限酵素 ¾伝子のみを導入されたプラスミ ド PACE 601- Ns p【II を構 築した。 該ブラスミ ドは、 Ns Ρ7524ΠΙ修飾酵素遗伝子を共存させていない大 腸菌 JM109に安定に保持された。
(10) s P7524III制限酵素遺伝子の発現システムの構築
上記のシステムプラスミ ド pFSP 6を導入された大腸菌 HB 101 (以下 H B 101/pFSP 6と略す) に N s ρ7524ΙΠ制限酵素遣伝子を含む上記のプ ラスミ ドpACE601- NspIΠ、 及び対照として用いるブラスミ ド pAC E 601をそれぞれ導入して得られる形質転換体 HB 10 lZpFSP6ZpA CE 601- Ns pill , および HB 101 Zp F S P 6 p A C E 601を L
3 Θ B培地で定常期まで培養し、 この培養液を新鮮な培地 2本に 1 %ずつ植菌して 3 7でで好気的に培養し、 生育が ODe«。 =0. 6に達した時点でその一方に I P TGを最終濃度が 0. 2 πίίになるように添加し、 さらに培養終了後、 集菌した菌 体を破砕用緵街液 A (20 DiM トリスー塩酸、 pH7. 5、 10 mM 2—メルカ ブトエタノール) に懸酒して超音波破砕した後、 遠心分離して粗抽出液を得た。
この粗抽出液 1 1 を 3 Ο zl の反応液 (10 mM トリスー塩酸、 pH7. 5 、 1 OnAI0MgC 12、 lmMの DTT、 5 OmM0NaC 1、 1 λ-DNA) に加え、 37· ( 、 1時間反応させた後、 反応液をァガロースゲル電気泳動に付し て λ— DNAの分解を調べ、 制限酵素活性を確認した。 第 14図に示されるよう に I PTGを添加して発現誘導を行った HB 10 1/pFSP 6/pACE 60 1 -Ns pill のみに制限醉素活性が認められ、 その λ- DNA切断のパターン は Ns p 75241 II のアイソシゾマーである A v a Iのものと一致した。
また、 上記の λ— DN A消化反応の反応時面を 2時間、 3時簡まで延長した場 合の結果を第 1 5図に示す。 この場合、 本実験に使用した粗抽出液には宿主大腸 菌由来のヌクレアーゼが含まれているため、 レーン 8〜1 0では Nsp7524【II 活性によって生じた D N A断片が大腸菌のヌクレアーゼによってさらに分解を受 け、 反応時間を延長するにつれてバンドが薄くなり、 レーン 1 0ではバンドが確 認できなくなつている。 レーン 1 1〜1 3では Nsp7524UI が誘等されないた め、 反応時簡を延長しても Ns P7524III 分解断片が生成せず、 λ— DNAが直 接大腸菌のヌクレアーゼの作用を受けて低分子化されていることが分かる。 なお 、 HB 1 01/pFSP 6/pACE 601の粗抽出液では制限酵素活性は検出 されなかった。 実施例 2
A c c Π〖 制限酵素遗伝子の単離及び発現
(1) Ac cIII 制限酵素タンパク質の N末 4Sアミノ酸配列の決定及び賅ァミノ 酸配列対応ブライマ一 DNAの合成
市販の Ac cll〖 制限酵素 (宝酒造社製) 約 2 kuを Sephacryl S300 (フアル マシア社製) カラムを用いたゲルろ過に供し、 Ac clil 制限酵素の分子量を測 定した。 活性の溶出位置より、 Ac cin制限酵素の分子量は約 7万であること を確認した。 制限酵素タンパク質はダイマーであることが多く、 従って SDSボ リアクリルアミ ドゲル髦気泳動で Ac c III 制 酵素タンパク質は分子量約 3万 5千の位 Sに泳動されることが推定された。
次に、 Ac cIII制限酵素タンパク質の N末端アミノ酸配列解析用のサンプル を得るため、 市販の Ac cIII制限酵素約 2 k uをタンパク質分子量マーカーと ともに SDSボリアクリルアミ ドゲル «気泳動に供した後、 ゲルから PVDF膜 にタンパク質を移し、 タンパク質位置の確認のために、 ブロモフエノールブルー によるタンパク質染色をおこなった。 10%酔酸一 50%メタノールで脱色後、 分子量約 3万 5千のタンパク質がブロッテイングされた P V D F膜の部分を切り 取り、 プロテイン シークェンサ一 G 1000 A (ヒューレッ ト ゾ、'ッカード 社製) を用いた自動エドマン分解により、 配列表の配列番号: 1 9で示す N末端 アミノ酸配列を決定した。 次にこの配列に基づき、 カセットプライマーの対とし て用いる、 配列表の配列番号: 20で表される Ac c il〖 ブライマー 1と配列番 号: 21で表される Ac cIII プライマー 2を合成した。
(2) Ac c菌ゲノム DNAの謂製
ヌクレイック ァシッズ リサーチ (Nucleic Acids Research)第 13巻、 第
8685〜 8694頁 (1985) に記載されている寥多らの方法に従い A c c 菌を培養し、 湿菌体を得た。 得られた湿菌体 2 gを 1 Omlの緩衝液 B [25m M トリスー HC 1 (pH8. 0) 、 5 OmM グルコース、 1 OmM EDT A] に懸濁し、 緩衝液 Bで 2mgZm 1に謂製した l m 1のリゾチーム溶液を加 えて撹拌し、 37でで 20分静置した。 つぎに、 当該溶液に 28mlの緩衝液 C [ 10 OmM Na C 10 OmM トリスー HC 1 (pH8. 0) ] を加え 、 «拌し、 さらに TE [1 OmM トリスー HC 1 1 mM EDTA (pH8 . 0) ] で 2 OmgZmlになるように謂製した lmlのプロティナーゼ K溶液 と 4mlの 1096SDS溶液を加え、 攬拌し、 37でで 1時間静置した。
当該溶液に 6mlの 5M NaC 1水溶液と 6m 1の緩衢液 D [ 10% CT AB (セチル トリメチル アンモニゥム ブロマイド) 、 0. 7M NaC l ] を加えて搜拌し、 60でで 20分静置した。 当該溶液をフ ノール処理、 被い てクロ口ホルム処理を行い水雇を分取した。 これに、 TEで 1 OmgZmlにな るように調製した RNa s e A (シグマ社製) 溶液 50 1を加えて擾拌し、 3 7'Cで 40分静 Sした。 当該溶液を静置後、 フヱノール処理、 クロ口ホルム処理 を行い水層を分取後、 これに等量の冷エタノールを加え、 析出した DN Aをガラ スキヤビラリーで巻き取って回収した。 DNAを 70%エタノールで洗浄し 3m
1の TEに溶解させ、 約 200 gのゲノム DNAを得た。
(3) Ac cゲノムカセットライブラリーの作製
以後の操作は、 基本的に、 宝酒造社の遺伝子工学ガイド ( 1995〜 1996 年版) F 16〜F 17頁記載の方法に従い、 実施した。
(2)で得られたゲノム DNAを、 EcoR I (宝酒造社製) で完全消化し、 得られた DN A断片と相補的な突出末端を有する E c oR Iカセット (宝酒造社 製) を連結し EcoR Iカセットライブラリーを作製した。 同様にゲノム DNA を制限酵素 Bg I II、 EcoT14 I、 Ec oT22 Κ Hi nd III、 Ps t I、 Sa l I、 Xba I (以上宝酒造社製) で個別に完全消化し得られたゲノム DNA断片と、 相補的な突出末端を有する数種のカセット (宝酒造社製) とを個 別に結合することにより Bg III、 EcoTl 4 Ec oT22 I、 Hi nd III、 Ps t I、 Sa l I、 Xba I各カセットライブラリーを作製した。 各力 セッ トライブラリーを総称し Ac cゲノムカセットライブラリーとした。 (4) A c c I Π制限酵素遺伝子の解析
(3)で得られた Ec oR Iカセットライブラリーを ϋ型とし、 Ac cUI ブ ライマー 1とカセットプライマー C 1 (宝酒造社製) をブライマー対として、 P C Rによる D N A増幅反応を行つた。 目的領域を効率よく特異的に増幅させるた めに、 該反応液の一部を用い、 Ac cin ブライマー 2とカセットブライマー C 2 (宝酒造社製) をブライマー対として、 2度目の PCRによる DNA堆幅反応 を行い、 堆轜 DN A断片を得た。 同様に、 Bgin、 EcoTl 4 K EcoT 22 I、 Hi ndIU、 Ps t I、 Sa l I、 Xba I各カセットライブラリー を铸型として、 個別に上記 2段階 PC Rによる DN A増幅反応を行い、 増幅 DN A断片を得た。 各增幅 DNAをァガロースゲル ¾気泳動により解析したところ、 Xba Iカセットライブラリー由来の增幅 DNA断片が、 特に高い増幅効率を示 し 7こ o
そこで、 Xba Iカセットライブラリーを鎵型とし、 Ac cin プライマー 1 とカセッ トブライマー C 1をプライマー対として、 再度、 PCRによる DNA增 幅反応を行った。 この反応液をァガロースゲル S気泳動に供し、 約 0. 5kbの 增輻 DNA断片をゲルから回収した。 得られた DNA断片を铸型とし、 Ac cU I ブライマー 2とカセットブライマー C 2をプライマー対として、 2度目の PC Rによる DNA增輻反応を行った。 得られた約 0. 5 kbの増幅 DNA断片の塩 基配列を決定したところ、 (1)で N末端のアミノ酸配列を決定したタンパク質 の一部をコードしていることが明らかとなった。 一方、 Ac cIII制限酵素タン パク質の推定分子量は約 3万 5千であり、 該タンパク質をコードしている遺伝子 は、 約 1 kbの長さと推定される。 従って、 今回埴基配列を一部决定した遺伝子 領域において、 上記約 0. 5 kbの塩基配列情報に加え、 Ac cIII ブライマー 1及び Ac cIII プライマー 2がアニーリングした 5' 端領域の正確な塩基配列 情報及び 3' 側領域の残り約 0. 5 kbの塩基配列情報が得られれば、 Ac cll 【 制限酵素遺伝子の塩基配列を決定できると考えられた。
そこで、 Ac c III制限酵素遺伝子領域の正確な 5 ' 端領域の塩基配列を確認 するために、 配列表の配列番号: 22で表される Ac c III プライマー 3、 配列 番号: 23で表される Ac cll〖 ブライマー 4を合成した。 次に、 (3)で得ら れた E c oR Iカセットライブラリーを铸型とし、 Ac cIII プライマー 4と力 セヅトブライマー C 1をブライマー対として、 PCRによる DNA堆幅反応を行 つた。 この反応液の一部を用い、 Ac cin プライマー 3とカセットプライマー C 2をブライマ一対として、 PCRによる DNA增幅反応を行った。 同様に、 B g l ll、 E c oT 14 K E c οΤ22 Hi nd III、 Ρ s t Κ Sa l I 、 Xb a I各カセットライブラリーを铸型として、 個別に上記 2段階 PCRによ る DNA增幅反応を行った。 增幅した DNA断片のうち最短の、 Ec oTl 4 I カセットライブラリーを練型として得られた、 約 1 kbの DNA断片の塩基配列 を決定した。
さらに Ac cIU制 IS酵素タンパク質をコードすると推定される遗伝子領域の 3' 側、 約 0. 5 kbを決定するために、 配列番号: 24で表される Ac c III ブライマー 5と、 配列番号: 25で表される Ac cIII プライマー 6を合成した 。 (3〉 で得られた E c oR Iカセットライブラリーを铸型とし、 Ac c III プ ライマー 5とカセットブライマー C 1をブライマー対として、 PCRによる DN A增幅反応を行った。 この反応液の一部を用い、 Ac clII プライマー 6とカセ ットプライマー C 2をプライマー対として、 PCRによる DNA増幅反応を行つ た。 同様に、 Bg l II、 EcoT14 I、 Ec oT22 Hi nd III、 P s t I、 Sa l I、 Xb a I各カセットライブラリーを錄型として、 捆別に上記 2 段喈 P C Rによる D N A増幅反応を行つた。
增輻した DN A断片のうち、 EcoT221、 Hi ndIU各カセットライブ ラリーを铸型として得られた、 約 0. 5kb、 約 0. 8kbの DNA断片の塩基 配列を決定した。 得られた 3つの塩基配列の結果を総合し、 約 1, 6kbの DN A断片の塩基配列情報を得た。 その塩基 S3列を配列表の配列番号: 26に示す。 さらに、 タンパク質をコードしうるオーブンリーディングフレーム (ORF) を 検索した結果、 塩基番号 558〜1442に ORF 1が、 塩基番号 1588〜1 434に ORF 2と考えられる一部、 及び塩基番号 1〜535に ORF3と考え られる一部が存在した。 0RF 1は、 (1)で N末端アミノ酸配列を決めたタン パク質をコードしており、 ORF 1が Ac cin制限酵素遺伝子であると推定さ れた。 Ac cUI 制限酵素 ¾伝子の塩基 列を配列表の配列番号: 2に示す。 但 し、 この場合 5' 端より 4番目の塩基は Cである。 また、 該塩基配列より推定さ れるアミノ酸配列を、 配列表の配列番号: 1に示す。 但し、 この場合 Ν末端より 2番目のアミノ酸は Leuである。 なお、 ORF2の翻訳方向は、 ORF 1及び ORF 3とは逆方向である。
(5) プラスミ ド PCRA19の構築
次に、 Ac clll 制限酵素遺伝子を発現させるためのブラスミ ドの構築を行つ た。 まず、 該遺伝子を得るため、 配列表の配列番号: 27で表される Ac c— R Lブライマーと配列番号: 28で表される Ac c— RRをブライマーを合成した 0 なお、 両プライマーに制限酵素 Nc 0 I認識配列部位を導入した。 上記ブライ マー対を用いると、 Ac c菌のゲノム DNAを铸型とした PCRにより得られる DNA增幅断片より、 Ac c【I〖 制限酵素遺伝子を制 I®酵素 N c o Iで切り出す ことが可能となり、 また PACE 61 1ベクターの Nc 0 I部位に揷入した K、 SP 6プロモーター支配下、 翻訳時のコドンフレームが合うようになる。 一方、 このブライマー対の使用により、 A c c Π I制 Κ酵素遺伝子の 5, 末端より 4番 目の塩基が Cから Gに、 該遺伝子にコードされるタンパク質の Ν末 ¾より 2番目 のアミノ酸が、 Leuから Va 1に置換される。 上記プライマー対を用い、 (2 ) で得られた Ac c菌のゲノム DNAを接型として、 PCRによる DNA增輟反 応を行った。 この DNA断片を制限酵素 Nc 0 I (宝酒造社製) で完全消化した 後、 ァガロースゲル «気泳動し、 約 900 bpの大きさの DNA断片を回収した 。 次に、 この DNA断片が SP 6プロモーターの下流に配置されるように、 pA CE 61 1ベクターの Nc 0 I部位に揷入した。
この組換え DNAを大腸菌 JM1 09に導入し形質転換体を得た。 適当に 30 個の形質転換体を選択し、 30 zg Zm 1のクロラムフエ二コール含む 5m 1の LB培地に個別に接種し、 37でで培養した。 菌体培養液の ODeo。 が 0. 6に なった時に、 ブラスミ ドコピー数堆加のために、 最終濃度が 2mMになるように I PTGを添加し、 さらに 37でで 2時間培養後、 各培養物より、 個別にブラス ミ ド DNAを謂製した。 得られた各プラスミ ド DNAを制 K醉素 Hi ndlll と Xb a Iで同時に切断し、 ァガロースゲル ¾気泳動により生じた DNA断片長を 確認したところ、 挿入した遺伝子領域が正方向に揷入されたと考えられるプラス ミ ドが、 1価存在した。 本プラスミ ド DNAを pCRAl 9と命名し、 pCRA 1 9を大腸菌 JM1 09に導入し、 得られた形質転換体を Escherichia coli JM1 09 I pCRA19 と命名した。
(6) Ac cIII 制限酵素 ¾伝子の発現システムの構築
まず、 大腸菌 JM1 09に SP 6RNAボリメラーゼ遗伝子を導入し、 さらに 発現させることが可能なファージベクターを構築した。 すなわち、 市販のファー ジベクター Ml 3mp 18 (宝酒造社製) のマルチクローニングサイト内の Ba mH I - H i n dill サイトに、 p S P 6— 2を B amH I— H i n d ΙΠ 消化 することにより得た SP 6RNAボリメラーゼ«伝子断片を挿入することにより 、 SP 6RNAボリメラーゼ発現ファージ Ml 3 s p 6を構築した。
次に、 形質転換体 (Esherichia coli JM109/pCRA19) をクロラムフエ二コール 30 /zg /m 1を含む 5mlの LB塔地に接種し、 37でで培養した。 菌体培養 液の OD,。。 が 0. 6になった時に、 プラスミ ドコピー数堆加のために、 I PT Gを最終濃度が 2 mMになるように添加し、 更に 37でで 2時閟培養した。 その 後、 Ac cIII 制 IS酵素遺伝子にコードされたタンパク質を発現させるため、 フ ァージ Ml 3 sp 6を感染させ、 更に、 37でで1 6時間培養した。 得られた湿 菌体 l lmgを、 180 tx 1の菌体破砕用緵衝液 E [2 OmM トリスー HC 1 (pH7. 5) 1 OmM 2—メルカブトエタノール] に懸獨し、 超音波処理に より菌を破砕し、 遠心分離 (18000g、 10分閎) により固液分離を行った。
上淸の活性を、 宝酒造社製の Ac cIH制 酵素に添付されたデータシートに 記載の活性測定条件で測定したところ、 湿菌体 1 g当り約 8000ュニットの A c cIII制限酵素が産生されていた。 これは Ac c菌の産生量と比較して、 湿菌 体重量あたり約 1 6倍の産生量であった。 また、 得られた上淸には Ac c III以 外の制限酵素活性、 及び Ac cIII修飾酵素活性は認められなかった。 ブラスミ ド PCRA19に挿入された Ac cIII制限酵素遺伝子は、 PCRによる DNA增幅 を行った際、 翻訳開始塩基より 4番目の塩基が Cから Gに置換され、 この結果、 翻訳タンパク質の N末端から 2番目のアミノ酸が、 1^ 611から 31に置換され ているが、 該タンパク質は Ac c【I〖 制限酵素活性を有することが明らかとなつ 以上、 Ac c III修飾酵素非共存下での Ac c III 制限酵素遺伝子の単離及び 大量生産系が完成した。
( 7 ) A c c 111 修飾酵素遺伝子の単離
修飾酵素遺伝子と制 IS酵素遗伝子は近傍に存在することが多い。 そこで (4) で推定された ORF 2領域の決定のために、 (4) において Ac cIII制限酵素 遺伝子 3' 側領域を決定するために ¾製した堆餳 DN A断片のうち、 Ec oR I カセットライブラリーを錄型として得られた、 約 1. 4 kbの DNA断片の塩基 配列を決定した。 さらに、 (4)で推定された ORF 3領域の決定のために、 ( 4) において Ac cIII制限酵素逮伝子 5' 端領域の塩基配列を決定するために 調製した «幅 DNA断片のうち、 Bg 1 IIカセットライブラリーを铸型として得 られた約 2. 2 kbの DNA 片の塩基配列を決定した。 これらと、 (4)で決 定した Ac c III制限酵素遺伝子領域を含む約し 6 kbの DNA断片の塩基配 列を総合して、 配列表の配列番号: 29に示す約 4. 2 kbの塩基配列情報が得 られた。 埴基番号 1913〜2797に Ac cIII制限酵素遺伝子が、 塩基番号 3712〜2789に ORF2が、 塩基番号 691〜1890に ORF3が存在 した。 各 ORFのうち、 既知の制眼修飾系における修飾酵素面の保存領域とホモ ロジ一の高い部分が、 OR F 2に存在することが判明した。
次に、 ORF2領域を得るため、 E c oR Iカセットライブラリーを錄型とし 、 Ac cブライマー 6、 カセットブライマー C 1及び Ac cブライマー 5、 カセ ッ トブライマー C 2を各々対としたニ段瞎 PCRにより得られた堆幅 DNA断片 より、 Ec oT22 I、 Hpa I (宝酒造社製) を用い OR F 2を含む約 1. 1 kbの部分を切り出し、 pUC 1 18ベクター 1 a cプロモーター下流の Smai 部位に揷入した。 本組み換えプラスミ ドが Ac cIII修飾酵素遺伝子を含んでお り、 大腸菌内で Ac cIII修飾酵素が発現可能であれば、 本組み換えプラスミ ド を大腸菌 JM109に導入し、 得られた形質転換体の培養物中の DNAは Ac c III修飾酵素によりメチル化を受け、 Ac cIII制限酵素による切断に耐性を示 すようになる。
しかし本組換えブラスミ ドの構築に用いられた pUC 1 18ベクターには Ac cIII制限酵素認戮配列が存在しないため、 本組換えプラスミ ド自体を Ac ell I 修飾酵素活性の確 Sに使用するのは不適当である。 一方、 大騰菌 JM109内 で本組み换ぇプラスミ ドと共存可能なブラスミ ド pSTV29には Ac cIII制 限醉素認織 K列が存在する。 そこで PSTV29を Ac cil〖 修飾薛素発現の指 標とするため、 先の組み換えプラスミ ドを pSTV29と共に大腸菌 JM109 に導入し形質転換体を得た。 3fflの形質転換体を 100/zg/mlのアンピシリ ン、 30 tfgZm 1のクロラムフエ二コール及び 2mM I PTGを含む 2ml の LB培地で個別に 37'C、 16時間培養し、 各培養物よりブラスミ ド DNAを 調製した。
調製した DNAは pSTV29と上記の組換えプラスミ ドの混合物として得ら れる。 各 DNAサンプルを Ac cin制限酵素による切断反応に供したところ、 全てのサンプル中の PSTV29が Ac cIII制限酵素切断に対する耐性を示し 、 該 DNAサンブルに含まれる組換えプラスミ ドに、 Ac cIII修飾酵素遺伝子 が挿入されていることが確 Sされた。 更に、 該 DNAサンプルを、 制限酵素 Hi ndlll と Xb a Iで同時に切断し、 ァガロースゲル電気泳動にて生じた DNA 断片長を解析したところ、 組換えブラスミ ド中に OR F 2が存在することが確 B された。 従って、 ORF2が Ac cH〖 修飾酵素遗伝子であることが判明した。 得られた Ac cIII修飾酵素遺伝子の塩基配列を配列表の K列番号: 14に、 ま た該配列より推定されるアミノ酸配列を配列番号: 13にそれぞれ示す。
本発明により明らかとなった Ac cIII制限修飾系遺伝子構造を第 16図に示 す。 図中、 Mは修飾酵素遗伝子、 Rは制限酵素遺伝子、 矢印は OR Fの方向を示 す。 産業上の利用可能性
本発明により、 宿主に対し致死的となる、 あるいは有害なタンパク質をコード する外来性の目的 ¾伝子を該宿主に導入しうるプラスミ ドベクター、 および該プ ラスミ ドベクターを用いて効率よく該タンパク質を発現させうる方法が初めて提 供される。 また、 従来技術では困難であった制限修飾系を構成する制 酵素遗伝 子を、 條飾酵素遺伝子の非共存下で可能な単離方法が提供される。 さらに、 本発 明により AccIII制限酵素遺伝子及び Acc〖〖【 修飾酵素遺伝子が単離され、 該遗 伝子を含有するブラスミ ドで形質転換した大腸菌により、 遺伝子工学において有 用な AccIII制限酵素あるいは修飾酵素を、 従来の精製方法と比べ、 容易に必要 な純度の酵素標品を得ることが可能となる。 配列番号: 1

Claims

請 求 の 範 囲
1. 宿主が本来有しない RNAボリメラーゼによって認識される、 目的遗伝子 の発現を制御するプロモータ一配列と、 外来因子による誘導によってコピー数を 增加させる複製起点とを有することを特敏とするプラスミ ドベクター。
2. プロモーター配列がバクテリオファージ由来の RN Aボリメラーゼによつ て認識されるものである請求項 1に記載のブラスミ ドベクター。
3. プロモーター配列が S P 6ファージ由来の RN Aボリメラーゼによって認 識されるものである請求項 2に記載のプラスミ ドベクター。
4. プロモーター配列が配列表の配列番号 30で示される塩基配列を含むもの である請求項 3に記載のプラスミ ドベクター。
5. 複製起点がプロモーターの制御下にある、 請求項 1〜請求項 4のいずれか 1項に記載のブラスミ ドベクター。
6. 複製起点が 1 a cプロモータの制御下にある、 請求項 1〜請求項 5のいず れか 1項に記載のプラスミ ドベクタ一。
7. 還択マーカーとして薬剤耐性遺伝子を有する、 箫求項 1〜睛求項 6のいず れか 1項に記載のプラスミ ドベクター。
8. PACE 60 U ACE 6 1 1、 pACE 70 1、 pACE 702から 選択される請求項 7に記載のブラスミ ドベクター。
9 . 請求項 1〜請求項 8のいずれか 1項に記載のプラスミ ドベクターに発現さ せようとする目的遺伝子を組み込んだプラスミ ドベクター。
1 0 . 請求項 1〜講求項 8のいずれか 1項に記載のブラスミ ドベクターに発現 させようとする目的遺伝子を組み込んだブラスミ ドベクターと、 該ブラスミ ドべ クタ一上のプロモーター配列を する RN Aボリメラーゼ遺伝子とを宿主に導 入し、 外来因子により該ブラスミ ドベクターのコピー数の増加を誘導すると共に 該 R N Αボリメラーゼの発現の誘導を行って目的遺伝子の転写、 翻訳を行うこと を特徴とする、 目的 ¾伝子の発現方法。
1 1 . プラスミ ドベクターのコピー数の増加と RNAボリメラーゼの発現とが 異なる外来因子により誘導されることを特徴とする請求項 1 0に記載の目的遗伝 子の発現方法。
1 2 . プラスミ ドベクターのコピー数の增加と RNAボリメラーゼの発現とが 同一の外来因子により誘導されることを特»とする請求項 1 0に記載の目的遗伝 子の発現方法。
1 3 . プラスミ ドベクターのコピー数の墙加を誘導する外来因子が、 イソプロ ビル一 8— D—チォガラクトシド ( I P T G) の添加、 ラクトースの添加、 ガラ クトースの添加、 ァラビノースの添加、 トリブトファン囊度の低減、 及び形質転 換体の培養温度の調整からなる群より選択されるものの 1種以上である、 請求項 1 0〜請求項 1 2のいずれか 1項に記載の目的遗伝子の発現方法。
1 4 . RN Aボリメラーゼの発現を誘導する外来因子が、 イソプロピルー ー D—チォガラクトシド ( I P T G) の添加、 ラクトースの添加、 ガラク トースの 添加、 ァラビノースの添加、 トリブトファン濃度の低滅、 及び形質転換体の培養 温度の調整からなる群より選択されるものの 1種以上である、 請求項 1 0〜請求 項 1 2のいずれか 1項に記載の目的遺伝子の発現方法。
1 5 . R N Aボリメラーゼ遗伝子が他のプラスミ ドベクター又はファージべク 夕一によって宿主に導入されていることを特敏とする請求項 1 0に記載の目的遗 伝子の発現方法。
1 6 . R N Aボリメラーゼ遗伝子が宿主の染色体上に組み込まれていることを 特徴とする請求項 1 0に記載の目的遺伝子の発現方法。
1 7 . R N Aボリメラーゼ遺伝子が S P 6ファージ由来のものであることを特 徴とする請求項 1 5又は 1 6に記載の目的遺伝子の発現方法。
1 8 . 目的遺伝子が宿主に対し致死的となる、 あるいは有害なタンパク質をコ —ドするものである、 請求項 1 0〜請求項 1 7のいずれか 1項に記載の目的遺伝 子の発現方法。
1 9 . 大腸菌を宿主に用いて行われることを特徴とする請求項 1 0〜請求項 1 8のいずれか 1項に記載の目的遗伝子の発現方法。
2 0 . 目的遺伝子の単離方法において、 講求項 1〜講求項 8のいずれか 1項に 記載のプラスミ ドベクターを用いることを特徴とする目的遺伝子の単雜方法。
2 1 . 目的遺伝子が宿主に対し致死的となる、 あるいは有害なタンパク質をコ 一ドする遗伝子である請求項 2 0に記載の目的遺伝子の単離方法。
22. 宿主に対し致死的となる、 あるいは有害なタンパク質をコードする遗伝 子が制限酵素遺伝子である、 請求項 21に記載の目的遺伝子の単讎方法。
23. 配列表の配列番号: 1に示されるァミノ酸配列の全部、 又はその一部を 含むものであって、 Ac cIII制限酵素活性を有するボリペプチド。
24. 配列表の配列番号: 1に示されるアミノ酸配列又はその一部のアミノ酸 配列において、 1偭若しくは a数のアミノ酸残基が欠失、 付加、 挿入又は置換の 少なくとも 1つがされており、 かつ Ac cIII制限酵素活性を有するボリべプチ
25. 配列表の配列番号: 1に示されるアミノ酸配列の全部、 叉はその一部を 含むボリペプチドであって、 Ac cIII制 IS酵素活性を示すボリペプチドをコー ドする DNA。
26. 配列表の配列番号: 2で示される DNAの全部、 又はその一部を含む D NAであって、 該 DNAの発現産物が Ac c III制限酵素活性を示す DNA。
27. 配列表の配列番号: 1で示されるアミノ酸配列又はその一部のアミノ酸 配列において、 1個若しくは複数のアミノ酸残基が欠失、 付加、 揷入若しくは置 換の少なくとも 1つがされており、 かつ Ac cIII制 酵素活性を示すボリべブ チドをコ一ドする DNA。
28. 講求項 25〜講求項 27のいずれか 1項に記載の DNAにハイブリダィ ズ可能な、 Ac cni制限酵素活性を有するボリペプチドをコードする DNA。
