技術分野
本発明は、 新規なポリペプチド、 その製造方法、 そのポリペプチドをコー ドする c D N A、 その c D N Aからなるベクター、 そのベクターで形質転換 された宿主細胞、 そのポリペプチドの抗体、 およびそのペプチドまたは抗体 を含有する薬学的組成物に関する。 背景技術
従来、 ある特定のポリペプチドまたはそれをコードする c D N Aを得よう とする場合、 組織や細胞培養液中に目的とする生物活性を確認し、 次いでポ リペプチドの単離精製を経て、 遺伝子をクロ一ニングするという方法、 ある いはその生物活性を指標として遺伝子を発現クローニングする方法が一般的 に用いられてきた。 しかし、 生体内生理活性ポリペプチドは、 多様な生物活 性を有している場合が多いので、 あるひとつの活性を指標にして遺伝子をク ローニングした結果、 それが既知のポリペプチドと同一であることが後にな つて判明するという事例が増えている。 また、 微量しか産生されなかったり、 特別な生理的条件でのみ発現する因子も多く、 そのことが単離、 精製および 生物活性の確認を困難なものとしている。
近年、 c D N Aの作製技術やシークェンス技術は急速に発展し、 大量の c D N Aのシークェンスを迅速に行うことができるようになった。 そこでこ れらの技術を利用して、 様々な細胞や組織から c D N Aライブラリーを作製 し、 ランダムに c D N Aをクローニングして塩基配列を決定し、 新規なポリ ペプチドをコードする遺伝子を単離する方法が発展している。 この方法は、 生化学的、 遺伝学的な解析を一切必要とせずに遺伝子をクローニングし、 その塩基配列の情報を得ることができるという特徴を有しているが、 目的と する遺伝子の発見は偶発的要素が大きい。
本発明者らは、 これまで造血系や免疫系で働く増殖分化因子の遺伝子のク ローニングを研究してきた。 そして、 増殖分化因子 (例えば、 各種サイ トカ イン等) のような分泌蛋白質やそのレセプ夕一のような膜蛋白質 (以下、 こ れらをまとめて分泌蛋白質等と呼ぶ。 ) の大部分がその N末端にシグナルべ プチドと呼ばれる配列を有していることに着目して、 シダナルぺプチドをコ ードする遺伝子を効率的かつ選択的にクローニングする方法を鋭意検討した。 その結果、 動物細胞を用いて、 シグナルペプチドの有無を簡単に検索できる 方法 (シグナルシークェンストラップ (S ST) 法) を見出した (特願平 6- 13951号参照) 。 さらに同じ概念のもとに、 酵母を用いてさらに大量かつ簡便 にシグナルペプチドをコードする遺伝子を単離する方法 (酵母 S ST法) も 開発された (米国特許 No. 5, 536, 637 参照) 。 発明の開示
本発明者らは、 治療、 診断、 あるいは研究上有益な新規な因子 (ポリぺプ チド) 、 特に分泌シグナルを有する分泌蛋白質および膜蛋白質に着目してそ れを見出すべく、 鋭意検討を行なった。
その結果、 多種多様な分泌蛋白質および膜蛋白質を産生していると予想さ れる細胞株および組織、 例えばヒト胎盤、 成人の脳組織および脳組織由来の 細胞株、 ヒト骨および骨髄由来の細胞株、 およびヒト臍帯静脈内皮細胞株が 産生している新規な分泌蛋白質あるいは膜蛋白質、 およびそれをコードする cDNAを見出すことに成功し、 本発明を完成した。
本発明が提供する c DN A配列は、 クローン ON 0 5 6, ON 0 34 , 〇X 003として同定され、 前記酵母 S ST法によりヒト胎盤組織から作製 した c DN Aライブラリーより単離された。 クローン ON 0 5 6 , ON 0 34, 〇X 0 0 3は分泌蛋白質 (ここではそれぞれ ON 0 5 6, ON 034, OX 003蛋白質として表される。 ) をコードする完全な c DNA 配列を含む全長鎖 c DN Aである。
核酸配列デ一夕ベースに登録されている既知の核酸配列に対して BLASTNお よび FASTAにより、 またアミノ酸配列デ一夕ベースに登録されている既知のポ リペプチドのアミノ酸配列に対して BLASTX、 BLAST?および FASTAにより検索し た結果、 本発明のポリペプチド〇N 056, ON 034, OX 003および それをコードする核酸配列と一致する配列はなかった。 このことから、 本発 明のポリぺプチドは、 新規な分泌蛋白質であることが判明した。
本発明が提供する c DN A配列は、 クローン OA 052, OC 004, OM01 7, OM101, OM 126, OM 160, OMA 016 a, OM A 016 b, OMB 130, OMB 142, 〇 V B 100として同定され、 前記酵母 S S T法により成人の脳組織および脳組織由来の細胞株 (Τ 98 G, I MR- 32, および CCF— STTG 1) から作製した c D N Aライブラ リーより単離された。 クローン OA 052, OC 004, OM017, OM 101, OM 126, OM 160 , OMA016 a, OMA 016 b, OM B 130 , OMB 142, OVB 100は分泌蛋白質 (ここではそれぞれ OA 0 52 , OC 004, OM 0 1 7 , OM 1 0 1 , OM 1 26 , OM 160, OMA 016 a, OMA 0 16 , OMB 130, OMB 142, OVB 100蛋白質として表される。 ) をコードする完全な c DNA配列を 含む全長鎖 c DN Aである。
核酸配列データベースに登録されている既知の核酸配列に対して BLASTNお よび FASTAにより、 またアミノ酸配列データベースに登録されている既知のポ リぺプチドのアミノ酸配列に対して BLASTX、 BLASTPおよび FASTAにより検索し た結果、 本発明のポリペプチド OA 052, 〇C 004, OM01 7, OM 101, 〇M126, OM 160, OMA016 a, OMA 0 16 b, OM B 130, OMB 142, O V B 100およびそれをコードする核酸配列と 一致する配列はなかった。 このことから、 本発明のポリペプチドは、 新規な 分泌蛋白質であることが判明した。
本発明が提供する cDN A配列は、 クロ一ン OAF 062, OAF 075, 〇AG 1 19として同定され、 前記酵母 S ST法によりヒト骨および骨髄由 来の細胞株 (HAS 303, LP 101から作製した c DNAライブラリー より単離された。 クローン OAF 062, OAF 075, OAG 1 19は分 泌蛋白質 (ここではそれぞれ OAF 062, OAF 075 , OAG 1 1 9蛋 白質として表される。 ) をコードする完全な c DNA配列を含む全長鎖 c DNAである。
核酸配列デー夕ベースに登録されている既知の核酸配列に対して BLASTNお よび FASTAにより、 またアミノ酸配列データベースに登録されている既知のポ リペプチドのァミノ酸配列に対して BLASTX、 BLASTPおよび FASTAにより検索し た結果、 本発明のポリペプチド OAF 062, OAF 075, 〇AG 1 19 およびそれをコードする核酸配列と一致する配列はなかった。 このことから、 本発明のポリペプチドは、 新規な分泌蛋白質であることが判明した。
本発明が提供する c DN A配列は、 クローン OAH 040, OAH058 として同定され、 前記酵母 S ST法によりヒト臍帯静脈内皮細胞株 (HUV -EC-C) から作製した c DNAライブラリーより単離された。 クローン OAH 040, OAH058は分泌蛋白質 (ここではそれぞれ O AH 040, OAH 058蛋白質として表される。 ) をコードする完全な c DNA配列を 含む全長鎖 c DN Aである。
核酸配列デ一夕ベースに登録されている既知の核酸配列に対して BLASTNお よび FASTAにより、 またアミノ酸配列デ一夕ベースに登録されている既知のポ リペプチドのアミノ酸配列に対して BLASTX、 BLASTPおよび FASTAにより検索し た結果、 本発明のポリペプチド OAH 040 , OAH 058およびそれを コードする核酸配列と一致する配列はなかった。 このことから、 本発明のポ リぺプチドは、 新規な膜蛋白質であることが判明した。
本発明が提供する c DN A配列は、 クローン OM0 1 1 , OM 028 , OMB 092, OMB 108, 0 T 007として同定され、 前記酵母 S ST 法により成人の脳組織および脳組織由来の細胞株 (I MR— 32) から作製 した c DNAライブラリ一より単離された。 クローン OM0 1 1 , OM 028, OMB 092, OMB 108, OT 007は膜蛋白質 (ここではそ れぞれ OM0 1 1, OM028, OMB 092, OMB 1 08, OT 007 蛋白質として表される。 ) をコードする完全な c DNA配列を含む全長鎖 c DNAである。
核酸配列デ一夕ベースに登録されている既知の核酸配列に対して BLASTNお よび FASTMこより、 またアミノ酸配列データベースに登録されている既知のポ リペプチドのァミノ酸配列に対して BLASTX、 BLASTPおよび FASTAにより検索し た結果、 本発明のポリペプチド OM0 1 1, OM02 8, OMB 0 92, OMB 1 08, OT 007およびそれをコ一ドする核酸配列と一致する配列 はなかった。 このことから、 本発明のポリペプチドは、 新規な膜蛋白質であ ることが判明した。
本発明が提供する c DN A配列は、 クローン〇 AGO 5 1, OUB 068 として同定され、 前記酵母 S ST法によりヒト骨および骨髄由来の細胞株 (LP 1 0 1および U— 2 OS) から作製した c DNAライブラリーより単 離された。 クローン OAG 05 1, OUB 068は膜蛋白質 (ここではそれ ぞれ O AG 05 1, OUB 068蛋白質として表される。 ) をコードする完 全な c DNA配列を含む全長鎖 c DNAである。

さらに、 本発明には、 本発明のポリペプチドを発現させるための条件下で、 本発明の宿主細胞を培養することからなる本発明のポリぺプチドの製造方法 も含まれる。 培養は、 本発明のポリペプチドが発現し、 宿主細胞より製造さ れる条件下で行なわれることが好ましい。
本発明の c DNAは、 前記のようなベクタ一のアンチセンス領域に挿入す ることでアンチセンス RN Aを製造することもできる。 このようなアンチセ ンス RN Aは、 細胞中の本発明のポリぺプチドのレベルを制御することに用 いることもできる。
本発明は、 本発明におけるポリぺプチドのモノク口一ナルまたはポリク口 —ナル抗体をも含む。 さらに本発明におけるポリペプチドのモノクローナル またはポリクローナル抗体の製造方法をも含む。 モノクローナル抗体は、 本 発明のペプチドまたは、 その断片を抗原として用い、 通常のハイプリドーマ の技術により製造することができる。 ポリクローナル抗体は、 宿主動物 (例 えば、 ラットやゥサギ等) に本発明のポリペプチドを接種し、 免疫血清を回 収する、 通常の方法により製造することができる。
本発明には、 本発明のポリペプチド、 その抗体と薬学的に許容される賦形 剤およびノまたは担体を含有する薬学的組成物も含まれる。
( 1) の本発明のポリペプチドとしては、 配列番号 1、 4、 7、 10、 13、 1 6、 19、 22、 25、 28、 3 1、 34、 37、 40、 43、 46、 49、 52、 55、 58、 61、 64、 67、 70、 73、 76また は 79で示されたアミノ酸配列を有するもの以外に、 その一部が欠損したも の (例えば、 配列番号 1中、 生物活性の発現に必須な部分だけからなるポリ ペプチド等) 、 その一部が他のアミノ酸と置換したもの (例えば、 物性の類 似したアミノ酸に置換したもの) 、 およびその一部に他のアミノ酸が付加ま たは挿入されたものも含まれる。
よく知られているように、 ひとつのアミノ酸をコードするコドンは 1〜6 種類 (例えば、 Me tは 1種類、 L e uは 6種類) 存在する。 従って、 ポリ ぺプチドのアミノ酸配列を変えることなく c DN Aの塩基配列を変えること ができる。
(2) で特定される本発明の c DN Aには、 (1) の配列番号 1、 4、 7、 10、 1 3、 16、 1 9、 22、 2 5、 28、 3 1、 34、 37、 40、 43、 46、 49、 52、 55、 58、 6 1、 64、 67、 70、 73、 7 6または 7 9で示されるポリペプチドをコードするすべての塩基配列群が 含まれる。 塩基配列を変えることによって、 ポリペプチドの生産性が向上す ることがある。
(3) で特定される cDNAは、 (2) で示される c DNAの一態様であ り、 天然型配列を表わす。
(4) に示される c DNAは、 (3) で特定される c D N Aに天然の非翻 訳部分を加えた配列を示す。
配列番号 3で示される塩基配列を有する c DNAの作製は、 以下の方法に 従って行われる。
はじめに酵母 S S T法 (米国特許 No.5, 536, 637に記載) の概要について説 明する。
サッカロマイセス 'セレヒシェ (Saccharomyces cerevisiae) などの酵母 がショ糖またはラフイノースをエネルギー源や炭素源として利用するために はィンベルターゼを培地中に分泌しなければならない (ィンベル夕一ゼはラ フィノースをショ糖とメリビオースに、 ショ糖をフルク 1 ^一スとグルコース に分解する酵素である。 ) 。 また数多くの既知の哺乳類のシグナルペプチド は酵母のィンベル夕一ゼを分泌させ得ることが知られている。 これらの知見 から、 酵母のインベルタ一ゼの分泌を可能にする新規のシグナルペプチドを 哺乳類の c DNAライブラリーからラフイノース培地上での酵母の生育を指 標にスクリーニングする方法として本方法は開発された。 翻訳開始点 ATG を除した非分泌型のインベル夕一ゼ遺伝子 S U C 2 (GENBANK accession No. V01311)を酵母の発現ベクター (発現用プロモーター (ADHプロモー ター) および夕一ミネ一ター (ADH夕一ミネ一夕一) は AAH5プラスミ ド (Gammerer, Methods in Enzyraol. χοχ, 192-201, 1983) 由来で、 酵母複製 起点は 2m o r i、 酵母選択マーカ一には TRP 1、 大腸菌複製起点は C o 1 E 1 o r i、 大腸菌 薬剤耐性マ一力一にはアンピシリンが使用されてい る。 ) に組み込んで酵母 S S T用べクタ一 p SUC 2を作製した。 その S UC 2遺伝子の上流に哺乳類の c DNAを組 み込んで、 酵母 S S T c DNAライブラリ一を調製した。 このライブラリ一を分泌型インベル夕一 ゼを欠損している酵母に形質転換した。 組み込まれた哺乳類 c DNAがシグ ナルぺプチドをコ一ドしている場合、 酵母で発現されたィンベルタ一ゼに対 しても分泌作用をもっと考えられ、 その結果ラフイノ一ス培地上での生育が 可能となる。 よって出現したコロニーから酵母を培養してプラスミドを調製 し、 インサート c DN Aの塩基配列を決定することによって、 新規なシグナ ルペプチドの検索を迅速かつ容易にした。
酵母 SST c DNAライブラリーの作製は、 - (1) 対象となる細胞より mRNAを単離し、 特定の制限酵素 (酵素 I) サ ィトを連結したランダムプライマーを用いて二本鎖 c DNAを合成し、
(2) 酵素 Iとは異なる特定の制限酵素 (酵素 II) サイトを含むアダプター を連結して、 酵素 Iで消化した後、 適当なサイズで分画し、
(3) 酵母発現べクタ一内のシグナルペプチドを削除したィンベルターゼ遺 伝子の上流に得られた c D N A断片を連結し、 形質転換する工程よりなる。 各工程を詳しく説明すると、 工程 (1) では、 対象となる哺乳類の臓器や 細胞株などより、 必要により適当な刺激剤で刺激した後、 公知の方法 (以下、 公知の方法は特に記載がなければ Molecular Cloning (Sambrook, J., Fr i tsch, E. F. および Maniat is, T. 著、 Cold Spring Harbor Laboratory Pressより 1989年に発刊) または Current Protocol in Molecular Biology (F.M. Ausubelら編、 〗ohn Wiley&Sons, Incより発刊) に記載の方法に従つ て行われる。 ) に従って mRN Aの単離が行われる。
対象となる細胞としては、 T 98 G (ヒトグリア芽細胞腫株: ATCC No. CRL-1690) 、 I MR— 32 (ヒト神経芽細胞腫株: ATCC No. CCL-127) 、 U - 2 OS (ヒト骨肉腫株: ATCC No. HTB-96) 、 CCF-STTG 1 (ヒトァ ストロ細胞 腫株: ATCC No. CRL-1718) 、 HAS 30 3 (ヒ卜骨髄ストロー マ細胞株:東京 医科大学第一内科外山圭助教授、 相沢信助手より供与。 J. Cell. Physiol. H8, 245-251, 1991および Experimental Hematol. 22, 482-487, 1994に記載) 、 LP 10 1 (ヒト骨髄ストローマ細胞株:東京医 科大学第一内科外山圭助教授、 相沢信助手より供与。 〗. Cell. Physiol. 148, 245-251, 1991および Experimental Hematol. 22, 482-487, 1994 に記載) ま たは HUV— EC— C (ヒ卜臍帯静脈血管内皮細胞: ATCC No. CRL-1730) が 挙げられる。 また組織としては、 ヒト胎盤およびヒト成人脳が挙げられる。 ランダムプライマ一を用いる二本鎖 c DN Aの合成は公知の方法により行わ れる。
アダプターに連結される制限酵素 (酵素 I) サイトと次の工程 (2) で用 いられる制限酵素 (酵素 II) サイトは、 互いに異なるものであれば何を用い てもよい。 好ましくは、 酵素 Iとして Xh o I、 酵素 IIとしては E c 0 R I が用いられる。
工程 (2) では T 4 DN Aポリメラーゼで末端を平滑化し、 酵素 Πァダプ 夕一を連結した後、 酵素 Iで消化し、 ァガロース電気泳動 (AGE) により 300〜800 b pの c DNAを分画する。 酵素 IIは、 前記したように酵素 Iと異なるものなら何でもよい。 工程 (3) は、 酵母発現用プラスミドベクターに連結されたシグナルぺプ チドを削除したインベル夕一ゼの遺伝子の上流に (2) で得られた c DNA 断片を組み込んで大腸菌に形質転換する工程である。 ここで酵母発現用ブラ スミドベクターとしては種々のものが知られているが、 例えば、 大腸菌内で も機能する YE p 24などが用いられるが、 好適には前述したプラスミド p SUC 2が用いられる。
形質転換のための宿主大腸菌株はすでに多くのものが知られており、 好ま しくは DH 1 0 Bのコンビテントセルである。 また形質転換方法は公知のい ずれを用いてもよいが、 好ましくはエレクトロポレーション法により行われ る。 形質転換体は常法により培養され、 酵母 S S T用の c DNAライブラ リーが得られる。
この c DNAライブラリ一は、 すべてのクローンが前記の c DNA断片を 含んでいるわけではないし、 またすべてが未知の (新規の) シグナルべプチ ドをコードする遺伝子断片とは限らない。 そこで、 次に前記ライブラリ一か ら未知のシグナルペプチドをコードする遺伝子断片をスクリーニングする必 要がある。
すなわち、 c DNAライブラリ一をィンベル夕ーゼ遺伝子をもたない酵母 Saccharomyces cerevisiae (例えば Y T 455株など) またはインベル夕一 ゼ遺伝子を人為的に欠損させた株 (公知の方法に従い作製可能) を用いるこ とができる。 酵母の形質転換は公知の方法、 例えば酢酸リチウム法によって 行われる。 形質転換体を選択培地で生育後、 ラフイノ一スを炭素源とする培 地に移し、 生育可能なコロニ一を選択し、 プラスミドを回収する。 ラフイノ —スを炭素源として酵母が生育したということは、 ライブラリ一中に何らか の分泌蛋白質のシグナルぺプチドが組み込まれていたことを示している。 次に、 単離した陽性クローンについて、 塩基配列を決定し、 未知の蛋白質 をコードすることが明らかになった c DNAについては、 それをプローブと して全長クローンを単離し、 全長の塩基配列を決定することができる。 これ らの操作は、 当業者にとってすベて公知の方法で行われる。 配列番号 2、 5、 8、 1 1、 14、 17、 20、 23、 26、 29、 32、 35、 38、 41、 44、 47、 50、 53、 56、 59、 62、 65、 68、 7 1、 74、 77、 80、 3、 6、 9、 12、 1 5、 1 8、 2 1、 24、 27、 30、 33、 36、 39、 42、 45、 48、 5 1、 54、 57、 60、 63、 66、 69、 72、 75、 78または 81で示される塩 基配列が、 一部、 好ましくは全てが確定されると哺乳類に存在する本発明の 蛋白質をコードする c DNAもしくは本発明蛋白質のホモローグおよびサブ セットをコ一ドする c DN Aを得ることができる。 適当な塩基配列を有する オリゴヌクレオチドを合成し、 それを用いて、 哺乳類由来の c DN Aライブ ラリーあるいは mRNAから P CR法により、 あるいは適当な塩基配列の断 片をプローブとしてハイブリダィズさせることにより、 他の哺乳類 c DNA ライブラリーあるいはそのゲノムライブラリ一から、 他の哺乳類型の本発明 蛋白質をコードする c DNAを得ることができる。
このようにして得られた c DNAが、 S STで得られた c DNA断片の塩 基配列 (またはその相同配列) を含んでいるならばシグナルペプチドをコー ドしていることになるので、 その cDNAが全長、 またはほぼ全長であるこ とは明らかである (シダナルぺプチドは例外なく蛋白質の N末端に存在する ことから、 cDNAのオープンリーディングフレームの 5 ' 末端にコードさ れている。 ) 。
さらに公知の方法に従い、 前記 c D NAをプローブとしてノザン (Northern)解析によつて全長の確認をしてもよい。 ハイプリダイズしたバン ドから得られる mRNAのサイズとその c DN Aのサイズを比較し、 ほぼ同 じであればその c DN Aはほぼ全長であると考えられる。
配列番号 2、 5、 8、 1 1、 14、 17、 20、 23、 26、 29、 32、 35、 38、 41、 44、 47、 50、 53、 56、 59、 62、 65、 68、 7 1、 74、 77、 80、 3、 6、 9、 12、 1 5、 1 8、 2 1、 24、 27、 30、 33、 36、 39、 42、 45、 48、 5 1、 54、 57、 60、 63、 66、 69、 72、 75、 78または 81で示される塩 基配列が一旦確定されると、 その後は、 化学合成によって、 あるいはこれら の塩基配列の断片を化学合成し、 これをプローブとしてハイブリダィズさせ ることにより、 本発明の c DNAを得ることができる。 さらに、 本 cDNA を含有するベクター cDN Aを適当な宿主に導入し、 これを増殖させること によって、 目的とする cDN Aを必要量得ることができる。
本発明のポリぺプチドを取得する方法としては、
(1) 生体または培養細胞から精製単離する方法、
(2) ペプチド合成する方法、 または
(3) 遺伝子組み換え技術を用いて生産する方法、
などが挙げられるが、 工業的には (3) に記載した方法が好ましい。
遺伝子組み換え技術を用いてポリペプチドを生産するための発現系 (宿主 一ベクター系) としては、 例えば、 細菌、 酵母、 昆虫細胞および哺乳動物細 胞の発現系が挙げられる。
例えば、 大腸菌で発現させる場合には、 成熟蛋白質部分をコードする cDNAの 5' 末端に開始コドン (ATG)を付加し、 得られた cDNAを、 適当なプロモータ一 (例えば、 t r pプロモーター、 l a cプロモーター、 λ PLプロモーター、 T 7プロモー夕一等) の下流に接続し、 大腸菌内で機 能するべクタ一 (例えば、 pBR322、 pUC 18、 pUC 19等) に挿 入して発現ベクターを作製する。
次に、 この発現ベクターで形質転換した大腸菌 (例えば、 E. Coli DH1、 E. Coli JM109、 E. Coli HB 101株等) を適当な培地で培養して、 その菌体より目的とするポリペプチドを得ることができる。 また、 パクテリ ァのシグナルペプチド (例えば、 p e 1 Bのシグナルペプチド) を利用すれ ば、 ペリブラズム中に目的とするポリペプチドを分泌することもできる。 さ らに、 他のポリペプチドとのフュージョン ·プロテイン (fusion protein) を生産することもできる。
また、 哺乳動物細胞で発現させる場合には、 例えば、 配列番号 3、 6、 9、 12、 1 5、 1 8、 2 1、 24、 27、 30、 33、 36、 39、 42、 45、 48、 5 1、 54、 57、 60、 63、 66、 69、 72、 75、 78または 81で示される塩基配列をコードする c DNAを適当なベクター (例えば、 レトロウイルスベクター、 パピローマウィルスベクター、 ヮクシ ニァウィルスベクター、 SV40系べクタ一等) 中の適当なプロモーター (例えば、 SV40プロモーター、 LTRプロモー夕一、 メタ口チォネイン プロモーター等) の下流に挿入して発現ベクターを作製する。 次に、 得られ た発現ベクターで適当な哺乳動物細胞 (例えば、 サル COS _ 7細胞、 チヤ ィニーズハムスター CHO細胞、 マウス L細胞等) を形質転換し、 形質転換 体を適当な培地で培養することによって、 本発明の蛋白質が分泌蛋白質の場 合と膜蛋白質の場合で、 次のように発現される。
本発明の蛋白質が分泌蛋白質の場合、 その細胞上清中に目的とするポリべ プチドが発現される。 さらに、 その他のポリペプチド、 例えば抗体の定常領 域 (Fc portion) をコードする c DNA断片と連結することによって、 フユ一 ジョン,プロテイン (fusion protein) を生産することもできる。
一方、 本発明の蛋白質が膜蛋白質の場合、 その細胞膜上に目的とするボリ ペプチドが発現される。 また配列番号 3、 6、 9、 12、 15、 18、 21、 24、 27、 30、 33、 36、 39、 42、 45、 48、 5 1、 54、 57、 60、 63、 66、 69、 72、 75、 78または 81で示される塩 基配列をコードする c DNAの膜貫通領域を欠いた欠失体を前記べクタ一に 挿入し、 これを用いて適当な哺乳類動物細胞を形質転換することによって、 その培養液中に目的とする可溶性ポリペプチドが分泌される。 さらにその膜 貫通領域を欠いた欠失体をコードする c DN A断片とその他のポリぺプチド、 例えば抗体の定常領域 (Fc portion) をコードする c D N A断片を連結するこ とによって、 フユ一ジョン ' プロテイン (fusion protein) を生産すること もできる。
以上のようにして得られたポリペプチドは、 一般的な生化学的方法によつ て単離精製することができる。 産業上の利用可能性
本発明のポリペプチドおよびそれをコードする c DN Aは、 一つあるいは それ以上の効果あるいは生物活性 (以下に列挙するアツセィに関連するもの を含む。 ) を示すことが考えられる。 本発明の蛋白質に関して記述される効 果あるいは生物活性は、 その蛋白質の投与あるいは使用により、 あるいは、 その蛋白質をコードする cDN Aの投与あるいは使用 (例えば、 遺伝子療法 や cDNA導入に適したベクター) により、 提供される。
[サイトカイン活性および細胞増殖 Z分化活性]
本発明の蛋白質は、 サイ ト力イン活性および細胞増殖 (誘導あるいは阻 害) Z分化活性 (誘導あるいは阻害) を示す可能性、 あるいはある細胞集団 に他のサイ トカインの産生を誘導あるいは抑制すると考えられる。 全ての既 知のサイト力インを含む、 現在発見されている多くの蛋白性因子は、 因子に 依存した一つあるいはそれ以上の細胞増殖アツセィ法で、 活性を示してきた ので、 それらのアツセィは、 サイト力イン活性の便利な確認法として機能す る。 本発明の蛋白質の活性は、 多くの従来の因子依存性の細胞株の細胞増殖 アツセィのうちのいずれかによつて証明され得る。
[免疫刺激ノ抑制活性]
本発明の蛋白質は、 免疫刺激活性および免疫抑制活性を示すと考えられる。 また、 ある蛋白質は、 例えば、 Tリンパ球および Bリンパ球あるいはどちら か一方の成長および増殖を制御 (刺激あるいは抑制) することや、 同様に N K細胞や他の集団の細胞傷害性活性に影響を与えることによって、 様々な免 疫不全および疾患 (severe combined immunodeficiency (S C I D) を含 む) の治療に効果を示すと考えられる。 これらの免疫不全は遺伝性である場 合もあるし、 例えば、 H I Vのようなウィルスや、 同様に細菌やカビの感染 が原因で起こる場合もある。 あるいは、 自己免疫疾患から由来する可能性も ある。 より特殊な場合に、 H I V、 肝炎ウィルス (hepatitis viruses), へ リレぺスゥイリレス (herpes viruses) 、 マイコノヾクテリア (mycobacteria) 、 リ一シュマニア ( leishmania) 、 マラリア (malaria) およびカンジダ (Cand ida) のような様々なカビ感染を含むウィルス、 細菌、 カビあるいは他 の感染による感染症の原因を、 本発明の蛋白質を用いることによって治療で きると考えられる。 もちろん、 この関連より、 本発明の蛋白質は、 免疫シス テムが増大していることが一般的に示唆される場所、 すなわち癌治療の箇所 において効果を示すと考えられる。
本発明の蛋白質は、 アレルギー反応および喘息や他の呼吸器系疾患のよう な状況の治療に効果にも効果を示すと考えられる。 免疫抑制が望まれるよう な、 例えば、 喘息や関連呼吸器疾患を含む他の状態にも、 本発明の蛋白質を 用いて治療できると考えられる。
本発明の蛋白質は、 例えば、 敗血病性のショックあるいは全身性炎症反応 症候群 (S I R S ) のような感染、 炎症性大腸炎、 クローン病に関連する、 あるいは、 I L _ 1 1により効果が証明された T N Fや I L一 1のようなサ ィ卜カインの過剰産生から由来する慢性あるいは急性の炎症を抑制する可能 性もある。
[造血細胞制御活性]
本発明の蛋白質は、 造血細胞の制御に、 またそれに応じて骨髄球様細胞あ るいはリンパ球様細胞の欠乏に対する治療にも効果を示すと考えられる。 コ ロニー形成細胞あるいは因子依存性細胞株の援助の下での極く弱い生物活性 でさえも、 造血細胞の制御に係わることを示唆する。 その生物活性とは、 次 に挙げる全てあるいはそのいずれかで例えられるようなものに係わるもので ある。 赤血球前駆細胞のみの成長および増殖を支持、 あるいは他のサイトカ インとの組み合わせ、 また、 それが示唆する有効性、 例えば様々な貧血の治 療、 あるいは赤血球前駆細胞および赤血球あるいはそのどちらかの産生を刺 激する放射線療法/化学療法と組み合わせての使用;顆粒球および単球 Zマ クロファ一ジのような骨髄球の成長および増殖を支持 (すなわち、 古典的な C S F活性) 、 化学療法に伴う骨髄抑制を防ぐための化学療法との併用;巨 核球の成長および増殖およびそれに続く血小板の成長および増殖の支持、 そ れによって血小板減少症のような様々な血小板障害を防御および治療を可能 とする血小板輸血の際あるいは相補的な一般的使用;前記造血細胞の幾つか あるいは全ての細胞へ成熟可能な造血幹細胞の成長および増殖の支持、 従つ て、 様々な幹細胞障害 (限定はされないが、 再生不良性貧血および発作性夜 間血色素尿症を含む、 移植で一般的に治療されるようなもの) に治療的効果 を見い出せる、 また、 正常細胞あるいは遺伝子療法のため遺伝的に操作され た細胞をイン · ビトロ (in vi t ro) あるいはェキソ · ビポ (ex vivo) (すな わち、 骨髄移植に伴う) どちらかで、 放射線療法ノ化学 療法後の幹細胞分画 の再構築を行うことも同様である。

[組織生成 Z修復活性]
本発明の蛋白質は、 損傷治癒および組織修復、 また、 火傷、 切開、 および 潰瘍の治療と同様に、 骨、 軟骨、 腱、 靭帯、 および神経組織成長あるいは再 生のいずれかに使用されると考えられる。
骨を正常に形成しない環境での軟骨および骨あるいはいずれかの成長を誘 導するような本発明の蛋白質は、 ヒトおよび他の動物の骨折および軟骨損傷 あるいは欠損の治癒に適用される。 また、 本発明の蛋白質を使用する製剤は、 開放骨折と同様に閉鎖骨折の整復、 また人工関節の固定の改良や、 予防的使 用にも有効であると考えられる。 骨形成剤により誘導された新生骨形成は、 先天性、 外傷性、 癌切除術により誘発した頭蓋顔面の欠損の修復に貢献する。 また、 美容形成外科分野にも有効である。
本発明の蛋白質は、 歯根膜症の治療および他の歯の修復にも使用されると 考えられる。 そのような薬品は、 骨形成細胞を引き寄せ、 その細胞の増殖を 刺激し、 その前駆細胞の分化を誘導する環境を提供すると考えられる。 本発 明の蛋白質は、 骨および軟骨あるいはいずれかの修復を刺激することを通し て、 あるいは炎症あるいは炎症過程で介される組織破壊 (コラゲナーゼ活性 や破骨細胞の活性) の過程を阻止することにより、 骨粗鬆症および骨関節炎 の治療に有効であると考えられる。 本発明の蛋白質に起因すると考えられる組織再生活性の別のカテゴリ一は 腱 靭帯形成である。 本発明の蛋白質は、 腱/靭帯様組織あるいは他の組織 が正常に形成されない環境でそのような組織形成を誘導するものであるが、 ヒトおよび他の動物における腱/靭帯の裂傷、 奇形、 および他の腱 靭帯の 障害の治癒に適用できる。 腱 靭帯様組織を誘導する蛋白質を使用する製剤 は、 骨あるいは他の組織への腱/靭帯の固定の改良、 および腱 Z靭帯組織の 欠損の修復での使用はもちろん、 腱あるいは靭帯の損傷の防御に対する予防 的使用も考えられる。 本発明の構成物により誘導された新生腱 靭帯様組織 形成は、 先天性、 外傷、 あるいは他の起源の腱あるいは靭帯欠損の修復に貢 献する。 また、 腱あるいは靭帯の貼付あるいは修復という美容形成外科でも 有効である。 本発明の構成物は、 腱 靭帯形成細胞を引き寄せ、 その細胞の 増殖を刺激し、 その前駆細胞の分化を誘導する環境を提供すると考えられる。 あるいは、 組織修復を果たすためイン · ビポ (in v ivo) への返還に備えてェ キソ · ビポ (ex v ivo) で腱/靭帯細胞あるいはその前駆細胞を誘導する。 本 発明の構成物は、 腱炎、 手根トンネル症候群 (Carpal tunne l syndrome) 、 および他の腱あるいは靭帯欠損の治療にも有効である。 本発明の構成物には、 適当なマトリックスおよびキャリアーと同様に当業者に良く知られている錯 化 (Seques ter ing) 剤も含まれる。
本発明の蛋白質は、 神経細胞の増殖、 および、 神経および脳組織の再生、 すなわち、 神経細胞あるいは神経組織の変性、 死、 あるいは外傷を含む機械 的および外傷的障害と同様に中枢および末梢神経系疾患および神経病の治療 に対しても効果を示すと考えられる。 より特異的には、 ある蛋白質は、 末梢 神経障害、 末梢神経症、 および局所的神経症のような末梢神経系の疾患、 お よびアルツハイマー病、 パーキンソン病、 ハンチントン病、 筋萎縮性側策症 (amyo t rophic l ateral) , およびシャイ一ドレ一ガー (Shy- Drager) 症候群 のような中枢神経系の疾患の治療に有効であると考えられる。 更に本発明に 応じて治療され得る条件には、 脊髄障害、 頭部外傷、 および脳卒中等の脳血 管疾患のような機械的および外傷的障害を含む。 化学療法あるいは他の治療 力 起因する末梢神経症も本発明の蛋白質を用いて治療可能である。
本発明の蛋白質は、 例えば、 塍臓、 肝臓、 腸、 腎臓、 皮膚、 内皮を含む臓 器、 平滑、 骨格あるいは心臓筋肉、 および血管内皮を含む血管組織のような 他の組織を生成する活性、 あるいはそのような組織を構成する細胞の増殖を 促進する活性を示す可能性も期待される。 望まれる効果の一部は、 正常組織 を再生させる繊維性瘢痕 (scarr ing) の阻害によっても担われると考えられ る。
本発明の蛋白質は、 消化管保護あるいは再生、 および肺あるいは肝臓の繊 維化、 様々な組織の再還流損傷、 および全身性サイト力イン障害に起因する 状態に対する治療にも有効であると考えられる。
[ァクチビン Zインヒビン活性]
本発明の蛋白質は、 ァクチビン/インヒビンに関連した活性を示すと考え られる。 ァクチビンは濾胞刺激ホルモン (F S H) の放出を刺激する活性に よって特徵づけられるが、 インヒビンは、 濾胞刺激ホルモン (F S H) の放 出を阻害する活性によって特徴づけられる。 よって、 本発明の蛋白質は、 単 独あるいはインヒビン aフアミリーのメンバーとのヘテロダイマーで、 哺乳 類動物の雌の受精率を減少させ、 雄の精子形成を減少させるインヒビンの活 性に基づく避妊調節剤として有効であると考えられる。 充分量の他のインヒ ビンの投与によって、 哺乳動物の不妊を誘導可能である。 一方、 本発明の蛋 白質は、 インヒビン bグループの他の蛋白質サブユニットとのホモダイマ一 あるいはヘテロダイマーで、 前脳下垂体の細胞から F S Hの放出を刺激する ァクチビン分子の活性に基づいた治療的な不妊誘導として有効であると考え られる (米国特許 4, 798, 885を参照) 。 本発明の 蛋白質は、 牛、 羊、 および 豚のような家畜の生涯出産能力可能な期間を延ばすために、 性的に未熟な哺 乳類動物における妊娠開始を早めることに有効であると考えられる。
[走化性/化学運動性活性]
本発明の蛋白質は、 例えば、 単球、 好中球、 T細胞、 マスト細胞、 好酸球、 および内皮細胞、 あるいはそのいずれかを含む、 哺乳動物の細胞に対して、 例えばケモカインとして働く走化性 z化学運動性活性を有すると考えられる。 走化性 Z化学運動性蛋白質は、 反応の望まれる部位へ、 望まれる細胞集団を 固定化あるいは引き寄せるため使用されることが可能である。 走化性 Z化学 運動性蛋白質は、 局所的な感染と同様に、 創傷および他の外傷の治療に特別 な優位性を提供する。 例えば、 リンパ球、 単球、 あるいは好中球を腫瘍ある いは感染部位へ引き寄せることは、 腫瘍あるいは感染部位に対する免疫応答 を改善する結果となると考えられる。
蛋白質やペプチドは、 もしそれが直接あるいは間接的に特殊な細胞集団に 対して指示された方向あるいは運動を刺激可能であれば、 そのような細胞集 団に対する走化性活性を保持している。 望ましくは、 その蛋白質やペプチド は、 細胞の指示された運動を直接的に刺激する活性を保持する。 特別な蛋白 質がある集団の細胞に対し走化性活性を保持するか否かは、 どんな既知の細 胞走化性のアツセィ法にそのような蛋白質あるいはペプチドを使用しても容 易に決定できる。
[凝血および血栓活性]
本発明の蛋白質は、 凝血あるいは血栓活性も示すと考えられる。 結果とし て、 そのような蛋白質は、 様々な凝固障害 (血友病のような遺伝性障害を含 む。 ) の治療に有効であると期待される。 あるいは、 外傷、 手術または他の 原因により生じた創傷の治療における凝固および他の凝血事象を促進させる ことが期待される。 本発明の蛋白質は、 血栓の形成の溶解あるいは阻害 (血 栓あるいは卒中等) 、 およびそれより生じる状態の治療および予防にも効果 があると考えられる。
[受容体 Zリガンド活性]
本発明の蛋白質は、 受容体、 受容体 リガンドあるいは受容体 リガンド のインヒビ夕一あるいはァゴニストとしての活性を示す可能性もある。 その ような受容体およびリガンドの例として、 サイトカイン受容体およびそのリ ガンド、 受容体キナーゼおよびそのリガンド、 受容体フォスファタ一ゼおよ びそのリガンド、 細胞間相互作用に関連した受容体 (Sel ec t in, Integur in, およびそのリガンド 、 受容体キナーゼ等の細胞接着分子を含む。 ) およびそ のリガンド、 および抗原提示、 抗原認識、 および細胞性および液性免疫反応 の発達に係わる受容体 Zリガンドの組み合わせが挙げられるが、 これらに制 限されるものではない。 受容体およびリガンドは、 その相互作用に対する可 能なぺプチドあるいは小分子のインヒビ夕一のスクリーニングにも有効であ る。 本発明の蛋白質は、 それ自身受容体 リガンド (受容体およびリガンド の断片を含むが、 制限されるものではない。 ) の相互作用のインヒビ夕一と して有効であると考えられる。
[その他の活性]
本発明の蛋白質は、 以下に示す付加的な活性あるいは効果の一つあるいは それ以上を示すと考えられる :細菌、 ウィルス、 カビ、 および他の寄生虫を 含む感染性の物質を殺傷する;身長、 体重、 髪の色、 目の色、 肌あるいは他 の組織の色素沈着、 あるいは器官の大きさ (例えば、 胸部増量あるいは減 量) 等の身体的特徴を抑制あるいは促進する効果を及ぼす;食餌脂肪、 蛋白 質、 あるいは炭水化物の分解に効果を及ぼす;食欲、 性欲、 ストレス、 認識 (認識障害) 、 鬱病、 暴力行動を含む行動特徴に効果を及ぼす;鎮痛効果あ るいは他の痛みを減少させる効果を及ぼす;胚性幹細胞の造血系以外の他の 系統への分化および増殖を促進する ;および、 酵素の場合、 その酵素の欠失 を補う、 また関連疾患を治療する。
前記活性を有する蛋白質は、 例えば、 B細胞、 T細胞、 肥満細胞の増殖ま たは細胞死、 免疫グロブリンのクラススイツチ促進によるクラス特異的誘導、 B細胞の抗体産生細胞への分化、 顆粒球前駆細胞の増殖または分化、 細胞死、 単球 ·マクロファージ前駆細胞の増殖または分化、 細胞死、 好中球、 単球, マクロファージ、 好酸球、 好塩基球の増殖または機能亢進、 細胞死、 巨核球 前駆細胞の増殖または細胞死、 好中球前駆細胞の増殖または分化、 細胞死、 Bまたは T前駆細胞の増殖または分化、 細胞死、 赤血球の産生促進、 赤血球、 好中球、 好酸球、 好塩基球、 単球 ·マクロファージ、 肥満細胞、 巨核球前駆 細胞の増殖支持、 好中球、 単球 ·マクロファージ、 B細胞または T細胞の遊 走促進、 胸腺細胞の増殖または細胞死、 脂肪細胞の分化抑制、 ナチュラルキ ラー細胞の増殖または細胞死、 造血幹細胞の増殖または細胞死、 幹細胞およ び各種造血前駆細胞の増殖抑制、 間葉系幹細胞からの骨芽細胞、 軟骨細胞へ の分化促進または増殖、 細胞死、 あるいは破骨細胞の活性化や単球から破骨 細胞への分化促進による骨吸収の促進の作用を本発明のポリペプチドのみで、 またリガンドーレセプ夕一間の結合を介して、 あるいは他の分子と相乗的に 働くことにより有すると考えられる。
また本発明のペプチドは神経系にも作用することが予測されるので、 各種 神経伝達物質作動性神経細胞への分化ならびにそれらの生存維持または細胞 死、 グリア細胞の増殖促進または細胞死、 神経突起の伸展、 神経節細胞の生 存維持または細胞死、 ァストロサイトの増殖または分化促進または細胞死、 末梢神経の増殖または生存維持、 細胞死、 シュワン細胞の増殖または細胞死、 運動神経の増殖または生存維持、 細胞死の作用もあると考えられる。
さらに、 本発明のポリペプチドは初期胚の発生過程において、 外胚葉誘導 作用による表皮、 脳、 背骨、 神経の器官形成、 中胚葉誘導作用による背索結 合組織 (骨、 筋肉、 腱) 、 血球細胞、 心臓、 腎臓、 生殖巣の器官形成、 ある いは内胚葉誘導作用による消化器系臓器 (胃、 腸、 肝臓、 塍臓) 、 呼吸器系
(肺、 気管) の形成に促進的または抑制的に作用すると考えられるとともに、 生体においても前記器官の増殖あるいは増殖抑制作用を有すると考えられる。 したがって、 本発明のポリペプチドはそれ自身で、 免疫系または神経系も しくは骨代謝の機能の低下または亢進に関する疾患、 または造血系細胞の発 育不全または異常増殖、 例えば、 炎症性疾患 (リウマチ、 潰瘍性大腸炎等) 、 骨髄移植後の造血幹細胞の減少症、 ガン、 白血病に対する放射線照射または 化学療法剤投与後の白血球、 血小板、 B細胞または T細胞の減少症、 貧血、 感染症、 ガン、 白血病、 A I D S、 骨代謝異常 (骨粗鬆症等) 、 各種変性疾 患 (アルツハイマー病、 多発性硬化症等) 、 あるいは神経損傷の予防または 治療薬として用いることが期待される。
また本発明のポリペプチドは、 外胚葉、 中胚葉または内胚葉由来器官の分 化または増殖作用を有すると考えられるので、 各器官 (表皮、 骨、 筋肉、 腱、 心臓、 腎臓、 胃、 腸、 肝臓、 滕臓、 肺、 気管等) の組織修復剤として用いる ことも期待される。
また、 本発明のポリペプチドのポリクロ一ナル抗体またはモノクローナル 抗体を用いて、 生体における本発明のポリペプチドの定量が行なえ、 これに よつて本ポリぺプチドと疾患との関係の研究あるいは疾患の診断等に利用す ることができる。 ポリクロ一ナル抗体およびモノクローナル抗体は本発明の ポリペプチドあるいはその断片を抗原として用いて常法により作製すること ができる。
また本発明のポリペプチド (好ましくは、 その細胞外ドメインのポリぺプ チド) を用いることにより、 例えばァフィ二ティーカラムを作製して、 本発 明のポリペプチドと結合する既知または未知の蛋白質 (リガンド) の同定、 精製あるいはその遺伝子クロ一ニングを行うことができる。
また本発明のポリペプチド (好ましくは、 その膜貫通領域または細胞内ド メインのポリペプチド) を用いて、 例えばウェスト—ウエスタン法により、 または前記 c D N A (好ましくは、 本発明ポリペプチドの膜貫通領域または 細胞内ドメインをコードする c D N A) を用いて、 例えば酵母 2—ハイブ リッド法により本発明のポリペプチドと細胞質内で相互作用する下流のシグ ナル伝達分子の同定、 遺伝子クロ一ニングを行うこともできる。
さらに本発明のポリペプチドを用いることによって、 本発明のポリべプチ ドレセプ夕一ァゴニスト、 アンタゴニストおよび受容体一シグナル伝達分子 間の阻害剤等のスクリーニングを行うこともできる。
本発明の c D N Aは、 多大な有用性が期待される本発明のポリペプチドを 生産する際の重要かつ必須の铸型となるだけでなく、 遺伝病の診断や治療 (遺伝子欠損症の治療またはアンチセンス D N A ( R N A) によって、 ポリ ペプチドの発現を停止させることによる治療等) に利用できる。 また、 本発 明の c D N Aをプローブとしてジエノミック (genomic) D NAを分離できる。 同様にして、 本発明 c D N Aと相同性の高いヒトの関連ポリペプチドの遺伝 子、 またマウス以外の生物における本発明ポリペプチドと相同性の高いポリ ぺプチドの遺伝子を分離することも可能である。
[医薬品への適用]
前記の疾患に適応するために、 本発明のポリペプチド、 あるいは本発明の ポリペプチドに対する抗体は通常、 全身的又は局所的に、 一般的には経口ま たは非経口の形で投与される。 好ましくは、 経口投与、 静脈内投与および脳 室内投与である。
投与量は、 年齢、 体重、 症状、 治療効果、 投与方法、 処理時間等により異 なるが、 通常、 成人一人あたり、 一回につき、 1 0 0 8から 1 0 0 111 8の 範囲で、 一日一回から数回経口投与されるか、 または成人一人あたり、 一回 にっき、 1 0 gから 1 0 O m gの範囲で、 一日一回から数回非経口投与さ れる。
もちろん前記したように、 投与量は、 種々の条件により変動するので、 前 記投与量より少ない量で十分な場合もあるし、 また範囲を越えて必要な場合 fcある。
本発明化合物を投与する際には、 経口投与のための固体組成物、 液体組成 物およびその他の組成物、 非経口投与のための注射剤、 外用剤、 坐剤等とし て用いられる。
経口投与のための固体組成物には、 錠剤、 丸剤、 カプセル剤、 散剤、 顆粒 剤等が含まれる。 カプセルには、 ソフトカプセルおよびハードカプセルが含 まれる。
このような固体組成物においては、 一つまたはそれ以上の活性物質が、 少 なくとも一つの不活性な希釈剤 (例えば、 ラクト一ス、 マンニトール、 ダル コース、 ヒドロキシプロピルセルロース、 微結晶セルロース、 デンプン、 ポ リビニルピロリドン、 メタケイ酸アルミン酸マグネシウム等) と混合される。 組成物は、 常法に従って、 不活性な希釈剤以外の添加物、 例えば、 潤滑剤 (ステアリン酸マグネシウム等) 、 崩壊剤 (繊維素グリコール酸カルシウム 等) 、 安定化剤 (ヒト血清アルブミン、 ラクトース等) 、 溶解補助剤 (アル ギニン、 ァスパラギン酸等) を含有していてもよい。
錠剤または丸剤は、 必要により白糖、 ゼラチン、 ヒドロキシプロピルセル ロース、 ヒドロキシプロピルメチルセルロースフタレ一ト等の胃溶性あるい は腸溶性のフィルムで被膜してもよいし、 また 2以上の層で被膜してもよい。 さらにゼラチンのような吸収されうる物質のカプセルも包含される。
経口投与のための液体組成物は、 薬学的に許容される乳濁剤、 溶液剤、 懸 濁剤、 シロップ剤、 エリキシル剤等を含み、 一般に用いられる不活性な希釈 剤 (例えば、 精製水、 エタノール等) を含んでいてもよい。 この様な組成物 は、 不活性な希釈剤以外に湿潤剤、 懸濁剤のような補助剤、 甘味剤、 風味剤、 芳香剤、 防腐剤を含有していてもよい。
経口投与のためのその他の組成物としては、 ひとつまたはそれ以上の活性 物質を含み、 それ自体公知の方法により処方されるスプレー剤が含まれる。 この組成物は不活性な希釈剤以外に亜硫酸水素ナトリゥムのような安定剤と 等張性を与えるような安定化剤、 塩化ナトリウム、 クェン酸ナトリウムある いはクェン酸のような等張剤を含有していてもよい。 スプレー剤の製造方法 は、 例えば米国特許第 2, 868, 691号および同第 3, 095, 355号明細書に詳しく記 載されている。
本発明による非経口投与のための注射剤としては、 無菌の水性または非水 性の溶液剤、 懸濁剤、 乳濁剤を包含する。 水性または非水性の溶液剤、 懸濁 剤としては、 一つまたはそれ以上の活性物質が、 少なくとも一つの不活性な 希釈剤と混合される。 水性の希釈剤としては、 例えば注射用蒸留水および生 理食塩水が挙げられる。 非水性の希釈剤としては、 例えばプロピレングリコ ール、 ポリエチレングリコール、 オリ一ブ油のような植物油、 エタノールの ようなアルコール類、 ポリソルべ一ト 8 0 (登録商標) 等が挙げられる。 このような組成物は、 さらに防腐剤、 湿潤剤、 乳化剤、 分散剤、 安定化剤 (例えば、 ヒト血清アルブミン、 ラクトース等) 、 溶解補助剤 (例えば、 ァ ルギニン、 ァスパラギン酸等) のような補助剤を含んでいてもよい。 発明を実施するための最良の形態
以下に本発明の実施例を挙げて本発明をより具体的に説明するが、 これら は本発明の範囲を制限するものではない。 実施例 1 :クローン ON 056
(1) o 1 y (A) +RNAの調製
ヒト胎盤組織より TR I z o 1試薬 (TRIzol reagent) (登録商標、 GIBC0 BRL社より販売) を用いて全 RNAを抽出し、 mRNAピユリフィケ一シヨン キット (mRNA Purification K ) (商品名、 Pharmacia社より販売) を用いて po l y (A) +RNAを精製した。
(2)酵母 S ST c DNAライブラリ一の調製
前記の p o l y (A) +RN Aを铸型に Xh o I部位を連結したランダム 9
NNNNNNNN- 3 ' (配列番号 82 ) をプライマーとして、 スーパ一 ' スクリプト ·プラスミドシステム (Super Script Plasmid System for cDNA Synthesis and Plasmid Cloning) (商品名、 GIBCO BRL社より販売) を用いて 2本鎖 c DNAの合成を行なった。 Ec oR Iアダプター (GIBCO BRL社より 販売) を DNAライゲーシヨンキット 2 (DNA ligation kit ver.2) (商品名 宝酒造 (株) より販売。 以後 c DNAの連結はすべて本キットを使用し た。 ) を用いて連結した後、 Xh o lで消化し、 ァガロース電気泳動で 300〜800 b pの c DNAを切り出して分画し、 pSUC2 (米国特許 5536637号参照) の E c o R I /N o t I部位に連結し、 大腸菌 DH 10 B株 にエレクトロポレーシヨン法で形質転換して酵母 S S T用の c DNAライブ ラリーを得た。
(3) S STによるスクリーニングおよび S ST陽性クローンの塩基配列の決定 この c DNAライブラリ一のプラスミ ドを調製し、 酢酸リチウム法 (Current Protocols In Molecular Biology 13.7.1を参照) により酵母 YT Kl 2株を形質転換し、 トリブトファン (T r p) を含まない酵母形質転換 体の選択培地 (CMD— T r p培地) のプレート上にまき、 30でで48時 間インキュベートした後、 ァクトラン · レプリカ · プレー夕一 (Accutran Replica Plater) (商品名、 Schleicher&Schuel 土より販売) を用いて得られ たコロニー (形質転換体) のレプリカ をラフイノースを炭素源とする YPR プレートにとり、 30 で 14日間インキュベートした。 3日目以降、 出現 してきた各々のコロニーを一つずつ再度 YP Rプレートにストリークして 30 で 48時間インキュベートした後、 シングルコロニーを YPD培地に 植菌し、 30でで 48時間インキュベートした後、 プラスミドを調製した。 続いて p SUC 2のクローニングサイ卜の両端の配列の 2種類のプライマ一 (センス鎖はピオチン化プライマ一) を用いて公知の方法に従って P CRを 行ない、 インサート c DN Aを増幅した後、 ダイナビーズ (Dynabeads) (商 品名、 DYNAL社より販売) を用いてピオチン化 1本鎖 cDNAを精製し、 塩基 配列の決定を行なった。 塩基配列の決定は DNAシ一ケンシングキット (DNA Sequencing kit) (Dye Terminator Cycle Sequencing Ready Reaction;商品 名、 A卯 lied Biosystems Inc. より販売) を用いた蛍光ダイ夕一ミネ一夕一 サイクルシークェンス法で反応を行ない、 自動 DNAシークェンサ一 373 (Applied Biosystems Inc. ) で読み取りを行なった (以下、 塩基配列決定は すべて本方法で行なった。 ) 。
得られた塩基配列および推定されるアミノ酸配列についてデータベースと の相同性検索を行ない、 データベースに登録されていない新規な c DN Aで あることが明らかとなったクローンについて、 全長 c DN Aのクロ一ニング を試みた。 また推定されるァミノ酸配列を既知のシグナルぺプチドと比較す ることにより各 c D N Aが機能的かつ構造的にもシグナルべプチドを有する ことを確認した。
(4)全長 c DNAのクロ一ニングおよび塩基配列の決定
全長 c DN Aのクロ一ニングはジーントラッパ一 · c DNAポジティブ · セレクションシステム (GENETRAPPER cDNA Positive Selection System) (GIBCO BRL社より販売) を用いて行なった。 まず Super Script Plasmid System for cDNA Synthesis and Plasmid Cloningを用いてヒト胎盤組織の p o l y (A) +RNAよりプラスミド p S PORT 1 (GIBCO BRL社より販 売) をベクターとして dT— primed c DN Aライブラリ一を作製した。 つぎ に S STで得られた塩基配列の情報に基づいて 27me rのピオチン化ブラ イマ一 ON 056— F 1 : 5 ' biot in- A A C AT G A AT C T T T C G C TCGTC CTGGCT— 3, (配列番号 83 ) を作製した後、 ジーントラ ッパー (GeneTrapper) キットの方法にしたがってピオチン化プライマ一 と 特異的にハイブリダィズするプラスミドを前記の c DNAライブラリ一から 回収し、 大腸菌 DH 1 0 Bに形質転換した。 さらにランダムプライマ一 DNAラベリングキット (Random Primer DNA Labeling kit) (商品名、 宝酒 造(株) より販売) を用いて32 P- d CTPでラベルした ON 056 S ST c DNAをプローブとして、 公知の方法によりコロニーハイブリダイゼ一シ ヨンを行ない、 陽性クローンを単離して、 プラスミ ドを調製した。 初めに 5 ' 側の塩基配列を決定して〇N 056 S ST cDNAの塩基配列が存在 することを確認した後、 全塩基配列を決定し、 配列番号 3に示す配列を得た。 さらにオープンリーディングフレームを決定し、 アミノ酸に翻訳して配列番 号 1および 2に示す配列を得た。
核酸配列デ一夕ベースに登録されている既知の核酸配列に対して BLASTNお よび FASTAにより、 またアミノ酸配列データベースに登録されている既知のポ リペプチドのアミノ酸配列に対して BLASTX、 BLASTPおよび FASTAにより検索し た結果、 本発明のポリペプチド〇N 056およびそれをコードする核酸配列 と一致する配列はなかった。 このことから、 本発明のポリペプチドは、 新規 な分泌蛋白質であることが判明した。
しかし、 BLASTX、 BLASTPおよび FASTAは、 クローン ON 056 (配列番号 1 のアミノ酸配列 1〜 3 3 4間の領域) とヒ ト ' カテブシン L (Human Cathepsin L(SwissProt Accession P07711)のアミノ酸配列 1〜 334間の領 域) の間に、 またクロ一ン QN 0 56 (配列番号 1のアミノ酸配列 2 2〜 3 3 4間の領域) とヒト ' カテブシン K (Human Cathepsin K(SwissProt Accession P43235)のアミノ酸配列 19〜329間の領域) の間に有為な相同 性があることを示した。 これらの相同性に基づいて、 クローン ON 0566 は、 少なくともカテブシン L (Cathepsin L) フアミ リーと同様な活性を保 持すると期待される。
(5)大腸菌を用いた蛋白質発現
得られた全長 c D N Aよりシグナルペプチドをコードする部分を除去した コーディング領域の c DN Aを PC Rにより増幅し、 大腸菌用発現ベクター p ET (Novagen社より販売) の翻訳開始点 AT Gの下流にフレームを合わせ て連結し、 発現用プラスミドを構築した。 得られたプラスミドを大腸菌 BL 21 (DE 3) に形質転換し、 培養を行ない I PTGを添加して蛋白質発現 を誘導した。 得られた大腸菌を集菌後、 超音波破碎または界面活性剤により 溶菌し、 不溶性画分は尿素により可溶化して SDS— PAGEを行ない、 ク マシ一染色にて ON 056蛋白質の発現を確認した (図 1中矢印部分) 。
(6)哺乳動物細胞を用いた蛋白質発現
得られた全長 c DN Aを哺乳動物細胞用発現ベクター p ED 6 (Kaufman et.al., Nucleic Acids Res. 19, 4485 - 4490 (1991)参照) の Xh o l (また は E c oR I) /No t I部位に連結し、 分泌蛋白質または膜蛋白質の発現 用プラスミドを構築した。 得られたプラスミドをリポフエクチン (商品名、 GIBCO BRL社より販売) を用いて Co s 7細胞に導入し、 24時間後に Me t および Cy sフリーの培地に交換した後、 35s—Me tおよび35 S— Cy sを 添加して 5時間培養を行なった。 細胞上清を回収後、 セントリコン— 1 0
(商品名、 Amicon社より販売) にて約 1 0倍に濃縮し、 SDS— PAGEを 行なった。 アクリルアミドゲルを乾燥させた後、 35sでラベルされた蛋白質の 発現を BAS2000 (富士フィルム社より販売) を用いて検出した。 実施例 2 :クローン ON 034
本発明のクローン ON 034に関する実施例は、 ON 056と同様な手法 を用いたが、 以下の点のみ異なる。 (1) o 1 y (A) +RNAの調製
ヒ ト胎盤組織より TRIzol reagentを用いて全 RNAを抽出し、 mRNA Purification Kitを用いて p o 1 y (A) TRNAを精製した。
(2)全長 c DNAのクロ一ニングおよび塩基配列の決定
全長 c D N Aのクローニングは GENETRAPPER cDNA Positive Selection Systemを用いて行なった。 まず Super Script Plasmid System for cDNA Synthesis and Plasmid Cloningを用いてヒト胎盤組織の p o 1 y (A) + RNAよりプラスミ ド p S P ORTlをベクターとして d T - primed c DNAライブラリ一を作製した。 つぎに S STで得られた塩基配列の情報 に基づいて 2 8 m e rのピオチン化プライマ一〇 N 0 34— F 1 : 5 ' biotin-TGAAGCCCATCACTACATCGC CATTACG- 3 ' (配列番号: 84) を作製した後、 GeneTrapperキットの方法にしたがつ てピオチン化プライマーと特異的にハイブリダィズするプラスミドを前記の c DNAライブラリーから回収し、 大腸菌 DH 10 Bに形質転換した。 さら に Random Primer DNA Labeling kitを用いて 32p-d C T Pでラベルした ON 034 S ST c DNAをプロ一ブとして、 公知の方法によりコロニーハイ ブリダィゼーシヨンを行ない、 陽性クローンを単離して、 プラスミドを調製 した。 以下〇N 056と同様に全塩基配列を決定し、 配列番号 6に示す配列 を得た。 さらにオープンリーディングフレームを決定し、 アミノ酸に翻訳し て配列番号 4および 5に示す配列を得た。
核酸配列データベースに登録されている既知の核酸配列に対して BLASTNお よび FASTAにより、 またアミノ酸配列デ一夕ベースに登録されている既知のポ リペプチドのァミノ酸配列に対して BLASTX、 BLASTPおよび FASTAにより検索し た結果、 本発明のポリペプチド ON 034およびそれをコードする核酸配列 と一致する配列はなかった。 このことから、 本発明のポリペプチドは、 新規 な分泌蛋白質であることが判明した。 実施例 3 : クローン OX 003
本発明のクローン〇X 003に関する実施例は、 ON 056と同様な手法 を用いたが、 以下の点のみ異なる。
(1) p o 1 y (A) TRNAの調製
ヒト胎盤組織より TRIzol reagentを用いて全 R N Aを抽出し、 mRNA Purification Kitを用いて po 1 y (A) +RNAを精製した。
(2)全長 c DNAのクローニングおよび塩基配列の決定
全長 c DN Aのクロ一ニングはマラソン c DN Aアンプリフィケ一シヨン キット (Marathon cDNA Amplification Kit) (商品名、 Clontech社より販売) による 3' RACE (Rapid Amplification of cDNA End) 法を用いて行なつ た。 2本鎖 c DNAの調製には、 各クローンの由来、 すなわちヒト胎盤組織 の po l y (A) +RNAより作製した。 S S Tで得られた塩基配列の情報に 基づいて推定翻訳開始点 AT G領域を含む 27 me rのプライマー OX 00 一 3' (配列番号 85) を作製して、 前記キットに添付されたアダプタープ ライマ一とで P CRを行なった。 また、 1回の P CRで c DNAが十分に増 幅されなかったので、 OX 003— F 1プライマーの 3 ' 側にさらに 23 me rのプライマ一 OX 003— F 2 : 5 ' 一 TCACCAAGGCTAA CATGGTGGCC- 3 ' (配列番号 86) を作製してネステッド P C R (nested P CR) を行なった。 クローン〇X 003に特異的に増幅された c DNAをァガ口一ス電気泳動で分画後、 pT 7ブル一 2 · Τベクター (ρΤ7 Blue-2 T-Vector) (商品名、 Novagen社より販売) に連結し、 大腸菌 DH5 a に形質転換してブラ スミドを調製した。 初めに 5' 側の塩基配列を決定して OX 003 S ST c DN Aの塩基配列が存在することを確認した後、 全塩 基配列を決定し、 配列番号 9に示す配列を得た。 さらにオープンリーデイン グフレームを決定し、 アミノ酸に翻訳して配列番号 7および 8に示す配列を 得た。
核酸配列データベースに登録されている既知の核酸配列に対して BLASTNお よび FASTAにより、 またアミノ酸配列データベースに登録されている既知のポ リペプチドのアミノ酸配列に対して BLASTX、 BLASTPおよび FASTAにより検索し た結果、 本発明のポリペプチド OX 003およびそれをコードする核酸配列 と一致する配列はなかった。 このことから、 本発明のポリペプチドは、 新規 な分泌蛋白質であることが判明した。 実施例 4 :クロ一ン OA052
本発明のクローン〇A052に関する実施例は、 ON 056と同様な手法 を用いたが、 以下の点のみ異なる。
(1) o 1 y (A) +RNAの調製
ヒトグリア芽腫細胞株 T 98 G (ATCC No. CRL-1690) より TRIzol reagent を用いて全 RNAを抽出し、 mRNA Purification kitを用いて p o 1 y (A) T
RNAを精製した。
(2)全長 c DN Aのクローニングおよび塩基配列の決定
全長 c D N Aのクローニングは Marathon cDNA Amplification kitによる 3 ' RACE法を用いて、 OX 003と同様の方法で行なった。 前記キット の方法に従って 各クローン由来、 すなわち T 98 G細胞株の p o 1 y (A) +
RNAよりアダプタ一を連結した 2本鎖 c DNAの調製した。 S STで得ら れた塩基配列の情報に基づいて推定翻訳開始点 AT G領域を含む 27me r のプライマー OA 052 -F 1 : 5 ' -ATGCCTAGAAGAGGAC TGATTCTTCAC- 3 ' (配列番号 87 ) を作製して、 前記キットに 添付されたアダプタープライマーとで P CRを行なった。 クロ一ン OA 05 2に特異的に増幅された c DNAを〇X 003と同様な手法でリクローニン グし、 全塩基配列を決定し、 配列番号 12に示す配列を得た。 さらにオーブ ンリーディングフレームを決定し、 アミノ酸に翻訳して配列番号 10および 11に示す配列を得た。
核酸配列データベースに登録されている既知の核酸配列に対して BLASTNお よび FASTAにより、 またアミノ酸配列デー夕べ一スに登録されている既知のポ リペプチドのァミノ酸配列に対して BLASTX、 BLASTPおよび FASTAにより検索し た結果、 本発明のポリペプチド OA 052およびそれをコードする核酸配列 と一致する配列はなかった。 このことから、 本発明のポリペプチドは、 新規 な分泌蛋白質であることが判明した。 実施例 5 :—クローン〇C 004
本発明のクローン〇C 004に関する実施例は、 ON 056と同様な手法 を用いたが、 以下の点のみ異なる。
(1) p o 1 y (A) +RNAの調製
ヒト成人脳組織より TRIzol reagentを用いて全 RN Aを抽出し、 mRNA Purification kitを用いて po 1 y (A) +RNAを精製した。
(2)全長 cDN Aのクローニングおよび塩基配列の決定
全長 c DNAのクロ一ニングは Marathon cDNA Amplification kitによる 3 ' RACE法を用いて、 OX 003と同様の方法で行なった。 2本鎖 c DNAの調製には、 各クローンの由来、 すなわちヒト成人脳組織の p o 1 y (A) +RNAより作製した。 S S Tで得られた塩基配列の情報に基 づいて推定翻訳開始点 AT G領域を含む 27me rのプライマー OC 004 - F 1 : 5 ' -ATGAGGAAAGGGAACCTTCTGCTGAGC 一 3' (配列番号 88) を作製して、 前記キットに添付されたアダプタープ ライマーとで PCRを行なった。 また、 1回の P CRで c DNAが十分に増 幅されなかったので、 OC 004— F 1プライマーの 3 ' 側にさらに 1 8 me rのプライマー〇C 004— F 2 : 5' — TGAGCTTCCAGAG CTGTC- 3 ' (配列番号 89) を作製して nested PCRを行なった。 ク ローン〇C 004に特異的に増幅された c DNAを、 〇X 003と同様な手 法でリクローニングし、 全塩基配列を決定し、 配列番号 15に示す配列を得 た。 さらにオープンリーディングフレームを決定し、 アミノ酸に翻訳して配 列番号 13および 14に示す配列を得た。
核酸配列データベースに登録されている既知の核酸配列に対して BLASTNお よび FASTAにより、 またアミノ酸配列デ一夕ベースに登録されている既知のポ リペプチドのアミノ酸配列に対して BLASTX、 BLASTPおよび FASTAにより検索し た結果、 本発明のポリペプチド OC 004およびそれをコードする核酸配列 と一致する配列はなかった。 このことから、 本発明のポリペプチドは、 新規 な分泌蛋白質であることが判明した。 実施例 6 : クローン〇M017
本発明のクローン OM0 17に関する実施例は、 ON 056と同様な手法 を用いたが、 以下の点のみ異なる。
(1) p o 1 y (A) +RNAの調製
ヒ ト成人脳組織より TRIzol reagentを用いて全 RN Aを抽出し、 mRNA Purification kitを用いて po 1 y (A) +RNAを精製した。
(2)全長 c DNAのクローニングおよび塩基配列の決定
全長 c DN Aのクロ一ニングは Marathon cDNA Amplification kitによる 3 ' RACE法を用いて、 〇X 003と同様の方法で行なった。 2本鎖 c DNAの調製には 、 各クローンの由来、 すなわちヒト成人脳組織の p o l y (A) +RNAより作製した。 S S Tで得られた塩基配列の情報に基 づいて推定翻訳開始点 AT G領域を含む 27 me rのプライマー OM017 - 3 ' (配列番号 90) を作製して、 前記キットに添付されたアダプタープ ライマーとで P CRを行なった。 また、 1回の P CRで c DNAが十分に増 幅されなかったので、 OM0 1 7— F 3プライマーの 3 ' 側にさらに 27 1116 1"のプラィマ一01^01 7—? 1 : 5 ' — ATGAAACATTTCT GT AAC CTGCTTTGT— 3 ' (配列番号 9 1 ) を作製して nested PCRを行なった。 クロ 一ン〇M017に特異的に増幅された c DNAを、 OX 003と同様な手法でリクローニングし、 全塩基配列を決定し、 配列番 号 18に示す配列を得た。 さらにオープンリーディングフレームを決定し、 アミノ酸に翻訳して配列番号 16および 17に示す配列を得た。 核酸配列データベースに登録されている既知の核酸配列に対して BLASTNお よび FASTAにより、 またアミノ酸配列デ一夕ベースに登録されている既知のポ リペプチドのァミノ酸配列に対して BLASTX, BLASTPおよび FASTAにより検索し た結果、 本発明のポリペプチド OM0 1 7およびそれをコードする核酸配列 と一致する配列はなかった。 このことから、 本発明のポリペプチドは、 新規 な分泌蛋白質であることが判明した。
しかし、 BLASTX、 BLASTPおよび FASTAは、 クローン OM0 1 7 (配列番号 1 6のアミノ酸配列 433〜 709、 42〜 225、 1 70〜 399、 1〜2
24間の領域) とヒト ' DXS 6673 E (Human DXS6673E (Candidate gene for Mental Retardation) (PRF Code 2218282A(Genbank Accession X95808)) のアミノ酸配列 1 083〜 1 358、 758〜 932、 850〜 108 1、 739〜965間の領域) の間にそれぞれ有為な相同性があることを示した。 これらの相同性に基づいて、 クローン OM 0 1 7は、 少なくとも Human DXS6673Eに関連した活性を保持すると期待される。 実施例 7 : クローン OM 101
本発明のクローン OM1 0 1に関する実施例は、 ON 056と同様な手法 を用いたが、 以下の点のみ異なる。
(1) p o 1 y (A) +RNAの調製

3 ' RAC E法を用いて、 〇X 0 0 3と同様の方法で行なった。 2本鎖 c DNAの調製には、 各クローンの由来、 すなわちヒト成人脳組織の p o l y (A) +RNAをより作製した。 S S Tで得られた塩基配列の情報に 基づいて推定翻訳開始点 AT G領域を含む 27 me rのプライマー〇M1 0 1 - F 3 : 5 ' - C- 3 ' (配列番号 92) を作製して、 前記キットに添付されたアダプター プライマーとで P CRを行なった。 また、 1回の P CRで c DNAが十分に 増幅されなかったので、 OM 1 0 1— F 3プライマーの 3 ' 側にさらに 27 me rのプライマ一〇M 1 0 1— F 1 : 5 ' -ATGAGGACTTACC ATTATATAC C ATTA— 3 ' (配列番号 9 3 ) を作製して nested P CRを行なった。 クローン〇M 1 0 1に特異的に増幅された c DNAを、 〇X 003と同様な手法でリクローニングし、 全塩基配列を決定し、 配列番 号 2 1に示す配列を得た。 さらにオープンリーディングフレームを決定し、 アミノ酸に翻訳して配列番号 19および 20に示す配列を得た。
核酸配列デ一夕ベースに登録されている既知の核酸配列に対して BLASTNお よび FASTAにより、 またアミノ酸配列データベースに登録されている既知のポ リぺプチドのアミノ酸配列に対して BLASTX、 BLASTPおよび FASTAにより検索し た結果、 本発明のポリペプチド OM 1 0 1およびそれをコードする核酸配列 と一致する配列はなかった。 このことから、 本発明のポリペプチドは、 新規 な分泌蛋白質であることが判明した。
しかし、 BLASTX、 BLASTPおよび FASTAは、 クローン OM10 1 (配列番号 1 9のァミノ酸配列 1〜 7 7間の領域) がヒト · カドヘリン— 6 (Human Cadherin-6(SwissProt Accession P55285)のアミノ酸配列:!〜 7 7間の領 域) およびヒ ト · 脳カ ドヘ リ ン (Human Brain-Cadherin(SwissProt Accession P55289)のアミノ酸配列 1〜78間の領域) をはじめとする多数の 力ドヘリンファミリーに有為な相同性があることを示した。 これらの相同性 に基づいて、 クローン OM1 0 1は、 少なくとも Human Cadherin- 6その他の カドヘリンフアミリーに関連した活性を保持すると期待される。 実施例 8 :クローン〇M 126
本発明のクローン〇M 1 26に関する実施例は、 ON 056と同様な手法 を用いたが、 以下の点のみ異なる。
(1) p o 1 y (A) +RNAの調製 ヒト成人脳組織より TRIzol reagentを用いて全 RN Aを抽出し、 mRNA Purification kitを用いて p o 1 y (A) +RNAを精製した。
(2)全長 c DN Aのクローニングおよび塩基配列の決定
2本鎖 c DN Aの調製には、 各クローンの由来、 すなわちヒト成人脳組織 の p o l y (A) +RNAをより作製した。 S S Tで得られた塩基配列の情報 に基づいて 推定翻訳開始点 AT G領域を含む 27 me rのプライマ一 OM1
TG- 3 ' (配列番号 94) を作製して、 前記キットに添付されたアダプタ 一プライマーとで P CRを行なった。 クローン OM1 26に特異的に増幅さ れた c DNAを、 OX 003と同様な手法でリクローニングし、 全塩基配列 を決定し、 配列番号 24に示す配列を得た。 さらにオープンリーディングフ レームを決定し、 アミノ酸に翻訳して配列番号 22および 23に示す配列を 得た。
核酸配列デー夕ベースに登録されている既知の核酸配列に対して BLASTNお よび FASTAにより、 またアミノ酸配列デ一夕ベースに登録されている既知のポ リぺプチドのアミノ酸配列に対して BLASTX、 BLASTPおよび FASTAにより検索し た結果、 本発明のポリペプチド OM1 26およびそれをコードする核酸配列 と一致する配列はなかった。 このことから、 本発明のポリペプチドは、 新規 な分泌蛋白質であることが判明した。
しかし、 BLASTX、 BLASTPおよび FASTAは、 クローン OM126 (配列番号 2 2のアミノ酸配列 25〜1 1 5間の領域) がィムノグロブリンドメインに有 為な相同性があることを示した。 これらの相同性に基づいて、—クローン OM 126は、 少なくともある種のィムノグロプリンスーパ一ファミリーに関連 した活性を保持すると期待される。 実施例 9 :クロ一ン〇M 160
本発明のクローン〇M 1 60に関する実施例は、 ON056と同様な手法 を用いたが、 以下の点のみ異なる。 (1) p o 1 y (A) +RNAの調製

全長 c DN Aのクローニングは GENETRAPPER cDNA Positive Selection Systemを用いて行なった。 まず Superscript Plasmid System for cDNA Synthesis and Plasmid Cloningを用いてヒト成人脳組織の p o 1 y (A) +
RNAよりプラスミ ド p S P ORT lをベクターとして d T - primed c DNAライブラリーを作製した。 つぎに S S Tで得られた塩基配列の情報 に基づいて 2 7 me rのピオチン化プライマ一 O M 1 6 0 - F 1 : 5 '
(配列番号 9 5) を作製した後、 Gene Trapperのキッ卜の方法にしたがって ピオチン化プライマーと特異的にハイプリダイズするプラスミ ドを前記の c DNAライブラリ一から回収し、 大腸菌 DH 1 0 Bに形質転換した。 さら に Random Primer DNA Labeling ki tを用いて 32 p-d C T Pでラベルした OM 1 60 S ST c DNAをプローブとして、 公知の方法によりコロニーハイ ブリダィゼ一シヨンを行ない、 陽性クローンを単離して、 プラスミドを調製 した。 以下〇N 056と同様に全塩基配列を決定し、 配列番号 27に示す配 列を得た。 さらにオープンリーディングフレームを決定し、 アミノ酸に翻訳 して配列番号 2 5および 2 6に示す配列を得た。
核酸配列データベースに登録されている既知の核酸配列に対して BLASTNお よび FASTAにより、 またアミノ酸配列デー夕ベースに登録されている既知の ポリぺプチドのァミノ酸配列に対して BLASTX、 BLASTPおよび FASTAにより検索 した結果、 本発明のポリペプチド OM1 60およびそれをコードする核酸配 列と一致する配列はなかった。 このことから、 本発明のポリペプチドは、 新 規な分泌蛋白質であることが判明した。
しかし、 BLASTX、 BLASTPおよび FASTAは、 クローン OM160 (配列番号 2 5のアミノ酸配列 1 53〜395間の領域) とキイ口ショウジヨウバエ ·神 経原性分泌シグナルプロテイ ン ( Drosophila neurogenic secreted signaling protein(Genepept Accession U41449)のァミ ノ酸配列 8 0〜 317間の領域) の間に有為な相同性があることを示した。 これらの相同性 に基づいて、 クローン OM 1 60は、 少なくとも Drosophila neurogenic secreted signaling proteinと同様な活性を保持すると期待される。 実施例 10 :クローン OMA016
本発明のクローン OMA016に関する実施例は、 OA052と同様な手 法を用いたが、 以下の点のみ異なる。
(1) o 1 y (A) +RNAの調製
ヒト成人脳組織より TRIzol reagentを用いて全 R N Aを抽出し、 mRNA Purification kitを用いて po l y (A) TRNAを精製した。
(2)全長 c DN Aのクローニングおよび塩基配列の決定
全長 c D N Aのクローニングは Marathon cDNA Amplification kitによる 3 ' RACE法を用いて、 〇X 003と同様の方法で行なった。 2本鎖 c DNAの調製には 、 各クローンの由来、 すなわちヒト成人脳組織の o l y (A) +RNAより作製した。 S S Tで得られた塩基配列の情報に基 づいて推定翻訳開始点 ATG領域を含む 2 7 m e rのプライマー OMA TGG- 3' (配列番号 96) を作製して、 前記キットに添付されたァダプ 夕一プライマーとで PCRを行なった。 クローン OMA016に特異的に増 幅された c DNAは 2種類存在したため OMAO 16 a, OMA016 bと 命名し、 OX003と同様な手法でリクローニングし、 全塩基配列を決定し、 それぞれ配列番号 30および 33に示す配列を得た。 さらにオープンリーデ イングフレームを決定し、 アミノ酸に翻訳して配列番号 28、 3 1および 29、 32に示す配列を得た。
核酸配列デ一夕ベースに登録されている既知の核酸配列に対して BLASTNお よび FASTAにより、 またアミノ酸配列データベースに登録されている既知のポ リペプチドのァミノ酸配列に対して BLASTX、 BLASTPおよび FASTAにより検索し た結果、 本発明のポリペプチド OMA0 1 6 a, bおよびそれをコードする 核酸配列と一致する配列はなかった。 このことから、 本発明のポリペプチド は、 新規な分泌蛋白質であることが判明した。 実施例 1 1 : クロ一ン OMB 130
本発明のクローン〇MB 1 30に関する実施例は、 ON 056と同様な手 法を用いたが、 以下の点のみ異なる。

TCC- 3 ' (配列番号 97) を作製して、 前記キットに添付されたァダプ 夕一プライマ一とで P CRを行なった。 クローン OMB 1 30に特異的に増 幅された cDNAを、 OX 003と同様な手法でリクロー ニングし、 全塩基 配列を決定し、 配列番号 36に示す配列を得た。 さらにオープンリーデイン グフレームを決定し、 アミノ酸に翻訳して配列番号 34および 35に示す配 列を得た。
核酸配列デー夕ベースに登録されている既知の核酸配列に対して BLASTNお よび FASTAにより、 またアミノ酸配列データベースに登録されている既知のポ リペプチドのアミノ酸配列に対して BLASTX、 BLASTPおよび FASTAにより検索し た結果、 本発明のポリペプチド OMB 1 30およびそれをコードする核酸配 列と一致する配列はなかった。 このことから、 本発明のポリペプチドは、 新 規な分泌蛋白質であることが判明した。 しかし、 BLASTX、 BLASTPおよび FASTAは、 クローン QMB 130 (配列番号 34のアミノ酸配列 10〜177間の領域) とサル A型肝炎ウィルスレセプ 夕一 (Monkey Hepatitis A virus receptor (PRF Code 2220266A(Genbank Accession X98252))のアミノ酸配列 6〜 173間の領域) の間に有為な相同 性があることを示した。 これらの相同性に基づいて、 クローン OMB 130 は、 少なくとも Monkey Hepatitis A virus receptorに関連した活性を保持す ると期待される。 実施例 12 :クローン〇MB 142
本発明のクローン〇MB 142に関する実施例は、 ON 056と同様な手 法を用いたが、 以下の点のみ異なる。

GAT— 3' (配列番号 98) を作製して、 前記キットに添付されたァダプ 夕一プライマ一とで P CRを行なった。 また、 1回の P CRで c DNAが十 分に増幅されなかったので、 〇MB 142— F 2プライマ一の 3 ' 側にさら に 28me rのプライマー OMB 142 - F 1 : 5 ' — GGAGATGGT ACGGATCTTAAGGACTGTG— 3 ' (配列番号 99 ) を作製し て nested PCRを行なった。 クローン OMB 142に特異的に増幅された cDNAを、 〇X 003と同様な手法でリクローニングし、 全塩基配列を決 定し、 配列番号 39に示す配列を得た。 さらにオープンリーディングフレー ムを決定し、 アミノ酸に翻訳して配列番号 37および 38に示す配列を得た。 核酸配列デー夕ベースに登録されている既知の核酸配列に対して BLASTNお よび FASTAにより、 またアミノ酸配列データベースに登録されている既知のポ リペプチドのアミノ酸配列に対して BLASTX、 BLASTPおよび FASTAにより検索し た結果、 本発明のポリペプチド OMB 142およびそれをコードする核酸配 列と一致する配列はなかった。 このことから、 本発明のポリペプチドは、 新 規な分泌蛋白質であることが判明した。 実施例 13 : クローン OTB 033
本発明のクローン OTB 033に関する実施例は、 ON 056と同様な手 法を用いたが、 以下の点のみ異なる。

ヒト神経芽腫細胞株 I MR— 32 (ATCC No. CCL-127) より TRIzol reagent を用いて全 RNAを抽出し、 mRNA Purification kitを用いて p o 1 y (A) +
RNAを精製した。
(2)全長 c DNAのクロ一ニングおよび塩基配列の決定
全長 c D N Aのクロ一ニングは Marathon cDNA Amplification kitによる 3 ' RACE法を用いて、 〇X 003と同様の方法で行なった。 2本鎖 c DNAの調製には、 各クローンの由来、 すなわち IMR— 32の p o l y (A) +RNAをより作製した。 S STで得られた塩基配列の情報に基づいて 推定翻訳開始点 AT G領域を含む 27me rのプライマ一 OTB 033 -
3' (配列番号 100) を作製して、 前記キットに添付されたアダプタープ ライマーとで PCRを行なった。 また、 1回の P CRで c DNAが十分に増 幅されなかったので、 OTB 033 _F 1プライマーの 3 ' 側にさらに 19 me rのプライマ一OTB 003 -F 2 : 5 ' — CCATGTACACAG TGGGGGC— 3, (配列番号 101 ) を作製して nested P C Rを行 な つた。 クローン OTB 033に特異的に増幅された c DNAを、 OX003 と同様な手法でリクローニングし、 全塩基配列を決定し、 配列番号 42に示 す配列を得た。 さらにオープンリーディングフレームを決定し、 アミノ酸に 翻訳して配列番号 40および 41に示す配列を得た。

本発明のクローン OVB 100に関する実施例は、 ON 056と同様な手 法を用いたが、 以下の点のみ異なる。

CAC- 3 ' (配列番号 102) を作製して、 前記キットに添付されたァダ プ夕一プライマ一とで P CRを行なった。 クローン OVB 100に特異的に 増幅された cDNAを、 〇X 003と同様な手法でリクローニングし、 全塩 基配列を決定し、 配列番号 45に示す配列を得た。 さらにオープンリ一ディ ングフレームを決定し、 アミノ酸に翻訳して配列番号 43および 44に示す 配列を得た。

本発明のクローン OAF 062に関する実施例は、 ON 056と同様な手 法を用いたが、 以下の点のみ異なる。

(2)全長 c DNAのクローニングおよび塩基配列の決定
全長 c D N Aのクロ一ニングは Marathon cDNA Amplification kitによる 3 ' RACE法を用いて、 〇X 003と同様の方法で行なった。 2本鎖 c DNAの調製には、 各クローンの由来、 すなわち HAS 303の p o 1 y

3' (配列番号 103) を作製して、 前記キットに添付されたアダプタープ ライマーとで P CRを行なった。 また、 1回の P CRで c DNAが十分に増 幅されなかったので、 OAF 062—F 2プライマ一の 3, 側にさらに 27 me rのプライマー OAF 062— F 3 : 5 ' -TAGAGGAC AGAA ATGCAGTTCATGAAC— 3 ' (配列番号 1 04) を作製して nested PCRを行なった。 クローン O A F 062に特異的に増幅された cDNAを、 OX003と同様な手法でリ クロ一ニングし、 全塩基配列を決 定し、 配列番号 48に示す配列を得た。 さらにオープンリーディングフレー ムを決定し、 アミノ酸に翻訳して配列番号 46および 47に示す配列を得た。 核酸配列デ一夕ベースに登録されている既知の核酸配列に対して BLASTNお よび FASTAにより、 またアミノ酸配列デ一夕ベースに登録されている既知のポ リペプチドのァミノ酸配列に対して BLASTX、 BLASTPおよび FASTAにより検索し た結果、 本発明のポリペプチド OAF 062およびそれをコードする核酸配 列と一致する配列はなかった。 このことから、 本発明のポリペプチドは、 新 規な分泌蛋白質であることが判明した。 実施例 16 :クロ一ン OAF 075
本発明のクローン OAF 075に関する実施例は、 ON 056と同様な手 法を用いたが、 以下の点のみ異なる。

しかし、 BLASTX、 BLASTPおよび FASTAは、 クローン OAF 075 (配列番号 49のアミノ酸配列 1〜42 1間の領域) とヒト ·力ルポキシぺプチダ一ゼ A 2 (Human Carboxypept idase A2 (SwissProt Accession P48052)のアミノ酸 配列 1〜4 1 7間の領域) 、 ヒト ·カルボキシぺプチダーゼ A 1 (Human Carboxypept idase Al (SwissProt Accession P15085)のアミノ酸配列 1〜 4 1 7間の領域) 、 ヒト '力ルポキシぺプチダーゼ B (Human Carboxypept idase B (SwissProt Accession P15086)のアミノ酸配列 5〜 416間の領域) および ヒ ト · マス ト細胞カルボキシぺプチダ一ゼ A ( Human Mas t Cell Carboxypept idase A (SwissProt Accession PI 5088)のアミノ酸配歹 ij 1〜 4 1 2間の領域) の間に有為な相同性があることを示した。 これらの相同性に基 づいて、 クロ一ン OAF 07 5は、 少なくとも前記のカルボキシぺプチ夕一 ゼ (Carboxypept idase) ファミリ一と同様な活性を保持すると期待される。 実施例 17 : クローン O AG 1 19

全長 c D N Aのクロ一ニングは Marathon cDNA Amplification kitによる 3 ' RACE法を用いて、 OX 003と同様の方法で行なった。 2本鎖 c DN Aの調製は、 各クローンの由来、 すなわち L P 1 0 1の p o l y (A) +RNAより作製した。 S STで得られた塩基配列の情報に基づいて推 定翻訳開始点 ATG領域を含む 28me rのプライマ一〇AG 1 19— F 3 ' (配列番号 106) を作製して、 前記キットに添付されたアダプタープ ライマ一とで P CRを行なった。 クローン〇AG 1 19に特異的に増幅され た cDNAを、 OX 003と同様な手法でリクローニングし、 全塩基配列を 決定し、 配列番号 54に示す配列を得た。 さらにオープンリーディングフレ ームを決定し、 アミノ酸に翻訳して配列番号 52および 53に示す配列を得 た。
核酸配列デ一夕ベースに登録されている既知の核酸配列に対して BLASTNお よび FASTAにより、 またアミノ酸配列データベースに登録されている既知のポ リペプチドのァミノ酸配列に対して BLASTX、 BLASTPおよび FASTAにより検索し た結果、 本発明のポリペプチド 0 AG 1 19およびそれをコードする核酸配 列と一致する配列はなかった。 このことから、 本発明のポリペプチドは、 新 規な分泌蛋白質であることが判明した。 実施例 18 :クローン OAH040
本発明のクローン〇AH 040に関する実施例は、 ON 056と同様な手 法を用いたが、 以下の点のみ異なる。
(1) o 1 y (A) TRNAの調製
ヒト臍帯静脈血管内皮細胞株 HUV— EC— C (ATCC No. CRL-1730) より TRIzol reagentを用いて全 RNAを抽出し、 mRNA Purification kitを用いて p o l y (A) +RNAを精製した。
(2)全長 c DNAのクローニングおよび塩基配列の決定
全長 c DNAのクローニングは Marathon cDNA Amplification kitによる 3, RACE法を用いて、 〇X 003と同様の方法で行なった。 2本鎖 c DNAの調製は、 各クロ一ンの由来、 すなわち HU V— E C— Cの p o l y (A) +RNAより作製した。 S S Tで得られた塩基配列の情報に基 づいて推定翻訳開始点 AT G領域を含む 28me rのプライマー O AH 040 - F 1 : 5 ' 一 TTAGC C C AC C CATGTTGATAGAAC ACCC-3' (配列番号 107) を作製して、 前記キットに添付されたァ ダプタープライマーとで P CRを行なった。 クローン OAH040に特異的 に増幅された cDNAを、 OX 003と同様な手法でリクローニングし、 全 塩基配列 を決定し、 配列番号 57に示す配列を得た。 さらにオープンリー ディングフレームを決定し、 アミノ酸に翻訳して配列番号 55および 56に 示す配列を得た。

本発明のクローン〇M 01 1に関する実施例は、 ON 056—と同様な手法 を用いたが、 以下の点のみ異なる。

全長 c D N Aのクロ一ニングは GENETRAPPER cDNA Positive Selection Systemを用いて行なった。 まず Superscript Plasmid System for cDNA Synthesis and Plasmid Cloningを用いてヒト成人脳組織の p o 1 y (A) + RNAよりプラスミ ド p S P ORT lをべクタ一として d T - primed c DN Aライブラリーを作製した。 つぎに S STで得られた塩基配列の情報 に基づいて 2 7 m e rのピオチン化プライマー OM 0 1 1— F 1 : 5 '
(配列番号 1 09) を作製した後、 GeneTrapperのキットの方法にしたがって ピオチン化プライマーと特異的にハイブリダィズするプラスミドを c DNA ライブラリーから回収し、 大腸菌 DH 10 Bに形質転換した。 さらに Random Primer DNA Labeling ki tを用いて 32 p - d C T Pでラベルした O M 0 1 1 S ST c DNAをプローブとして、 公知の方法によりコロニーハイブリダィ ゼーシヨンを行ない、 陽性クローンを単離して、 プラスミドを調製した。 以 下 ON 056と同様に全塩基配列を決定し、 配列番号 63に示す配列を得た。 さらにオープンリーディングフレームを決定し、 アミノ酸に翻訳して配列番 号 61および 62に示す配列を得た。
核酸配列データベースに登録されている既知の核酸配列に対して BLASTNお よび FASTAにより、 またアミノ酸配列デ一夕ベースに登録されている既知のポ リぺプチドのアミノ酸配列に対して BLASTX、 BLASTPおよび FASTAにより検索し た結果、 本発明のポリペプチド OM01 1およびそれをコードする核酸配列 と一致する配列はなかった。 このことから、 本発明のポリペプチドは、 新規 な膜蛋白質であることが判明した。
しかし、 BLASTX、 BLASTPおよび FASTAは、 クローン OM0 1 1 (配列番号 6 1のアミノ酸配列 26〜 3 96間の領域) とヒト · プラズマ細胞ダリコ プロテイ ン P C— 1 ( Human Plasma-cell Glycoprotein PC-1 (Alkal ine Phosphodiesterase I) (SwissProt Accession P22413)のアミノ酸配列 1 5 8 〜543間の領域) の間に有為な相同性があることを示した。 これらの相同 性に基づいて、 クロ一ン〇M 0 1 1は、 少なく とも Human Plasma-cell Glycoprotein PC-1と同様な活性を保持すると期待される。 実施例 21 :クローン〇M028
本発明のクローン OMO 28に関する実施例は、 ON 056と同様な手法 を用いたが、 以下の点のみ異なる。

全長 c DN Aのクローニングは GENETRAPPER cDNA Positive Selection Systemを用いて行なった。 まず Superscript Plasmid System for cDNA Synthesis and Plasmid Cloningを用いてヒト成人脳組織の p o 1 y (A) + RNAよりプラスミ ド p S P ORT lをべクタ一として d T - primed c DN Aライブラリーを作製した。 つぎに S STで得られた塩基配列の情報 に基づいて 2 7 me rのピオチン化プライマー〇 M 0 2 8— F 1 : 5 '
(配列番号 1 1 0) を作製した後、 GeneTrapperのキットの方法にしたがって ピオチン化プライマーと特異的に八イブリダィズするプラスミドを c DNA ライブラリーから回収し、 大腸菌 DH 1 0 Bに形質転換した。 さらに Random Primer DNA Labeling ki tを用いて 32 p - d C T Pでラベルした O M 0 2 8 S ST c DNAをプロ一 ブとして、 公知の方法によりコロニーハイブリダ ィゼーシヨンを行ない、 陽性クローンを単離して、 プラスミドを調製した。 以下 ON056と同様に全塩基配列を決定し、 配列番号 66に示す配列を得 た。 さらにオープンリーディングフレームを決定し、 アミノ酸に翻訳して配 列番号 64および 65に示す配列を得た。

しかし、 BLASTX、 BLASTPおよび FASTAは、 クローン OM028 (配列番号 6 4のアミノ酸配列 1〜708間の領域) とマウス · ロイシンリツチ · リピー トプロテイン (Mouse Leu-rich repeat protein(PRF Code 2212307ACGENBANK Accession D49802))のアミノ酸配列 1〜 707間の領域) の間をはじめとし て、 多数のロイシンリッチ · リピート (Leu-rich repeat) を有する蛋白質と 有為な相同性があることを示した。 これらの相同性に基づいて、 クローン〇 M028は、 少なくともある種の Leu- rich repeatを有する蛋白質と同様な活 性を保持すると期待される。 実施例 22 :クローン OMB 092
本発明のクローン OMB 092に関する実施例は、 ON 056と同様な手 法を用いたが、 以下の点のみ異なる。
(1) o 1 y (A) +RNAの調製
ヒト成人脳組織より TRIzol reagentを用いて全 RN Aを抽出し、 mRNA Purification kitを用いて p o 1 y (A) TRNAを精製した。
(2)全長 c DNAのクローニングおよび塩基配列の決定
全長 c DN Aのクローニングは Marathon cDNA Amplification kitによる 3 ' RAC E法を用いて、 〇X 0 0 3と同様の方法で行なった。 2本鎖 c DNAの調製には、 各クローンの由来、 すなわちヒト成人脳組織の o l y (A) +RNAをより作製した。 S S Tで得られた塩基配列の情報に 基づいて 27me rのプライマ一〇MB 092—F 1 : 5, — ACTCAC CTGGATC C CTAAGGGC AC AGC— 3 ' (配列番号 1 1 1 ) を 作製して、 前記キットに添付されたアダプタープライマ一とで PCRを行な つた。 また、 1回の P CRで c DNAが十分に増幅されなかったので、 OM B 092 -F 1プライマーの 3 ' 側にさらに推定翻訳開始点 AT G領域を含 む 28me rのプライマ一 OMB 092 - F 2 : 5, — AGAATGAGC TATTACGGCAGCAGCTATC— 3 ' (配列番号 1 12 ) を作製 して nested PCRを 行なった。 クローン OMB 092に特異的に増幅され た cDNAを、 OX003と同様な手法 でリクローニングし、 全塩基配列を 決定し、 配列番号 69に示す配列を得た。 さらにオープンリーディングフ レームを決定し、 アミノ酸に翻訳して配列番号 67および 68に示す配列を 得た。

しかし、 BLASTX、 BLASTPおよび FASTAは、 クロ一ン OMB 092 (配列番号 67のアミノ酸配列 1〜254間の領域) とラット内在性整流型カルシウム チ ャ ンネル B I R 9 ( Rat Inward Rectifier Potassium Channel BIR9(SwissProt Accession P52191)のアミノ酸配列 1〜 254間の領域) の 間をはじめとする多数の力リウ ムチャンネルファミリ一と有為な相同性があ ることを示した。 これらの相同性に基づいて、 クローン OMB 092は、 少 なくともカリゥムチヤンネルファミリーと同様な活性を保持すると期待され る。 実施例 23 : クローン OMB 108

GCC- 3 ' (配列番号 1 13) を作製して、 前記キットに添付されたァダ プタープライマ一とで P CRを行なった。 また、 1回の PCRで cDNAが 十分に増幅されなかったので、 OMB 108— F 1プライマ一の 3 ' 側にさ らに 22me rのプライマ一OMB 108— F 2 : 5 ' 一 TGGTTATG GCCTGTCGCTGGAG— 3 ' (配列番号 1 14) を作製して nested PCRを行なった。 クローン〇MB 108に特異的に増幅された c DN Aを、 〇X003と同様な手法でリクローニングし、 全塩基配列を決定し、 配列番 号 72に示す配列を得た。 さらにオープンリーディングフレームを決定し、 アミノ酸に翻訳して配列番号 70および 71に示す配列を得た。

しかし、 BLASTX、 BLASTPおよび FASTAは、 クローン OMB 108 (配列番号 70のアミノ酸配列 164〜256間および 373〜487間の領域) とヒ ト低密度リポ蛋白質レセプター関連蛋白質 1 ( Human Low-Density Lipoprotein Receptor Related Protein 10(SwissProt Accession Q07954) ) をはじめとする LDLレセプ夕一ファミリ一の LDLリピートの領域の間に 有為な相同性があること、 またクローン〇MB 108 (配列番号 70のアミ ノ酸配列 47〜 158間および 259〜370間の領域) とヒト骨形成蛋白 質 1 Human Bone Morphogenet ic Protein 1 (SwissProt Access ion P13497)) 等が有する CUBドメインの間に有為な相同性があることを示した。 すなわち OMB 108は細胞外ドメインに 2ケ所の CUBドメインと 5ケ所 の LDLリピートを有していることが判明した。 これらの相同性に基づいて、 クローン〇MB 108は、 LDLリピートを有する蛋白質ならびに C U Bド メインを有する蛋白質と同様な活性の少なくとも幾つかを保持すると考えら れる。 実施例 24 :クローン OT 007
本発明のクローン〇T 007に関する実施例は、 ΟΝ 056と同様な手法 を用いたが、 以下の点のみ異なる。

RNAよりプラスミド p SPORT lをべクタ一として d T— primed c DN Aライブラリーを作製した。 つぎに S S Tで得られた塩基配列の情報に基づ いて 27me rのピオチン化プライマ一 O T 007 -F 1 : 5 ' biotin— A AAATGACTCCCCAGTCGCTGCTGCAG- 3 ' (配列番号 1 15) を作製した後、 GeneTrapperのキットの方法にしたがってピオチン化 プライマーと特異的にハイブリダィズするプラスミドを c DNAライブラリ —から回収し、 大腸菌 DH10Bに形質転換した。 さらに Random Primer DNA Labeling kitを用いて32 P- d CTPでラベルした〇T 0 0 7 S S T c DNAをプローブとして、 公知の方法によりコロニーハイブリダィゼーシ ヨンを行ない、 陽性クローンを単離して、 プラスミドを調製した。 以下 ON 056と同様に全塩基配列を決定し、 配列番号 7 5に示す配列を得た。 さら にオープンリーディングフレームを決定し、 アミノ酸に翻訳して配列番号 73および 74に示す配列を得た。

全長 c D N Aのクローニングは Marathon cDNA Amplification kitによる 3 ' RAC E法を用いて、 〇X 0 0 3と同様の方法で行なった。 2本鎖 c DNAの調製には 、 各クローンの由来、 すなわち L P 1 0 1の p o 1 y (A) +RNAより作製した。 S STで得られた塩基配列の情報に基づいて推 定翻訳開始点 AT G領域を含む 27me rのプライマー〇 AG 05 1— F 1 : 5 ' 一 GGAAATGTTTAC ATTTTT GTTGACGTG - 3' (配列番号 1 1 6) を作製して、 前記キットに添付されたアダプタープ ライマ一とで P CRを行なった。 クロ一ン OAG 0 5 1に特異的に増幅され た c DNAを、 OX 003と同様な手法でリクローニングし、 全塩基配列を 決定し、 配列番号 7 8に示す配列を得た。 さらにオープンリーディングフ レームを決定し、 アミノ酸に翻訳して配列番号 76および 77に示す配列を 得た。

しかし、 BLASTX、 BLASTPおよび FASTAは、 クローン OAG05 1 (配列番号 7 6のアミ ノ酸配列 4〜 7 0 3間の領域) と Mouse Frizzled - 6 (PRF Code2208383E (GenebankAccess ion U43319)のアミノ酸配列 6〜 7 0 8間の領 域) の間、 またクローン〇 AG O 5 1 (配列番号 7 6のアミノ酸配列 1〜 6 2 7 間の領域) と Mouse Frizzled- 3 (PRF Code 2208383E (Genebank Accession U43205)のアミノ酸配列 7〜 6 1 8間の領域) の間をはじめとする 多数の Frizzledファミリ一と有為な相同性があることを示した。 これらの相 同性に基づいて、 クロ一ン OAG 0 5 1は、 少なくとも Frizzledファミリ一 と同様な活性を保持すると期待される。 実施例 26 : クローン OUB068

66、 69、 72、 75、 78または 81で示される塩基配列を有する請求 の範囲第 3項記載の c DNA、 またはその配列に選択的にハイブリダィズす るフラグメントからなる c DNA。
6. 請求の範囲第 3項から第 5項のいずれかの項に記載の c DN Aからな る複製または発現べクタ一。
7 . 請求の範囲第 6項記載の複製または発現べクタ一で形質転換された宿 主細胞。
8 . 請求の範囲第 1項または第 2項に記載されたポリぺプチドを発現させ るための条件下で請求の範囲第 7記載の宿主細胞を培養することからなる請 求の範囲第 1項または第 2項記載のポリぺプチドの製造方法。
9 . 請求の範囲第 1項または第 2項に記載されたポリペプチドのモノ クロ一ナルまたはポリクローナル抗体。
