遊離型ィンスリン様成長因子の濃度上昇方法 技術分野
本発明は、 生体内に大量に存在している内在性のインスリン様成長因子を有効 に利用する方法に関する。 背景技術
インスリン様成長因子 (以下、 IGFと略する。) には、 分子種が 2種類存在し、 それぞれ IGF - 1と IGF - IIと呼ばれている。 ヒ トの IGF-Iと IGF - IIはそれぞれァミノ 酸 70個と 67個からなり、 このうち IGF- 1は、 インスリンの部分構造である Cぺプ チドに相当する部分のァミノ酸が IGF- IIに比べると 3個多い。 IGF - 1と IGF - II間 のァミノ酸配列の相同性は約 60%であり、 IGF-Iとインスリンとの間では約 40%の 相同性がある。 生体内における IGF-Iの産生場所は、 肝臓 ·腎臓が主と考えられ ていたが、 その後 m R N Aを用いたノーザンプロッティング法による解析の結果 から、 IGF-Iは体組織のほとんどの部位で作られていることが明らかになった (D ' Ercole, A. J., et al. , Proc. Natl. Acad. Sci . USA. , 81, 935 (1984); Hum bel, R. E. , et al. , Eur. J. Biochem. , 190, 445 (1990) )。 すなわち、 IGF - 1 はェンドクリンとして作用するばかりでなく、 パラクリンゃォ一トクリンとして も作用している。
IGF- 1と IGF-IIには、 これらにそれぞれ特異的なレセプター、 IGF-Iレセプタ一 および IGF- 11/カチオン非依存性マンノース 6 リン酸レセプターが存在する。 但 し IGF-I Iは IGF-Iレセプターとも結合し、 IGF- IIの種々の生物活性は主に細胞表 面上の IGF-Iレセプターを介した現象であると考えられている (Casella, S. J. , et al. , J. Biol. Chem. , 261, 9268 (1986); Sakano, K. , et al., J. Biol. Chem. , 266, 20626 (1991) )。
IGF- 1レセプターはァミノ酸配列から見ると、 インスリンレセプターと相同性 が高く、 その細胞内情報伝達機構も類似している (Shemer, J., et al. , J. Bio
1. Chem. , 262, 15476 (1987); Myers, M. G. Jr. , et al. , Endocrinology, 13 2, 1421 ( 1993) )。 動物実験の結果から、 IGFの糖代謝活性はインスリンと異な り、 末梢組織で活発であるので、 レセプターの発現部位の違いによってインスリ ンと IGFの作用部位が異なるものと考えられている (Laager, R. , et al. , J. CI in. Invest. , 92, 1903 (1993) )。
ヒ トの成人の血中には、 IGFは平均約 100nM、 インスリンは約 ΙΟΟρΜ存在してい ると われてレヽる (Baxter, R. C. , in Modern Concepts of Insul in-Like Grow th Factors (Spencer, E. M. , ed) pp. 371, Elsevier Science Publishing Co. ,
New York-Amsterdam (1991) )。 生体内に存在する IGFのほとんどはその特異的 結合タンパク質である IGF結合タンパク質 (以下、 IGFBPと略する。) と複合体を 形成している。 IGFBPと結合していない遊離型 IGFの血糖降下作用はィンスリンの 約 5〜10¾>であるので (Guler, H. P. , et al. , New Engl. J. Med. , 317, 137 (1987) )、単純に血糖降下作用だけをみると、 インスリンの約 50〜100倍のィンス リン様物質が内在していることになる (Baxter, R. C. , in Modern Concepts of
Insulin-Like Growth Factors (Spencer, E. M. , ed) pp. 371, Elsevier Science
Publ ishing Co. , New York-Amsterdam (1991) )。
糖尿病は、 現在その臨床像から WH Oの分類に従って、
(1)ィンスリン依存性糖尿病 (insul in-dependent diabetes mellitus;以下、 IDDMと略する。)、
(2)インスリン非依存性糖尿病 (non insul in dependent diabetes mell itus ; 以下、 NIDDMと略する。)、
(3)その他の糖尿病 (膝疾患に伴うもの、 内分泌疾患に伴うものなど) と 3つに大別されている。
IDDMに関しては、 インスリン療法が主要な治療法であるが、 NIDDMに関しては、 食事療法、 運動療法、 経口血糖降下剤およびインスリン療法等が主に採用されて いる。 近年では、 インスリンを投与しても無効なインスリン抵抗性糖尿病におい て、 IGFによる治療効果が検討されている。 (Kuzuya, H. , et al . , Diabetes, 4
2, 696 ( 1993) )。 また、 NIDDMにおいても IGFによる治療効果が検討されている (Zenobi, P. D., et al. , J. Cl in. Invest. , 90, 2234 (1992); Moses, A. C., et al. , Diabetes, 45, 91 (1996) )。
Gulerらは、 IGF-I 100 μ g/kgを成人に静脈注射したところ、 血糖が経時的に低 下し、 20分後に最低値となる血糖降下作用を認めた (Guler, H. P. , et al. , Ne w Engl. J. Med. , 317, 137 (1987) )。
Takanoらは、 成人に IGF-I 60〜120 g/kgを皮下注射にて投与して血糖降下作 用を認めた。 また、 IGF-I lOO ju g/kgを 6日間連続投与し、 血中尿酸値およびク レアチニン値の低下を認めた (Takano, K., et al. , Endocrinol. Jpn., 37. 30 9 (1990) )。
その他、 IGF- 1の投与による血中遊離脂肪酸の低下 (Turkalj. I., et al. , J. Clin. Endocrinol. Metab. , 75, 1186 (1992) ) およびトリグリセライ ド等の中 性脂肪の低下 (Turkalj, I. , et al. , J. Cl in. Endocrinol. Metab. , 75, 1186 (1992); Zenobi, P. D. , et al. , J. Cl in. Invest. , 90, 2234 (1992) )、 総コ レステロールの低下 (Zenobi, P. D. , et al., Diabetologia, 36, 465 (199 3) ) や、 腎血流量および糸球体ろ過率の増加 (Elahi, D. , et al. , in Modern C oncepts of Insul in-Like Growth Factors (Spencer, E. M. , ed) pp219, Elsevie r Science Publ ishing Co. , New York-Amsterdam (1991) ) につレヽての報告もあ る。
また、 IGF- IIの投与が、 難治性糖尿病に対して有効であったという報告もある (Usara, A. , et al. , Diabetes, 44, Suppl. 1, 33A, (1995) )。 さらに、 出血 性ショック時の糖代謝を含めたストレス状態を緩和、 並びに糖輸液の使用による 副作用の軽減、 およびエネルギー供給を目的とした IGF- 1の投与の有効性がモデ ル動物を用いた実験から示唆されている (特開平 7-242565号公報)。
IGFの作用については、 血糖降下作用だけではなく、 細胞増殖作用、 細胞分化 作用、 タンパク質同化作用が動物およびヒ トに投与して検討されている。 損傷し た末梢神経に IGF-Iを局所投与すると、 関連する非神経細胞の増殖とともに神経 細胞も刺激されることが分かった。 脊髄細胞に IGF-Iレセプターが認められ、 IGF - 1投与は運動ニューロンの細胞死を減少させることが報告されている。 また、 筋 肉の終板を増大させたり、 座骨神経損傷時の機能回復を促進したり、 化学療法時 にみられる末梢運動麻痺を予防することが認められている (Sjoberg, J., et a 1. , Brain Res. , 485, 102 (1989) ) 0
これらの実験結果により、 筋萎縮性側索硬化症や運動ニューロンの変性疾患に IGF- 1の臨床治験が行われている (Lewis, M. E. , et al. , Exp. Neurol. , 124. 73 (1993) )。 同様に、 アルツハイマー病、 卒中、 筋萎縮性側索硬化症およびパー キンソン病等に認められるニューロン細胞の生存を促進するために IGFの作用を 利用することも考えられている (特表平 6-510305号公報)。 さらに筋ジストロフ ィ一についても IGF-Iの有効性が報告されている (Vlachopapadopoulou, E. , et al. , J. Clin. Endocrinol. Metab. , 80, 3715 (1995) )。
糖尿病性神経症に対する IGFの作用についても検討がなされており、 IDDMモデ ノレラッ卜である STZ—rat (streptozotocin— diabetic rat) iこおレヽて、 血糖降下を伴 わない IGF投与量により糖尿病性神経症の改善が見られることが報告されている (Zhuang, H- X. et al. , Exp. Neurol. , 140, ppl98-205 (1996) )。 また NIDDM モデルラットである diabetic obese Zuker (fa/ fa) ratの座骨神経、 脊髄神経及び 脳神経では IGF_IImRNAレベルの低下が見られ、 血糖降下を伴わない量の IGF- Π投 与により糖尿病性神経症の改善が見られることも報告されている (Zhuang, H-X, et al. , J. Pharmacol. Exp. Ther., 283, pp366 - 374 (1997) )。 これらのこと から糖尿病性神経症治療への I GF療法の有効性が示唆されている。
心機能に関しても IGF-I投与が心筋障害を改善するか否か検討されている。 す なわち、 ラッ トにドキソルビシンを投与して心筋障害を作成し、 心拍出量を検討 したところ、 IGF - 1の投与は障害を受けた心筋の機能を改善したと報告されてい る (Ambler, G. R. , et al. , Cardiovasc. Res. , 27. 1368 (1993) )。 同様に、 心筋炎または心筋梗塞を含む心筋障害、 心臓病または急性発作の予防または治療 のため、 並びに心臓拍動量の増加による拍出量の改善を目的として IGF- 1が使用 されている (特表平 6-504286号公報)。
また、 虚血による急性腎不全の実験動物を作成し、 IGF- 1の効果を検討した報 告がある。 虚血 5日目より IGF- 1を皮下注射で 1 日 3回、 3日間投与したところ、 対照に比し腎機能の改善、 尿細管細胞増殖促進、 タンパク質分解抑制およびタン パク質合成促進が認められ、 全体としてカタボリズムを減少させたと報告されて いる (Ding, H. , et al., J. Cl in. Invest. , 91, 2281 (1993) )。
皮膚損傷 (外傷、 火傷等) 時に IGF- 1を局所に投与すると、 治癒に至る期間が 縮小することも報告されている。 火傷を受けたラッ トの実験では、 ラット IGF-I の投与により体重、 腸粘膜重量、 粘膜 D N A、 タンパク質量の増加を認め、 腸内 細菌の腸内リンパ腺への移動も減少し、 従って腸管機能も改善し全体として生命 予後に良好な結果をもたらしたと報告されている (Huang, K. F., et al. , Arch. Surg. , 128, 47 (1993) )。
IGF- 1は血小板由来成長因子 (以下、 PDGFと略する。) と一緒になって間葉培 養細胞の有糸分裂およびタンパク質合成を促進し、 皮膚障害に対する PDGFまたは IGF- 1単独適用では治癒促進しないが、 両因子を一緒に適用すると、'結合組織お よび上皮組織の成長が促進する (Stiles, C. D. , et al. , Proc. Natl. Acad. S c i. USA, 76, 1279 ( 1987) )。 しかし、 他の報告では、 傷への個々の成長因子の 適用によって傷治癒が刺激されたとされている (Tsuboi, R., et al. , J. Exp. Med. , 172, 245 (1990) )。 そこで、 傷の治癒の促進を目的とした IGFの利用が試 みられている (特開昭 63- 233925号公報、 特表平 3-505870号公報、 特表平 6- 50619 1号公報、 特開平 7- 316066号公報)。
また、 IGF-Iには免疫能力を改善する効果がある。 胸腺または炎症部位などの 組織中で局所的に産生された IGF- 1は、 IGF- 1レセプターを有している Tリンパ球 の増殖および機能の向上に関与していると考えられている (Tapson, V. F. , et al. , J. Cl in. Invest. , 82, 950 (1988) )。 また、 IGF- 1はナノモル濃度では、 リンパ球に対する増殖促進因子であることが報告されている (Schimpff, R. M. , et al. , Acta Endocrinol . 102, 21 (1983) ) 0 従って、 エイズ患者をはじめと する免疫不全患者への IGF- 1の使用も検討されている (特表平 6-508830号公報)。 さらに、 IGF-Iには骨量増加作用があるので骨粗鬆症の治療に効果があると考 えられている (Bennett, A. E., et al. , J. Cl in. Endocrinol. etab. , 59, 7 01 (1984); Brixen, K. , et al. , J. Bone. Miner. Res. , 5, 609 (1990); Joha nnsson, A. G. , et al., J. Intern. Med. , 234, 553 (1993); Johannsson, A. G. , et al. , Lancet, 339, 1619 (1992); Riggs, B. し., Am. J. Med. , 95, Sup pl. 5A, 62S, (1993); 特開平 4- 235135号公報、 米国特許第 4, 861, 757号)。
0 一 しかしながら、 IGFはもともと生体内ではほとんどが IGFBPと複合体を形成し、 それらの生理作用が調節されていることが知られている (Rechler, . M. , Vita m. Horm., 47, 1 (1993); Clemmons, D. R., Growth Regul. , 2, 80 (1992) )。
IGFBPは、 今までに 6種類存在することが知られており、 それらは各々 IGFBP- 1 から IGFBP- 6と命名されている。 これら 6種類の IGFBPはアミノ酸配列では、 相互 に高い相同性を示し、 特に N末端側と C末端側のいずれもシスティンを多く含む ドメインでは相同性が顕著であり、 IGFBPのアミノ酸配列上での中間部分では相 同性があまり認められない。 ヒ トの場合、 これら 6種類の IGFBPでは 16個のシス ティンの位置が保存されている (なお、 IGFBP-1から IGFBP - 5の 5種類の IGFBPで は 18個のシスティンが位置的に保存されている。) (Shimasaki, S., et al., Pr og. Growth Factor Res. , 3, 243 (1991) )。
ヒ トの成人の血中における IGFBP-1、 IGFBP- 2、 IGFBP- 3の濃度はそれぞれ約 2 n Μ、 5 ηΜ、 ΙΟΟηΜであり、 血中では IGFBP-3が、 IGFに対する主な結合タンパク質で める (Baxter, R. C., in Modern Concepts of Insul in- Like browth Factors (Spencer, E. M. , ed) pp371, Elsevier Science Publ ishing Co. , New York-Am sterdara ( 1991) )。 しかし、 健常人の血清を中性条件下でゲルろ過し、 IGFの溶 出位置を確認すると、 ほとんどの IGFは 150kDa付近に溶出され 3つの分子の複合 体として存在する (Baxter, R. , et al. , Proc. Natl. Acad. Sci. USA, 86, 6898 (1989) )。 この 3つの分子の複合体は、 分子量約 7. 5KDaの IGF- 1 (又は IGF - Π) と、 酸に安定な 53KDaの IGFBP-3に加え、 さらに酸に不安定な 84KDaのサブュ ニット蛋 白 (Acid Labi le Subunit;以下、 ALSと略する。 α -サブユニッ トとも 呼ばれる) から構成されている。 そして、 IGFが血中の主要な結合タンパク質で ある IGFBP - 3と結合したときにのみ、 これに ALSが結合して、 150KDaの複合体を形 成するものと考えられている。
遊離型 IGFや、 IGFと IGFBPの 2者の複合体は毛細血管壁を通過できるが、 IGFと IGFBP - 3と ALSの 3者の複合体は通過できないとされている (Rechler, M. M. , Vi tarn. Horm. , 47, 1 ( 1993) )。 一方、 IGFのヒ ト血中内半減期は、 遊離型 IGFの場 合は約 10分と短いが、 IGFと IGFBPの複合体では約 3 0分、 また IGF、 IGFBP-3およ び ALSの 3者の複合体では約 15時間である (Zapf, J. , et al. , in Modern Conce pts of Insul in -し ike Growth Factors (Spencer, E. M. , ed) pp. 591, El sevier
Science Publ ishing Co. , New York-Amsterdam (1991) )。
従って、 ALSは、 IGFBP- 3と共に、 IGF、 IGFBP- 3および ALSの 3者の複合体を形 成することで、 IGFの血中半減期を延長させると共に、 IGFの生理活性を抑制する ものと考えられ、 また IGFBPは、 IGFと IGFBPの 2者の複合体を形成することで IGF の血中半減期の延長を図るとともに、 IGFの生理活性を抑制または促進するもの と考えられている (Baxter, R. C. , et al. , Prog. Growth Factor Res. , 1, 49
(1989) )。
ALSの種差はほとんどなく、 ヒ トとラットの ALSの相同性は 78%である (Dai,J., et al. , Biochem. Biophys. Res. Commun. , 188, 304 (1992) )。 ALSは、 単独では I GFまたは IGFBP-3と結合しないと言われているが、 近年、 ラット血清中では、 ALS と IGFBP - 3の複合体として存在するという報告もある (Lee, C. Y., Endocrinolo gy, 136, 4982 (1995) )。
生体内に IGFを投与した場合、 主に活性を発現する遊離型 IGFの血中濃度はさほ ど上昇せずに、 IGFBP - 2の血中濃度の上昇が認められることが知られており、 こ のことから生体内には IGFによる IGFBPの発現調節機構が存在すると考えられてい る (Zapf, J. , et al. , in Modern Concepts of Insul in -し i Ke Growth Factors (Spencer, E. M. , ed) pp. 591, Elsevier Science Publ i shing Co. , New York-Ams terdam (1991) )。
また、 IGF、 IGFBP- 3および ALSの 3者の複合体による調節機構の変化として、 低血糖を呈する膝外腫瘍 (IGF - I I産生腫瘍:以下、 NICTHと略する。) の例があ る。 NICTH患者では通常の 7. 5kDaの IGF- I Iの代わりに IGF- I Iの E—ドメイン部位 に糖鎖が付加したいわゆる big IGF- IIが主要に存在している。 この big IGF- IIは IGFBP- 3と複合体を形成するものの、 IGFBP - 3と ALSとの 3者の複合体は形成でき レヽ (Baxter, R C. , in Modern Concepts of Insul in-Like Growth Factors (Spencer, E. M. , ed) pp. 371, Elsevier Science Publ ishing Co. , New York-Am sterdam ( 1991) )。 それにより、 biglGF- I Iによる血糖降下作用が ALSによって妨 害されることなく発現されると考えられている。 big IGF - IIは、 血中では主とし て IGFBP- 3との複合体として存在するものであるが、 この複合体は毛細血管壁を 通過できると言われている (Rechler, M. M., Vi tam. Horm. , 47, 1 (1993) )。 従 つて、 big IGF - IIと IGFBP-3の複合体は標的部位に到達できるものと考えられる。 また、 IGF- 1と IGFBP-3の複合体を脳下垂体除去ラッ トに投与しても、 IGF-I単独 投与の場合よりも弱いが、 IGF-Iの作用が認められたとの報告もある (Zapf, J., et al. , J. Clin. Invest. , 95, 179 (1995) )。 従って、 血中で存在する IGFと IGFBP- 3の複合体は標的部位へ移行して IGFとしての活性を発現する能力があるも のと考えられる。
さらに、 IGFと IGFBPの複合体の調節機構の変化として IGFBPのプロテアーゼの 存在が挙げられる。 妊娠後期の妊婦血中 IGFBP-3濃度を分析すると、 抗 IGFBP- 3抗 体を用いた RIAでは軽度上昇が認められるのに対し、 1 2 5 I- IGFを用いたゥエスタ ン リガンド プロッ ト法では、 逆に IGFBP-3濃度は低下する。 この測定法によ る差が何に起因するものかを調べた結果、 妊婦血中には IGFBP- 3を分解するプロ テア一ゼが存在することが明らかにされた (Hossenlopp, P. , et al. , J. Clin. Endocrinol. Metab. , 71, 797 (1990); Giudice, L. C. , et al., J. Cl in. End ocrinol. Metab. , Π , 806 (1990) )。 このように血中に存在するプロテア一ゼ 力 IGFBP- 3を分解するため、 IGFと IGFBP-3との親和性が低下し、 その結果 IGFのァ べィラピリティーを増加するものと考えられている。
このように、 もともと生体内に存在する IGFは、 IGFBPおよび ALSによりその活 性が調節されている。 また、 生体外から投与された IGFも遊離型であれば速やか に代謝される力、 あるいは新たに発現してきた IGFBPとの 2者の複合体や IGFBPお よび ALSとの 3者の複合体を形成するものと考えられる。 生体外から IGFまたは IG F様活性を持つ物質を投与しても、 生体內における IGFの IGFBPおよび/または ALS による調節機構により、 その活性の発現は制御されている。
そこで、 本発明者らは鋭意検討した結果、 生体外から IGFあるいは IGF様活性を 持つ物質を投与するのではなく、 生体内において遊離型 IGFを増やす物質や、 IGF 様活性を示す IGFと IGFBPの 2者の複合体を増やす物質を投与することにより、 IG F作用を発現させることができることを見いだした。
すなわち、 IGFと IGFBPの 2者の複合体や IGF、 IGFBPおよび ALSの 3者の複合体 から遊離型 IGFに変換する物質、 3者の複合体から IGFと IGFBPの 2者の複合体に 変換するような物質、 3者の複合体から IGFや IGFと IGFBPの 2者の複合体を遊離 させる物質、 または IGFと IGFBPの 2者の複合体形成や IGFと IGFBPと ALSの 3者の 複合体形成を阻害する物質を投与することにより、 IGF作用を発現させることが できることを見いだした。
本発明はこのような知見に基づきなされたものであり、 生体内に大量に存在し ている内在性の IGFを有効に利用することを目的とする。
すなわち、 本発明は、 生体内に内在している IGFを、 十分な IGF活性を有する遊 離型 IGFに変換する遊離型 IGFの濃度上昇方法を提供することを目的とする。
また本発明は、 遊離型 IGFよりは IGF活性は低いものではあるが、 IGFと IGFBPと ALSの複合体に比べ高レ、 IGF活性を有する IGFと IGFBPとの複合体の濃度上昇方法を 提供することを目的とする。 発明の開示 本発明は、 生体内に内在している IGFを、 十分な IGF活性を有する遊離型 IGFに 変換する遊離型 IGFの濃度上昇方法を提供する。
この遊離型 IGFへの変換は、
IGFと IGFBPの複合体、 又は IGF、 IGFBPおよび ALSの複合体を IGFに変換するこ とにより ;
IGFと IGFBPの複合体から、 又は IGF、 IGFBPおよび ALSの複合体から、 IGFを遊 離させることにより ;或いは、
IGFと IGFBPとの結合、 又は IGFと IGFBPと ALSとの結合を阻害することにより、 行うことができる。
また本発明は、 遊離型 IGFよりは IGF活性は低いものではあるが、 IGFと IGFBPと ALSの複合体に比べ高い IGF活性を有する IGFと IGFBPとの複合体の濃度上昇方法を 提供する。 この IGFと IGFBPとの複合体への変換は、
IGF、 IGFBPおよび ALSの 3者の複合体を、 IGFと IGFBPの 2者の複合体に変換 することにより ;
3者の複合体から、 IGFと IGFBPの 2者の複合体を遊離させることにより ;或 いは
IGFと IGFBPの 2者の複合体と、 ALSとの結合を阻害することにより
行うことができる。
さらに本発明は、 生体内の遊離型 IGF又は IGFと IGFBPとの複合体の濃度を上昇 させる以下の物質を提供する。
1)生体内の IGFと IGFBPの 2者の複合体を遊離型 IGFに変換する物質。

本発明の遊離型 IGFの濃度上昇方法は、 生体外から IGFまたは IGF様活性を持つ 物質を投与することにより達成するものではなく、 もともと生体内に存在する遊 離型 IGFや、 IGFと IGFBPの 2者の複合体および/または IGF、 IGFBPおよび ALSの 3 者の複合体中に内在する IGFを利用するものである。 ここでいう生体とは、 ヒ ト やゥシ、 ゥマ、 ヒッジ、 ブタといったヒ ト以外の哺乳類等の血液や肝臓、 腎臓と いった臓器等の組織のことをいう。 また、 遊離型 IGFとは、 生体内において 2者 の複合体または 3者の複合体から変換、 あるいは遊離された IGFのみならず、 IGF BPとの 2者の複合体または IGFBPと ALSの 3者の複合体を形成していない IGFのこ とをいう。
本'発明の遊離型 IGFの濃度上昇方法は、
IGFと IGFBPの 2者の複合体中において不活性な IGFを活性型 IGFに変換する、 IGFと IGFBPの 2者の複合体から IGFを遊離させる、
IGFと IGFBPとの結合を阻害する、
IGF、 IGFBPおよび ALSの 3者の複合体中において不活性な IGFを活性型 IGFを 活性型 IGFに変換する、
IGF、 IGFBPおよび ALSの 3者の複合体から IGFを遊離させる、 あるいは IGFと IGFBPと ALSとの結合を阻害する
こと等により、 遊離型 IGFの濃度を上昇させるものである。
また、
IGF、 IGFBPおよび ALSの 3者の複合体を IGFと IGFBPの 2者の複合体に変換す る、
IGF、 IGFBPおよび ALSの 3者の複合体から IGFと IGFBPの 2者の複合体を遊離 させる、 あるいは
IGFと IGFBPの 2者の複合体と ALSとの結合を阻害する
こと等により、 いったん IGFと IGFBPの 2者の複合体の濃度を上昇させ、 次いで IGFと IGFBPの 2者の複合体を IGFに変換する、
IGFと IGFBPの 2者の複合体から IGFを遊離させる
こと等によっても、 遊離型 IGFの濃度を上昇させることができる。 遊離型 IGFの濃 度が上昇し、 IGF作用が発現するので、 本方法は、 IGFの作用により、 予防および /または治療が可能な疾患の予防 ·治療の方法となる。
本発明の IGFと IGFBPの 2者の複合体の濃度上昇方法は、
IGF、 IGFBPおよび ALSの 3者の複合体を IGFと IGFBPの 2者の複合体に変換す る、
IGF、 IGFBPおよび ALSの 3者の複合体から IGFと IGFBPの 2者の複合体を遊離 させる、 あるいは
IGFと IGFBPの 2者の複合体と ALSとの結合を阻害する
こと等により、 IGFと IGFBPの 2者の複合体の濃度を上昇させるものである。 従つ て、 本方法は IGFと IGFBPの 2者の複合体の示す IGF様活性により、 予防および/ま たは治療が可能な疾患の予防 ·治療の方法となる。
以下、 本発明の物質について説明する。
本発明の物質は、 インスリンレセプタ一および/または IGFレセプターに結合し て、 その活性を発現させる IGFまたは IGF様活性を持つ物質とは異なり、 インスリ ンレセプターおよび IGFレセプターの両レセプターには実質的に結合しないもの である。
本発明における IGFと IGFBPの 2者の複合体を IGFに変換する物質とは、 2者の 複合体に作用して、 IGFにかえてしまう物質を意味する。 したがって、 複合体を I GFに変換する物質を投与すると、 遊離型 IGFは増える。
本発明における IGFと IGFBPの 2者の複合体から、 IGFを遊離させる物質とは、 2者の複合体に作用して、 IGFを遊離させる物質を意味する。 したがって、 遊離 させる物質を投与すると、 遊離型 IGFは増える。
本発明における IGFと IGFBPとの結合を阻害する物質とは、 IGFと IGFBPの 2者の 複合体の形成を阻害する物質を意味する。 したがって、 結合を阻害する物質を投 与すると、 遊離型 IGFは増える。
本発明における IGF、 IGFBPおよび ALSの 3者の複合体を、 IGFと IGFBPの 2者の 複合体に変換する物質とは、 3者の複合体に作用して、 IGFと IGFBPの 2者の複合 体にかえてしまう物質を意味する。 したがって、 IGFと IGFBPの 2者の複合体に変 換する物質を投与すると、 IGFと IGFBPの 2者の複合体は増える。
本発明における IGF、 IGFBPおよび ALSの 3者の複合体から、 IGFと IGFBPの 2者 の複合体を遊離させる物質とは、 3者の複合体に作用して、 IGFと IGFBPの 2者の 複合体を遊離させる物質を意味する。 したがって、 IGFと IGFBPの 2者の複合体を 遊離させる物質を投与すると、 IGFと IGFBPの 2者の複合体は増える。
本発明における IGFと IGFBPの 2者の複合体と ALSとの結合を阻害する物質とは、 IGF、 IGFBPおよび ALSの 3者の複合体の形成を阻害する物質を意味する。 したが つて、 結合を阻害する物質を投与すると、 IGFと IGFBPの 2者の複合体は増える。 本発明における IGF、 IGFBPおよび ALSの 3者の複合体を IGFに変換する物質とは、 複合体に作用して、 IGFにかえてしまう物質を意味する。 したがって、 3者の複 合体を IGFに変換する物質を投与すると、 遊離型 IGFは増える。
本発明における IGF、 IGFBPおよび ALSの 3者の複合体から、 IGFを遊離させる物 質とは、 3者の複合体に作用して、 IGFを遊離させる物質を意味する。 したがつ て、 遊離させる物質を投与すると、 遊離型 IGFは増える。
本発明における IGFと IGFBPと ALSとの結合を阻害する物質とは、 IGF、 IGFBPお よび ALSの 3者の複合体の形成を阻害し、 遊離型 IGFを増やす物質を意味する。 し たがって、 結合を阻害する物質を投与すると遊離型 IGFは増える。
なお、 本発明において、 IGFと IGFBPの 2者の複合体おょぴ IGFと IGFBPと ALSの 3者の複合体は、 静電的相互作用、 水素結合、 疎水的相互作用等により形成され ているものをいう。 また、 IGFの種類、 IGFBPの種類等に特に限定はなく、 すべて の種類の複合体が含まれる。 例えば IGF- 1と IGFBP - 3の 2者の複合体および IGF - 1、 IGFBP- 3および ALSの 3者の複合体等を挙げることができる。
本発明の物質を投与すると、 生体内で遊離型 IGFまたは IGFと IGFBPの 2者の複 合体が増え、 IGF作用が発現するので、 本発明の物質は医薬として有用なもので あり、 さらには、 IGFにより予防および/または治療が可能な疾患の予防 ·治療剤 として有用なものである。
したがって、 本発明の物質は、 例えば糖尿病、 糖尿病性神経症、 筋萎縮性側索 硬化症および骨粗鬆症等の予防剤および/または治療剤となり得る。
また疾患によっては、 IGFBPの組織特異的発現および/または IGFの組織特異的 作用を考慮に入れて、 本発明の物質の種類の選択ができる。 すなわち、 筋、 骨と いった各種臓器に選択的で、 局所における遊離型 IGF濃度を調節し、 IGFによる効 果を組織特異的に引き出すことが可能な物質を調製することもできる。
本発明の物質が有する機能は上述のとおりだが、 本発明において物質とは、 特 に限定はなく、 例えば、 微生物、 植物もしくは動物等の生物が産生する物質、 植 物もしくは動物の細胞あるいは組織培養物が産生する物質、 生物等の抽出物また は化学合成した化合物等を挙げることができる。 なお、 これらは単独でもよいし、 2種以上の混合物であってもよい。
本発明の物質は、 インスリン様成長因子誘導体および抗ィンスリン様成長因子 結合たんぱく質抗体を包含する。 インスリン様成長因子誘導体とは、 インスリン 様成長因子の誘導体であって、 例えば、 インスリン様成長因子に化学修飾等して 合成したもの、 或いは遺伝子工学的手法を用いて、 インスリン様成長因子のアミ ノ酸配列において 1つまたは 2つ以上のアミノ酸残基を付加、 欠失もしくは置換 させたもの等を挙げることができる。 アミノ酸残基の付加、 欠失もしくは置換は、 又献 (Genetic Engineering, 3, 1 (1981); Nucleic Acid Research, 10, 6487 (1982) ) 等に記載されている技術により実施することができる。
本発明のインスリン様成長因子誘導体としては、 ヒ ト インスリン様成長因子 - IIのァミノ酸配列の 27位のチロシン残基および 43位のバリン残基をそれぞれ口 イシン残基に置換したものが好ましい。
本発明の物質を投与する方法は、 非経口投与、 経口投与どちらでも構わないが、 使用時の簡便性等を考慮すると経口投与が好ましい。 投与するためには、 通常知 られた方法により製剤化すればよい。 製剤化にあたっては、 賦形剤、 崩壊剤、 結 合剤、 滑沢剤、 流動化剤、 分散剤、 懸濁化剤、 乳化剤、 防腐剤および安定化剤等 の通常知られた添加物を適宜、 適当量加えることができる。
非経口投与するための製剤の形態としては、 軟膏剤、 硬膏剤、 坐剤、 注射剤、 点眼剤、 点鼻剤、 点耳剤、 吸入剤、 酒精剤、 パップ剤、 リニメント剤およびロー シヨン剤等を挙げることができる。 また、 経口投与するための製剤の形態として は、 エリキシル剤、 カプセル剤、 顆粒剤、 細粒剤、 丸剤、 懸濁剤、 乳剤、 散剤、 錠剤、 シ口ップ剤、 ト口ーチ剤、 ドライシロップ剤およびリモナ一デ剤等を挙げ ることができる。
本発明の物質の投与量は、 投与方法、 対象疾患、 患者の状態等を考慮して適宜 検討すればよい。
本発明者らは、 本発明に係る物質を 2種類見いだし、 これらを正常ラットおよ ぴ糖尿病モデルラッ トに in vivo投与することにより、 遊離型 IGF- 1の血中濃度が 上昇することおよび IGFの作用が発現されることを確認した。
以下、 本発明の物質をスクリーニングするための試験方法を説明する。
IGF、 IGFBP、 ALSのいずれか一つを直接又は間接に検出可能に標識し、 当該標 識物 (例えば、 IGF) と他の結合相手 (例えば、 IGFBP) との結合が、 試料添加に よって阻害されるかどう力、 或いは、 試料添加により、 当該標識物 (例えば、 IG F) と結合相手 (例えば、 IGFBP) との結合物から当該標識物が遊離するかどうか によって、 本発明の物質を探索することができる。
直接に検出可能な標識とは、 その標識物の量が物理的測定により直接検出でき るできるものをいい、 例えばラジオアイソトープ標識の場合には放射能測定によ り、 標識物を直接検出することができる。 また、 Scinti l lation Proximity Assa y (Cook, N. D. , Drug Discovery today, 丄, 287 (1996) ) のような方法でも検出 できる。 また色素や蛍光色素などによって標識した場合には、 光学的に直接標識 物の量を検出できる。
間接に検出可能な標識とは、 それ自体は物理的に検出できないが、 化学的反応 を経ることにより直接検出可能な分子種 (例えば色素や蛍光色素など) を化学量 論的に生成し、 これを定量することのより標識物の量を定量できるようなものを いう。 例えば、 酵素標識した場合には、 試料 (被検物) 共存下の反応後、 酵素基 質を添加して検出可能な色素などを生成させて、 それらを検出すればよい。
ラジオアイソトープを用いたスクリ一二ングを行うには、 従来より汎用されて いる液相法に準拠して行うことができる。 例えば、 1 2 5 1で標識した IGF ( 1 2 51-1 GF) と IGFBPとを反応させた後、 複合体として存在する' 2 51 - IGF (結合型) と複 合体を形成していない' 2 5 I-IGF (遊離型) を、 1 ) 活性炭を用いて分ける方法 (Moses, A. , et al. , Endocrinology, 104, 536 (1979); Binoux, M. , et al. , J. Cl in. Endocrinol. Metab. , 59, 453 (1
984); Scott, C. D., et al., Endocrinology, U6, 1094 (1985); Szabo, L. , et al. , Biochem. Biophys. Res. Commun. , 151. 207 (1988); Gelato, M. C. , et al. , J. Clin. Endocrinol. Metab. , 70, 879 (1990); Oh, Y. , et al. , Bio chem. J. , 278, 249 (1991)等)、
2 ) レクチンタンパク質が IGFBPの糖鎖部分を認識することを利用して分ける 方法 (Martin, J. L. , et al. , J. Biol. Chem. , 261, 8754 (1986) )、
3 ) 一次抗体および二次抗体を用いて免疫沈降して分ける方法 (Martin, J. し, et al. , J. Biol. Chem. , 261, 8754 (1986); Baxter, R. C., et al. ' J. Biol. Chem. , 264, 11843 (1989); Baxter, R. C. , Biochem. J., 271, 773 (19 90)等) 等の汎用方法で分離することができる。
これらの方法を利用して、 1 2 5 I-IGF、 1 2 5 I- IGFBP、 または1 2 5 I- ALSとそれぞれ 適切な一次抗体の組み合わせにより、 IGFと IGFBPの 2者の複合体または IGF、 IGF BPおよび ALSの 3者の複合体の形成能の評価を行うことができる。
ただし、 これらの従来方法は、 取扱いが煩雑で危険であり、 また半減期が短い ラジオァイソ トープを標識物質として用い、 また結合型/遊離型を分離する際に 遠心操作を行うので、 多検体のスクリーニングには不向きである。
高速で多検体のスクリーニングを行うには、 Scinti l lation Proximity Assay のような方法も利用できるが、 標識物質としてラジオアイソトープを使用せずに、 酵素を用いた以下のような固相法によるものも便宜である。
本発明の IGFと IGFBPの 2者の複合体を遊離型 IGFに変換する物質または 2者の 複合体から遊離型 IGFを遊離させる物質は、 例えば、 予め固相化した IGFBPに、 予 め酵素で標識した IGFを加えて、 2者の複合体を形成させた後に、 被験体を添加 して反応を行い、 洗浄後、 標識に用いた酵素の活性を測定することでスクリー二 ングが可能である。 また、 IGFを固相化し、 IGFBPを酵素で標識してもよい。
本発明の IGFと IGFBPとの結合を阻害する物質は、 固相化した IGFBPに、 予め酵 素で標識した IGFおよび被験体を同時に添加して反応を行い、 洗浄後、 標識に用 いた酵素の活性を測定することでスク リーニングが可能である。 また、 IGFを固 相化し、 IGFBPを酵素で標識して酵素活性を測定することによりスクリーニング 本発明の IGF、 IGFBPおよび ALSの 3者の複合体を IGFと IGFBPの 2者の複合体に 変換する物質、 IGF、 IGFBPおよび ALSの 3者の複合体から IGFと IGFBPの 2者の複 合体を遊離させる物質、 IGF、 IGFBPおよび ALSの 3者の複合体を遊離型 IGFに変換 する物質、 または IGF、 IGFBPおよび ALSの 3者の複合体から IGFを遊離させる物質 は、 例えば、 予め固相化した ALSに、 IGFおよび予め酵素で標識した IGFBPを加え て、 3者の複合体を形成させた後に、 被験体を添加して反応を行い、 洗浄後、 標 識に用いた酵素の活性を測定することでスクリーニングが可能である。 また、 酵 素で標識するものを IGFにかえてもよい。
あるいは、 予め固相化した IGFと IGFBPの 2者の複合体に、 予め酵素で標識した ALSを加えて、 3者の複合体を形成させた後に、 被験体を添加して反応を行い、 洗浄後、 標識に用いた酵素の活性を測定することによりスクリーニングしてもよ レ、。
さらには、 予め固相化した IGFに、 IGFBPおよび予め酵素で標識した ALSを加え て、 3者の複合体を形成させた後に、 被験体を添加して反応を行い、 洗浄後、 標 識に用いた酵素の活性を測定することによりスクリーニングしてもよい。 なお、 スクリーニングにより得られた物質が、 IGF、 IGFBPおよび ALSの 3者の複合体を I GFと IGFBPの 2者の複合体に変換する物質、 IGF、 IGFBPおよび ALSの 3者の複合体 から IGFと IGFBPの 2者の複合体を遊離させる物質、 IGF、 IGFBPおよび ALSの 3者 の複合体を遊離型 IGFに変換する物質または IGF、 IGFBPおよび ALSの 3者の複合体 から遊離型 IGFを遊離させる物質のうち、 どれに属するかを確認するには、 前述 の IGFと IGFBP 2者の複合体を遊離型 IGFに変換する物質または 2者の複合体から 遊離型 IGFを遊離させる物質をスクリーニングする方法を適用することにより可 能である。
本発明の IGFと IGFBPの 2者の複合体と ALSとの結合を阻害する物質または IGFと IGFBPと ALSとの結合を阻害する物質は、 例えば、 予め固相化した ALSに、 IGFと予 め酵素で標識した IGFBPの 2者の複合体および被験体を同時に添加して反応を行 い、 洗浄後、 標識に用いた酵素の活性を測定することでスク リーニングが可能で ある。 また、 酵素で標識するものを IGFにかえてもよい。
あるレ、は、 予め固相化した IGFと IGFBPの 2者の複合体に、 予め酵素で標識した ALSおよび被験体を同時に添加して反応を行い、 洗浄後、 標識に用いた酵素の活 性を測定することでスク リ一二ングが可能である。
さらには、 予め固相化した IGFに IGFBPと予め酵素で標識した ALSおよび被験体 を同時に添加して反応を行い、 洗浄後、 標識に用いた酵素の活性を測定すること によりスク リーニングしてもよレ、。 なお、 スクリーニングにより得られた物質が、 IGFと IGFBPの 2者の複合体と ALSとの結合を阻害する物質または IGFと IGFBPと ALS との結合を阻害する物質のどちらに属するかを確認するには、 前述の IGFと IGFBP との結合を阻害する物質をスクリーニングする方法を適用することにより可能で ある。
IGF、 IGFBPまたは ALSを固相化するには、 通常知られた方法で行えばよい。 す なわち、 直接固相法でもよく、 またアビジン一ピオチン、 ハプテン一抗ハプテン 抗体等を用いた間接固相法でもよい。 固相の素材としては、 例えばガラス、 ポリ スチレンを主としたプラスチック、 ポリアクリルアミ ドおよび酢酸セル口一ス等 を挙げることができる。 固相の形態としては、 これらの素材を用いて成型した試 験管、 ビーズ、 マイクロタイタープレート、 ディスクおよびチップ等を挙げるこ とができる。 機器を用いて高速で多検体をスクリーニングすることを考慮すると、 市販されている多穴マイクロタイタープレートを利用するのが好ましい。
酵素活性を測定するには、 標識に用いる酵素、 基質、 緩衝液、 pH、 温度等を適 宜検討し、 通常知られた測定法で行えばよく、 測定法としては、 例えば比色法、 蛍光法および発光法等を挙げることができる。
IGF、 IGFBPまたは ALSを酵素標識するには、 通常知られた方法で行えばよく、 例えば、 マレイミ ド法、 過ョーソ酸法およびダルタルアルデヒ ド法等を挙げるこ とができる。
また、 標識に用いる酵素と基質は適宜組み合わせを考慮すればよく、 例えば、 酵素として ]3 - D-ガラク トシダーゼを用いる場合には、 基質としては、 2-ニトロ フェニル- β -D-ガラク トシド、 4 -メチルゥムべリフエリノレ- - D -ガラク トシドぉ よび 5-ブロモ -4 -ク口ロ- 3-ィンドリノレ- )3 - D-ガラク トシド等を挙げることができ る。 また、 酵素としてペルォキシダーゼを用いる場合には、 基質としては、 2,2' -アジノ-ビス (3-ェチルベンズチアゾリン- 6-スルフォニック アシッド)、 3, 3 ' , 5, 5' -テトラメチルベンジジンおよび 1, 2 -フエ二レンジアミン等を挙げること ができる。 さらに、 酵素としてアルカリフォスファターゼを用いる場合には、 基 質としては、 4-メチルゥムベリフェリルフォスフエ一トおよび N-二ト口フエニル フォスフエ一ト等を挙げることができ 。
スクリ一二ングは緩衝液中で行われるのが好ましく、 緩衝液は酵素活性の測定 において通常用いられるものでよく、 例えば、 リン酸ナトリウム緩衝液、 グリシ ン—水酸化ナトリウム緩衝液およびトリスー塩酸緩衝液等を挙げることができる。 緩衝液の pHは適宜検討すればよいが、 pHが IGFBPの溶解性に影響を与えることが あるので、 pH6. 0〜7. 4が好ましい。 また、 緩衝液には、 塩や界面活性剤を添加し てもよく、 例えば塩化ナトリウムを添加する場合には、 その添加濃度としては 0. 15M程度までが好ましい。
産業上の利用可能性 本発明の方法によれば、 生体外から IGFあるいは IGF様活性を持つ物質を投与す るのではなく、 生体内に内在している IGFを遊離型 IGFや IGFと IGFBPの 2者の複合 体に変換等することにより、 IGF作用を発現させることができる。
本発明の物質を in vivo投与することにより、 遊離型 IGFの濃度が上昇し、 IGF の作用が認められた。 したがって、 本発明の物質は IGFにより予防および/または 治療可能な疾患 (糖尿病、 筋萎縮性側索硬化症および骨粗鬆症等) の予防剤およ び/または治療剤として有用なものである。
以下に、 本発明を実施例により詳細に説明するが、 本発明はこれらに限定され るべきものではない。 実 施 例 実施例 1 ヒ ト IGF - 1およびヒ ト IGF-I Iの調製
ヒ ト IGF - 1は GroPep社より購入したものを用いた。 ヒ ト IGF-IIは、 Sakanoら の方法 (Sakano, K. , et al. , J. Biol. Chem. , 266, 20626 (1991) ) と同じ操 作により得た。 すなわち、 遺伝子組換え手法により、 大腸菌でヒ ト IGF-IIを発現 させた。 菌体から抽出したヒ ト IGF-Πは、 リフォールデイングの後、 逆相 H P L Cカラムを用いたクロマトグラフィ一にて精製した。 実施例 2 ラット IGFBP-3遺伝子のクローニング
ラッ卜 IGFBP - 3遺伝子は文献 (Shimasaki, S., et al. , Biochem. Biophys. R es. Commun. , ^65, 907 (1989) ) に従い、 クローニングした。 すなわち、 P C R 法によりラッ ト滕臓 c D N Aライブラリーを铸型とし、 IGFBP - 3遺伝子の上流領 域 (5'側) と下流領域とをそれぞれ増幅して各 P C Rフラグメントを得た。 5'側 クローニング用の P C Rプライマーには、
5' -CGCCATGCATCCCGCGCGCC-3'、
5' -ACGCCGCACGCGTCGCCTTC-3'
を使用した。
3'側クローニング用の P C Rプライマーには
5' -GCGCGGGCCCCGTGGTGCGCTGCGAACCGT-3'

を使用した。
回収された各 P C Rフラグメントは、 それぞれ平滑化、 リン酸化した後、 pUCl 9 (Sall/Blunting/BAP) に組み込み、 クローニングした (5'側: BP- 3 up/pUC19, 3,側: BP- 3 downん UC 19)。
5'側クローン BP-3 up/pUC19について、 転写開始コ ドン (A T G ) よりさらに 上流にある Sphl部位を削除して、 プラスミ ド BP- 3 up/pUC19/ A SphIを得た (図 1 参照)。 次に 3'側クロ一ン BP- 3 down/pUC19を BamHI- Pstlで消化後、 BP - 3 downを 含むフラグメントを回収し、 平滑化し pTV119N (HincII/BAP) に組み込み、 lac Z プロモーターに対し同一方向に組み込まれたプラスミ ドを得た (BP- 3 down/pTVl 19N: 図 2参照)。
5'側クローン (BP- 3 up/pUC19/ A SphI) を Mlul- Hindl llで切断し、 BP- 3 upを 含む 250bpのフラグメントを回収した。 3'側クローン (BP- 3 down/pTV119N) を Ml ul- HindIII/ΒΑΡ処理し、 このプラスミ ドに BP-3 upを含む 250bpフラグメントを組 み込み、 ラッ ト IGFBP- 3クローンを作製した (BP- 3/pTV119N:図 3参照)。 実施例 3 ラット IGFBP- 3遺伝子の動物細胞発現用ベクターの構築
動物細胞発現用として、 得られたラット IGFBP- 3クローンを、 RSV- LTR (raus s arcoma virus long terminal repeat) ¾r¾ τるプラスミ ド (Nawa, K. , et al. , Biochem. Biophys. Res. Comraun. , 171, 729 (1990) ) に組み込んだ。 すなわち、 プラスミ ド BP - 3/pTV1 19Nを Xbal- Hindlllで切断し、 約 900bpのフラグメントを得 た。 これを発現用プラスミ ド (SV2-Term/SV40- Pro/neo/SV2- Term/+Amp) に組み 込みんだ後、 さらに Hindlll部位に、 RSV - LTRを Hindlllにてフラグメント化した 約 600bpを組み込んだ (RSV- LTR/rat IGFBP- 3/SV2_Term/SV40-Pro/neo/SV2- Term/ +Amp) (図 4参照)。 実施例 4 ラッ ト IGFBP- 3遺伝子の大腸菌発現用ベクターの構築
大腸菌発現には、 PhoAシグナル配列を用いた分泌発現プラスミ ドを作製して、 これを用いた。 すなわち、 PhoAシグナル配列を合成 D N Aオリゴマーを用いて作 製し、 これを大腸菌用発現プラスミ ド pTrc99A (Pharmacia社製) の Ncol- Hindll l 部分に組み込んで分泌用発現ベクターとした (図 5参照)。 大腸菌でラッ ト IGF BP-3を発現させるためにはラット IGFBP-3前駆体からシグナル配列を削除する必 要があるので、 実施例 2で得たプラスミ ド BP- 3/pTV119を Nael-Xbalで切断するこ とにより、 シグナル配列を削除したラット IGFBP- 3遺伝子フラグメントを回収し た。
得られたフラグメントを、 プラスミ ド BP - 3/pTV119を NcoI/Klenow/Xbal/BAP処 理したものに、 組み込んで、 図 6のプラスミ ドを作製した。 このプラスミ ドは Nc ol部位が復活しているので、 このプラスミ ドを Ncol - EcoRIで切断し、 次いで Mun g Bean Nucleaseで平滑化し、 シグナル配列を削除したラッ ト IGFBP- 3遺伝子フ ラグメントを回収した。 これを図 5の分泌用発現ベクター (Hindlll/lQenow/BA P) に導入してラッ ト IGFBP- 3分泌用発現プラスミ ドを作製した (図 7参照)。 実施例 5 ラット IGFBP- 3の動物細胞での発現おょぴ精製
ラット IGFBP-3の動物細胞での発現には CH0-K1細胞を用いた。 実施例 3で得た 図 4の動物細胞発現用プラスミ ドをリン酸カルシウム法を用いて CH0-K1細胞に導 入した。 組み換え細胞は 0. 4mg/ml G-418を含む DMEM/F- 12培地にてクローニング を行った。 優良発現株を選定し、 1% ITESを含む T培地にて培養した。 培養上清 を回収し、 これに酵素阻害剤 (2mMへ"ンス"アミに I lmM PMSF / lOOU/ml Trasylol I 2mM EDTA) を加え、 フィルターろ過 (CAPSULE FILTER 0. 2 m steri l ized, G elman Science社製) した後、 1M 酢酸ナトリウム溶液を加えて pH6. 0に調整した。 これを 0. 15M 塩化ナトリウムを含む 10mM 酢酸ナトリウムバッファー (pH6, 0) で 平衡化した SP-Sepharose F. F. カラム (Pharmacia社製) にアプライした。 同バ ッファー、 続けて 0. 5M 塩化ナトリウムを含むバッファーで洗浄後、 1M塩化ナト リゥムを含むバッファーで溶出した。 この溶出液を 2倍に希釈してリン酸ナトリ ゥム溶液にて最終的に pH7. 0となるよう調整した。
IGF- IIを固定化したリガンドアフィニティーカラムは HiTrap affinity column NHS-act i vated (Pharmacia社製) を用い、 常法に従って調製した。 このカラム を 0. 45M 塩化ナトリウムを含む 50mM リン酸ナトリウムバッファー (pH7. 0) で平 衡化した後、 先の SP- Sepharose F. F. 溶出サンプルをアプライした。 同バッファ 一、 1M 塩化ナトリ ウムを含む lOmM 酢酸ナトリ ウムバッファー (pH7. 0)、 そし て水で洗浄後、 0. 5M 酢酸にて溶出した。 得られた溶出液を回収し、 凍結乾燥後、 0. 1% トリフルォロ酢酸 (以下、 TFAと略することもある。) に溶解し、 逆相 HPLC に供した。
逆相 HPLCカラム (CAPCELLPAK C18 SG300、 250 X 4. 6mm I. D.、 SHISEID0社製) を用いて、 流速 l ml/min、 ァセトニトリル直線濃度勾配にて溶出し、 ラッ ト IGF BP - 3画分を分取した。 最後に、 分取したラッ ト IGFBP- 3 (以下、 RBP- 3CH0と略す る。) を凍結乾燥して使用するまで保存した。 実施例 6 ラット IGFBP- 3の大腸菌での発現および精製
ラッ ト IGFBP- 3分泌発現大腸菌 (UT5600) の各コロニーを L B培地にて、 37°C、 250rpmといいう条件で、 培養し、 Abs=3. 45に達したときに、 IPTGを終濃度 0. 3mM となるように添加して発現誘導を行った。 集菌した菌体から、 浸透圧ショック法 (Nossal, N. G. et al., J. Biol. Chem. , 241, 3055 (1966) ) によりペリプラ ズムを回収し、 リン酸バッファ一にて pH7. 2に調整した。
次に、 IGF- IIを固定化したリガンドアフィニティ一カラムに、 pH7. 2に調整し たサンプルをアプライし、 前述した RBP- 3CH0の精製 (実施例 5参照) と同じ操作 を行い、 0. 5M 酢酸にて溶出した画分を凍結乾燥した。 凍結乾燥にて得られた粉 末を 0, 1% TFA 10mlに溶解し、 逆相 HPLCカラム (YMC-PACK PR0TEIN_RP、 250 X 4. 6 mm I. D.、 YMC社製) を用いて流速 1 ml/min、 ァセトニトリル直線濃度勾配にて ラット IGFBP-3を溶出し、 分取した。 最後に、 分取したラット IGFBP- 3 (以下、 RBP-3E. col iと略する。) を凍結乾燥して使用するまで保存した。 実施例 7 ラット血清からのラット ALSの精製
ラット Aし Sは Baxter, R. C.らの方法 (Baxter, R. C. , et al. , Endocrinology, 134, 848 (1994) ) にてラッ ト血清から精製した。 ただし、 リガンドアフイエテ ィーカラムとして前述の実施例 5の IGF- IIを固定化した HiTrap afinity column NHS- activated (Pharmacia社製) と前記実施例 6の RBP- 3E. col iを用いた点と、 最終精製段階で DEAE- 5PWカラム (75 X 7. 5mm I . D .、 T0S0H社製) を用いた点 で、 文献とは異なる。
DEAE- 5PWカラムによる最終精製は、 まずカラムを 50mM塩化ナトリゥムを含む 1 OmM リン酸ナトリウムバッファー (pH8. 0) で平衡化し、 サンプルをアプライし た後、 同バッファーの塩化ナトリウム直線濃度勾配にてラッ ト ALS (以下、 RALS と略する) を溶出し、 分取して使用するまで凍結保存した。 実施例 8 ヒ ト IGF-Π誘導体 ([Leu27, Leu43] rIGF-II) の作製
ヒ ト IGF-I Iの 27位のチロシン残基をロイシン残基に置換した [Leu27] rIGF - I I およびヒ ト IGF- IIの 43位のパリン残基をロイシン残基に置換した [Leu43] rIGF- 1 Iは、 IGF-II/カチオン非依存性マンノース 6リン酸レセプターには結合するが、 生物活性発現に重要な IGF-Iレセプターおよびィンスリンレセプターへの結合活 性をほぼ消失しており、 さらに実際細胞増殖活性も大きく減弱していることが知 られている (Sakano, K. , et al., J. Biol. Chem. , 266, 20626 (1991) )。 さ らに、 これらの誘導体の IGFBP- 3との結合能は野生型とほぼ同等であることが知 られている (Bach, L. A. , et al. , J. Biol. Chem. , 268. 9246 (1993) )。
そこで、 27位のチロシン残基と 43位のパリン残基を同時にロイシン残基に置換 し、 IGF-Iレセプターおよびィンスリンレセプタ一^ ·の親和性をさらに低下させ、 且つ、 IGFBP- 3への親和性は保持した誘導体 ([Leu27, Leu43] rIGF-II) を作製し た。
すなわち、 Sakanoらのヒ ト IGF - Π作製方法 (Sakano, K. , et al. , J. Biol. C hem. , 266, 20626 (1991) ) に準拠して、 誘導体 ([Leu27, Leu43] rIGF- II) を作 製した。 すなわち、 変異導入用合成 D N Aオリゴマーとして [Leu27] rIGF- IIおよ び [Leu43] rIGF-IIを作製したときに使用するものと同じもの (5' - CTGGAAAAGAGGA AACCTCTG-3' , 5, - TTCTTCGAGGATACCTC - 3' ) を用いる他は、 Sakanoらの方法と同様 に、 ヒ ト IGF - IIをコ一ドする D N Aクローンを铸型として常法に従って変異を導 入後、 大腸菌にて発現させ、 精製した。 実施例 9 抗ラッ ト IGFBP-3抗体の作製
RBP-3CH0 (実施例 5で得たラッ ト IGFBP- 3) を抗原とし、 ゥサギ (日本白色種、 雄、 5羽) に免疫した。 免疫は 2週間間隔で計 5回行った。 得られた抗血清から ポリクローナル抗体を精製するために Protein Aカラムクロマトグラフィー (PRO SEP- A、 Bioprocessing社製) を行った。 まず、 抗血清を PBSで 2倍に希釈し、 pH7. 4となるように調製し、 PBSにて平衡化した Protein Aカラムにアプライした。 PBS で洗浄後、 0. 1M グリシン一塩酸バッファー (pH3. 0) で溶出し、 得られた溶出画 分を直ちに限外ろ過により濃縮と同時に PBSへバッファー交換した。 最終的に 5 種類のロッ 卜のポリクロ一ナル抗体 (ant i- RBP- 3 pAb #35、 #36、 #88、 S89 #9 0) が得られた。 実施例 1 0 固相系における IGFと IGFBP- 3との結合試験
次の基本条件のもとに試験を行った。
96穴マイクロタイタープレートは Costar社製のものを使用した。 基本バッファ 一を 50mM リン酸ナトリウムバッファー (pH6. 5) とし、 固相化には基本バッファ 一、 洗浄には 0. 03% Tween 20を添加したバッファー、 ブロッキングには 1% BSAを 添加したバッファ一、 そして反応には 0. 25% BSAおよび 0. 03% Tween 20を添加し たバッファーをそれぞれ用いた。 各反応毎の洗浄は 400 μ 1/ゥュルで 2回行うこ ととした。 西洋ヮサビペルォキシダーゼラベル IGF - II (以下、 HRP-IGF - IIと略す ることもある。) は市販の標識用キッ ト (PIERCE社製) を用い、 常法に従って作 製した。
この基本条件は、 特に断らない限り後述の実施例 1 1〜1 7 (いずれも in vit ro系) でも同じである。
まず、 実施例 6で得た RBP-3 E. col i (150ng/ml) をマイクロタイタ一プレート に 50 // 1/ゥエル添加し、 4 °Cでー晚以上静置して固相化した。 ブロッキング後、 HRP-IGF-II (終濃度 12, 000倍希釈) を 50 μ 1/ゥュル添加し、 25°Cで 2時間反応さ せた。 最後に 2,2' -アジノ-ビス (3-ェチルベンズチアゾリン- 6-スルフォニック ァシッ ド) (以下、 ABTSと略する) 溶液 (Kirkegaard & Perry Laboratories社 製) を 100 1/ゥュル添加して 20分間、 室温静置後にプレートリーダー (Vmax、 M olecular Devices社製) にて吸光度 (Abs405nm) を測定した。
検体による競合阻害実験の場合は、 ブロッキング後、 HRP- IGF- II (終濃度 12, 0 00倍希釈) と各種検体 (競合阻害物質) (各濃度) を 50 /^ 1/ゥュルとなるように 同時に添加して、 同様の反応を行った。
阻害物質を加えないときの吸光度値を Boとし、 阻害物質を加えたときの吸光度 値を Bとした。 RBP- 3E. col iを固相化しないゥエルに検体 (阻害物質) を加えず にアツセィしたときの吸光度を非特異的結合量 (NSB) とした。 検体添加時の固 相化 IGFBP- 3に対する標識 IGF-IIの結合量 (%) は
(B-NSB) I (Bo - NSB) X 100
の計算式から算出した。 検体として、 IGF - 1、 IGF-I I及び RBP - 3E. col i (IGFBP - 3) を用いてトリプリケートで測定した。 その平均値をプロットして、 SD値をバーで 示した (図 8参照)。
この結合試験を用いれば、 例えば各種 IGFおよび IGFBP誘導体の IGFBPに対する 親和性の評価、 さらに IGFと IGFBPの結合を阻害する物質等のスクリーニングが可 能である。 実施例 1 1 IGFと IGFBP- 3との結合に対する各種化合物の阻害実験
下記の表 1に記載の各化合物を検体として、 実施例 1 0に従い結合阻害実験を 行った。 各化合物は、 5%メタノール含有基本バ 00000ッファーにて lmg/mlとな るように溶解又は懸濁し、 これを反応バッファ一にて 5倍希釈して、 200jug/ml とした。 これを検体として添加した時の、 IGF-IIと IGFBP-3 (RBP-3 E.coli) の 結合量(%)を測定し、 「100(%) — 結合量(%)」 を検体の阻害活性 (%) とし た。 測定は 2回行い、 その阻害活性の平均値を求めた。 結果を図 9A, 9 Bに示 す。 測定時の各化合物の最終濃度は lOOj g/mlである。 化合物 27 (Ellagic aci d)、 化合物 29(Aclacinomycin A), 化合物 31 (Heparin)において高い阻害活性が 認められた。

34 Vitamin Be 実施例 1 2 固相系における IGFと IGFBP- 3と ALSとの結合試験 (1 ) 実施例 1 0に記載の基本条件下で、 まず、 ストレプトアビジン (l i g/ml) を マイクロタイタープレートに 50 μ 1/ゥエル添加し、 4 °Cでー晚以上静置して固相 化した。 ブロッキングバッファーでブロックした後、 ピオチン化キット (Amersh am社製) を用いて、 常法通りにピオチン化した RALS (実施例 7のラット ALS: 50η g/ml) を 50 μ 1/ゥヱル添加して 25°Cで 2時間反応させた。 次に RBP- 3E. coli (終 濃度 25ng/ml) と、 検体として各種濃度の IGF - 1又は IGF- IIとを計 50 1/ゥ-ルと なるように同時に添加し、 4 °Cでー晚反応させた。 これらの反応により形成され た IGFと IGFBP - 3と ALSの 3者の複合体に、 実施例 8で得た ant i-RBP- 3 pAb #35 ( 3 g/ml) を 50 /ζ 1/ゥエル添加し、 25°Cで 2時間反応させ、 続いて標識 2次抗 体 (anti-rabbit IgG, horseradish peroxidase l inked whole antibody, 1, 000 倍希釈、 Amersham社製) を 50 / 1/ゥ ル添加し、 25°Cで 2時間反応させた。 最後 に ABTS溶液を 100 /Z 1/ゥュル添加して 20分間、 室温静置後に吸光度 (Abs405nm) を測定した。
検体として IGF-I又は IGF - IIを加えたときの値 (吸光度) を Bとし、 検体として IGF-II 300pM加えたときの値 (吸光度) を total bound ( 100%) とした。 ビォチ ン化 RALSを加えずにアツセィしたときの値 (吸光度) を非特異的結合量 (NSB) とした。 結合量 (%) は

の計算式から算出した。 測定は n = 3で行い、 その平均値をプロットし、 SD値を バーで示した (図 1 0参照)。
この結合試験を用いれば、 例えば IGFや各種 IGF誘導体等による、 IGFBPと ALSと の 3者の複合体の形成能を評価することができる。 また IGFと IGFBPと ALSの複合 体形成を阻害する物質は、 上記測定系で、 RBP - 3E. col iと、 IGF - 1又は IGF- IIと を添加する際に同時に (又はその前後に) 添加することにより、 スク リーニング することも可能である。 実施例 1 3 固相系における IGFと IGFBP - 3と ALSとの結合試験 (2 )
実施例 1 0に記載の基本条件下で、 まず、 ストレプトアビジン (l g/ml) を マイクロタイタ一プレートに 50 / 1/ゥエル添加し、 4 °Cで一晚以上静置して固相 化した。 ブロッキングバッファーでブロックした後、 ピオチン化 RALS (200ng/m 1) を 50 μ 1/ゥ-ル添加して 25°Cで 2時間反応させた。 次に HRP-IGF- II (終濃度 2, 000倍希釈) と RBP - 3E. col i (終濃度 25ng/ml) を 50 μ 1/ゥエルとなるように同時 に添加し、 25°Cで 2時間反応させた。 洗浄後、 最後に ABTS溶液を 100 z 1/ゥエル 添加して 20分間室温静置した後に吸光度 (Abs405nm) を測定した。
競合阻害実験として、 ピオチン化 RALSの反応の後に HRP- IGF- II (終濃度 2, 000 倍希釈)、 RBP-3E. col i (終濃度 25ng/ml)、 そして競合阻害物質として RALS (各 濃度) を 50 μ 1/ゥエルとなるように同時に添加して、 同様の反応を行った。
ビォチン化 RALSを加えないときの吸光度を非特異的結合量 (NSB) とし、 阻害 物質を加えないときの吸光度を Boとし、 阻害物質を加えたときの吸光度を Bとし た。 競合阻害物質 (RALS) 添加時の、 固相化 RALSへの IGFBP- 3及び IGF- IIの結合 量 (%) は
(B-NSB) I (Bo -NSB) X 100
の計算式から算出した。 測定は n = 3で行い、 その平均値をプロットし、 SD値を バーで示した (図 1 1参照)。
この結合試験を用いれば、 例えば ALSや各種 ALS誘導体の IGFと IGFBP - 3との複合 体に対する親和性の評価、 さらに IGF、 IGFBP- 3および ALSの 3者の複合体の形成 を阻害する物質、 IGFと IGFBP- 3の 2者の複合体と ALSとの結合を阻害する物質等 のスクリーニングが可能である。
実施例 1 4 固相系における IGFBP - 3と ALSとの結合試験
実施例 1 0に記載の基本条件下で、 まず、 ストレプトアビジン (l ju g/ml) を マイクロタイタ一プレー卜に 50 μ 1/ゥエル添加後、 4 °Cでー晚以上静置して固相 化した。 ブロッキングバッファーでブロックした後、 ピオチン化 RALS (200ng/m 1) を 50 / 1/ゥヱル添加して 25。Cで 2時間反応させた。 次に RBP- 3E. col i ( 100ng/ ml) 50 μ 1/ゥュル添加し、 4 °Cでー晚反応させた。 これらの反応により形成され た IGFBP-3と ALSの複合体を、 実施例 9で得た anti-RBP- 3 pAb #35 ( 3 μ g/ml) を 50 μ 1/ゥエル添加し、 25°Cで、 2時間、 続いて 2次抗体を 50 μ 1/ゥ: ル添加し、 25°Cで、 2時間反応させた。 最後に ABTS溶液を 100// 1/ゥュル添加して 20分間、 室温静置後に吸光度 (Abs405nm) を測定した。 競合阻害実験の場合は、 ピオチン 化 RALSの反応の後に RBP- 3E. coli (終濃度 100ng/ml) と競合阻害物質として RALS (各濃度) を 50μ1/ゥエルとなるように同時に添加して、 同様の反応を行った。 ピオチン化 RALSを加えないときの値を非特異的結合量 (NSB) とし、 阻害物質 を加えないときの値を Boとし、 阻害物質を加えたときの値を Bとした。 競合阻害 物質 (RALS) 添加時の、 固相化 RALSに対する RBP- 3 E. coliの結合量 (%) は

の計算式から算出した。 測定は n= 3で行い、 その平均値をプロットし、 SD値を バーで示した (図 1 2参照)。
この結合試験を用いれば、 例えば ALSや ALS誘導体の IGFBP - 3に対する親和性の 評価および IGFBP- 3と ALSの複合体を阻害する物質等のスクリ一二ングが可能であ る。 実施例 1 5 in vitroにおけるヒ ト IGF - II誘導体 ([Leu27, Leu43]rIGF-II)
の特性 (1 )
実施例 8で得た [Leu27, Leu43]rIGF- IIが、 IGF-Iレセプター及びインスリンレ セプターへの親和性をどの程度消失しているかを、 ヒ ト胎盤から精製した IGF- 1 レセプターまたはィンスリンレセプターを用いたラジオレセブターアツセィ (Le Bon, T. R. , et al. , J. Biol. Chem. , 261. 7685 (1986); Fujita-Yamaguchi, Y., et al. , J. Biol. Chem. , 258, 5045 (1983)) にほぼ準拠して、 評価した。 まず、 125I- IGF- 1 (または1251-インスリン) (Amersham社製) 2 X 104cpm量 と各レセプターをィンキュベーションし、 50%の結合が得られるアツセィ系に添 加する各レセプタ一量を求めた。 これらレセプターと I 25I- IGF-Iまたは1251-ィ ンスリン (2 X l04cpm) に、 各濃度の [Leu27, Leu43]rIGF- IIを加え、 0.1% BSA および 0.075% Triton X- 100を含む 50mM トリス—塩酸バッファー (pH7.4) で反 応液を 300//1とし、 4°Cで、 一晚反応させた。 反応後、 75 /1のヒ ト γ _グロブ リン (4mg/ml) と 375 μ 1の 20% PEG 6000 (pH7.0) を同時に加え、 さらに 4°Cで、 1時間反応させ、 遠心 (3, 000rpmX30min、 4°C) した。 遠心上清を除去し、 沈 殿中に含まれる y - count (CPM)を測定した。
比較のため、 IGF- 1レセプタ一アツセィの場合には、 [Leu27, Leu43] rIGF- IIの 代わりに、 各濃度の IGF- 1又は IGF - IIを検体として添加した他は上記と同じ操作 を行った。 またインスリンレセプターアツセィの場合には、 [Leu27, Leu43] rIGF -IIの代わりに、 各濃度のィンスリン又は IGF- IIを検体とした他は上記と同じ操 作を行った。
レセプターを加えないときの値を非特異的結合量 (NSB) とし、 検体を加えな いときの値を Boと し、 検体を加えたときの値を Bと した。 レセプター結合量 (%) は

の計算式から算出した。 測定は n = 3で行い、 その平均値をプロットし、 SD値を ノく一で示した。
図 1 3は、 1 2 5 I- IGF- 1と IGF- 1レセプタ一を用いた IGF- 1レセプターアツセィの 結果を示す。 IGF- IIには、 IGF- 1と同程度の IGF-Iレセプターに対する親和性が認 められるのに対し、 [Leu27, Leu43] rIGF-IIの IGF-Iレセプターへの親和性は、 そ の 1〜 2 %にまで減少していたことが確認できた。
図 1 4は、 1 2 51-ィンスリンとインスリンレセプターを用いたインスリンレセ プタ一アツセィの結果を示す。 IGF- IIには、 インスリンと同程度のインスリンレ セプターに対する親和性が認められるのに対し、 [Leu27, Leu43] rIGF- I Iのイン スリンレセプターへの親和性は、 その 1〜 2 %にまで減少していたことが確認で きた。 実施例 1 6 in vitroにおけるヒ ト IGF- II誘導体 (「し eu27, Leu43] rIGF-II) の 特性 (2 )
[Leu27, Leu43] rIGF-IIが、 IGFと IGFBP-3の結合を阻害するか否かを実施例 9 にしたがって、 評価した。
その結果、 [Leu27, Leu43] rIGF-IIは、 ほぼ IGF- IIと同程度の濃度で、 標識 IGF - IIと固相化 IGFBP - 3の結合を阻害することが確認された (図 1 5参照)。 実施例 1 7 抗 IGFBP-3抗体の特性
実施例 9で得た抗 IGFBP- 3抗体 (ant i - RBP- 3 pAb) 、 IGFと IGFBP-3の結合を どの程度阻害するかを実施例 1 0にしたがって、 評価した。 すなわち、 検体 (競 合阻害物質) として、 抗 IGFBP - 3抗体を用いた他は実施例 1 0と全く同様にして 測定を行った。
その結果、 抗体 #35と #90が最も結合阻害活性が強く、 RBP-3E. col i ( IGFBP - 3) の活性を 100%としたとき、 モル比で 0. 2632% (重量比で 2 %) であった。 抗体 #36と S89はほぼ同じ活性を示してモル比で 0. 0875%、 抗体 #88はやや活性が弱くモ ル比で 0. 0525%であったが、 いずれの抗体も IGFと IGFBP - 3の結合を阻害すること が確認された (図 1 6参照)。 実施例 1 8 in vivoにおけるヒ ト IGF- II誘導体及び抗 IGFBP-3抗体の特性
本発明の方法または本発明の物質が、 実際に in vivoで IGFの有効性を発現でき るか否かを調べる必要がある。 そのために
( 1 ) 本発明の物質の投与により、 IGF投与と同様な効果が認められるか、
( 2 ) 本発明の物質の投与により、 ラットでの血中主要 IGFである遊離型 IGF- 1の 濃度の上昇が認められる力 の 2つの点について評価を行った。
IGF投与時に認められる効果として、 血中脂質レベルの低下が知られている。 具体的には、
( a ) 遊離脂肪酸レベルの低下 (Turkalj. I. , et al., J. Cl in. Endocrinol. etab. , 75, 1186 (1992) )、
( b ) トリグリセライ ドレベルの低下 (Turkalj. I. , et al. , J. Clin. Endocr inol. Metab. , 75, 1186 (1992); Zenobi, P. D. , et al. , J. Clin. Invest. , 90, 2234 (1992) )、
( c ) LDL-コレステロールレベルの低下に伴う総コレステロールの低下 (Kazumi, Τ· , Metabol ism, 35, 1024 (1986); Zenobi, P. D., et al. , Diabetologia, 3
6, 465 (1993) ) 等である。
そこで、 実施例 8および実施例 9で得た結合阻害物質の投与により、 このよう な効果が認められるか否かを調べた。 実施例 1 8— 1 血中脂質レベルの評価 ( 1 )
SDラッ ト (n = 3、 8週齢、 雄、 平均体重 300g、 日本チャールズ . リバ一社よ り購入) に非絶食条件下、 実施例 8で得た [Leu27, Leu43]rIGF- II (1,000// g/k g) を皮下投与した。 また別の SDラッ ト群に、 実施例 9で得た anti- RBP- 3 pAb #3 5 (20mg/head) を腹腔内投与した。 陰性対照群としては生理食塩水 (500 l/hea d) を皮下投与した。 陽性対照群としては IGF- 1 (50/ g/kg、 200Mg/kg) を皮下 投与した。 経時的に尾静脈から EDTAをコートしたガラスチューブに約 500μ 1の血 液を採取し、 採血後 30分以上経過した時点で遠心 (3, 500rpmX12min、 25°C) し、 上清の血漿を回収してアツセィまで— 80°Cにて保存した。 血漿総コレステロール、 総トリグリセライ ド濃度はォ一トセラ CH02および TG2 (いずれも第一化学薬品社 製) を用いた酵素法により、 日立 7150形自動分析装置で標準測定法に準じて測定 した。
その結果、 投与 6時間後の総トリグリセライ ド濃度は、 陰性対照群と比較して IGF-I (50Mg/kg) と anti- RBP - 3 pAb #35投与により有意に低下し、 [Leu27, Leu 43]rIGF-IIでも低下傾向が認められた (図 1 7参照)。 また、 総コレステロール 濃度は、 陰性対照群と比較して、 IGF-I (200^g/kg) 投与 6および 24時間後で有 意に低下した。 [Leu27, し eu43]rIGF-IIでは 24時間後で有意に低下しており、 ant i-RBP-3 pAb #35投与でも低下傾向が認められた (図 1 8参照)。
以上の結果から、 これらの結合阻害物質を投与することにより IGF-I投与と同 様の脂質レベル低下作用が確認された。
なお、 図 1 7および図 1 8は、 投与前 (0時間) の種々濃度を 100%としたとき の、 投与後の値 (%) を示し、 SEM (Standard Error Means) をバーで示した。 実施例 1 8— 2 血中脂質レベルの評価 (2)
ィンスリン抵抗性モデルとして知られている Zucker fatty rat (n = 7〜 9、 6〜11週齢、 雄、 東京実験動物社より購入) を投与開始前日より 20時間絶食後、 実施例 9で得た anti- RBP-3 pAb #36 (40mg/head、 n = 5) を皮下投与した。 陰 性対照群としては生理食塩水 (500/i l/head、 n =18) を皮下投与した。 陽性対 照群として IGF - 1 (300 M g/kg, n=8) または IGF - II (1, 200 μ §Λ§, η = 8 ) およ び (600 // g/kg、 n = 8 ) を皮下投与した。 いずれの群も投与 6時間後に採血し た。 血漿総コレステロールおよび総トリグリセライ ド濃度測定は、 実施例 1 8—
1に準拠して行った。 血漿遊離脂肪酸濃度は NEFAC-テス トヮコー (和光純薬社 製) を用いた酵素法により、 標準操作法の 1/20スケールで 96穴マイクロタイター プレート (Costar社製) 内で反応後、 プレートリーダー (Vmax、 Molecular Devi ces社製) で吸光度 (Abs550nm) を測定することにより求めた。
その結果、 総コレステロール濃度では陰性対照群に比べ、 投与 6時間後におい て全ての投与群で有意な低下がみられた (図 1 9参照)。 また総トリグリセライ ド濃度でも投与 6時間後において全ての投与群で有意な低下がみられた (図 2 0 参照)。 同様な傾向は遊離脂肪酸濃度変化にも観察された (図 2 1参照)。
以上の結果から、 結合阻害抗体 (すなわち、 IGFと IGFBP- 3の結合を阻害する抗 IGFBP - 3抗体) を投与することにより IGF投与と同様の脂質レベル低下作用が確認 された。
なお、 図 1 8、 図 1 9および図 2 0は、 投与前 (0時間) の種々濃度を 100%と したときの、 投与後の値 (%) を示し、 SEM値をバーで示した。 実施例 1 8— 3 血中遊離型 IGF- 1レベルの評価
SDラット (n = 7、 8週齢、 雄、 平均体重 300g、 日本チャールズ . リバ一社よ り購入) に非絶食条件下、 実施例 9で得た anti- RBP- 3 pAb #90 (40mg/head) を 腹腔内投与した。 陰性対照群としては生理食塩水 (500 /x l/head) を皮下投与し た。 経時的採血は実施例 1 8— 1に準拠して行った。
血漿遊離型 IGF-Iは逆相カートリッジ (Sep-Pak C18 cartridge) 法 (Hizuka, N. , et al. , Growth Regulat ion, 1 , 51 (1991) )、 血漿総 IGF- 1はギ酸/ァセト ン抽出法 (Bowsher, R. R. , et al . , Endocrinology, 128, 805 (1991) ) に準じ てそれぞれ調製した。
' " I-IGF-Iとソマトメジン - C抗血清 (抗 IGF- 1抗体) は栄研化学社製を、 また スタンダードのラッ ト IGF- 1は GroP印社製を用い、 常法に従ってラッ ト IGF- 1ラジ オイムノアツセィを行った (Moses, A. に, et al., Eur. J. Biochem. , 103, 4 01 (1980); Daughaday, W. H. , Methods Enzymol. , 146, 248 (1987))。 まず、 12 5I- IGF- 1 (7.77X 103cpm/100M 1) と、 抗血清 50 目当量と、 スタンダードまた は検体とを混合して、 アツセィバッファー (0.25% BSA、 0.05% Tween 20および 0. 1% NaN3を含む 25mM リン酸ナトリウムバッファ一 (pH7.5)) にて 300μ1に調製 し、 4°Cでー晚反応させた。 次に 75μ 1の 4mg/ml ヒ ト γ—グロブリンと 375 1 の 25% PEG 6000を加えて 4 °C、 1時間反応後に遠心 (3, OOOrpm X 20min、 4°C) した。 遠心上清を除去し、 沈殿中に含まれる γ -count (CPM)を測定した。
その結果、 遊離型 IGF - 1と総 IGF-I濃度の比率は、 陰性対照群と比較して anti - R BP - 3 pAb #90投与後 6時間において有意な上昇が認められ、 結合阻害抗体投与に より遊離型 IGF-I濃度が上昇していることが確認された (図 22参照)。
なお、 図 22は、 投与前 (0時間) の総 IGF- 1量に占める遊離型 IGF-I量 (free /total) の比率を 100%とし、 投与 1および 6時間後の値を示し、 SEM値をバーで 示した。

2者の複合体から、 インスリン様成長因子を遊離させることによる遊離型ィ ンスリン様成長因子の濃度上昇方法。
生体内のインスリン様成長因子とインスリン様成長因子結合たんばく質と の結合を阻害することによる遊離型ィンスリン様成長因子の濃度上昇方法。 生体内のインスリ ン様成長因子、 インスリ ン様成長因子結合たんぱく質お よび酸に不安定なサブュニットの 3者の複合体を、 インスリン様成長因子と インスリン様成長因子結合たんぱく質の 2者の複合体に変換することによる、 インスリン様成長因子とインスリン様成長因子結合たんぱく質の 2者の複合 体の濃度上昇方法。
