Abstract

Die vorliegende Erfindung betrifft ein in vitro-Selektionsverfahren, mit dem Ribozyme selektiert werden können, die 2'-OH-Gruppen von Ribonucleinsäuren in trans kovalent modifizieren können. Die vorliegende Erfindung betrifft außerdem Ribozyme, die durch dieses Verfahren erhältlich sind. Außerdem betrifft die vorliegende Erfindung diese enthaltende Arzneimittel, die vorzugsweise zur Hemmung der Genexpression - beispielsweise in der Gentherapie - verwendet werden können. Die erfingungsgemäßen Ribozyme können darüber hinaus zur Herstellung von Muteinen und nucleaseresistenten Ribonucleinsäuren - beispielsweise ribonucleaseresistenten 'Antisense'-Oligonucleotide - verwendet werden.
